Abstract:Prunus mume is a small deciduous arbour tree species with a weak trunk and a shallow and broad root system. The flower buds of P. mume are covered with scales, and usually, one bud has only one flower. Branching is weak and gradually weakens with increasing tree age; the majority of the long twigs are located in the middle and upper parts of the branches. Adaxial leaf surfaces in P. mume bear hairs on young leaves, which gradually fall as the leaves mature. The fruiting branches of P. mume trees can be divided into six categories: water sprouts, long branches, middle branches, short branches, tiny branches and needle branches. According to the characteristics of morphological differentiation and biochemical changes, and based on the differentiation period of the sepal, in the stage of morphological differentiation, we distinguish flower primordium differentiation and the differentiation of flower organs. The pistil of P. mume consists of a single carpel, and the ovary is superior. There are generally three peak fruit-drop periods in the early stage, and some varieties show fruit-drop before harvest. The phenological period of P. mume varies with region and variety. Its main feature is early flowering, first flowering and then leafing; the fruit growth and development periods are short, and leaf drop is early.

Abstract:Prunus mume (P. mume Sieb. et Zucc., 2n = 2x = 16) is a popular ornamental plant widely cultivated in East Asia, where it was domesticated over 3000 years ago. As an important member of the genus Prunus, it plays a pivotal role in phylogenetic studies of the Rosaceae family. To understand the origin of P. mume, it is important to know how the proto-species of P. mume did form and where it has its origin. Specimens of wild P. mume have been discovered in Yunnan Sichuan, Hubei, Guizhou and Tibet, China.

Abstract:Prunus is an economically important fruit tree genus and includes many stone fruit trees such as peach (Prunus persica Batsch), apricot, Japanese apricot (Prunus mume), sweet cherry (Prunus avium L.), European plum (Prunus domestica L.), Japanese plum (Prunus salicina Lindl.) and almond [Prunus dulcis (Mill.) D. A. Webb.]. The characterisation of genes associated with agriculturally important traits such as fruit ripening, dormancy, self-incompatibility, fruit quality and various other developmental processes is important to improve Prunus breeding programmes. Based on high- density molecular genetic maps, many genes are located on the genome, and subsequently, most of them were fine-mapped and further identified by positional cloning. Currently, with the availability of the P. mume genome sequence, the identification of new genes is significantly accelerated.

Abstract:Microarray analysis is a technique that can be employed to provide expression profiles of single genes and new insights to elucidate the biological mechanisms responsible for fruit development. To evaluate expression of genes mostly engaged in fruit development between Prunus mume and Prunus armeniaca, we first identified differentially expressed transcripts along the entire fruit life cycle by using microarrays spotted with 10,641 ESTs collected from P. mume and other Prunus EST sequences. A total of 1418 ESTs were selected after quality control of microarray spots and analysis for differential gene expression patterns during fruit development of P. mume and P. Armeniaca. From these, 707 up-regulated and 711 down-regulated genes showing more than two-fold differences in expression level were annotated by GO based on biological processes, molecular functions and cellular components. These differentially expressed genes were found to be involved in several important pathways of carbohydrate, galactose, and starch and sucrose metabolism as well as in biosynthesis of other secondary metabolites via KEGG. This could provide detailed information on the fruit quality differences during development and ripening of these two species. With the results obtained, we provide a practical database for comprehensive understanding of molecular events during fruit development and also lay a theoretical foundation for the cloning of genes regulating in a series of important rate-limiting enzymes involved in vital metabolic pathways during fruit development.

Abstract:Abstract Prunus mume Sieb. et Zucc. is an important fruit crop of the subtropical region, originating in China. It blooms earlier than other deciduous fruit trees, but different regions have different blooming periods. The time of anthesis is related to the dormancy period, and a certain amount of chilling promotes bud break and blooming. To identify the relationship between blooming time and the climatic adaptation of P. mume cultivars in China, the nuclear and chloroplast genomes of 19 cultivars from the main cultivation areas of P. mume in China were resequenced. The average depth of coverage was 34X–76X, and a total of 388,134 single nucleotide polymorphisms were located within the coding regions of the gene (CDs). Additionally, the 19 cultivar accessions were divided into three groups based on their blooming time: early, mid, and late. Associated with the blooming time groups, 21 selective sweep regions were identified, which could provide evidence supporting the possible model of P. mume domestication originating due to natural selection. Furthermore, we identified a flowering gene, FRIGIDA‐LIKE 3 (FRL3), seems to affect the blooming time and the climatic adaptation of P. mume cultivars. This study is a major step toward understanding the climatic adaptation of P. mume cultivars in China.

