Abstract:The development of a highly selective and sensitive optical method for quantitative enantioselective sensing of chiral amino acids in the presence of other amino acid interferences has remained a major challenge to date. Herein, a homochiral metal–organic framework (MOF), [Zn(L)(2,2′‐bipy)]·H2O (LNNU‐1) (H2L = HOOCC6H4CH2PO(OH)(OC2H5), 2,2′‐bipy = 2,2′‐bipyridine), is designed to interact with tryptophan via multiple noncovalent interactions. The interactions result in the emergence of a strong induced circular dichroism signal. This enables accurate determination of the absolute configuration and enantiomeric ratio of chiral tryptophan in water, mixed natural amino acids, and blood plasma components without any interference from other coexistent species. A simple device with advantageous characteristics of reusability and simple manipulation is designed for enantioselective sensing assay. The obtained results suggest that the rationally designed homochiral MOF is a promising candidate for enantioselective sensing of chiral species.

Abstract:Chiral recognition of enantiomers with identical mirror-symmetric molecular structures is important for the analysis of biomolecules, and it conventionally relies on stereoselective interactions in chiral chemical environments. Here, we develop a magneto-electrochemical method for the enhanced detection of chiral amino acids (AAs), that combines the advantages of the high sensitivity of electrochemiluminescence (ECL) biosensors and chirality-induced effects under magnetic field. The ECL difference between L- and D-enantiomers can be amplified by over 35-fold under a field of 3.5 kG, and the chiral discrimination can be achieved in dilute AA solutions down to nM level. The field-dependent ECL and chronocoulometry measurements suggest that chiral AAs can lock the spins on their radicals and thus enlarge the ECL change under applied magnetic field (magneto-ECL, MECL), which explains the field-enhanced chiral discrimination of AA enantiomers. Finally, a detailed protocol is demonstrated for the identification of unknown AA solutions, in which the species, chirality and concentration of AAs can be determined simultaneously from the 2D plots of the ECL and MECL results. This work benefits the development of field-assisted detection methods and represents a promising and universal strategy for the comprehensive analysis of chiral biomolecules.

Abstract:Chiral channels or pores are a fundamental feature of chiral metal-organic frameworks (CMOFs). However, the roles of chiral channels and pores have always been overlooked in enantioselective sensing based on the CMOFs. Aiming at the chiral sensing platforms of enantioselectivity with efficient electrochemical recognition, the L-His, L-Glu, and L-Lys have been introduced into the metal-organic framework (MOF) as CMOFs. Taking advantage of postsynthetic modifications (PSM), the resulting CMOFs (MOF-His/MOF-Glu/MOF-Lys) exhibit chiral channel dependence for different amino acid enantiomers. Ascribed to the introduction of chiral linkers, the nanomaterials with chiral channels of different sizes exhibit electrochemical recognition behaviors for electrochemical recognition of glutamate (Glu) enantiomers, tryptophan (Trp) enantiomers, and phenylalanine (Phe) enantiomers, which are evaluated in detail by differential pulse voltammetry (DPV). In addition, ascribed to the regulation of chirality information transmission through introducing chiral molecules of different sizes, the resolved nanomaterials reveal a unique adjustable chiral channel upon the addition of amino acid molecules, leading to passage of amino acid enantiomers of different sizes with excellent recognition efficiency. This work provides not only a design strategy for developing CMOFs with channel tunability but also an idea for the construction of an electrochemical sensing platform.

Abstract:As the most important small molecules revealing the origins of life, amino acids (AAs) play essential roles in living organisms and their facile enantiodiscrimination has long been a great challenge for analytical chemists. Inspired by the specific stereomatching effect between biomolecules and AA enantiomers, herein, we first developed a bio-inspired highly sensitive platform based on an extended-gate metal-oxide-semiconductor field-effect-transistor (EG-MOSFET) for highly sensitive AA enantiodiscrimination. Bovine serum albumin (BSA) was self-assembled on deposited Au surfaces to afford the extended gate (EG) sensing unit, and its enantiorecognition ability was initially verified using common electrochemical techniques. The EG was thereafter installed to a MOSFET to build the desired BSA-EG-MOSFET highly sensitive chiral sensing platform, which realized the efficient enantiodiscrimination of essential AAs with high sensitivity, where effective chiral resolution was achieved at the femtomole level to phenylalanine (Phe). Combining molecular docking and circular dichroism spectroscopy, the weak intermolecular interactions between BSA and AAs enantiomers were investigated and the mechanism for signal amplification was proposed. Our results demonstrate that the as-fabricated biosensor has great potential in highly sensitive chiral sensing fields and can also afford a potential tool for biomolecular interaction investigations.

