Abstract:Heterocyclic ketene aminals reacted with ethyl 2-(bromomethyl)benzoate in refluxing acetonitrile to afford the C-benzylated products that underwent intramolecular cyclocondensation reaction to produce epsilon-lactam fused heterocyclic compounds. The heterocyclic ring size effect on the reaction was discussed.

Abstract:The regiospecific allylation of benzoyl-substituted heterocyclic ketene aminals 2 is reported. Compound 2 reacted with allyl bromide in refluxing acetonitrile to give, exclusively, C-allylated products 5 in moderate to goad yields, while only N-allylated compounds 7 were obtained in dimethylformamide with the use of sodium hydride. Under neutral conditions, reaction between 2 and allyl bromide proceeded via a six-membered transition state involving the secondary enamine segment (HNC=C) and carbon-bromine bond (C-Br). Without the secondary amino group, C-allylation was inhibited, which was illustrated by the fact that 1,3-dimethyl-2-(aroylmethylene)imidazolidines did not react with allyl bromide. No 3-aza-Cope rearrangement of N-allylated heterocyclic ketene aminals 7 was observed at temperatures up to 140 degrees C. Only with the use of zinc chloride did the rearrangement take place at 140 degrees C to give allyl-shifted products 5. N-Benzylated heterocyclic ketene aminal analogues did not undergo the rearrangement under the same conditions, indicating that the 3-aza-Cope rearrangement proceeds through a charge-accelerated concerted mechanism.

Abstract:A novel protocol for the facile synthesis of chiral 3-substituted 1,3,4,5-tetrahydrobenzo[e] [1,4]diazepin-2-ones via a domino reaction has been developed, which involves a cascade of S(N)2 nucleophilic substitution and copper-catalyzed C-N coupling reactions of a-amino acid amides and orthohalobenzyl halides. This protocol has some merits, such as one-pot process, easy operation, a wide scope of substrates. Furthermore, no heavy racemization occurs during the reaction.

Mechanism and regio- and stereoselectivity in NHC-catalyzed reaction of 2-bromoenals with β-ketoamides

Abstract:We have investigated an N-heterocyclic carbene (NHC)-catalyzed annulation reaction in order to determine the mechanisms and origins of stereoselectivity. DFT results show that the catalytic formation of spiro-glutarimides involves coordination of 2-bromoenal to NHC, followed by a bicarbonate aided proton transfer to form the Breslow intermediate. Subsequent debromination and proton transfer forms the α,β-unsaturated acylazolium intermediate. Then the reaction proceeds via Michael addition, proton transfer and intramolecular cyclization to deliver the cyclic intermediate, which finally releases NHC affording the spiro-glutarimide product. The Michael addition resulting in a new carbon−carbon bond is found to be the stereochemistry-controlling step and generates the SR-configurational product observed in experiment. The important role of noncovalent interactions (such as C−H•••O, C−H•••π, π•••π and lone pair (LP)•••π) in controlling stereoselectivity is elucidated by noncovalent interaction (NCI) analysis. Moreover, analysis of the global reactivity indexes (GRIs) and the computed C−Br bond lengths revealed that NHC plays an important role in activating C−Br bond.

Abstract:A series of β-ketoenamines was synthesized from various phenacyl sulfoxides bearing 1-methyl-1H-tetrazole and oximes in moderate to excellent yields. The proposed mechanism involved the generation of α-sulfines from sulfoxides through thermolytic elimination, regiospecific formal [3 + 2] annulations, and elimination of SO(2). This protocol provides convenient access to a variety of synthetically valuable N-unprotected β-enaminones with absolute Z selectivity.

Abstract:In the presence of triethyl amine, the reaction of 2,4-disubstituted-2,3-dihydro- 1,5-benzothiazepine with chloro and dichloroacetyl chlorides produced not only the expected beta -lactam derivative of the benzothiazepine, but also the ring opening product. Different results were obtained when the substituent at 2-position of the bexuothiazepine varied from methyl to aryl, and the substituent on the chloroacetyl chloride varied from H to Cl, or when carrying out the reaction at different temperatures. The structures of the obtained products and the reaction mechanism are discussed.

