Abstract:Aptamer is an oligonucleotide sequence that can bind to a specific target molecule, which is screened from a random oligonucleotide library. It has shown promising prospects in biomedical applications. Due to its decisive advantages such as simple preparation, easy modification, good stability and wide range of targets compared with traditional antibodies, aptamer has been widely applied in the development of biosensors, which can not only be used to realize the identification and detection of target molecules, but also be utilized as the bridge between sensitive elements and transducers to improve the coupling efficiency. In addition, in recent years, the introduction of various nanomaterials such as metal nanoparticles, carbon materials and functional nanospheres to aptamer-based biosensors has contributed to their improvement in analytical performances and commercial applications. First, this review briefly introduces the methods for aptamer screening based on the SELEX technique and summarizes basic properties of aptamers. Second, the basic principle and classification of aptamer-based biosensors are introduced in detail. Third, based on the most recent progress in the field of aptamer-based biosensors, we focus on the applications of aptamers in biosensor development, including the applications in the field of biomimetic olfactory and taste receptor biosensors as well as applications in the detection of small molecules and proteins. Finally, we analyze and discuss the problems and development trends of applications of aptamers in biosensors.

Abstract:Cell-based biosensor is a hotspot in research of biosensors recently, which takes living cell as sensitive unit and analyzes the functional information of analytes. Cell-based biosensor characterizes with high sensitivity, excellent selectivity and fast response. So it can be applied in many areas, such as biomedical, environmental detection and drugs discovery, etc. This paper reviews the development of surface processing technique of cell-based biosensor and our recent some research results. Besides, this paper also indicates the recent problems of this kind of biosensors and their future development trends.

Abstract:This paper reviews a novel cell-based biosensor and Bio-MEMS which incorporate living cells as sensing elements that convert a change in immediate environment to signals conducive for processing.It is characterized with high sensitivity,excellent selectivity and fast response and have been implemented for a number of applications ranging from pharmaceutical screening to environmental pollutant detection.This paper also introduces our recent work about Light-Addressable Potentiometric Sensors （LAPS）,Field Effect Transistor （FET）,Micro-Electrode Array Sensors （MEAS） and Bio-MEMS for detecting the changes of concentration of extracellular ions and the action potential of living cell under effect of drugs and environmental parameters.Finely, the paper gives some prospects of cell-based biosensors in the future.

Abstract:Receptor and ion channel are natural bio-sensing units on the membrane of mammalian cells.On the basis of analyzing the advantages of receptor and ion channel and specific obstacles for the development of biosensors,some biomedical sensor developments are introduced in detail,such as bilayer lipid membrane platforms supported by nano-porous,electrifying cell receptors with ion channels coupled to receptors,and ion channels and receptors naturally expressed on cell-based biosensors.Based on these new techniques and methods,the development and application of receptor and ion channel based biosensors was prospected.

Abstract:Biosensors utilizing living tissues and cells have recently gained significant attention as functional devices for chemical sensing and biochemical analysis. These devices integrate biological components (i.e. single cells, cell networks, tissues) with micro-electro-mechanical systems (MEMS)-based sensors and transducers. Various types of cells and tissues derived from natural and bioengineered sources have been used as recognition and sensing elements, which are generally characterized by high sensitivity and specificity. This review summarizes the state of the art in tissue- and cell-based biosensing platforms with an emphasis on those using taste, olfactory, and neural cells and tissues. Many of these devices employ unique integration strategies and sensing schemes based on sensitive transducers including microelectrode arrays (MEAs), field effect transistors (FETs), and light-addressable potentiometric sensors (LAPSs). Several groups have coupled these hybrid biosensors with microfluidics which offers added benefits of small sample volumes and enhanced automation. While this technology is currently limited to lab settings due to the limited stability of living biological components, further research to enhance their robustness will enable these devices to be employed in field and clinical settings.

Cell-based biosensors have been implemented for a number of applications ranging from pharmaceutical screening to environmental pollutant detection. This work demonstrate a novel cell-based biosensor following the principle of light-addressable potentiometric sensor and field effect transistor for detecting the action potential of single living cell under drugs. The experimental results agree with our expectation.

Single-cell based biosensor characterizes with high sensitivity, excellent selectivity and fast response. So it is able to be applied rapidly in many areas, such as biomedical, enviromental detection and drugs discovery. This paper presents a novel single-based biosensor, from the ground of light-addressable potentiometric sensor. We develope the structure and analyzed the possibility of single-cell based biosensor used to detect electric characters of living cell. The results confirm that this kind of blosensor is a potential tool for research of electrophysiology of living cell and evaluation of drugs.

Abstract:In this chapter, the enzyme biosensors and microorganism biosensors are introduced as catalytic biosensors. The antibody and antigen biosensors (immune sensors), nucleic acid biosensors (DNA sensors), as well as receptor and ion-channel sensors are introduced as affinity biosensors. Then, sensors for measurement of cellular metabolism, impedance, and electrophysiology are discussed in cell and tissue biosensors. Finally, some novel techniques such as biochips for microarrays and nanomaterials are also described, including typical sensing applications for biosensors.