Abstract:For protecting a normal reference for bloom,guaranteeing high-quality produce to provide a theory of Prunus mume on the production,and lay a foundation for further research pollen germination and cross-fertilize.The authors have investigated biological traits of flowers and pollen germination among 47Prunus mume cultivars,including aroma,the rate of incomplete flower and pollen quantity and measured the rate of pollen germination.Subsequently,the authors have got some results as follows through the correlative statistics analysis:Chinese cultivars’aroma is more fragrant than Japanese cultivars’;the rate of incomplete flower covers 1.17%-59.65%;there was an obvious diversity among the pollen quantity of different cultivars,and the coverage was from 0 to 1265;there was an obvious diversity among the rate of pollen germination of different cultivars,the highest rate was 84.76%and the lowest rate was 0;there was an obvious correlativity between pollen quantity and the rate of pollen germination.Incomplete flowers occupy a high proportion and flowering periods were easily endangered by low temperature,which resulted in Prunus mume yield low and unsteady.This experiment showed that different species of Prunus mume presented a marked diversity among the rate of incomplete flower,pollen quantity and germination.

Abstract:Mei (Prunus mume) is an ornamental woody plant that has been domesticated in East Asia for thousands of years. High diversity in floral traits, along with its recent genome sequence, makes mei an ideal model system for studying the evolution of woody plants. Here, we investigate the genetic architecture of floral traits in mei and its domestication history by sampling and resequencing a total of 351 samples including 348 mei accessions and three other Prunus species at an average sequencing depth of 19.3×. Highly-admixed population structure and introgression from Prunus species are identified in mei accessions. Through a genome-wide association study (GWAS), we identify significant quantitative traits locus (QTLs) and genomic regions where several genes, such as MYB108, are positively associated with petal color, stigma color, calyx color, and bud color. Results from this study shed light on the genetic basis of domestication in flowering plants, particularly woody plants.

Abstract:Mei, Prunus mume Sieb. et Zucc., is an ornamental plant popular in East Asia and, as an important member of genus Prunus , has played a pivotal role in systematic studies of the Rosaceae. However, the genetic architecture of botanical traits in this species remains elusive. This paper represents the first genome-wide mapping study of quantitative trait loci (QTLs) that affect stem growth and form, leaf morphology and leaf anatomy in an intraspecific cross derived from two different mei cultivars. Genetic mapping based on a high-density linkage map constricted from 120 SSRs and 1,484 SNPs led to the detection of multiple QTLs for each trait, some of which exert pleiotropic effects on correlative traits. Each QTL explains 3-12% of the phenotypic variance. Several leaf size traits were found to share common QTLs, whereas growth-related traits and plant form traits might be controlled by a different set of QTLs. Our findings provide unique insights into the genetic control of tree growth and architecture in mei and help to develop an efficient breeding program for selecting superior mei cultivars.

Abstract:Prunus mume is one of the most ancient medicinal herbs and health foods commonly used in Asian countries. It is widely used as a constituent of many medicinal preparations and as a food ingredient for its beneficial health effects. In this review, we retrieved reports from PubMed, embase, Scopus, and SciFinder databases, to collect extensive scientific evidence on the phytochemical constituents, pharmacological properties, and clinical applications of Prunus mume . The literature review revealed that approximately 192 compounds have been isolated from different parts of the plant, and their molecular structures have been identified. The pharmacological properties of the plant, including anti-diabetic, liver-protective, antitumor, antimicrobial, antioxidant, and anti-inflammatory activities, as well as their underlying mechanisms, have been clarified by in vitro and in vivo studies. Clinical studies, although very limited, have been highlighted in this review to provide a reference for further exploration on therapeutic applications of the plant.

