As an important member of the genus Prunus, it plays a pivotal role in phylogenetic studies of the Rosaceae family. Abstract:Prunus mume Sieb. Et Zucc (P. mume) is an acidic fruit native to China (named Chinese Mei or greengage plum). It is currently cultivated in several Asian countries, including Japan ("Ume"), Korea (Maesil), and Vietnam (Mai or Mo). Due to its myriad nutritional and functional properties, it is accepted in different countries, and its characteristics account for its commercialization. In this review, we summarize the information on the bioactive compounds from the fruit of P. mume and their structure-activity relationships (SAR); the pulp has the highest enrichment of bioactive chemicals. The nutritional properties of P. mume and the numerous uses of its by-products make it a potential functional food. P. mume extracts exhibit antioxidant, anticancer, antimicrobial, and anti-hyperuricaemic properties, cardiovascular protective effects, and hormone regulatory properties in various in vitro and in vivo assays. SAR shows that the water solubility, molecular weight, and chemical conformation of P. mume extracts are closely related to their biological activity. However, further studies are needed to evaluate the fruit's potential nutritional and functional therapeutic mechanisms. The industrial process of large-scale production of P. mume and its extracts as functional foods or nutraceuticals needs to be further optimized.

Abstract:Microarray analysis is a technique that can be employed to provide expression profiles of single genes and new insights to elucidate the biological mechanisms responsible for fruit development. To evaluate expression of genes mostly engaged in fruit development between Prunus mume and Prunus armeniaca, we first identified differentially expressed transcripts along the entire fruit life cycle by using microarrays spotted with 10,641 ESTs collected from P. mume and other Prunus EST sequences. A total of 1418 ESTs were selected after quality control of microarray spots and analysis for differential gene expression patterns during fruit development of P. mume and P. Armeniaca. From these, 707 up-regulated and 711 down-regulated genes showing more than two-fold differences in expression level were annotated by GO based on biological processes, molecular functions and cellular components. These differentially expressed genes were found to be involved in several important pathways of carbohydrate, galactose, and starch and sucrose metabolism as well as in biosynthesis of other secondary metabolites via KEGG. This could provide detailed information on the fruit quality differences during development and ripening of these two species. With the results obtained, we provide a practical database for comprehensive understanding of molecular events during fruit development and also lay a theoretical foundation for the cloning of genes regulating in a series of important rate-limiting enzymes involved in vital metabolic pathways during fruit development.
Antioxidant Activity of Chinese Mei (prunus Mume) and Its Active Phytochemicals

Abstract:Prunus mume fruits have been used as a healthy food and traditional drug in China. The present study investigated the phenolic compounds and antioxidant activity of ethyl acetate extract from fruits of P. mume in China. Total phenol content was determined as gallic acid equivalents by the Folin-Ciocalteu method. The antioxidant activity was measured by DPPH and ABTS(+) radicals scavenging and lipid peroxidation inhibition activities. Three chlorogenic acid isomers, namely, chlorogenic acid, neochlorogenic acid and cryptochlorogenic acid, were isolated and purified by preparative HPLC from the extract and identified by ultra-violet (UV), MS and NMR and the latter two compounds were identified in P. mume fruits for the first time. The contents of these isolated compounds were quantified by HPLC. Results showed that chlorogenic acid was of the highest level in these three isomers. The ethyl acetate extracts demonstrated activity to some degree in all the antioxidant assays. In all tested assays, all of the isolated chlorogenic acid isomers exhibited strong antioxidant activities, which were almost the same. The results showed that chlorogenic acid isomers are the key phenolic compounds which are responsible for antioxidant activity of the ethyl acetate extract from P. mume fruits in China.

Abstract:Prunus mume is one of the most ancient medicinal herbs and health foods commonly used in Asian countries. It is widely used as a constituent of many medicinal preparations and as a food ingredient for its beneficial health effects. In this review, we retrieved reports from PubMed, embase, Scopus, and SciFinder databases, to collect extensive scientific evidence on the phytochemical constituents, pharmacological properties, and clinical applications of Prunus mume . The literature review revealed that approximately 192 compounds have been isolated from different parts of the plant, and their molecular structures have been identified. The pharmacological properties of the plant, including anti-diabetic, liver-protective, antitumor, antimicrobial, antioxidant, and anti-inflammatory activities, as well as their underlying mechanisms, have been clarified by in vitro and in vivo studies. Clinical studies, although very limited, have been highlighted in this review to provide a reference for further exploration on therapeutic applications of the plant.

