Abstract:A new steroidal glycoside (1), (25R)-14α, 17α-hydroxyspirost-5-en-3β-yl 3-O-α-L-rhamnpyranosyl-(1 → 2)-β-D-glucopyranosyl-(1 → 3)-β-D-glucopyranoside, together with three known steroidal glycosides, (25R)-3β-hydroxyspirost-5-en-1β-yl-3-O-α-L-rhamnopyranosyl-(1 → 2)-O-β-D-xylopyranosyl-(1 → 3)-α-L-arabinopyranoside (2), Cixi-ophiopogon B (3) and Cixi-ophiopogon A (4), were obtained from the tuberous roots of Ophiopogon japonicus (Liliaceae). Compound 2 was isolated from the Ophiopogon genus for the first time. Their structures were identified on the basis of extensive mass and nuclear magnetic resonance spectroscopic analysis.

Abstract:Two new cis-eudesmane sesquiterpene glycosides, liriopeoside A (1) and ophiopogonoside A (2), were extracted and purified from tubers of Liriope muscari and Ophiopogon japonicus, respectively, along with three known compounds. Their structures were elucidated as 1beta,6beta-dihydroxy-cis-eudesm-3-ene-6-O-beta-D-glucopyranoside (1) and 1beta,4beta,6beta-trihydroxy-cis-eudesmane-6-O-beta-D-glucopyranoside (2) by spectral data analysis. The structure and the relative configuration of compound 1 were confirmed by X-ray crystallographic analysis. This is the first time that cis-eudesmane-type sesquiterpenes have been reported from the genera Ophiopogon and Liriope.
Determination and fingerprint analysis of steroidal saponins in roots of Liriope muscari (Decne.) L. H. Bailey by ultra high performance liquid chromatography coupled with ion trap time-of-flight mass spectrometry

Abstract:Liriope muscari (Decne.) L. H. Bailey is a well-known traditional Chinese medicine used for treating cough and insomnia. There are few reports on the quality evaluation of this herb partly because the major steroid saponins are not readily identified by UV detectors and are not easily isolated due to the existence of many similar isomers. In this study, a qualitative and quantitative method was developed to analyze the major components in L. muscari (Decne.) L. H. Bailey roots. Sixteen components were deduced and identified primarily by the information obtained from ultra high performance liquid chromatography with ion-trap time-of-flight mass spectrometry. The method demonstrated the desired specificity, linearity, stability, precision, and accuracy for simultaneous determination of 15 constituents (13 steroidal glycosides, 25(R)-ruscogenin, and pentylbenzoate) in 26 samples from different origins. The fingerprint was established, and the evaluation was achieved using similarity analysis and principal component analysis of 15 fingerprint peaks from 26 samples by ultra high performance liquid chromatography. The results from similarity analysis were consistent with those of principal component analysis. All results suggest that the established method could be applied effectively to the determination of multi-ingredients and fingerprint analysis of steroid saponins for quality assessment and control of L. muscari (Decne.) L. H. Bailey.

Abstract:Two new steroidal glycosides ponasteroside C (1) and ponasteroside D (2) were isolated from the aerial parts of Lepidogrammitis drymoglossoides. Their structures were elucidated by various spectroscopic techniques (IR, HRESIMS, 1D and 2D NMR). All compounds were evaluated for their cytotoxicity against HeLa and HCT-8 cell lines, and compounds 1 and 2 showed mild activity against all the test cell lines.

Abstract:Four new steroidal glycosides ( 1–4 ), including two steroidal saponins named lililancifoloside B and C ( 1 – 2 ), one pregnane glycoside named lililancifoloside D ( 3 ), and one C22-steroidal lactone glycoside named lililancifoloside E ( 4 ), together with five known ones ( 5–9 ), were isolated from the bulbs of Lilium lancifolium Thunb By using spectroscopic analysis, including 1D, 2D NMR, and HR-ESI-MS, the structures of 1–4 were elucidated. All isolates were tested for their cytotoxic potential against the MCF-7, MDA-MB-231, HepG2, and A549 cell lines. Compound 6 distinguished out among them, IC 50 values of 3.31, 5.23, 1.78, and 1.49 μM against the four cell lines, respectively. Other compounds such as compound 3 , 5 , and 9 have also shown specific cytotoxic activity. Next, studies showed that compound 6 might cause HepG2 cells to undergo a cell cycle arrest during the G2/M phase and apoptosis.

Abstract:Four new C-21 steroidal glycoside, stemucronatosides D (1), E (2), F (3), and G (4) were isolated from the roots of Stephanotis mucronata. Their structures were determined on the basis of chemical evidence and extensive spectroscopic methods including one-dimensional and two-dimensional NMR. These isolated compounds were assayed for their immunological activities in vitro against concanavalin A (Con A)- and lipopolysaccharide (LPS)-induced proliferation of mice splenocytes. Compounds 1, 2, and 4 showed immunosuppressive activities in a dose-dependent manner, while compound 3 showed immunomodulating activities.

