Abstract:Chirality is an important property of nature and it regulates fundamental phenomena in nature and organisms. Here, we develop a chiral electrochemical sensor based on copper–amino acid mercaptide nanorods (L-CuCys NRs) to discriminate tryptophan (Trp) isomers. The chiral L-CuCys NRs are prepared in alkaline solution based on the facile coordination reaction between the sulfhydryl groups of L-Cys and copper ions. Since the stability constant ( K ) of L-CuCys NRs with L-Trp (752) are much higher than that of L-CuCys NRs with D-Trp (242), the cross-linking bonds between L-CuCys NRs and L-Trp are more stable than those between L-CuCys NRs and D-Trp. Consequently, this electrochemical sensor can selectively recognize the Trp isomers with an enantiomeric electrochemical difference ratio ( I L-Trp / I D-Trp ) of 3.22, and it exhibits a detection limit of 0.26 μM for L-Trp. Moreover, this electrochemical sensor can quantitatively measure Trp isomers in complex samples. Importantly, this electrochemical sensor has the characteristics of high stability, good repeatability, easy fabrication, low cost, and efficient discrimination of tryptophan (Trp) isomers.

Abstract:In this work, an electrochemical chiral sensor of a nanowire-like chiral metal-organic framework/multiwalled carbon nanotube-chitosan/glassy carbon electrode( D -MOF/MWCNTs-CS/GCE) was proposed for the enantiorecognition of L/D -tryptophan( L/D -Trp). The asymmetrical spatial structure of D -MOF provides the feasibility for the enantiorecognition of Trp enantiomers. Moreover, differential pulse voltammetry(DPV) was carried out to be the detection method and the DPV peak potential difference(Δ E p ) between L -Trp and D -Trp was referred as the index of the enantiorecognition performance. Several parameters, such as mass ratios and drop-coated volume of MWCNTs-CS, drop-coated volume and concentration of D -MOF, pH and detection temperature of D -MOF/MWCNTs-CS/GCE were optimized for the largest Δ E p value. And the molecular dynamics(MD) simulation was used to elucidate the enantiorecognition mechanism. Furthermore, the proportions of D -Trp( D -Trp%) in Trp mixtures were detected in a good linear relationship with the DPV peak potentials( E p ), and the proposed electrochemical chiral sensor exhibited good reproducibility, stability and enantiorecognition ability. Additionally, the proposed electrochemical chiral sensor( D -MOF/MWCNTs-CS/GCE) has a good application prospect in the fields of biomedicine, clinical diagnosis, chemical production, pharmaceuticals safety and so on.

Abstract:Due to the widespread application of chiral substances in current society, the electrochemical strategy to assemble chiral metal-organic frameworks (CMOFs) as sensors has shown great potential for sensitive enantiorecognition. In this paper, an efficient and highly selective electrochemical sensor was proposed based on multi-walled carbon nanotube (MWCNT) and CMOF, and successfully achieved sensitive chiral recognition of Tryptophan (Trp) enantiomers. Numerous MOFs in multiple synthesis conditions were investigated for a regular process to access the electrochemical chiral recognition. A variety of amino acids were introduced into the MOFs skeleton by a simple hydrothermal method for the enantio-specificity. Meanwhile the MWCNT smoothly integrated with the CMOF to form a functional electrosensing interface with the increase of conductivity. Based on the current ratio (Rcp) of tryptophan enantiomers by different modified electrodes, a series of experimental conditions were optimized for the optimal chiral recognition performance of the CMOF/MWCNT-based sensor, finally reaching 2.9 times of ΔIL/D between enantiomers. In addition, D/L-Trp showed a good linear relationship with the current in the range from 0.4 μM to 19 μM with the detection limit of 0.11 μM for L-Trp and 0.16 μM for D-Trp. The prepared composite-functionalized sensor owned the desired reproducibility and selectivity to identify Tryptophan enantiomers with robust chiral recognition. The research on the structure-activity relationship of chiral MOFs to Tryptophan enantiomers provides a new path to construct more electrochemical sensors for the application of enantiorecognition in pharmaceutical analysis.