Abstract:Self-assembly of zinc salt with rationally designed chiral ligand, (1R,2R)-2-(pyridine-4-ylcarbamoyl) cyclohexanecarboxylic acid (RR-PCCHC) generated 2D homochiral metal-organic framework [Zn(RR-PCCHC)] (HMOF-1) that is composed of DNA-like right-handed double-helix structure. HMOF-1 shows high solvent and thermal stability and is also stable in neutral, weak acidic and weak basic aqueous solution. Emulsified HMOF-1 shows strong inherent circular dichroism (CD) signal in aqueous solution, which can show regular intensity change by induction of amino acids. On the basis of the measuring of CD signal intensity, a chemosensor for unmodified amino acids is fabricated, which differ from reported those in which CD signal is amplified by a complicated chemical reaction of originally CD-silent molecule with probed amino acids. This chemosensor can be used for rapid, convenient and sensitive detection of micro amount of amino acids. Most remarkably, 3 × 10 mol of l-aspartic acid and 4 × 10 mol of d-aspartic acid in aqueous solution can completely quench CD signal of emulsified HMOF-1 in HO. It is found that the difference of recognition ability between d- and l-proline is the largest in all probed amino acids. The LOD (limit of detection) of the proposed sensor for the determination of aspartic acid is 13.31 ppm. The recognition efficiency η = [Formula: see text] × 100% for l-aspartic acid is as high as 92.1%. The interacting mechanism of DNA-like HMOF-1 with probed amino acids is similar to that of groove binding of targeting drug with DNA.
Chiral MOFs encapsulated by polymers with poly-metallic coordination as chiral biosensors

Abstract:Chiral materials have drawn the widespread attention for their its chiral recognition ability. The design and synthesis of chiral material are of importance owing to the unpredictability in controlling chirality during the synthesis process. To circumvent problems, a chiral MOF (D-His-ZIF-8) was synthesized by ligand exchange of 2-methylimidazole (Hmim) on ZIF-8 by D-histidine (D-His), which can be treated as chiral host to distinguish amino acid enantiomers. The obtained D-His-ZIF-8 can provide chiral nanochannels for amino acid guests. Meanwhile, polynary transition-metal ion (Co2+ and Fe3+) coordinating with polydopamine (PDA) wrapped on the surface of D-His-ZIF-8 can increase the active sites. The electrochemical chiral recognition behavior showed that D-His-ZIF-8@CoFe-PDA exhibited good recognition of the tryptophan enantiomer (L/D-Trp) (working potential of -0.2 V vs. Hg/HgCl2). The LOD and LOQ of L-Trp were 0.066 mM and 0.22 mM, respectively, while the LOD and LOQ of D-Trp were 0.15 mM and 0.50 mM, respectively. Finally, the usefulness of D-His-ZIF-8@CoFe-PDA/GCE was evaluated with a recovery of 94.4-103%. The analysis of real samples shows that D-His-ZIF-8@CoFe-PDA/GCE is a feasible sensing platform for the detection of L-Trp and D-Trp.
Electrochemical chiral sensor for recognition of amino acid enantiomers with cyclodextrin-based microporous organic networks

Abstract:Background: Chirality is a ubiquitous phenomenon in nature, but enantiomers exhibit different pharmacological activities and toxicological effects. Therefore, Chiral recognition plays a pivotal role in various fields such as life sciences, chemical synthesis, drug development, and materials science. The synthesis of novel chiral composites with well-defined loading capabilities and ordered structures holds significant potential for electrochemical chiral recognition applications. However, the design of selective and stable electrochemical chiral recognition materials remains a challenging task. Result: In this work, we construct a simple and rapid electrochemical sensing platform for tryptophan (Trp) enantiomer recognition using cyclodextrin-modified microporous organic network as chiral recognition agent. CD-MON with chiral microenvironment was prepared by Sonogashira-Hagihara coupling reaction of the chiral molecule heptyl-6-iodo-6-deoxyβ-cyclodextrin and 1, 4-Diethynylbenzene. The adhesion of BSA makes CD-MON firmly fixed on the electrode surface, and as a chiral protein, it can improve the chiral recognition ability through synergistic effect. Chiral amino acids are in full contact with the chiral microenvironment during pore conduction of MON, and L-Trp is more stably bound to CD-MON/BSA due to steric hindrance, host-guest recognition and hydrogen bonding. Therefore, the electrochemical sensor can effectively identify tryptophan enantiomers (IL-Trp/ID-Trp = 2.02), and it exhibits a detection limit of 2.6 μM for L-Trp. UV-Vis spectroscopy confirmed the adsorption capacity of CD-MON towards tryptophan enantiomers in agreement with electrochemistry results. Significance: The prepared chiral sensor has excellent stability, reproducibility (RSD = 3.7%) and selectivity, realizes the quantitative detection of single isomer in tryptophan racemic and quantitative analysis in real samples with 94.0%-101.0% recovery. This work represents the first application of MON in chiral electrochemistry which expands the application scope of chiral sensors and holds great significance in separation science and electrochemical sensing.