Abstract:Three new phenolic glycosides, carvacrol-2-O-beta-D-apiofuranosyl-(1 -> 6)-beta-D-glucopyranoside (1), 1-methyl-3-isopropylphenol-4-O-beta-D-apiofuranosyl-(1 -> 6)-beta-D-glucopyranoside (2), p-methoxythymol-5-O-beta-D-apiofuranosyl-(1 -> 6)-beta-D-glucopyranoside (3), and a pair of new 8-O-4' neolignan enantiomers (5a/5b), together with 26 known compounds (4, 6-30) were isolated from the roots of Lilium dauricum. The structures of the new compounds were elucidated based on extensive spectroscopic and chemical methods, and the absolute configurations of 5a and 5b were established by electronic circular dichroism analysis. Nine compounds (1, 3, 4, 8, 9, 17, 25, 29, and 30) exhibited potent a-glucosidase inhibitory activity with IC50 values ranging from 73.4 mu M to 988.2 mu M. Besides, compound 19 displayed strong anticomplementary activity (CH50: 71.6 mu M).

Abstract:Four new diastereoisomeric neolignan glycosides ( 1 – 4 ) along with nine known lignan glycosides ( 5 – 13 ) were isolated from the root bark of Lycium chinense Mill. Their structures with absolute configurations were elucidated on the basis of NMR spectroscopy, ECD, Mo 2 (OAc) 4 -induced ECD spectra, enzymatic hydrolysis and acid hydrolysis. The isolated compounds were evaluated for their α -glucosidase inhibitory activity. Compounds 8 and 13 exhibited moderate inhibitory activities against α -glucosidase with IC 50 values of 26.82 ± 2.71 and 43.14 ± 2.81 μg/mL.

Abstract:AIM To investigate the chemical constituents of the bulbs ofLilium lancifolium Thunb.. METHODS Various chromatographic techniques were used to isolate the chemical constituents. MALDI-TOF-MS, HR-SI-MS, IR, ~1HNMR, ~ 13CNMR, DEPT, ~1H- ~1H COSY, HMQC and HMBC were used to determine the structures of the isolated constituents. RESULTS Ophiopogonin D′ and lililancifoloside A were isolated from the bulbs ofLilium lancifolium Thunb., and were identified as diosgenin 3-O-{O-α-L-rhamnopyranosyl-(1→2)-O-[β-D-xylopyranosyl(1→3)]-β-D-glucopyranoside} and diosgenin 3-O-{O-α-L-rhamnopyranosyl-(1→2)-O-[α-L-arabinopyranosyl(1→3)]-β-D-glucopyranoside}, respectively. CONCLUSION Ophiopogonin D′ was isolated from the bulbs ofLilium lancifolium Thunb. for the first time. Lililancifoloside A is a new steroidal saponin.

Abstract:A new sesquiterpene lactone (1) and a new aromatic glycoside (2), together with three known compounds (3–5) were isolated from the stem bark of Illicium difengpi K. I. B et K. I. M. Their structures were determined by spectroscopic methods, including 1D and 2D NMR, HRESIMS, and chemical methods. The absolute configuration of the secondary alcohol in 1 was confirmed by Mosher's method. Compound 2 exhibited significant anti-inflammatory activity with IC50 value of 6.72μmol/L.

Their structures were elucidated by spectroscopic analysis. Compounds 2-8 and 11-13 were first isolated from Rutaceae and the others were isolated for the first time from Z. armatum.

Abstract:Liriogerphines A–D (1–4, respectively), an unprecedented class of hybrids of germacranolide-type sesquiterpenoids and aporphine-type alkaloids, were isolated from the rare medicinal plant Liriodendron chinense. Their structures were elucidated by comprehensive spectroscopic analyses combined with electronic circular dichroism calculations and X-ray crystallographic data. Biosynthetically, an aza-Michael addition reaction is proposed to be involved in the assemblies of this class of hybrids. Compound 4 exhibited cytotoxicity against leukemia cells via inducing apoptosis and inhibiting Bcl-2 expression.

Abstract:The aerial parts of Artemisia sieversiana afforded, in addition to β-sitosterol, stigmasterol and daucosterol, two novel lignans as well as one known and three new guaianolides. The roots of Inula racemosa gave β-sitosterol, daucosterol and isoalantolactone. The structures were determined by a combination of spectral methods (IR, EIMS, 1H and 13C NMR, DEPT, COSY, NOESY and HETCOR). All isolates were subjected to antifungal tests. Isoalantolactone, a major sesquiterpene lactone of I. racemosa, was found to be active against the human pathogenic fungi, Aspergillus flavus, A. niger, Geotrichum candidum, Candida tropicalis and C. albicans at concentrations of 50, 50, 25, 25 and 25 μg\ml, respectively. The taxonomic significance of the characterized constituents is discussed briefly.