Abstract:Chiral discrimination plays an essential role in the fields of medicine, food science, life science, and materials science. So we synthesized CMC-MIL-88(Fe). A chiral material was synthesized by the amidation reaction between MIL-88(Fe)NH2 and sodium carboxymethyl cellulose (CMC). The post-modified chiral material CMC-MIL-88 (Fe) was successfully prepared as revealed by X-ray diffraction (XRD), Infrared spectroscopy (FT-IR), scanning electron microscopy (SEM) and X-ray photoelectron spectroscopy (XPS), which was then modified on a glassy carbon electrode (GCE) to form an electrochemical chiral platform for the detection of tryptophan (Trp). The method then used to describe the electrochemical behaviour of the chiral platform is cyclic voltammetry (CV). The method used to explore the stereoselective identification of the Trp enantiomer is differential pulse voltammetry (DPV). The results showed that CMC-MIL-88(Fe)/GCE exhibited better selectivity towards l-Trp, with an enantioselectivity coefficient of 2.04. The effects of incubation time and pH on sex recognition were also investigated to optimise the parameters. This strategy has excellent sensitivity and high selectivity, which opens a new way for the recognition of Trp enantiomers.

Abstract:A novel electrochemical chiral sensor was constructed for the discrimination of Tryptophan (Trp) by the modification of Au@Pt bimetallic nanoparticles, poly(cysteine) (PL-Cys), and Cu2+-modified β-cyclodextrin (Cu2-β-CD) on the surface of glassy carbon electrode. The constructed sensor was characterized by transmission electron microscopy, infrared spectroscopy, UV–vis adsorption spectra, and the electrochemical experiments. The modified Au@Pt endowed the sensor with high surface area and good conductivity. The electropolymerized PL-Cys facilitated the loading of Cu2-β-CD based on the electrostatic interaction. As a chiral selector, Cu2-β-CD enabled the sensor to effectively identify the Trp enantiomers based on the host–guest interaction with a high the oxidation peak current ratio (IL/ID, IL and ID were the oxidation peak currents of l-Trp and d-Trp measured on the sensor, respectively) of 2.38. Under the optimized conditions, IL/ID increased with the increase of l-Trp in the raceme solution, which was further employed for the detection of Trp in serum samples with satisfactory results. With the good selectivity, stability, and reproducibility, the present electrochemical sensor can be used for the simple, rapid, and efficient chiral recognition and real samples determination.

Abstract:A graphene quantum dots (GQDs)–chitosan (CS) composite film was prepared via successive electrodeposition of GQDs and CS on the surface of a glassy carbon electrode (GCE). The strong interactions between GQDs and CS resulted in the formation of a regular and uniform film, which can be applied in the electrochemical chiral recognition of tryptophan (Trp) enantiomers. CS in the composite film provides a chiral microenvironment, meanwhile, GQDs can amplify the electrochemical signals and improve the recognition efficiency. Due to the synergetic effect of GQDs and CS, chiral recognition of Trp enantiomers is achieved successfully. Compared with previous reports utilizing GQDs in photoluminescent research, this work opens a new avenue for broadening the applications of GQDs in the electrochemically chiral sensors.
Chiral voltammetric sensor for tryptophan enantiomers by using a self-assembled multiwalled carbon nanotubes/polyaniline/sodium alginate composite