Abstract:Due to the widespread application of chiral substances in current society, the electrochemical strategy to assemble chiral metal-organic frameworks (CMOFs) as sensors has shown great potential for sensitive enantiorecognition. In this paper, an efficient and highly selective electrochemical sensor was proposed based on multi-walled carbon nanotube (MWCNT) and CMOF, and successfully achieved sensitive chiral recognition of Tryptophan (Trp) enantiomers. Numerous MOFs in multiple synthesis conditions were investigated for a regular process to access the electrochemical chiral recognition. A variety of amino acids were introduced into the MOFs skeleton by a simple hydrothermal method for the enantio-specificity. Meanwhile the MWCNT smoothly integrated with the CMOF to form a functional electrosensing interface with the increase of conductivity. Based on the current ratio (Rcp) of tryptophan enantiomers by different modified electrodes, a series of experimental conditions were optimized for the optimal chiral recognition performance of the CMOF/MWCNT-based sensor, finally reaching 2.9 times of ΔIL/D between enantiomers. In addition, D/L-Trp showed a good linear relationship with the current in the range from 0.4 μM to 19 μM with the detection limit of 0.11 μM for L-Trp and 0.16 μM for D-Trp. The prepared composite-functionalized sensor owned the desired reproducibility and selectivity to identify Tryptophan enantiomers with robust chiral recognition. The research on the structure-activity relationship of chiral MOFs to Tryptophan enantiomers provides a new path to construct more electrochemical sensors for the application of enantiorecognition in pharmaceutical analysis.

Abstract:Chiral recognition of l-amino acids is of significant importance due to the crucial role of l-amino acids in life sciences and pharmaceutics. In this work, a chiral sensor with capability of probing two chiral amino acids by an attractive single-template molecular imprinting strategy is introduced and used in the simultaneous chiral recognition of d/l-alanine (d/l-Ala) and d/l-tyrosine (d/l-Tyr). The assay relies on the hydrolysis of l-alanyl-l-tyrosine dipeptide doped in silica/polypyrrole (SiO2/PPy) under acidic conditions, resulting in l-Ala and l-Tyr coimprinted chiral sensor. This work opens up a new avenue for simultaneous chiral sensing of two or more chiral amino acids by incorporating only one template, circumventing the shortcomings encountered with multitemplate molecularly imprinted technology.

Abstract:In this work, an electrochemical chiral sensor of a nanowire-like chiral metal-organic framework/multiwalled carbon nanotube-chitosan/glassy carbon electrode( D -MOF/MWCNTs-CS/GCE) was proposed for the enantiorecognition of L/D -tryptophan( L/D -Trp). The asymmetrical spatial structure of D -MOF provides the feasibility for the enantiorecognition of Trp enantiomers. Moreover, differential pulse voltammetry(DPV) was carried out to be the detection method and the DPV peak potential difference(Δ E p ) between L -Trp and D -Trp was referred as the index of the enantiorecognition performance. Several parameters, such as mass ratios and drop-coated volume of MWCNTs-CS, drop-coated volume and concentration of D -MOF, pH and detection temperature of D -MOF/MWCNTs-CS/GCE were optimized for the largest Δ E p value. And the molecular dynamics(MD) simulation was used to elucidate the enantiorecognition mechanism. Furthermore, the proportions of D -Trp( D -Trp%) in Trp mixtures were detected in a good linear relationship with the DPV peak potentials( E p ), and the proposed electrochemical chiral sensor exhibited good reproducibility, stability and enantiorecognition ability. Additionally, the proposed electrochemical chiral sensor( D -MOF/MWCNTs-CS/GCE) has a good application prospect in the fields of biomedicine, clinical diagnosis, chemical production, pharmaceuticals safety and so on.