Abstract:A novel rhodium-catalyzed highly selective N-2-alkylation of benzotriazoles with diazo compounds/enynones is achieved, providing N-2-alkylated benzotriazoles in good to excellent yields and with excellent N-2 selectivities. Importantly, different to traditional carbene insertion into X - H (X = N, O etc) bonds, DFT calculations disclose that this selective N-2-alkylation probably proceeds through a formal 1,3- rather than 1,2-H shift to give the final products.

Abstract:A Co( III )-catalyzed site-selective C5 and C6 benzannulation of substituted pyridones with 1,6-diynes via dual C–H bond activation has been reported. The scope of the benzannulation reaction was examined with various substituted 2-pyridyl pyridones and 1,6-diynes. The combination of cuprous acetate and silver carbonate plays a crucial role in the success of the reaction. A plausible reaction mechanism was proposed and supported by deuterium labelling studies and radical trapping experiments.

Abstract:The regioselectivity in addition of nucleophiles to the p-benzyne intermediates derived from unsymmetrical aza-substituted enediynes via Bergman cyclization was studied. Computational studies [using UB3LYP/6-31G(d,p) level of theory] suggest that the p-benzyne intermediate retains its similar electrophilic character at the two radical centers even under unsymmetrical electronic perturbation, thus supporting the predicted model of nucleophilic addition to p-benzyne proposed by Perrin and co-workers (Perrin et al. J. Am. Chem. Soc. 2007, 129, 4795-4799) and later by Alabugin and co-workers (Peterson et al. Eur. J. Org. Chem. 2013, 2013, 2505-2527). However, observed experimental results suggest that there was small but definite regioselectivity (∼5-25%), the extent varying with the electronic nature of the substituents. Differential solvated halide ion concentrations around the vicinity of two radical centers arising due to surrounding surface electrostatic potential (computationally calculated) may be one of the possible factors for such selectivity in some of the examined p-benzynes. However, other complicated dynamical issues like the trajectory of the attacking nucleophile to the radical center which can be influenced by electronic and/or steric perturbation of starting enediyne conformation cannot be ruled out. The overall yield of the anionic addition was in the range of 80-99%.

Abstract:Diastereo- and enantioselective N-heterocyclic carbene catalyzed 1-azadiene Diels-Alder reactions of (E)-2-styrylbenzothiazoles with α-chloro aldehydes are reported. This annulation strategy provides an efficient access to medicinally important dihydrobenzothiazolopyridin-1-ones in good to excellent yields (44-97%) with very good to excellent stereoselectivities (up to 9:1 dr, 98% ee) and tolerates quite a range of substituents.

Abstract:A palladium-catalyzed C-H alkenylation of imidazoles has been developed. High C5 selectivity was achieved for C2-unsubstituted and C2-substituted imidazoles using oxygen and copper(ii) acetate, respectively, as oxidants. The obtained products were applied to benzannulation through a sequence involving transposition of N-alkyl groups to give C4-alkenyl imidazoles, alkenylation, thermal 6π-electrocyclization, and oxidation, affording unsymmetrically substituted benzimidazoles.

Abstract:The annulation reactions of N-allylbenzamides with N-sulfonylaminopyridinium salts were developed under metal-free photoinduced mild conditions. Substituent-controlled sulfonaminoarylation and sulfonaminooxylation of benzamides were realized: N-allylbenzamides lead to benzosultams, while N-(2-phenylallyl)benzamides give sulfonamidylated oxazoline derivatives. Control experiments indicated that those reactions undergo a radical pathway with arylsulfonamidyl radicals as the intermediates. The aryl C-H bond functionalization in arylsulfonamidyl was involved for the first time to give benzosultams.

Abstract:Benzyl bromides and related molecules are among the most common substrates in organic synthesis. They are typically used as electrophiles in nucleophilic substitution reactions. These molecules can also be activated via single-electron-transfer (SET) process for radical reactions. Representative recent progress includes α-carbon benzylation of ketones and aldehydes via photoredox catalysis. Here we disclose the generation of (nitro)benzyl radicals via N-heterocyclic carbene (NHC) catalysis under reductive conditions. The radical intermediates generated via NHC catalysis undergo formal 1,2-addition with ketones to eventually afford tertiary alcohol products. The overall process constitutes a formal polarity-inversion of benzyl bromide, allowing a direct coupling of two initially electrophilic carbons. Our study provides a new carbene-catalysed reaction mode that should enable unconventional transformation of (nitro)benzyl bromides under mild organocatalytic conditions.