Abstract:Due to the crucial role of amino acids in life sciences and pharmaceutics, identification of optical amino acid molecules is of great significance. In this study, the two materials (CNT and PANI) were combined together to obtain the magnification of electrochemical signal by substrate material (CNT/PANI). Then a self-assembled multiwalled carbon nanotubes/polyaniline/sodium alginate (CNT/PANI/SA) nanocomposite with chiral sites and conductive material was synthesized as the electrochemical sensing interface. Next, a novel electrochemical sensing interface was fabricated via modifying the as-prepared chiral material on a polished glassy carbon electrode (CNT/PANI/SA/GCE) for precisely, efficiently, and rapidly differentiation of tryptophan (Trp) enantiomers. It was observed that CNT/PANI/SA/GCE showed desirable stereoselective recognition effect in the variety of signal strength to peak current (Ip) to the different optical activity of Trp enantiomers. In the case of optimal conditions, the peak current ratio in the solution of l-Trp and d-Trp (ID /IL ) was observed to be 2.1 at CNT/PANI/SA/GCE by differential pulse voltammogram (DPV). UV-visible spectroscopy further showed that CNT/PANI/SA had a greater binding energy to l-Trp. Also different factors affecting the enantioselectivity of CNT/PANI/SA/GCE, such as the incubation time, pH, and dropcoating volume of CNT/PANI/SA were optimized. Moreover, the proposed CNT/PANI/SA/GCE showed excellent specific stereoselectivity and anti-interference ability. Besides, the proposed chiral sensing platform can be effectively applied in real samples to detect Trp enantiomers sensitively. This work inspires us a new path for the preparation of substrate material with excellent electrical conductivity, as well as extend its application potential in chiral recognition.

Abstract:Chiral recognition of amino acid plays a significant role in pharmaceutical, medical, and food science. This study describes a chiral sensing system of β-cyclodextrin (β-CD)-coated sulfur quantum dots (CD-SQDs) for the selective fluorescence recognition of tryptophan (Trp) enantiomers. CD-SQDs were prepared by a facile assembly fission method and could selectively recognize L-Trp by the different binding ability between L/D-Trp and β-CD. The inclusion of L-Trp and the stereoselective catalysis of CD-SQDs enzyme mimics cause the increased fluorescence intensity of CD-SQDs, which has a linear response ranging from 10 to 500 nM and the detection limit as 2.3 nM. CD-SQDs also show great selectivity for L-Trp from the commercial compound amino acid injection. The study could provide an effective method for the chiral recognition of amino acid enantiomers based on the catalytic activity of nanoenzymes.

Electrochemical chiral sensing of tryptophan enantiomers by using 3D nitrogen-doped reduced graphene oxide and self-assembled polysaccharides

Abstract:A chiral sensor is described for the enantioselective recognition of L- and D-tryptophan (Trp). The sensor is based on the use of (a) Cu(II) ions coordinated with β-cyclodextrin (Cu-β-CD) that was self-assembled with carboxymethyl cellulose (CD-Cu-CMC) as a chiral selector, (b) of N-doped reduced graphene oxide (N-rGO) as substrate materials, and (c) of differential pulse voltammetry that was used for enantiorecognition. The 3D N-rGO was prepared by using reduced graphene oxide and pyrrole as the starting materials. Electrostatic interaction occurs between the carboxy groups of CMC and Cu(II) ions in Cu-β-CD. The FT-IR, SEM, XRD and XPS techniques showed that 3D N-rGO and the CD-Cu-CMC composite were successfully synthesized. The 3D N-rGO enabled the immobilization of the chiral selector (CD-Cu-CMC) and improves the active areas. A glassy carbon electrode was modified with N-rGO/CD-Cu-CMC and then showed a stronger electrochemical signal for L-Trp than for D-Trp, typically at a working potential of around 0.78 V (vs. SCE). UV-vis spectroscopy proved that CD-Cu-CMC has a higher affinity for D-Trp. The enantioselectivity for D-Trp over L-Trp is 4.72. The modified electrode had a limit of detection of 0.063 μM and 0.0035 μM for L-Trp and D-Trp, respectively, with a linear response range of 0.01 mM to 5 mM. The sensor was used to detect Trp (D- or L-Trp) in spiked real human urine and human serum protein samples. Chiral MOFs encapsulated by polymers with poly-metallic coordination as chiral biosensors

