Abstract:A new jatrophane diterpenoid ester (2S, 3S, 4R, 5R, 7S, 8R, 13R, 15R) - 3, 5, 7, 8, 15-pentaacetoxy-9, 14-dioxojatropha-6(17), 11E-diene was isolated from the whole plant of Euphorbia turczaninowii Kar. & Kin. Its structure was characterized by spectral analysis and confirmed by X-ray crystallographic analysis.

Abstract:From the fruits of Celastrus orbiculatus Thunb. a new beta-dihydroagarofuran sesquiterpene ester named 1beta,2beta,13-triacetoxy-9alpha-cinnamoyloxy-beta-dihydroagarofuran (1) has been isolated along with two known compounds, 6alpha-acetoxy-1beta, 9beta-dibenzoyloxy-8beta-hydroxy-beta-dihydroagarofuran (2), and 1beta, 6alpha-diacetoxy-9beta-benzoyloxy-8beta-hydroxy-beta-dihydroagarofuran ( 3). Abstract:Two novel diterpenoids, one with a rearranged trans,trans-fused tricyclo[10.3.0.04,6]pentadecane framework (1) and the other with an unprecedented 15S configuration (2), were isolated from Euphorbia helioscopia. Their structures were elucidated by extensive analysis of HR-ESI-MS, NMR, quantum-chemical calculation, and X-ray crystallographic data. Biosynthetically, 1 has a unique "cyclopropane-shift-like" biogenesis involving an oxa-di-π-methane (ODPM) rearrangement, which inspired us to accomplish the biomimetic conversion of 3 to 1. Moreover, compound 1 displayed a potent immunosuppressive effect by inhibiting Kv1.3 voltage-gated channels.

Abstract:A new sesquiterpene polyol ester, 1-beta-acetoxy-2-beta, 8-beta, 9-alpha-tribenzoyloxy-6-alpha-hydroxy-beta-dihydroagarofuran, was isolated from the root bark of Celastrus rosthornianus. Its structure was elucidated mainly on the basis of 1D and 2D NMR spectrometry. The complete assignment of the C-13 NMR chemical shifts, and the determination of H-1-H-1 weak couplings observed in the H-1 NMR spectrum were also carried out on the basis of the 2D NMR spectra.

Abstract:Compounds 1-3, the rare examples of 9,11-seco euphane or lanostane triterpenoids featuring an enol-hemiacetal functionality, were isolated from Euphorbia stracheyi. Their structures were elucidated by a combination of spectroscopic, computational, chemical, and single-crystal X-ray diffraction means, which enables the structure of previously published euphorol J to be revised as 1. 1-3 showed significant cytotoxicities on the breast cancer cell line MDA-MB-468 with IC50 values in the range of 2.9-3.9 μM.

Abstract:From an ethanol extract of Euphorbia micractina roots, seven steroids fifteen aromatic derivatives were isolated by a combination of various chromatographic techniques, including column chromatography over macroporous resin, silica gel, and Sephadex LH-20 and reversed-phase HPLC. Their structures were elucidated by spectroscopic data analysis as stigamast-5-ene-3beta, 7alpha-diol(1), stigamast-5-ene-3beta,7beta-diol(2), stigmast-5-en-3beta-ol-7-one(3), stigmast-4-en-6beta-ol-3-one(4), stigmast-1, 4-dien-3-one(5), stigmast-3,6-dione(6), beta-sitosterol(7), scopoletin(8), aesculetin(9), 6-hydroxy-5,7-dimethoxycoumarin(10), quercetin(11), 3,3', 4'-tri-O-methylellagic acid(12), p-hydroxyphenylethyl anisate(13), m-hydroxyphenylethyl alcohol(14), (E)-cinnamic acid(15), (E)-ferulic acid(16), 3,4-dihydroxybenzoic acid(17), vanillic acid(18), p-hydroxybenzoic acid(19), ethyl 3,4-dihydroxybenzoate (20), ethyl gallate(21), and methyl gallate(22). Abstract:From the dried roots of Euphorbia fischeriana, seven new diterpenoids, 3 alpha, 17-dihydroxy-ent-pimara-8(14), 15-diene (1), 7 beta, 11 beta, 12 beta-trihydroxy-ent-abieta-8(14), 13(15)-dien-16,12-olide (2), 17-acetoxyjolkinolide B (3), 13 beta-hydroxy-ent-abiet-8(14)-en-7-one (4), 12-deoxyphorbaldehyde-13-acetate (5) 12-deoxyphorbaldehyde-13-hexadecacetate (6), and 12-deoxyphorbol-13-(9Z)-octadecanoate-20-acetate (7), and two known compounds, 12-deoxyphorbol-13-decanoate (8) and prostratin (9), were isolated. The structures of the new compounds were elucidated on the basis of spectroscopic analysis. The structure of compound 1 was confirmed by single-crystal X-ray crystallography. Compounds 3 and 8 exhibited potent cytotoxic activity to Ramos B cells with IC50 values of 0.023 and 0.0051 mu g/mL, respectively.

