Sensitive and Selective Whole Cell Detection of Escherichia Coli from Waters by a Paper-Based Immunosensor with Laser-Induced Fluorescence Detection

Abstract:In this work, a method of immunoassay on a paper-based chip was developed for the determination of whole cells of Escherichia coli ( E. coli ). Laser-induced fluorescence (LIF) detection was firstly applied in the detection of bacteria on the paper-based chip. A paper chip-LIF interface was applied to perform high-throughput detection. E. coli were immobilized on the paper by the antibodies. Aptamers connected with initiator DNA were applied to selectively bind with E. coli and triggered hybridization chain reaction to amplify the fluorescence signals. The function of the aptamers connected with initiator DNA at different ends was studied. Under the optimized conditions, the detection limit for E. coli was as low as 4.50 × 10 2 CFU/mL. Only 5 μL solution of E. coli was needed for each test and the analysis time was less than 2.5 h much quick than that of conventional methods’. The study of specificity showed the method had good selectivity. Finally, the method was applied to determine E. coli in samples from waters of Fuzhou City. E. coli was successfully found from a tributary of Minjiang river and a suburb pond. The recoveries for E. coli ranged from 92.9% to 114%, which indicated the method was reliable. The developed method was fast, sensitive, selective, low-cost and reagent-saving in the determination of E. coli in samples.