Abstract:Chiral materials have drawn the widespread attention for their its chiral recognition ability. The design and synthesis of chiral material are of importance owing to the unpredictability in controlling chirality during the synthesis process. To circumvent problems, a chiral MOF (D-His-ZIF-8) was synthesized by ligand exchange of 2-methylimidazole (Hmim) on ZIF-8 by D-histidine (D-His), which can be treated as chiral host to distinguish amino acid enantiomers. The obtained D-His-ZIF-8 can provide chiral nanochannels for amino acid guests. Meanwhile, polynary transition-metal ion (Co2+ and Fe3+) coordinating with polydopamine (PDA) wrapped on the surface of D-His-ZIF-8 can increase the active sites. The electrochemical chiral recognition behavior showed that D-His-ZIF-8@CoFe-PDA exhibited good recognition of the tryptophan enantiomer (L/D-Trp) (working potential of -0.2 V vs. Hg/HgCl2). The LOD and LOQ of L-Trp were 0.066 mM and 0.22 mM, respectively, while the LOD and LOQ of D-Trp were 0.15 mM and 0.50 mM, respectively. Finally, the usefulness of D-His-ZIF-8@CoFe-PDA/GCE was evaluated with a recovery of 94.4-103%. The analysis of real samples shows that D-His-ZIF-8@CoFe-PDA/GCE is a feasible sensing platform for the detection of L-Trp and D-Trp.
Magneto-Electrochemical Method for Chiral Recognition of Amino Acid Enantiomers

Abstract:Chiral recognition of enantiomers with identical mirror-symmetric molecular structures is important for the analysis of biomolecules, and it conventionally relies on stereoselective interactions in chiral chemical environments. Here, we develop a magneto-electrochemical method for the enhanced detection of chiral amino acids (AAs), that combines the advantages of the high sensitivity of electrochemiluminescence (ECL) biosensors and chirality-induced effects under magnetic field. The ECL difference between L- and D-enantiomers can be amplified by over 35-fold under a field of 3.5 kG, and the chiral discrimination can be achieved in dilute AA solutions down to nM level. The field-dependent ECL and chronocoulometry measurements suggest that chiral AAs can lock the spins on their radicals and thus enlarge the ECL change under applied magnetic field (magneto-ECL, MECL), which explains the field-enhanced chiral discrimination of AA enantiomers. Finally, a detailed protocol is demonstrated for the identification of unknown AA solutions, in which the species, chirality and concentration of AAs can be determined simultaneously from the 2D plots of the ECL and MECL results. This work benefits the development of field-assisted detection methods and represents a promising and universal strategy for the comprehensive analysis of chiral biomolecules.

Abstract:Recognition and separation of chiral isomers are of great importance in both industrial and biological applications. However, owing to identical molecular formulas and chemical properties of enantiomers, signal transduction and amplification are still two major challenges in chiral sensing. In this study, we developed an enantioselective device by integrating chiral covalent organic framework nanosheets (CONs) with nanochannels for sensitive identification and quantification of enantiomers. Using 3,4-dihydroxyphenylalanine (DOPA) as the model analyte, the as-prepared chiral nanofluidic device exhibits a remarkable chiral recognition ability to l -DOPA than d -DOPA. More importantly, due to the chelation of DOPA with Fe 3+ ions, it can efficiently block the ion transport through channel and shield the channel surface charge, which will amplify the difference in the electrochemical response of l -DOPA and d -DOPA. Therefore, a sensitive chiral recognition can be achieved using the present nanofluidic device coupled using electrochemical amplification strategy. Notably, using this method, an ultra-low concentration of l -DOPA (as low as 0.21 pM) can be facilely and successfully detected with a linear range of 1 pM–10 μM. This study provides a reliable and sensitive approach for achieving highly selective detection of chiral molecules.

Abstract:Abstract Chirality exists everywhere in nature, so chiral recognition is also an extremely important field. This paper explores the recognition of amino acid configuration by an electrochemical chiral sensing platform constructed with carbon nanomaterials (carbon quantum dots (CQDs) as the substrate material) combined with chiral selectors (chitosan (CS)). CQDs/CS, which is bonded by intermolecular force, and CQDs‐CS, which is formed by covalent grafting through amidation, are selected as electrochemical chiral recognition materials, and a series of characterization methods are carried out to indicate the successful preparation of the materials, and then attached to the glass carbon electrode (GCE), forming a chiral sensing platform for amino acid enantiomers. The differential voltammetry (DPV) method was used to convert the chemical signal into electrical signal. The results showed that compared with CQDs/CS, CQDs‐CS had better enantiomer resolution to tryptophan (Trp), and the peak current ratio (I L /I D ) was 2.28.

