Abstract:A new naphthopyrone, pleuropyrone A (1), together with (-)-lyoniresinol 3a-O-beta-D-glucopyranoside (2) and (+)-lyoniresinol 3a-O-beta-D-glucopyranoside (3) was isolated from the roots of Pleuropterus ciliinervis. The structure of pleuropyrone A (1) was determined to be 2,5-dimethyl-8-hydroxynaphthopyrone 10-O-beta-D-glucopyranoside by spectroscopic methods including 2D-NMR. Compounds 2 and 3 showed moderate antioxidant activity.

Abstract:Three new a-pyrones, scirpyrones AC (13), and four new pyranes, scirpyranes AD (47), were isolated from solid cultures of the plant pathogen Pestalotiopsis scirpina. Their structures were elucidated primarily by NMR experiments. The absolute configurations of C-7 and C-2' in 1, C-7 in 2, and C-6 and C-7 in 3 were deduced on the basis of circular dichroism (CD) data, whereas those of the secondary alcohols in 47 were determined by a modification of Mosher's method. Compound 1 is the first described 6-(5-oxotetrahydrofuran-2-yl)-2H-pyran-2-one. Compounds 46 showed significant cytotoxicity towards MCF-7 cells.

Abstract:The root of Pyrola japonica Klenze ex Alefeld (Pyrolaceae) has been used for the treatment of arthritic disease (Kosuge et al., 1985) and hemostasis (Kagawa et al., 1992) in Chinese folk medicine. We have isolated chimaphilin from the root of P. japonica (Bae et al., 1996), a 1,4-naphthoquinone derivative, which was shown to have significant cytotoxicities against L1210 and K-562 tumor cell lines with EDs0 values of 1.1 7 and 0.88 p.g/ml, respectively (Bae et al., 1996). This paper reports the isolation and characterization of two naphthoquinone compounds (1, 2) based on spectroscopic and chemical evidence.

Abstract:Five new alpha-pyrone derivatives, necpyrones A-E (1-5), together with seven known compounds (6-12), were isolated from the extract of an endolichenic fungus Nectria sp. Chemical structures of these compounds were elucidated by spectroscopic analyses (HRESIMS and NMR). The absolute configurations of C-6 in the pyrone ring as well as the chiral carbons at the aliphatic side chain resulted from hydroxyl substitute were finally determined on the basis of measurement of ECD and Kusumi-Mosher method. New Pyrones from the Mangrove Endophytic Fungus Pestalotiopsis Sp

Abstract:Pestalotiopsis is generally found as endophytes of tropical plants and as prolific producers of structurally unusal natural products [1]. Our previous chemical investigations on fungus species Pestalotiopsis JCM2A4 isolated from the Chinese Mangrove plant Rhizophora mucronata Lam. have led to the isolation of a series of natural products and yielded over twenty different compounds with seventeen of them being new natural products including chromones, cytosporones and coumarins [2–3]. Following cultivation of the fungus now yielded eight new pyrone derivatives (1) as well as a known compound. The structures of all compounds were unambiguously established from their spectroscopic data, that included HR-ESIMS and 1- and 2-dimensional NMR spectroscopy, and by comparison with the literature [4]. Our findings proved endophyte genus Pestalotiopsis to be particularly productive.

Abstract:The new naphthalenone derivatives perenniporides A-D (1-4) were isolated from solid cultures of a fungus Perenniporia sp. inhabiting the larva of Euops chinesis, a phytophagous weevil with high host specificity to the medicinal plant Fallopia japonica. The structures of 1-4 were elucidated primarily by NMR experiments, and 1 was confirmed by X-ray crystallography. The absolute configuration of 1 and 2 was assigned by electronic circular dichroism (ECD) calculations, whereas that of the C-10 tertiary alcohol in 3 was deduced via the CD data of the in situ formed [Rh-2(OCOCF3)(4)] complex and supported by the ECD data Compound 1 showed antifungal activity against five plant pathogens.

The structure of 1 was elucidated by extensive spectroscopic analyses, including 1D and 2D NMR spectra and MS data. Abstract:Pyrenocines are better known as phytotoxic metabolites been found in Alternaria, Curvularia, Penicillium and Phomopsis strains. The class of compounds showed several biological activities, reportedly being phytotoxic, antifungal, cytotoxic, antitrypanosomal and antimalarial.1, 2, 3 Fungi of the genus Colletotrichum, a class of the widely distributed endophytic fungi, were reported as prolific producers of bioactive metabolites.4, 5, 6, 7 However, there are few reports on pyrenocines found in genus Colletotrichum. During our ongoing chemical investigations of endophytic fungi as sources of new cytotoxic natural products, the organic solvent extract of solid substrate fermentation culture of a Colletotrichum sp. HCCB03289 strain displayed cytotoxicity against cancer cell lines. Bioassay-guided fractionation of the extract led to the isolation of two novel pyrenocines N–O (1–2), and five known pyrenocines macommelin-9-acetate (3),8 pyrenocine A (4),9 pyrenocine B (5),9 pyrenocine E (6)10 and novae-zelandin A (7).11 Details of the isolation, structure elucidation and the cytotoxic activities of these compounds are presented here.

Abstract:Nakiterpiosin and nakiterpiosinone are two related C-nor-D-homosteroids isolated from the sponge Terpios hoshinota that show promise as anticancer agents. We have previously described the asymmetric synthesis and revision of the relative configuration of nakiterpiosin. We now provide detailed information on the stereochemical analysis that supports our structure revision and the synthesis of the originally proposed and revised nakiterpiosin. In addition, we herein describe a refined approach for the synthesis of nakiterpiosin, the first synthesis of nakiterpiosinone, and preliminary mechanistic studies of nakiterpiosin's action in mammalian cells. Cells treated with nakiterpiosin exhibit compromised formation of the primary cilium, an organelle that functions as an assembly point for components of the Hedgehog signal transduction pathway. We provide evidence that the biological effects exhibited by nakiterpiosin are mechanistically distinct from those of well-established antimitotic agents such as taxol. Nakiterpiosin may be useful as an anticancer agent in those tumors resistant to existing antimitotic agents and those dependent on Hedgehog pathway responses for growth.

Abstract:Four undescribed pymne derivatives, pestalopyrones A-D, containing unusual tricyclic 5/6/6 polycyclic skeletons, were isolated from the ethyl acetate extract of the endophytic fungus Pestalotiopsis neglecta 53 derived from the fresh stems of Rubia podantha Diels (Rubiaceae). Their planar structures were elucidated mainly by NMR and HRESIMS. Pestalopyrones A-D contained six contiguous chiral carbons, and the relative configurations of C-4, C-5, and C-8 in tricyclic 5/6/6 polycyclic skeletons were determined by ROESY spectra. For pestalopyrone B, the absolute configuration of C-16 was determined by the Mosher's method. All isolated compounds were tested for their cytotoxicity against cancer cell lines, antibacterial activity, and inhibitory effect on the lipopolysaccharide (LPS)-induced nitric oxide (NO) production, and the results showed that pestalopyrone A inhibited LPS-induced NO production with an IC50 value of 35.8 mu M.