Abstract:Prunus mume is a small deciduous arbour tree species with a weak trunk and a shallow and broad root system. The flower buds of P. mume are covered with scales, and usually, one bud has only one flower. Branching is weak and gradually weakens with increasing tree age; the majority of the long twigs are located in the middle and upper parts of the branches. Adaxial leaf surfaces in P. mume bear hairs on young leaves, which gradually fall as the leaves mature. The fruiting branches of P. mume trees can be divided into six categories: water sprouts, long branches, middle branches, short branches, tiny branches and needle branches. According to the characteristics of morphological differentiation and biochemical changes, and based on the differentiation period of the sepal, in the stage of morphological differentiation, we distinguish flower primordium differentiation and the differentiation of flower organs. The pistil of P. mume consists of a single carpel, and the ovary is superior. There are generally three peak fruit-drop periods in the early stage, and some varieties show fruit-drop before harvest. The phenological period of P. mume varies with region and variety. Its main feature is early flowering, first flowering and then leafing; the fruit growth and development periods are short, and leaf drop is early.

Abstract:Prunus mume flowers are used as traditional edible and medicinal materials in China. Abstract:Abstract Prunus mume Sieb. et Zucc. is an important fruit crop of the subtropical region, originating in China. It blooms earlier than other deciduous fruit trees, but different regions have different blooming periods. The time of anthesis is related to the dormancy period, and a certain amount of chilling promotes bud break and blooming. To identify the relationship between blooming time and the climatic adaptation of P. mume cultivars in China, the nuclear and chloroplast genomes of 19 cultivars from the main cultivation areas of P. mume in China were resequenced. The average depth of coverage was 34X–76X, and a total of 388,134 single nucleotide polymorphisms were located within the coding regions of the gene (CDs). Additionally, the 19 cultivar accessions were divided into three groups based on their blooming time: early, mid, and late. Associated with the blooming time groups, 21 selective sweep regions were identified, which could provide evidence supporting the possible model of P. mume domestication originating due to natural selection. Furthermore, we identified a flowering gene, FRIGIDA‐LIKE 3 (FRL3), seems to affect the blooming time and the climatic adaptation of P. mume cultivars. This study is a major step toward understanding the climatic adaptation of P. mume cultivars in China.

Meizi-Consuming Culture That Fostered the Sustainable Use of Plum Resources in Dali of China: An Ethnobotanical Study

Abstract:Prunus mume has been cultivated for more than three millennia with important edible, ornamental, and medicinal value. Due to its sour taste, the Prunus mume fruit (called Meizi in Chinese and Ume in Japanese) is not very popular compared to other fruits. It is, however, a very favorite food for the Bai people living in Eryuan County, Dali of Yunnan, China. The local people are masters of making various local products with plum in different ways. In this research, we conducted field investigations in Eryuan County using ethnobotanical methods from August 2019 to July 2021, focusing on the Prunus mume (for its edible fruits). A total of 76 key informants participated in our semi-structured interviews. The survey recorded 37 species (and varieties) belonging to 11 families related to the Bai people's Meizi-consuming culture. Among them, there are 14 taxa of plum resources, including one original species and 13 varieties. These 37 species are either used as substitutes for plum due to their similar taste or as seasonings to improve the sour taste of plum. The higher Cultural Food Significance Index value implies that Prunus mume, Chaenomeles speciosa, Phyllanthus emblica, Prunus salicina, and Chaenomeles cathayensis have high acceptance and use value in the Bai communities. Among the various local products traditionally made by the Bai people, carved plums, preserved plums, perilla-wrapped plums, and stewed plums are the most famous and popular categories in the traditional markets. Currently, the plum business based on the traditional Meizi-consuming culture of the Bai people is already one of Eryuan's economic pillars. This study showed that plums play an important role in expressing the local cultural diversity, and they also help the local people by improving their livelihood through their edible value. In turn, for the sustainable use of plum resources, the Bai people positively manage local forests through a series of measures to protect the diversity of plum resources and related plant communities.

Abstract:Prunus is an economically important fruit tree genus and includes many stone fruit trees such as peach (Prunus persica Batsch), apricot, Japanese apricot (Prunus mume), sweet cherry (Prunus avium L.), European plum (Prunus domestica L.), Japanese plum (Prunus salicina Lindl.) and almond [Prunus dulcis (Mill.) D. A. Webb.]. The characterisation of genes associated with agriculturally important traits such as fruit ripening, dormancy, self-incompatibility, fruit quality and various other developmental processes is important to improve Prunus breeding programmes. Based on high- density molecular genetic maps, many genes are located on the genome, and subsequently, most of them were fine-mapped and further identified by positional cloning. Currently, with the availability of the P. mume genome sequence, the identification of new genes is significantly accelerated.