Abstract:In this work, enhanced tryptophan (Trp) isomers recognition was successfully demonstrated on (CS/PAA) 3.5 @PEDOT:PSS/GCE, a multilayer chiral sensor with good stability and reproducibility. The (CS/PAA)n multilayers chiral interface was first fabricated via alternating self-assembly of chiral chitosan (CS) and achiral polyacrylic acid (PAA). Conductive PEDOT:PSS was then compounded with (CS/PAA) n multilayers to obtain the chiral sensor for the electrochemical recognition of Trp isomers. The structure of the sensor and its chirality properties for Trp isomers were characterized by fourier transform infrared spectroscopy (FT-IR),scanning electron microscopy (SEM) and electrochemical methods. The SEM images showed uniform distribution of PEDOT:PSS in the multilayer films, which changed the internal structure of the (CS/PAA) 3.5 . Consequently, (CS/PAA) 3.5 @PEDOT:PSS multilayers rendered more chiral centers in addition to improved good conductivity, which significantly amplified the oxidation peak current ratio of D-Trp to L-Trp (I D /I L ) up to 6.71 at 25 °C. In addition, a linear relationship was observed between the peak current and Trp enantiomer concentration in the range of 0.002–0.15 mM, and the detection limits of D-Trp and L-Trp were 0.33 and 0.67 μM, respectively. More importantly, the percentage of D-Trp in non-racemic Trp enantiomers mixture solutions were successfully determined on the chiral interface, showing its effectiveness and promising potential in practical applications.

Abstract:A range of electrochemical sensors capable of enantiomer discrimination have been developed, but the creation of their electrochemical interfaces remains complex. This often entails combining externally introduced chiral centers with conductive materials. This study marks a significant departure from this approach by exploiting the inherent chirality of single-walled carbon nanotubes (SWCNTs) for enantiomer discrimination. We have crafted a miniaturized, enantioselective electrochemical microsensor by seamlessly integrating high-purity SWCNTs, possessing inherent chirality, with an ultra-sensitive carbon fiber microelectrode. Using differential pulse voltammetry, we observed a notable difference in peak current, highlighting the sensor's precise ability to distinguish between enantiomers of 3,4-dihydroxyphenylalanine (DOPA). Furthermore, we investigated the intricacies of the interplay between enantioselective interfaces and the molecules under discrimination, offering deeper insights into the mechanisms governing this phenomenon. This study provides substantial momentum into the realm of enantioselective electrochemical sensing and sheds light on the unique attributes of inherently chiral carbon nanotubes in such applications. This paves the way for a new breed of chiral sensors that are simpler, more efficient, and cost-effective.

Abstract:A novel chiral sensing platform, 6-O-α-maltosyl-β-cyclodextrin (Mal-βCD)-based film, is proposed for selective electrochemical recognition of tyrosine (Tyr) enantiomers. Black phosphorus nanosheets (BP NSs) and Mal-βCD modified glassy carbon electrode (Mal-βCD/BP NSs/GCE) were prepared by a layer-to-layer drop-casting method, and the platform was easy to fabricate and facile to operate. It is proposed that the amino and hydroxyl groups of the Tyr enantiomers and the chiral hydroxyl groups of Mal-βCD selectively form intermolecular hydrogen bonds to dominate effective chiral recognition. Two linear equations of Ip (μA) = 11.40 CL-Tyr (mM) + 0.28 (R2 = 0.99147) and Ip (μA) = 7.96 CD-Tyr (mM) + 0.22 (R2 = 0.99583) in the concentration range 0.01-1.00 mM have been obtained. The limits of detection (S/N=3) for L-Tyr and D-Tyr were 4.81 and 6.89 µM, respectively. An interesting phenomenon was that the value of IL-Tyr/ID-Tyr (1.51) in this work was slightly higher than the value of IL-Trp/ID-Trp (1.49) reported in our previous study, where tryptophan (Trp) enantiomers were electrochemically recognized by Nafion (NF)-stabilized BPNSs-G2-β-CD composite. The two similar sensors fabricated by different methods showed different recognition ability toward either Tyr or Trp enantiomers, and the underlying mechanism was discussed in detail. More importantly, the proposed chiral sensor enables prediction of the percentages of D-Tyr in racemic Tyr mixtures. The chiral sensor may provide a novel approach for the fabrication of novel chiral platforms in the practical detection of L- or D-enantiomer in racemic Tyr mixtures.Graphical abstract.