Abstract:A high-performance liquid chromatography(HPLC)method for the determination of organic acids in flower,branch and leaf extract of Prunus mume is proposed here.Agilent TC-C18 column(250 mm×4.6 mm i.d.,5μm) was used at 30℃ and mobile phase was 0.01mol/L KH2PO4 solution containing 3% methanol with pH2.85 at 1.0 mL /min flow speed,detection wavelength was at 210 nm.The relative standard deviations were 0.36%~1.61% and the recoveries were 92.7%~104.2%.The correlation coefficients for the linear equations were 0.9943~1.0000.The method was rapid and accurate for quantitative analysis of ten organic acids including oxalic,tartaric,malic,L-ascorbic,lactic,acetic,citric,maleic,fumaric,succinic acids.Studies have showed that the total amount of organic acids in branch extract was 5.75% and they were mainly were citric acid and tartaric acid,and in leaf extract was 5.86% and mainly were oxalic acid and citric acid.Flower extract contained much less organic acids.

Abstract:Prunus mume flowers are used as traditional edible and medicinal materials in China. In this study, phenolic compounds and antioxidant activity of ethanolic extract from flowers of P. mume in China were investigated for the first time. The total phenol content was estimated as gallic acid equivalents by the Folin–Ciocalteu reagent method. The antioxidant activity was measured by DPPH, ABTS+, and OH free radicals scavenging and ferric-reducing antioxidant power (FRAP). Three chlorogenic acid isomers, namely, 3-O-caffeoylquinic, 4-O-caffeoylquinic and 5-O-caffeoylquinic acids, were isolated and purified by preparative HPLC from the ethanolic extract and identified by UV, MS and NMR. The contents of these isolated compounds were quantified by HPLC. Results showed that 5-O-caffeoylquinic acid was of the highest level in these three isomers. The ethanolic extract demonstrated activity to some degree in all the antioxidant assays. In all tested assays, all of the isolated chlorogenic acid isomers exhibited strong antioxidant activities, which were almost the same. The results showed that chlorogenic acid isomers are the key phenolic compounds which are responsible for antioxidant activity of the ethanolic extract from Chinese P. mume flowers.

Abstract:Backgrounds: Prunus mume in the Rosaceae and commonly referred to as mei or Chinese plum is widely used as a traditional ornamental flowering plant and fruit tree in China. Although some population and genetic analyses have been conducted for this species, no extensive comparisons of genetic variation from plastomes have yet been investigated. Methods: We de novo assembled a total of 322 complete P. mume plastomes in this study and did a series of comparative analyses to better resolve pan-plastomic patterns of P. mume . To determine the phylogeny and domestication history of this species, we reconstructed the phylogenetic tree of Prunus genus, and resolved the population structure of P. mume . We also examined the nucleotide variation of P. mume to find potential DNA barcodes. Results: The assembled plastomes exhibited a typical quadripartite structure and ranged from 157,871 bp to 158,213 bp in total size with a GC content ranging from 36.73 to 36.75%. A total of 112 unique genes were identified. Single nucleotide variants (SNVs) were the most common variants found among the plastomes, followed by nucleotide insertions/deletions (InDels), and block substitutions with the intergenic spacer (IGS) regions containing the greatest number of variants. From the pan-plastome data six well-supported genetic clusters were resolved using multiple different population structure analyses. The different cultivars were unevenly distributed among multiple clades. We also reconstructed a phylogeny for multiple species of Prunus to better understand genus level diversity and history from which a complex introgressive relationship between mei and other apricots/plums was resolved. Conclusion: This study constructed the pan-plastome of P. mume , which indicated the domestication of P. mume involved multiple genetic origins and possible matrilineal introgression from other species. The phylogenetic analysis in Prunus and the population structure of P. mume provide an important maternal history for Prunus and the groundwork for future studies on intergenomic sequence transfers, cytonuclear incompatibility, and conservation genetics.

