The self-assembled films of a-amylase on the negatively charged PET (PET-CO2-) is fabricated and the AFM results are described in this paper. The surface of PET-CO2- was covered by high-density layer of alpha-amylase, whose thickness was 5 - 10 nm. The molecules of alpha-amylase aggregated in the layer which was obtained from the pH 6.0 solution, and the shape of molecules became sharper, which resulted in lager RMS. On the lower-charged substrate (PET-CO2-), the aggregating molecules could be found. With raising the charge density of substrate the shape of molecules became sharper.

Abstract:The PET-CO-2 film was prepared and the lipase was assembled on the surface of the PET-CO-2 substrate. The structure of the surface and activity of lipase/PET monolayer were studied by FTIR-ATR and AFM and other methods. As shown in the results, AFM images of lipase/PET monolayer indicated that the surface fluctuation is in the range of nanometer scale and the depth of monolayer is less than 15 nanometers. The performance activity of lipase/PET monolayer self-assembly film is above 2.0.

Abstract:The self-assembly adsorption of function protein on crystal surfaces, as a common phenomenon, broadly takes place in many applications of biosensors, biocapsules and bioMEMS/bioNEMS. To systematically investigate the different adsorption characteristic of the same function protein on two different crystal surfaces under the identical environment, a hybrid surface composing silica and discontinuous Gold Nano Film (GNF) was fabricated by Physical Vapor Deposition (PVD) and ultrasonic cleaning method, where the dynamic process of the self-assembly adsorption of Bovine Serum Albumin (BSA) was in situ observed by Atomic Force Microscope (AFM). The variations on the junction area of the two different surfaces were studied in the aqueous solution before and after injecting BSA with 0.05 mg/ml concentration. It was found that silica, compared with same hydrophilic GNF, took on a fairly weak adsorption force. The results indicated that the adsorption strength of BSA on the hydrophilic crystal surface was determined not only by hydrophilic property, but also other interaction forces, like Van der Waals and so on. Moreover, observed under the contact mode of AFM, BSA adsorbed on GNF had great tendency to forming a ridge-like topography. These results may be helpful in the application of immunosensors and other areas.

Abstract:Domain structures were studied with atomic force microscopy (AFM) in binary phase-separated monolayer films composed of 5, 10, 15-triphenyl-20-(4-dl-α-phenylalanylamindo) phenyl porphyrin (TPPP) and one of a series of fatty acids which are arachidic acid (AA), palmitic acid (PA), and lauric acid (LA). The liquid-condensed (LC) domain structures of AA and PA were observed in their corresponding mixed monolayers. However, instead of the fatty acid domain, a liquid-expanded (LE) domain structure of TPPP appears in the mixed monolayer of LA/TPPP.

Abstract:Elongational effects on polymer chains in the polyamic acid alkylanine salt monolayer on the air-water interface are studied. Polymer chains elongated by a flow transferred onto graphite are observed by atomic force microscopy. We found that when the polymers are compressed to a condensed state by the elongational flow their chains are aligned in the flow direction.
Direct AFM force mapping of surface nanoscale organization and protein adsorption on an aluminum substrate

Abstract:We investigate the nanoscale organization of a superficially hydroxylated Al substrate and its effect on subsequent protein adsorption using atomic force microscopy (AFM). For this purpose we used a mode which allows a direct mapping of a variety of surface properties (adhesion, elasticity, dissipation, etc.) to be probed simultaneously with topographical images. The hydroxylation treatment leads to a drastic modification of the surface morphology, owing to the formation of AlOOH compounds. In air, AFM images revealed the formation of regular nanorod-like structures randomly distributed, inducing the appearance of nanoporous domains on the surface. In buffer solution, prior to the adsorption of proteins, the surface nanoscale organization is preserved, mainly due to the chemical stability of AlOOH compounds under these conditions. The adsorption of proteins on the obtained nanostructured surface was performed using either a globular (β-lactoglobulin) or a fibrillar (collagen) protein and by modulating the adsorbed amount through the incubation time or the concentration of proteins in solution. At low amounts, collagen adsorbs on the whole surface without preferential localization. The surface topography remains similar to the bare surface, while significant changes were evidenced on adhesion and elasticity maps. This is due to the fact that the surface became adhesive and less stiff, owing to the presence of a soft and hydrated protein layer. By contrast, β-lactoglobulin tends to diffuse into the nanoporous domains, leading to their filling up, and the surface is blurred with a thick and dense protein layer upon increasing the amount of adsorbed molecules. Our findings demonstrate the interest in using AFM for surface mapping to investigate the mechanism of protein adsorption at the nanoscale on materials with high surface roughness.

