Moreover. revised structures are proposed for decipinone B (1) and C (2) on the basis of X-ray-diffraction analyses. The structures of the compounds were elucidated by means of different spectroscopic methods, including 1D- and 2D-NMR studies.

Abstract:Ingenol esters, one type of diterpenoids isolated from genus Euphorbia, were reported to possess cytotoxic activities. In our previous study, a number of ingenol esters were isolated from Euphorbia esula L. (Ru-Jiang-Da-Ji in Chinese). And as a part of the ongoing program, a study of fragmentation pathway of ingenol esters was carried out using mass spectrometry. The fragmentation behaviors of 17 standard compounds were investigated with ion trap (IT-MSn) mass spectrometry and hybrid quadrupole-time of flight-mass spectrometer (Q-TOF-MS/MS) in positive ion mode. Electrospray ionization (ESI) was used as ion source in analysis. According to the IT-MSn spectra, the elimination order of the acyloxy was R1OH>R2H>R3H>R4H, which was hard to determine in Q-TOF-MS/MS spectra. The Q-TOF-MS preferred to eliminate all the acyloxys along with some hydroxyls and produce three characteristic ions (x0, y0, z0), which indicate the numbers of the oxygenated groups. The Q-TOF-MS also provide high resolution mass spectra of the parent and product ions, which allowed the determination of the chemical formulas and characterization of the ion structures. These two instruments gave complementary information, which were very useful for structural characterization of the studied compounds.

Abstract:From the ethanolic extract of the roots of Euphorbia ebracteolata Hayata four compounds were isolated. They are 24-methylenecycloartanone, tirucallol, procesterol and a new isopimarane diterpene, namely yuexiandajisu C. The structure of yuexiandajisu C was elucidated by spectral analysis. The bioassay in vitro showed yuexiandajisu C exhibited immunomodulatory activity.

Abstract:Thirteen new Euphorbia diterpenoids, euphylonanes A-M (1-13), and eight known ones were isolated from the whole plants of Euphorbia hylonoma. Compounds 1 and 2 are two rearranged ingenanes bearing a rare 6/6/7/3-fused ring system. Compound 3 represents the first example of a 9,10-epoxy tigliane, while 4-21 are typical ingenanes varying with substituents. Structures were elucidated using a combination of spectroscopic, computational, and chemical methods. Most ingenanes exerted a significant antiadipogenic effect in 3T3-L1 adipocytes, among which 4 was the most active with an EC50 value of 0.60 ± 0.27 μM. Mechanistic study revealed that 4 inhibited the adipogenesis and lipogenesis in adipocytes via activation of the AMPK signaling pathway.

Abstract:Two new triterpenoids, namely 24-methylene-5,24-dien-19(10→9)- abeo -8 α ,9 β ,10 α -eupha-3 β -ol ( 1 ) and 24-methyl-5,23-dien-19(10→9)- abeo -8 α ,9 β ,10 α -eupha-3 β -ol ( 2 ) were isolated from the stems of Euphorbia royleana , together with three known analogs. The structures of the new compounds were elucidated by extensive 1 H NMR, 13 C NMR, HSQC, HMBC, 1 H- 1 H COSY, ROESY and HR-MS spectroscopic analyses.

Abstract:A phytochemical investigation on the constituents from Euphorbia humifusa Willd. has resulted in the isolation of three new alpha-pyrrolidinonoidal compounds, 5beta-methoxy-4beta-hydroxy-3-methylene-alpha-pyrrolidinone (1), 5beta-methoxy-4alpha-hydroxy-3-methylene-alpha-pyrrolidinone (2), and 5beta-butoxy-4alpha-hydroxy-3-methylene-alpha-pyrrolidinone (3), and three new glycosides including an indole glycoside, 3-(2-hydroxyethyl)-5-(1-O-beta-glucopyranosyloxy)-indole (4), an ionone glycoside, 3-oxo-7,8-dihydro-alpha-ionone-11-O-beta-glucoside (5), and a hemiterpene glycoside, 1-(4-hydroxy-2-methyl-2-buten-1-yl)-6- (3,4,5-trihydroxybenzoyl)-beta-d-glucose (6), along with 10 known compounds. Their structures were elucidated by analysis of 1D and 2D NMR spectral data. The structure of 1 was further confirmed by a single-crystal X-ray diffraction analysis.

Abstract:Three novel rearranged tigliane-type diterpenoids, euphebrananes A-C (1–3), and one precursor tigliane-type diterpenoid, euphebrananes D (4), were isolated from the roots of Euphorbia ebracteolata. The structures of isolates were identified by comprehensive spectroscopy, HRESIMS, quantum chemical calculations (ECD and NMR), and X-ray crystallography. Euphebrananes A-C represented a tigliane-type diterpenoid, and a plausible biosynthetic pathway of 1–4 was proposed. In vitro, anti-inflammatory and estrogen-like activity screening showed that 1–4 exhibited weak related biological activities.

Abstract:In the present study,the constituents in the petroleum ether fraction from the extract of Nerviliae fordii(Hance) Schltr were separated by using various kinds of chromatography techniques and recrystallization,and their structures were identifed on the basis of spectral and physicochemical analysis.Ten compounds were isolated from this fraction,and identified as β-sitosterol(1),stigmaterol(2),(9R,10R,7E)-6,9,10-trihydroxyoctadec-7-enoic acid(3),(20S,22E,24R)-ergosta-7,22-dien3β,5α,6β-triol(4),stigmast-22-en-3β,6β,9β-triol(5),ergosta-7,22-dien-3β,5α,6α-triol(6),3β-hydroxystigmasr-5,22-dien-7-one(7),3β-hydroxystigmasr-5-en-7-one(8),aurantiamide acetate(9),and aurantiamide benzoate(10).Among them,compounds 3,5 ～ 8 and 10 were obtained from this plant for the first time.

Abstract:Compounds 1-3, the rare examples of 9,11-seco euphane or lanostane triterpenoids featuring an enol-hemiacetal functionality, were isolated from Euphorbia stracheyi. Their structures were elucidated by a combination of spectroscopic, computational, chemical, and single-crystal X-ray diffraction means, which enables the structure of previously published euphorol J to be revised as 1. 1-3 showed significant cytotoxicities on the breast cancer cell line MDA-MB-468 with IC50 values in the range of 2.9-3.9 μM.