Abstract:Japanese apricot (Prunus mume Sieb. et Zucc) is an important economic fruit crop in China and Japan, with more than 200 cultivars in China. It is one of the most valuable materials used in the food and winemaking industries and is believed to contain many physiochemicals beneficial for human health. MicroRNAs (miRNAs) are a class of endogenous, small, noncoding RNAs that regulate gene expression by mediating gene silencing at transcriptional and post-transcriptional levels in higher plants. High-throughput sequencing is used to identify and quantitatively profile small RNAs in Japanese apricot. Previous studies have shown comparative identification of miRNAs between perfect and imperfect Japanese apricot flowers and some small RNAs related to the flower opening process.
A Comparative Analysis of Floral Scent Compounds in Intraspecific Cultivars of Prunus mume with Different Corolla Colours

Abstract:Prunus mume is the only fragrant flowering species of Prunus. According to the previous studies, benzyl acetate and eugenol dominate its floral scent. However, the diversity of its floral scents remains to be elucidated. In this work, the floral volatiles emitted from eight intraspecific cultivars of P. mume with white, pink and red flowers, were collected and analyzed using headspace solid-phase microextraction combined with gas chromatograms-mass spectrometry (HS-SPME-GC-MS). In total, 31 volatile compounds were identified, in which phenylpropanoids/benzenoids accounted for over 95% of the total emission amounts. Surprisingly, except for benzyl acetate and eugenol, several novel components, such as benzyl alcohol, cinnamyl acohol, cinnamy acetate, and benzyl benzoate were found in some cultivars. The composition of floral volatiles in cultivars with white flowers was similar, in which benzyl acetate was dominant, while within pink flowers, there were differences of floral volatile compositions. Principal component analysis (PCA) showed that the emissions of benzyl alcohol, cinnamyl alcohol, benzyl acetate, eugenol, cinnamyl acetate, and benzyl benzoate could make these intraspecific cultivars distinguishable from each other. Further, hierarchical cluster analysis indicated that cultivars with similar a category and amount of floral compounds were grouped together. Our findings lay a theoretical basis for fragrant plant breeding in P. mume.

Palynological Study on Prunus Domestica and Its Related Species

Abstract:Thirteen plum accessions representing Prunus domestica and its related species were investigated with scanning electron microscopy(SEM), and the cluster analysis using average linkage means was conducted with nine quantitative and two qualitative characters based on palynological data. The observed results showed that all pollen grains of P. domestica tested were isopolar, radially symmetrical, small to medium in size, circular or sub-cricular in polar views and elliptical in equatorial views, the ornamentation characterized by small perforations and irregular ridges, this was significantly different from that of its related species. Pollen was mostly tri-colporate and the aperture belonged to N3P4C5 type, except the tetra-colporate observed in the individual pollen gains of Mt. Royal. The results of variance analysis for quantitative characters showed that there was significant difference in the pollen morphological characters between P. domestica and its related species. Cluster analysis divided all tested materials into two groups, e. g. the polyploid group and the diploid group. It was suggested that wild P. domestica had close relationship with P. spinosa, might contain the germplasm of P. spinosa, and the cultivaled P. domestica might originate from Wild P. domestica Finally, the evolution of Prunus species tested were discussed based on pollen morphology. This study demonstrated that palynology data incorporated with cluster analysis would have taxonomic value and be useful for elucidating the interspecies relationships of Prunus.

Abstract:Prunus mume Sieb. Et Zucc (P. mume) is an acidic fruit native to China (named Chinese Mei or greengage plum). It is currently cultivated in several Asian countries, including Japan ("Ume"), Korea (Maesil), and Vietnam (Mai or Mo). Due to its myriad nutritional and functional properties, it is accepted in different countries, and its characteristics account for its commercialization. In this review, we summarize the information on the bioactive compounds from the fruit of P. mume and their structure-activity relationships (SAR); the pulp has the highest enrichment of bioactive chemicals. The nutritional properties of P. mume and the numerous uses of its by-products make it a potential functional food. P. mume extracts exhibit antioxidant, anticancer, antimicrobial, and anti-hyperuricaemic properties, cardiovascular protective effects, and hormone regulatory properties in various in vitro and in vivo assays. SAR shows that the water solubility, molecular weight, and chemical conformation of P. mume extracts are closely related to their biological activity. However, further studies are needed to evaluate the fruit's potential nutritional and functional therapeutic mechanisms. The industrial process of large-scale production of P. mume and its extracts as functional foods or nutraceuticals needs to be further optimized.