Abstract:Biosensors, devices capable of detecting biomolecules or bioactive substances, have recently become one of the important tools in the fields of bioanalysis and medical diagnostics. A biosensor is an analytical system composed of biosensitive elements and signal-processing elements used to detect various biological and chemical substances. Biomimetic elements are key to biosensor technology and are the components in a sensor that are responsible for identifying the target analyte. The construction methods and working principles of biosensors based on synthetic biomimetic elements, such as DNAzyme, molecular imprinted polymers and aptamers, and their updated applications in biomedical analysis are summarised. Finally, the technical bottlenecks and future development prospects for biomedical analysis are summarised and discussed.

Abstract:Aptamers are oligonucleotides with distinct conformational shapes that allow them to bind a wide range of targets with high affinity and specificity. Compared with intact parent aptamers, split aptamers show advantages of small size and less unfavorable secondary structures, thus avoiding high background, false positive or non-specific signals well. Moreover, they are favorable to design label-free strategies since some fluorescent dyes are known to intercalate into DNA structure rather than covalent conjugation. So far, split aptamer fragments have shown good application prospects in the development of various high-performance biosensors. In this review, recent advances of biosensors based on split aptamers in biological analysis are summarized and elucidated. It includes the detection of nucleotides/nucleosides, alkaloids, proteins, antibiotics, exosomes, and tumor cells. Potential general platforms for the assay of more than one targets based on multifunctional biosensor are also included. Challenges and prospects for future development are discussed to provide valuable insights for the research of new split aptamers and aptasensors for more targets analysis.

Abstract:Cell-based electrochemical biosensors have contributed tremendously to the fields of biology, medicine, chemistry, pharmacology, and environmental science. With electrochemical transducers and whole cells as the recognition elements, these biosensors provide new horizons for biosensing and life science research. This review focuses on the research accomplishments on this topic over the last three years, and is divided into three sections according to the types of cellular responses. Our aim is to highlight how simple and sensitive electrochemical methods can be coupled with cells by virtue of the integration of interface control, nanotechnology and genetic engineering to generate new enabling technologies. Some specific examples to demonstrate how these sensors are useful in medicinal chemistry and drug design have also been discussed. It is hoped that this review can provide inspiration for the development of fast, selective, sensitive, and convenient detection and diagnosis platforms.

Abstract:Biosensors have been flourishing in the field of drug discovery with pronounced developments in the past few years. They facilitate the screening and discovery of innovative drugs. However, there is still a lack of critical reviews that compare the merits and shortcomings of these biosensors from a pharmaceutical point of view. This contribution presents a critical and up-to-date overview on the recent progress of tailored biosensors, including surface plasmon resonance, fluorescent, photoelectrochemical, and electrochemical systems with emphasis on their mechanisms and applications in drug screening, efficacy assessment, and toxicity evaluation. Multiple functional nanomaterials have also been incorporated into the biosensors. Representative examples of each type of biosensors are discussed in terms of design strategy, response mechanism, and potential applications. Abstract:Biosensors have achieved significant progress in the past decades and are now widely used for diagnostic and therapeutic purposes. A biosensor is defined as a device for analyte detection or physiological signal measurements, which usually combines selective biological elements and transducers that convert signals resulting from the interaction between the analyte and biological elements to other easily measured outputs (eg, electrical, optical, and thermal signals)[1]. A broad range of biomaterials have been involved in building various biosensors. The major components usually include a probe that selectively responds to a certain biological element, such as deoxyribonucleic acid (DNA), aptamer, antibody, enzyme, cell, and so on, and a transducer that transforms the physiochemical information into optical, electrical, magnetic, electrochemical, and thermal signals. Due to the advantages of high surface area, miniaturized size, and associated new physical properties, nanomaterials have drawn a great deal of attention as components for biosensors. Abstract:Biosensors have rapidly emerged as a high-sensitivity and convenient detection method. Among various types of biosensors, optical and electrochemical are the most commonly used. Conventionally, antibodies have been employed to ensure specific interaction between the transmission material and analytes. However, there has been increasing recognition of peptides as a promising recognition element for biosensor development in recent years. The use of peptides as recognition elements provides high level of specificity, sensitivity, and stability for the detection process. The combination of peptide designs and optical or electrochemical detection methods has significantly improved biosensor efficacy. These advancements present opportunities for developing biosensors with diverse functions that can be used to lay a strong scientific foundation for the development of personalized medicine and various other fields. This paper reviews the recent advancements in the development and application of peptide-based optical and electrochemical biosensors, as well as their prospects as a sensor type.