Abstract:Nine jatrophane diterpenoids were isolated from the whole plant Euphorbia helioscopia , including two new ones, helioscopnins A ( 1 ) and B ( 2 ). Comprehensive spectroscopic data analysis and ECD calculations elucidated their structures, including absolute configurations. All compounds were evaluated for bioactivity towards autophagic flux by flow cytometry using HM mCherry-GFP-LC3 cells. Compounds 1 , 3 , 4 , 5 , 8 , and 9 significantly increased autophagic flux.

Abstract:Euphorhelipanes A (1) and B (2), two Euphorbia diterpenoids with a new 4-(5,5-dimethylheptan-2-yl)-2,7-dimethylbicyclo[4.3.0]nonane skeleton, were isolated from a 95% ethanol extract of the whole plants of Euphorbia helioscopia. Their structures were elucidated by spectroscopic data analysis, quantum chemical calculations, and single-crystal X-ray diffraction data. Compounds 1 and 2 represent the first examples of Euphorbia diterpenoids with a 5/6 fused carbon ring system, and their plausible biosynthetic pathways originating from jatrophanes are proposed. Compounds 1 and 2 showed a triglyceride-lowering effect in oleic-acid-stimulated HuH7 cells at concentrations of 1-50 μM.

Abstract:Two new triterpenoids, namely 24-methylene-5,24-dien-19(10→9)- abeo -8 α ,9 β ,10 α -eupha-3 β -ol ( 1 ) and 24-methyl-5,23-dien-19(10→9)- abeo -8 α ,9 β ,10 α -eupha-3 β -ol ( 2 ) were isolated from the stems of Euphorbia royleana , together with three known analogs. The structures of the new compounds were elucidated by extensive 1 H NMR, 13 C NMR, HSQC, HMBC, 1 H- 1 H COSY, ROESY and HR-MS spectroscopic analyses.

Abstract:Twelve undescribed jatrophane diterpenoids, euphpepluones A-L (1-12), together with seven known analogues (13-19), were isolated from the whole plant of Euphorbia peplus, and their structures were elucidated by spectroscopic studies. The absolute configurations of 1 and 4 were assigned by X-ray crystallographic analysis. All isolates were investigated for their inhibitory effects against the ATR-Chk1 pathway using a Western blotting assay. As a result, 1, 2, 5, 8, 10, and 16 were found to suppress the camptothecin (CPT)-induced phosphorylation of Chk1, indicating that these compounds inhibit the activation of the ATR-Chk1 pathway. A preliminary structure-activity relationship (SAR) study of the isolates was conducted. When compound 10 and CPT were combined, apoptosis was induced in A549 cells with PARP cleavage, while there was no apoptotic effect by treatment with CPT or 10 alone. The data obtained indicate that 10 potentiates the chemotherapeutic sensitivity of A549 cells to CPT.

Abstract:Three novel rearranged tigliane-type diterpenoids, euphebrananes A-C (1–3), and one precursor tigliane-type diterpenoid, euphebrananes D (4), were isolated from the roots of Euphorbia ebracteolata. The structures of isolates were identified by comprehensive spectroscopy, HRESIMS, quantum chemical calculations (ECD and NMR), and X-ray crystallography. Euphebrananes A-C represented a tigliane-type diterpenoid, and a plausible biosynthetic pathway of 1–4 was proposed. In vitro, anti-inflammatory and estrogen-like activity screening showed that 1–4 exhibited weak related biological activities.

