Abstract:Heterocyclic ketene aminals 5 or 3H-1,5-benzodiazepines 4 were obtained from the reaction of benzoyl substituted ketene dithioacetals 1 with various diamines 2. The mechansim of formation of these two different products are discussed.

Abstract:An unexpected process from tetrahydrodiazirino[3,1-a]isoquinolines and diversified sulfonyl chlorides RSO2Cl (R=aryl, alkyl) to deliver highly yields 4,5-dihydro-3H-benzo[d][1,2]diazepine skeleton compounds was disclosed via N-insertion ring-opening reaction. The structural framework of 2,3-benzodiazepines is unambiguously confirmed by single-crystal X-ray crystallographic analysis. The present protocol features mild reaction conditions, high yields and feasibility of large-scale synthesis and catalyst-free access to benzo[2,3]diazepines skeleton. In contrast, azomethine imines were produced by employing tetrahydrodiazirino[3,1-a]isoquinoline reacts with aroyl chlorides under the same conditions.

Abstract:A novel protocol for the facile synthesis of chiral 3-substituted 1,3,4,5-tetrahydrobenzo[e] [1,4]diazepin-2-ones via a domino reaction has been developed, which involves a cascade of S(N)2 nucleophilic substitution and copper-catalyzed C-N coupling reactions of a-amino acid amides and orthohalobenzyl halides. This protocol has some merits, such as one-pot process, easy operation, a wide scope of substrates. Furthermore, no heavy racemization occurs during the reaction.

Abstract:Heterocyclic ketene aminals reacted with ethyl 2-(bromomethyl)benzoate in refluxing acetonitrile to afford the C-benzylated products that underwent intramolecular cyclocondensation reaction to produce epsilon-lactam fused heterocyclic compounds. Abstract:The first isolation of benzimidazoline ring substituted ketene aminals 7a-c and/or their amidine tautomers 7'b-d, which were synthesized by the reactions: (1) benzoyl substituted ketene mercaptals 3 with N-methyl-o-phenylenediamine 4, and (2) 1,2-dimethylbenzimidazole 5 with ethyl benzoates 6, were disclosed. The tautomeric equilibrium between 7 and 7', along with benzothiazoline and benzoxazoline ring substituted ketene N,S-acetals 8 and 8' and ketene N,O-acetals 9 and 9', were discussed.

Abstract:AbstractReaction of 2, 3‐dihydro‐1H‐1. 5‐benzodiazepines with dichlorocarbene generated in situ using benzyltriethylammonium chloride (TEBA) as a phase transfer catalyst in chloroform‐aqueous sodium hydroxide mixture gave mainly 1,2‐cycloadducts, cis and trans‐1a, 3‐disubstituted‐1, 1‐dichloro‐1a, 2,3,4‐tetrahydro‐1H‐azirino[1,2‐a][1,5]benzodiazepines (2.3), and formylated 1,2‐cycloadducts, trans‐1a,3‐disubstituted‐1, 1‐dichloro‐4‐formyl‐1a, 2, 3, 4–1 H‐azirino[1, 2‐a][1, 5]benzodiacepines (4). The stereo‐structures of cycloadducts and the mechanism are also discussed.

Abstract:A selective copper(I)-catalyzed benzylic C(sp3)–H geminal difunctionalization reaction of a benzylic-type sp3 carbon was developed. This novel strategy allowed simultaneous introduction of amide and hydroxyl group in a highly selective and efficient way via successive oxidative intramolecular amidation and hydroxylation. This method was also applied to the synthesis of 3-hydroxy-2,3-dihydro-1H-pyrrolo[3,4-b]quinoxalinones from readily available 3-methyl-N-substituted quinoxaline-2-carboxamides in moderate to good yields. The five-membered cyclic hemiaminal moiety of the pyrrolo[3,4-b]quinoxalinones can serve as an intermediates for subsequent transformations into other useful functional groups.

Abstract:2-Trifluoromethyl and 2-difluoromethyl substituted benzimidazole, benzoxazole and benzothiazole derivatives were efficiently prepared through a one-pot reaction of trifluoroacetic acid and difluoroacetic acid, respectively, with commercially available o-phenylenediamines, 2-aminophenols, and 2-aminobenzenethiols in good to excellent yields. Subsequential bromination of 2-difluoromethyl groups by photolysis with NBS led to the formation of bromodifluoromethyl benzo-1,3-diazoles which may be utilized to prepare the new generation of gem-difluoromethylene linked identical or non-identical twin molecules for drug synthesis.

Abstract:Benzodiazepine and benzothiazepine derivatives have been well known as therapeutically important compounds. Four new tricyclic heterocyclic compounds, 3a,4,5,11-tetrahydro-3H-1,2,4-triazolo[4,3-d] [1, 5]benzothiazepines (3), 3a,4,5,11-tetrahydro-3H,6H-1,2,4-triazolo[4,3-d] [1,5]benzodiazepine (4), 3a, 4,5,11-tetrahydro-1,2,4-oxadiazolo[4,5-d] [1,5]benzothiazepines (5, 6) and 3a,4,5, 11-tetrahydro-6H-1,2,4-oxadiazolo[4, 5-d] [1,5] benzodiazepines (7,8), have been synthesized by 1,3-dipolar cycloaddition reactions of 2, 3-dihydro-1,5-benzothiazepines and 2, 3-dihydro-1H-1, 5-benzodiazepine with benzonitrile N-phenylimine and benzonitrile oxides, respectively. Abstract:Two new series of tricyclic heterocycles, namely 5,6-dihydro-4H-benzo[b][1,2,4]triazolo[1,5-d][1,4]diazepinium salts 10 and the related neutral, free bases 13 were synthesized from 4-acetoxy-1-acetyl-4-phenylazo-1,2,3,4-tetrahydroquinolines 8 and nitriles 9 in the presence of aluminium chloride by the [3+ + 2]-cycloaddition reaction of the in situ generated azocarbenium intermediates 14 followed by a ring-expansion rearrangement. In the rearrangement reaction, the phenyl substituent in the initially formed spiro-triazolium adducts 16 underwent a [1,2]-migration from C(3) to the electron-deficient N(2). This led to the ring expansion from 6-membered piperidine to 7-membered diazepine furnishing the tricyclic 1,2,4-triazole-fused 1,4-benzodiazepines.

Abstract:The synthesis of a series of unprecedented tricyclic heterocycles, 1,2,4-triazolo[3,2-d][1,5]benzothiazepine 6-oxides (5a-i), was accomplished via cycloaddition of the allene-like cations 3, derived from thiochroman-4-one arylhydrazones, to the triple bond of nitriles and ensuing rings expansion annulation. The structure of one product (5h) has been unambiguously determined by X-ray diffraction analysis.