Abstract:In recent years, taste cell- and receptor-based biosensors have gained considerable attention as an emerging technology for chemical sensing as well as research on taste signal transduction mechanisms. Taste cells and receptors can recognize the specific chemical signals presented by various taste substances and compounds offering unique performance characteristics which cannot be achieved by current artificial devices. As a result, taste cells and receptors have been increasingly recognized as promising candidates for the development of chemical sensors. Currently, one of the main challenges of this technology is the preparation of functional taste cells and receptors to satisfy the need of building sensing components for chemical sensors. Additionally, the coupling of taste cells and receptors with transducers is a key step to develop chemical sensors which can meet the specific performance requirements for biomedical applications. Therefore, current efforts are mainly focused on the preparation of functional taste cells and receptors and their coupling with transducers. This review will summarize the most recent advances in taste cell- and receptor-based biosensors. We will describe the basis of biological taste sensing systems, current approaches for preparing functional taste cells and receptors, and basic design principles and approaches for coupling taste cells and receptors with transducers. Furthermore, several major applications of taste cell- and receptor-based biosensors will be presented. We will conclude by discussing the current and future challenges of taste cell- and receptor-based biosensors and offer possible strategies to overcome these limitations.

Abstract:Synthetic biology has promoted the development of biosensors as tools for detecting trace substances. In the past, biosensors based on synthetic biology have been designed on living cells, but the development of cell biosensors has been greatly limited by defects such as genetically modified organism problem and the obstruction of cell membrane. However, the advent of cell-free synthetic biology addresses these limitations. Biosensors based on the cell-free protein synthesis system have the advantages of higher safety, higher sensitivity, and faster response time over cell biosensors, which make cell-free biosensors have a broader application prospect. This review summarizes the workflow of various cell-free biosensors, including the identification of analytes and signal output. The detection range of cell-free biosensors is greatly enlarged by different recognition mechanisms and output methods. In addition, the review also discusses the applications of cell-free biosensors in environmental monitoring and health diagnosis, as well as existing deficiencies and aspects that should be improved. In the future, through continuous improvement and optimization, the potential of cell-free biosensors will be stimulated, and their application fields will be expanded.

Abstract:In recent decades, as a subclass of biomaterials, biologically sensitive nanoparticles have attracted increased scientific interest. Many of the demands for physiologically responsive nanomaterials in applications involving the human body cannot be met by conventional technologies. Due to the field’s importance, considerable effort has been expended, and biologically responsive nanomaterials have achieved remarkable success thus far. This review summarizes the recent advancements in biologically responsive nanomaterials and their applications in biosensing and molecular imaging. The nanomaterials change their structure or increase the chemical reaction ratio in response to specific bio-relevant stimuli (such as pH, redox potentials, enzyme kinds, and concentrations) in order to improve the signal for biologically responsive diagnosis. We use various case studies to illustrate the existing issues and provide a clear sense of direction in this area. Furthermore, the limitations and prospects of these nanomaterials for diagnosis are also discussed.

Abstract:With recent advances in point-of-care testing, wearable electronics, and implantable medical devices, there is a growing interest in developing state-of-the-art methods to monitor human health conveniently and dynamically. Among these methods, electrochemical biosensors have offered a unique opportunity to transform lab-in-hospital to lab-around-body and lab-on-body. In an electrochemical biosensing system, appropriate bioreceptors that could specifically recognize and capture target molecules pose a significant impact on the performance of the biosensors. Thus, this review systematically explores different types of bioreceptors and their immobilization methods utilized in electrochemical biosensors for the detection of a wide range of biomarkers in clinical applications. Moreover, it delves into different principles behind the operation of electrochemical biosensors and provides insights into the challenges and limitations associated with the use of different bioreceptors in practical settings. Furthermore, this review proposes possible strategies to overcome existing challenges in order to stimulate further research and clinical implementation of biosensing methods on a larger scale.

Abstract:The fields of biosensing have been transformed by the discovery of extraordinary molecular recognition components, such as aptamers and biomimetic receptors. Systematic Evolution of Ligands by Exponential Enrichment (SELEX) is a method used to select aptamers, or short sequences of single-stranded DNA (ssDNA) or RNA (ssRNA), based on their unique binding affinity to target molecules. Molecularly imprinted polymers (MIPs) are a type of biomimetic receptor that mimics the selectivity of natural receptors inside a synthetic matrix. They make it possible to identify pathogens, and illness biomarkers with accuracy. Aptamers and biomimetic receptors play crucial roles in various fields including diagnostics, therapeutics, and biosensing. Their high specificity, versatility, and adaptability enable targeted detection, drug delivery, and biomolecule manipulation, thereby contributing to advancements in personalized medicine, biotechnology, and disease diagnosis. Aptamers and biomimetic receptors have been combined with cutting-edge technologies, like nanotechnology and lab-on-a-chip systems, to create biosensors that are quick, portable, and extremely sensitive. These recognition features are anticipated to become more important as technology develops, helping to address global issues, advance biosensing capabilities, and raise people's standard of living everywhere. Abstract:Nature has inspired the development of biomimetic membrane sensors in which the functionalities of biological molecules, such as proteins and lipids, are harnessed for sensing applications. This review provides an overview of the recent developments for biomembrane sensors compatible with either bulk or planar sensing applications, namely using lipid vesicles or supported lipid bilayers, respectively. We first describe the individual components required for these sensing platforms and the design principles that are considered when constructing them, and we segue into recent applications being implemented across multiple fields. Our goal for this review is to illustrate the versatility of nature's biomembrane toolbox and simultaneously highlight how biosensor platforms can be enhanced by harnessing it.