Abstract:We report here the first AFM studies of phase-separated monolayer Langmuir-Blodgett films composed of a mixture of L-alpha-dipalmitoylphosphocholine (DPPC) and peptide bond bridged dl-alpha-phenylalanine-tetraphenylporphyrin (PTPP) under different surface pressures. The shapes of DPPC domains as well as microstructures within domains were also directly observed by AFM.
A Novel Atomic Force Microscope Operating in Liquid for in Situ Investigation of Electrochemical Preparation of Porous Alumina.

Abstract:A novel atomic force microscope (AFM) operating in liquid is described in this article. The specially designed AFM probe involves a tip attached to a cantilever, a tip holder, and a circular Plexiglas window. When the probe dives into the fluid, a circular meniscus is established around the Plexiglas window, preventing the tip from being affected or destroyed by surface tension of the liquid. In this setup, the whole scanning probe and the sample can completely dive into fluid. Meanwhile, the probe tip scans over the sample surface when the instrument works. These advantages enable the instrument to scan comparatively large or heavy samples with a high speed. The highest scan rate is about 30 lines/s or 14 s for a 400 x 400-pixel, 3 x 3 Pin image. Using the new AFM, we carry out in-situ investigation of the formation processes of porous alumina during electrochemical anodic oxidation. A lead ring and an aluminum foil serve as cathode and anode, respectively. They are entirely immersed in the bath electrolyte, which is oxalic acid solution. During anodic oxidation, the AFM images of the sample surface are successively acquired without elevating the sample out of the solution. Experiments reveal that electrochemical reactions take place soon after the power supply is switched on, and with the progression of anodization, nanostructures of porous alumina gradually occur on the aluminum substrate, finally yielding ordered arrays of nanopores. As a typical example of applications, the results of this work show that the new AFM is an ideal and powerful tool for in-situ observation and study of materials or samples in aqueous solutions. Abstract:In this study, light/pH responsive methoxy poly (ethylene glycol)-(5-propargylether-2-nitrobenzyl bromoisobutyrate)-poly methylacrylic acid-b-polystyrene (mPEG-ONB-PMAA-b-PS) polymers were synthesized, and successfully utilized to fabricate micelles and immobilize α-amylase. The critical micelle concentrations (CMC) of the polymers were measured with Pyrene Fluorescent Probe Technique. The morphology and diameter of micelles were characterized by transmission electron microscopy (TEM) and dynamic light scattering (DLS). In addition, the effects of pH, temperature and light-responsive on the catalytic activity were investigated. The optimized fabrication conditions of α-amylase-loaded micelles which α-amylase gave the higher activity were as follows: Immobilization time, 60min; Immobilization temperature, 50°C; enzyme concentration, 10UmL−1; PBS buffer, pH=5.4. α-Amylase immobilized in these micelles was much more stable than that free α-amylase.

Abstract:Array like domain structures were prepared in the mixed monolayer films composed of fatty acid and 5,10,15-triphenyl-20-(4-dl-alpha-phenylalanylamindo)phenyl porphyrin (TPPP) and investigated by atomic force microscopy (AFM). The mixtures exhibit the fatty acid domains in the liquid-condensed (LS) state on the condition of long alkyl chain. By shortening the length of the hydrocarbon chain, TPPP domains in the liquid-expanded (LE) state occur in the mixed monolayer films.

Abstract:The Langmuir-Blodgett monolayers of 10,12-tricosadiynoic acid molecules were deposited onto the basal plane of highly oriented pyrolytic graphite (HOPG) by Schaefer's method and then observed with the scanning tunnelling microscope (STM). With a view to achieving a parallel molecular arrangement on the graphite surface, the deposition was deliberately conducted at a relatively low surface pressure. As exhibited by the STM images, by this approach the 10,12-tricosadiynoic acid molecules could constitute an ordered structure with their molecular chains lying parallel to the substrate. The model of molecular dimer is put forward for the interpretation of the observed phenomena.
Bottom-Up Approach to Explore Alpha-Amylase Assisted Membrane Remodelling