Abstract:Backgrounds: Prunus mume in the Rosaceae and commonly referred to as mei or Chinese plum is widely used as a traditional ornamental flowering plant and fruit tree in China. Although some population and genetic analyses have been conducted for this species, no extensive comparisons of genetic variation from plastomes have yet been investigated. Methods: We de novo assembled a total of 322 complete P. mume plastomes in this study and did a series of comparative analyses to better resolve pan-plastomic patterns of P. mume . To determine the phylogeny and domestication history of this species, we reconstructed the phylogenetic tree of Prunus genus, and resolved the population structure of P. mume . We also examined the nucleotide variation of P. mume to find potential DNA barcodes. Results: The assembled plastomes exhibited a typical quadripartite structure and ranged from 157,871 bp to 158,213 bp in total size with a GC content ranging from 36.73 to 36.75%. A total of 112 unique genes were identified. Single nucleotide variants (SNVs) were the most common variants found among the plastomes, followed by nucleotide insertions/deletions (InDels), and block substitutions with the intergenic spacer (IGS) regions containing the greatest number of variants. From the pan-plastome data six well-supported genetic clusters were resolved using multiple different population structure analyses. The different cultivars were unevenly distributed among multiple clades. We also reconstructed a phylogeny for multiple species of Prunus to better understand genus level diversity and history from which a complex introgressive relationship between mei and other apricots/plums was resolved. Conclusion: This study constructed the pan-plastome of P. mume , which indicated the domestication of P. mume involved multiple genetic origins and possible matrilineal introgression from other species. The phylogenetic analysis in Prunus and the population structure of P. mume provide an important maternal history for Prunus and the groundwork for future studies on intergenomic sequence transfers, cytonuclear incompatibility, and conservation genetics.

Abstract:The ethanolic extract of Prunus mume (EPM) is a novel polyphenol preparation derived from branches (with leaves) of Prunus mume, which could be used as a functional ingredient for antioxidant and antiobesity therapy. The purpose of this study was to evaluate the safety of EPM. An EPM was prepared and evaluated for oral acute and subacute toxicity in Sprague-Dawley rats, while its mutagenic potential was assessed by a reverse mutation test using Salmonella typhimurium, by a bone marrow cell micronucleus test using ICR mice, and by a sperm abnormality test using ICR mice. The results showed no acute lethal effects at the maximal tested EPM dose of 20 g/kg bw in either rats or mice, suggesting that EPM can be regarded as virtually nontoxic. Administration at levels of 0.84, 1.67, and 3.33 g/kg bw to rats for 30 d did not induce any significant hematological, clinical, chemical, or histopathological changes. No mutagenicity evidence was detected in any of the 3 mutagenic tests. The level of "no observed adverse effect" (NOAEL) for EPM was above 3.33 g/kg bw for the subacute toxicity study.

Abstract:Mei (Prunus mume) is a traditional tree of Rosaceae rich in flavonoids that are important for flower color, medicinal use, food, and skincare applications in East Asia. However, a systematic evaluation of flavonoid resources in mei germplasms is lacking. Here, 81 flavonoid metabolites from 56 representative mei cultivars were identified and compared with those of three closely related species for the first time. It was found that flavonoids were more varied and abundant in mei germplasms compared to its congeneric species. Additionally, the flavonoids were diverse among the nine mei cultiver groups and its three relatives. It was further suggested that four flavonols could be used as biomakers to distinguish the Apricot Mei cultivars from other cultivar groups, providing new insights for systematic classification based on metabolites. On the other side, flavonoids of mei germplasms also displayed distinct flower color characteristics, and cyanidin branch derivatives affected the formation of different flower colors. Furthermore, flavonoids also exhibited some specific distributions among mei germplasms. Cultivars such as WYY, BXZS, DBLE, ELE, and JNTG, which were rich in functional flavonoids, may serve as valuable resources for exploiting the flavonoids of mei. These fundamental data and results were supposed to laid a solid foundation for the further application of mei flavonoid resources in the fields of flower color, medicinal use, food and cosmetic.
Anticancer properties of Prunus mume extracts (Chinese plum, Japanese apricot)