Abstract:OBJECTIVE:To study macrocyclic jatrophane diterpenes of Euphorbia kansui.METHOD:The compounds were isolated and purified by various column chromatographic methods. Structures were elucidated by spectroscopic analyses.RESULT:Three macrocyclic jatrophane diterpenes were isolated from E. kansui and were characterized as kansuinin D1 (1), kansuinin D (2), kansuinin A (3).CONCLUSION:Kansuinin D1 (1) was a new compound.

Abstract:Jatrofolianes A (1) and B (2), which are two highly modified lathyrane diterpenoids, were isolated from Jatropha gossypiifolia. 1 incorporates an unusual transannular 1,3-dioxolane moiety, forming a unique 5/6/5/8/3 ring system, while 2 possesses a new 10,11:13,14-diseco-lathyrane skeleton with a 12-membered macrocyclic lactone ring. Their structures were determined by spectroscopic analysis, quantum chemical calculations, and single-crystal X-ray diffraction. 2 showed significant multidrug resistance (MDR) reversal activity to cancer cells HepG2/ADR and HCT-15/5-FU at 10 μM.

They are 24-methylenecycloartanone, tirucallol, procesterol and a new isopimarane diterpene, namely yuexiandajisu C. The structure of yuexiandajisu C was elucidated by spectral analysis. The bioassay in vitro showed yuexiandajisu C exhibited immunomodulatory activity.

Abstract:Euphohyrisnoids A (1) and B (2), two highly rearranged lathyrane diterpenoids featuring a unique tetracyclo[10.2.2.0(1,10).0(3,7)]cetane and tricyclo[8.4.1.0(3,7])pentadecane skeleton, respectively, were isolated from the seeds of Euphorbia lathyris. Their structures were determined by detailed spectroscopic analysis and were further confirmed by single-crystal X-ray diffraction. 1 significantly inhibited adipogenesis in 3T3-L1 adipocytes by retarding cell differentiation at the early stage.

Abstract:Seven new triterpenoids including two cycloartanes (1-2), a lanostane (3), a tirucallane (4), a dammarane (5), an ursane (6), and an oleanane (7), along with nineteen known triterpenoids (8-26), have been obtained from the roots of Euphorbia fischeriana. Their structures were established by NMR, HRESIMS, single-crystal X-ray diffraction analysis, Mosher's method, NMR calculations, ECD analysis, and comparison with structurally related known analogues. Among them, compounds 1 and 8 were a pair of cycloartane-type triterpenoids epimers. Our bioassays have established that compounds 1-5 and 10 displayed moderate cytotoxic effects, and the structure-activity relationships of cycloartane-type triterpenoids (CTTs) were further examined. Notably, some triterpenoids displayed moderate inhibitory effects against AChE by an in vitro screened experiment. Triterpenoid 7 (Euphorfistrine G, ETG) displayed the potent inhibitory effect with IC50 = 2.45 and Ki = 2.30 μM (inhibition kinetic). And, in silico docking analyses have been performed to investigate the inhibitory mechanism of compound 7.

Abstract:Euphylonoids A (1) and B (2), two highly modified jatrophane diterpenoids, were isolated from Euphorbia hylonoma. 1 represents a new 9(10 -> 18)-abeo-8,12-cyclojatrophane skeleton containing a cage-like 3,8-dioxatricyclo[5.1.2.0(4,9)]decane core, while 2 is a 14(13 -> 20)-abeo-8,12-cyclojatrophane featuring an unusual 17-oxatetracyclo[12.2.1.0(1,5).0(9,13)]-heptadecane framework. Their structural elucidation was completed by spectroscopic, chemical, computational, and single-crystal X-ray diffraction means. 2 significantly inhibited early adipogenesis in 3T3-L1 adipocytes via activating AMP-activated protein kinase signaling.

Abstract:Four new lanostane triterpenoids, namely (3 ss)-3-hydroxy-24-methylenelanost-8-ene-7,11-dione (1), (3 ss)-3-hydroxylanosta-8,24-diene-7,11-dione (2), (3 ss,7 alpha)-3,7-dihydroxylanosta-8,24-dien-11-one (3), and (3 ss,11 ss)-3,11-dihydroxylanosta-8,24-dien-7-one (4) were isolated from Euphorbia humifusa, together with 2 known compounds. The structures of these new compounds were elucidated on the basis of extensive spectroscopic analysis and comparison with the related known compounds.