Abstract:Soluble alpha-amylases play an important role in the catabolism of polysaccharides. In this work, we show that the enzyme can interact with the lipid membrane and further alter its mechanical properties. Vesicle fluctuation spectroscopy is used for quantitative measurement of the membrane bending rigidity of phosphatidylcholines lipid vesicles from the shape fluctuation based on the whole contour of Giant Unilamellar Vesicles (GUVs). The bending rigidity of the lipid vesicles of 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine in water increases significantly with the presence of 0.14 micromolar alpha-amylase in the exterior solution. However, as the concentration increases above 1 micromolar, the bending rigidity decreases but remains higher than estimated without the protein. Contact between the alpha-amylase in the outer solution and the outer leaflet leads to spontaneous membrane curvature and the corresponding morphological changes of the GUVs. The presence of outbuds directly demonstrates that AA has a preferable interaction with the membrane, giving a positive spontaneous curvature of $C_0 \leq 0.05 \ \mu \rm{m}^{-1}$ at $18 \ \mu$M $(\approx$ 1 mg/ml) of AA concentration. Above 1 mg/ml of AA concentration the shape of GUVs collapse completely suggesting a highly convoluted state.

Abstract:The miscibility behavior and surface morphology of a mixed monolayer, stearic acid (SA) with 5,10,15-triphenyl-20-(4-DL-phenylalanylamindo)phenyl porphyrin (TPPP), were studied by analyzing the surface pressure vs. molecular area isotherms and atomic force microscopy (AFM) images of the transferred monolayer films. The mixed monolayer is a two-phase system with a liquid-condensed phase consisting of SA and a liquid-expanded phase consisting of TPPP with a trace of SA. The different domain structures of SA sensitive to surface pressure, compression rate, annealing, etc. were also studied with AFM in detail.

Abstract:Summary The structure of a self-organized polymer film prepared by slow evaporation of organic solvents is studied by atomic force microscopy, and compared with that of polymer film prepared by natural evaporation of organic solvents. AFM studies clearly indicates that there exists a self-organizing process of alkyl chains resulting in the partial interdigitated layer structure under the condition of slow evaporation of organic solvents.

Abstract:The Lipase/PET-NH 3 +monolayer self-assembly functional films were fabricated by the means of molecular depositing (MD). The content of lipase at the PET-NH 3 +substrate was quantitatively analyzed by XPS. The topographies of monolayer at the surface were studied by AFM. As showed in results, the amount of lipase depositing on the PET-NH 3 +substrate increases with the increase of ionic strength, time and the concentrations of lipase. In different conditions, the performance activity of lipase/PET-NH 3 + is different.

Abstract:The magnetic heads overcoated by tetrahedral amorphous carbon (ta-C) films have been immersed in the 1H, 1H, 2H, 2H-perfluorodecyltrichlorosilane (FDTS) solvent at different predetermined dipping time. The growth of FDTS self-assembled monolayer (SAM) on the ta-C surface has been confirmed by time-of-flight secondary ion mass spectrometry. Four distinct stages were observed in the growing process: (1) low coverage (nucleation), (2) growing up (3) coalescence and (4) complete monolayer, similar to the theoretical models of epitaxial film growth at low temperatures. Contact angle measurement using de-ionized water is carried out to study the time dependence of contact angle during the growth process of FDTS monolayer. The surface morphology, profiles and roughness (rms) of FDTS monolayer were determined by atomic force microscopy (AFM). Furthermore, the variation of monolayer thickness and distribution was revealed by transmission electron microscopy, which is in agreement with the AFM and contact angle results.

Abstract:Atomic Force Microscopy (AFM) has been used to show the formation of solid-phase domains from fluid-phase domains on compression of DiPalmitoyl-PhosphatidylCholine (DPPC) monolayer Langmuir-Blodgett (LB) films. The chiral structures on the solid substrates were observed for the first time. By applying the friction force technique, we were able to distinguish the different regions of LB films according to their elastic properties. The influence of rates of compression on the domain shape as well as the microstructure within the domain were also studied.

Abstract:Phase separation was observed by atomic force microscopy in the mixed monolayer films composed of arachidic acid (AA) and 5,10,15-triphenyl-20-( . 4-dl-alpha-phenylalanyiamindo)phenyl porphyrin (TPPP). Under different preparation conditions, the mixtures exhibit only AA domains or a coexistence structure of AA domains, TPPP domains and downy structures constructed by AA and TPPP together. Different UV-Vis spectra were obtained in the two kinds of phase-separated structures.