Abstract:Chiral transition metal oxides with tunable structures and multiple physicochemical features have been increasingly applied for chiral sensing and detection. In this work, chiral zinc oxide (ZnO) was first applied as selector to construct quartz crystal microbalance (QCM) sensor for enantioselective recognition of amino acids. The chiral ZnO was prepared by a methionine-induced self-assembly strategy and its high topological chirality was confirmed by several techniques such as circular dichroism spectrum. The chiral discrimination factors were calculated by frequency shifts in response to aspartic acid, phenylalanine, lysine and arginine on L-ZnO surface, achieving 1.89 ± 0.04, 1.76 ± 0.11, 1.66 ± 0.07 and 1.54 ± 0.09, respectively. Notably, L-enantiomers preferred stronger absorptions on L-ZnO surface as compared to D-forms. It was further found that this sensor was appropriate for quantitative analysis and enantiomer excess analysis and adsorption kinetics study. Furthermore, molecular docking revealed the recognition mechanism, where chiral distinction was caused by the different steric interactions between enantiomers and chiral ZnO. This method enjoyed merits of high enantioselectivity, simple preparation and low cost, offering newly chiral sensing method for other molecules.

Abstract:the vital role of Cysteine (Cys) in biological processes and the hazardous effect of its D-enantiomer, discriminating Cys enantiomers without auxiliary enzyme is highly wanted. In this work, a pair of UiO-MOF enantiomers (UiO-tart) have been fabricated through post-modification, which could be further fabricated into enantiomeric sensing devices (UiO-tart@Au). By employing the Quartz Crystal Microbalance (QCM) technology, gravimetric discrimination of Cys enantiomers could be achieved. UiO-tart@Au is highly enantioselective, and the afforded enantioselective factor (5.97 +/- 0.54) represents the best performance reported ever. In the fabricated device, MOF layer acts as the chiral selector for specific Cys enantiomer, and the reaction between the captured Cys enantiomer and Au results in the mass growth of the system. Solid-phase extraction (SPE) gives an e.e. value of 71.6 +/- 3.8%, substantially confirming the chiral-selector role of UiO-tart. DFT calculations indicate that enantiomeric H-bonding effect and greater reaction enthalpy should be the reason. To the best of our knowledge, this work represents the first example of chiral tartaric acid derived MOF sensors for enantioselective discrimination of Cys, suggesting a promising potential of developing chiral MOFs based devices for enhanced enantioselective application.

Abstract:Chiral recognition of amino acid plays a significant role in pharmaceutical, medical, and food science. This study describes a chiral sensing system of β-cyclodextrin (β-CD)-coated sulfur quantum dots (CD-SQDs) for the selective fluorescence recognition of tryptophan (Trp) enantiomers. CD-SQDs were prepared by a facile assembly fission method and could selectively recognize L-Trp by the different binding ability between L/D-Trp and β-CD. The inclusion of L-Trp and the stereoselective catalysis of CD-SQDs enzyme mimics cause the increased fluorescence intensity of CD-SQDs, which has a linear response ranging from 10 to 500 nM and the detection limit as 2.3 nM. CD-SQDs also show great selectivity for L-Trp from the commercial compound amino acid injection. The study could provide an effective method for the chiral recognition of amino acid enantiomers based on the catalytic activity of nanoenzymes.

Monitoring of the yogurt fermentation process based on a rapid bio-luminescent chiral pattern recognition of amino acids

Abstract:The analysis of chiral α-amino acids is of great significance in asymmetric synthesis, nutrition, food science, and microbiology. However, the ability of chiral recognition is difficult to achieve. Due to the demand for expensive equipment and skilled operators, traditional methods such as high-performance liquid chromatography are limited. The previously reported methods based on chemical sensor arrays usually cannot carry out the chiral analysis. Here, we developed a novel biosensor array based on the interaction between a suite of host-based luminescent bacteria and amino acids and used linear discriminant analysis to reflect their luminescence response patterns. This biosensor array could effectively discriminate chiral amino acids, including 19 L-amino acids, their corresponding D-enantiomers, and the achiral glycine. In addition, the determination of enantiomeric purity and quantitative ability has been proved. The successful identification of a complex system containing multiple chiral amino acids further demonstrates the superiority of the bioluminescent sensor array. Moreover, this sensor array could efficiently monitor the dynamic composition of free amino acids in the process of milk fermentation. Finally, the bioluminescence response mechanism of the luminescent bacteria for the recognition of chirality was clarified. This approach possessed the advantages of facile construction, high throughput, easy operation, high accuracy and fast response.