Abstract:This work reports an enantioselective electrochemical sensor toward tyrosine (Tyr) enantiomers based on macroporous carbon embedded with sulfato-β-cyclodextrin (MPC-SCD) hybrid. First, MPC-SCD hybrid was synthesized via adsorption of sulfato-β-cyclodextrin (SCD) by macroporous carbon (MPC) in aqueous solution, and characterized using transmission electron microscopy (TEM), scanning electron microscopy (SEM), thermogravimetric analysis (TGA), and Fourier transform infrared spectroscopy (FTIR). Subsequently, an enantioselective electrochemical sensor was fabricated by modifying glassy carbon electrode (GCE) with MPC-SCD hybrid, and its electrochemical performances were characterized by electrochemical impedance spectroscopy (EIS) and cyclic voltammograms (CV). Furthermore, the electrochemical sensor was utilized for chiral analysis toward Tyr enantiomers by differential pulse voltammograms (DPV). The peak currents of Tyr enantiomers increased linearly with increasing concentrations over the range of 1–500 μM. In addition, the sensitivity were calculated as 84.59 and 73.02 μA/mM for D-Tyr and L-Tyr, respectively, and the detection limits were 0.20 and 0.26 μM. Furthermore, multiple linear regression (MLR) chemometrics modeling method was used for simultaneously analysis of Tyr enantiomers. The presented sensor could provide a promising protocol for designing novel electrochemical platforms in the field of chiral analysis.

Abstract:The development of a highly selective and sensitive optical method for quantitative enantioselective sensing of chiral amino acids in the presence of other amino acid interferences has remained a major challenge to date. Herein, a homochiral metal–organic framework (MOF), [Zn(L)(2,2′‐bipy)]·H2O (LNNU‐1) (H2L = HOOCC6H4CH2PO(OH)(OC2H5), 2,2′‐bipy = 2,2′‐bipyridine), is designed to interact with tryptophan via multiple noncovalent interactions. The interactions result in the emergence of a strong induced circular dichroism signal. This enables accurate determination of the absolute configuration and enantiomeric ratio of chiral tryptophan in water, mixed natural amino acids, and blood plasma components without any interference from other coexistent species. A simple device with advantageous characteristics of reusability and simple manipulation is designed for enantioselective sensing assay. The obtained results suggest that the rationally designed homochiral MOF is a promising candidate for enantioselective sensing of chiral species.

Abstract:Polysaccharide materials have wide applications in the field of chiral recognition. Abstract:Chiral recognition, especially rendering specificity in biomolecular recognition, is a basic property of many biomolecules. Being closely related to the chirality of biomolecules, it has been regarded as one of the most important areas in biological and medical sciences due to the different effects in biological systems. Based on the possible interactions between chiral selectors and the enantiomers, various methods including chromatographic techniques such as gas or liquid chromatography, electromigration techniques such as capillary electrophoresis and so on were developed for the chiral separation and recognition of different optical isomers. Recently, chemical sensors and biosensors have been gradually designed and developed for the analysis of chiral compounds. Based on the difference in electrical response to different isomers, chiral identifications can be successfully implemented. Major successes in enantiomer recognition based on electrochemical analysis are reviewed. The research data available for highly enantio -selective recognition are categorized into several subgroups according to specific topics and critically discussed for the period since 1994, and the latest techniques for electrochemical chiral recognition of enantiomers were also reviewed. Simultaneously, a brief conclusive summary and future perspectives are presented, and the challenges and scientific prospects of the newest generation of electrode modifiers in electrochemical sensing are also proposed.