Abstract:Ethnopharmacological relevance: Extracts of the fruit of Prunus mume (Rosaceae) have been used for a long time in Eastern Asia, in many culinary and medicinal preparations. The plant originates from the south of mainland China (named méi) and was introduced later in Japan (ume), Korea (maesil) and Vietnam (mai or mo). Extracts of the fruits (Chinese plum or Japanese apricot, 'Nanko' mume cultivar of Prunus mume Sieb. et Zucc.) are used in traditional Chinese and Japanese medicine, and various Korean medical preparations, for more than 2000 years. The medicinal use of the flesh of the fruits is cited in ancient Japanese monographies (such as Shokokukodenhiho published in 1817). Aim of the study: To analyze the anticancer activities of P. mume extracts and their potential use to prevent or treat cancers. The use of P. mume extracts to alleviate the side effects of chemotherapy, notably drug-induced gastro-intestinal toxicities, is also reviewed. Methods: Extensive database retrieval, such as SciFinder and PubMed, was performed by using keywords such as "Prunus mume", "Chinese plum", "Japanese apricot", and "cancer". In addition, relevant textbooks, patents, reviews, and digital documents (in English) were consulted to collate all available scientific literature and to provide a complete science-based survey of the topic. Results: P. mume extracts display hepatoprotective, anti-inflammatory, antioxidative and antibacterial effects, as well as anticancer properties. A survey of the antitumor activities of MK615 and other P. mume extracts is provided here, with information about the natural products found in the extracts (such as ursolic acid and oleanic acid) and the mechanisms of action of these extracts. MK615 inhibits proliferation and induces apoptotic death of different types of cancer cells from both solid and hematological tumors. Conclusion: The pool of in vitro data and signs of anticancer activities in mice models and in Human, although very limited, support the use of this extract to treat cancer, notably gastro-intestinal tumors. However, more robust evidence of anticancer activity in Human are awaited. Beyond cancer treatment, the use of P. mume extracts to prevent or to treat mucositis and other gastro-intestinal damages induced by anticancer drugs is underlined. The woody plant Prunus mume, a member of the Rosaceae family, has a long plantation history in China, and has widely been planted in Asia due to its high ornamental value (colorful corollas, pleasant fragrance, weeping trait) and the culinary, nutritional and medicinal potential of the fruits from the specie Prunus mume Sieb. et Zucc (Mei). Over the past 20 years the therapeutic potential of the extract of Japanese apricot "Ume" has been regularly reported. Anti-bacterial, anti-oxidative, anti-inflammatory and anti-cancer properties have been described. A complete analysis of the published scientific literature on Ume and cancer is presented here.

The isolated phenolic compounds also exhibited inhibition activity against bacteria significantly, but showed weak or no inhibition activity against yeasts and mold. The results exhibited that the antimicrobial activity of P. mume seeds may be partly due to the phenolic compounds.

Abstract:Japanese apricot (Prunus mume Sieb. et Zucc) is an important economic fruit crop in China and Japan, with more than 200 cultivars in China. It is one of the most valuable materials used in the food and winemaking industries and is believed to contain many physiochemicals beneficial for human health. MicroRNAs (miRNAs) are a class of endogenous, small, noncoding RNAs that regulate gene expression by mediating gene silencing at transcriptional and post-transcriptional levels in higher plants. High-throughput sequencing is used to identify and quantitatively profile small RNAs in Japanese apricot. Previous studies have shown comparative identification of miRNAs between perfect and imperfect Japanese apricot flowers and some small RNAs related to the flower opening process.
Comparison of Biological Traits of Flowers and the Rate of Pollen Germination among 47 Prunus mume Cultivars

Abstract:For protecting a normal reference for bloom,guaranteeing high-quality produce to provide a theory of Prunus mume on the production,and lay a foundation for further research pollen germination and cross-fertilize.The authors have investigated biological traits of flowers and pollen germination among 47Prunus mume cultivars,including aroma,the rate of incomplete flower and pollen quantity and measured the rate of pollen germination.Subsequently,the authors have got some results as follows through the correlative statistics analysis:Chinese cultivars’aroma is more fragrant than Japanese cultivars’;the rate of incomplete flower covers 1.17%-59.65%;there was an obvious diversity among the pollen quantity of different cultivars,and the coverage was from 0 to 1265;there was an obvious diversity among the rate of pollen germination of different cultivars,the highest rate was 84.76%and the lowest rate was 0;there was an obvious correlativity between pollen quantity and the rate of pollen germination.Incomplete flowers occupy a high proportion and flowering periods were easily endangered by low temperature,which resulted in Prunus mume yield low and unsteady.This experiment showed that different species of Prunus mume presented a marked diversity among the rate of incomplete flower,pollen quantity and germination.