Development of highly efficient chiral recognition systems is crucial for the precise application of these chiral molecules. Herein, a homochiral zeolitic imidazolate frameworks (c-ZIF) functionalized nanochannel device that exhibits an ideal platform for electrochemical enantioselective recognition is reported. Its distinct chiral binding cavity enables more sensitive discrimination of tryptophan (Trp) enantiomer pairs than other smaller chiral amino acids owing to its size matching to the target molecule. It is found that introducing neighboring aldehyde groups into the chiral cavity will result in an inferior chiral Trp recognition due to the decreased adsorption-energy difference of D- and L-Trp on the chiral sites. This study may provide an alternative strategy for designing efficient chiral recognition devices by utilizing the homochiral reticular materials and tailoring their chiral environments.
Construction of an enzyme-based all-fiber SPR biosensor for detection of enantiomers

Abstract:Chiral analysis of amino acids (AAs) is of great importance in medical science due to the distinctive effect of AA isomers on human health. Although various chiral recognition techniques have been developed, the quantitative chiral recognition of low-level AA isomers remains challenging. Here, we combined the fiber optic SPR with an enzyme-substrate recognition mechanism to construct a direct-assay-type chiral AA biosensor. As a proof-of-concept attempt, a recently discovered Rasamsonia emersonii D-amino acid oxidase (ReDAAO) with a wide substrate spectrum and high stability was immobilized on the graphene oxide and gold nanorods composites (GO-AuNRs), using both EDC/NHS coupling and the gold-binding peptide (GBP) method. Such a biosensor can distinguish two AA isomers at the same concentration. It achieved specific detection of D-amino acids (D-AAs) with a linear range from 5x10-4 mM to 30 mM. Furthermore, it showed good resistance to enantiomeric interference. When the percentage of D-AA increases in the isomer mixture, a good linear relationship between the D/(D + L)-AA ratio and SPR spectral shift was obtained. This unique combination of the enzyme, nanocomposite, and SPR taps into the rich reservoir of proteins for chiral receptors. It lays the foundation for protein-based chiral recognition of other clinically important small molecules in future biosensor designs.

Preparation and application of achiral UiO-66-NH2 MOFs for enantioselective fluorescent detection of lysine enantiomers

Abstract:Lysine (Lys) is an essential nutrient that plays a crucial role in the growth and development of living organisms. Chiral analysis of Lys holds significant importance for ensuring the safety of food, pharmaceuticals, and health products. In this work, an achiral Zr-based metal organic frameworks (MOFs), UiO-66-NH2, was proposed as a fluorescent probe to achieve rapid response to l-lysine (L-Lys) in solution. Additionally, Zr4+ in the skeleton of UiO-66-NH2 framework exhibited different binding capacities towards Lys enantiomers, leading to distinct fluorescence responses for L-Lys and d-lysine (D-Lys). Leveraging these properties, the UiO-66-NH2 probe enabled accurate determination of L-Lys concentrations in solution, as well as the enantiomeric excess (ee) values of Lys solutions. Notably, in the detection of Lys enantiomers, the achiral UiO-66-NH2 acted as both a chiral selector and a fluorescent indicator, greatly improving the efficiency and stability of the detection system. The probable mechanism was further elucidated by pH titration experiments and density functional theory calculations. Additionally, the general applicability of this mechanism was validated by similar amino MOFs. The application of the UiO-66-NH2 fluorescent probe in analysis of infant formula milk powders and liquid milk samples confirmed the reliability of the method. Moreover, the construction of fluorescence test paper by immobilizing UiO-66-NH2 onto filler papers enabled the rapid identification of Lys enantiomers. Compared to previous fluorescent analyses of chiral amino acids assisted by additional metal ions, this study presented a novel approach and methodology that offers high efficiency, stability, reproducibility and reusability for the identification and detection of Lys enantiomers, highlighting the potential of the UiO-66-NH2 fluorescent probe in advancing analytical techniques.

Abstract:Electrochemical enantiorecognition of tyrosine (Tyr) isomers using a MOF@CCQDs/NiF electrode prepared by electrodepositing a metal-organic framework (MOF) and chiral carbon quantum dots (CCQDs) on Ni foil is reported. MOF@CCQDs/NiF not only shows highly selective, sensitive and quantitative analysis towards Tyr enantiomers but also presents the ability to determine l-Tyr% in racemic mixtures. This proposed that chiral sensors could be considered for practical applications in the field of Tyr related medical recognition.
