Abstract:A convenient and efficient synthesis of 2-substituted benzo[b]thiophenes is described. This one-step reaction of alkylthiobenzaldehyde and activated electrophiles proceeded smoothly in toluene at 120 degrees C with magnesium oxide as a catalyst. The target 2-substituted benzo[b]thiophenes are obtained in moderate yields and the side reaction is inhibited via azeotropic distillation. According to the experiments, a plausible mechanism is presented.

Abstract:A Lewis acid-catalyzed tandem nucleophilic addition/C-S cleavage/intramolecular annulation of ketene 1,3-dithietanes with aldehydes and photo-induced cycloaddition of ketene 1,3-dithietanes with alkynes were developed in this work. The readily available ketene 1,3-dithietanes were demonstrated as a promising sulfur-containing synthon for the facile construction of the thiophene ring. Moreover, these processes feature simple operation, good functional tolerance and mild reaction conditions. Also, the possible mechanisms are proposed in this study.

Abstract:Pyridinium 1,4-zwitterionic thiolates were applied to a formal [3+2] annulation reaction with modified activated alkynes, affording various tetrasubstituted thiophenes with aryl, alkenyl, alkyl or silyl group at the special position. The structural modification of alkyne substrates enabled the synthesis of diverse thiophenes to be achieved using the pyridinium 1,4-zwitterionic thiolates as the sulfur-containing build-ing blocks. This approach is metal-free and catalyst-free.

Abstract:Several aromatic aldehydes and sodium bisulfite were mixed to prepare hydroxyl sulfonates,which were added to o-phenylenediamine or substituted o-phenylenediamine in refluxing ethanol to afford 2-substi-tuted benzimidazoles through oxidation by air directly.One of the fifteen synthesized compounds,2-(4-chlorophyenyl)-6-methoxyl-1H-benzimidazole has not been reported.All the compounds were identified by IR,1H NMR spectra and elemental analysis.The method is simple,mild,eco-friendly and requires no catalyst.

Abstract:We developed a novel method for the synthesis of 2‐substituted‐4,5‐dihydrothiazol‐4‐ols compounds through cyclization of thioamides and 1,4‐dithiane‐2,5‐diol in the presence of Et3N. Stable hydroxy‐substituted thiazole compounds were isolated of substituted thioamides. The reaction conditions are simple, the substrate range is wide, and the target compounds were obtained in up to 82% yield.

Abstract:Herein, we reported the straightforward synthesis of thiomethylnaphthalenes via the reaction of propargyl alcohols with DMSO/SOCl2, which also provided easy access to deuterated thiomethyl and other thioalkyl scaffolds when deuterated dimethyl sulfoxide and dialkyl sulfoxides were used, respectively. In addition, the thiomethyl group can be readily derivatized to the corresponding sulfoxide, sulfone, and sulfoximine of interesting photophysical properties.

Abstract:The Cu(I)-catalyzed intramolecular cyclization of ortho-halogenated phenylthioacetamides to form 2-amino benzo[b]thiophenes in water was studied. This method employed water instead of organic solvent, and the catalyst-loading can be as low as to 5 mol% and the yield of products is up to 98%, which overcomes the drawbacks such as harsh reaction conditions and low yields in the previous methods. In addition, the reaction can be scaled up to gram-scale and catalyst can be reused with a maintained yield of products. This reaction provided a simple and environmentally friendly protocol for the preparaion of 2-amino benzo[b]thiophene and their derivatives.

Abstract:A facile and practical method for the synthesis of 2,3,4-trisubstituted 2-aminothiophenes by the cyclization of gem-dibromoalkenes or gem-dichloroalkenes with β-keto tertiary thioamides has been developed. The cyclization reaction proceeded chemoselectively and regioselectively under metal-catalyst-free conditions, providing various structurally diverse 2,3,4-trisubstituted N,N'-dialkyl 2-aminothiophenes in good to excellent yields.

Abstract:Trifluoromethylthiolation/oxidation of styrenes with easiy-handling AgSCF3 used as the trifluoromethylthiolating reagent has been described, which furnished a series of α-trifluoromethylthio acetophenons. A variety of substituted styrene could be bifunctionalized via radical process in a mixed solvent of dimethyl sulfoxide (DMSO) and CHCl3, with potassium persulfate used as the oxidant under mild conditions. A simple and efficent method for the facile construction of various α-trifluoromethylthio acetophenons is thus demonstrated.

Abstract:Photoredox-catalyzed aminoarylation and thioamination of unactivated alkenes have been developed, providing novel synthetic routes to access synthetically challenging quaternary carbon-centered benzoindolizidinones and trifluoromethylthiolated piperidines using readily available starting materials. Notably, these transformations were enabled by merging amidyl radical generation from N-alkyl benzamides with oxidant incorporation. Density functional theory calculations were performed to understand the reaction mechanism and to rationalize the regioselectivities. Moreover, the newly developed catalytic aminoarylation provided a convenient synthetic route for natural product tylophorine and its gem-dimethyl analogues with greatly improved drug-like properties such as enhanced solubility and stability. Abstract:A facile synthetic method for the construction of 3-aminothiophenes from readily available thioamides and alllenes in the presence of a TBAI/TBHP catalyst system was developed. Abstract:Functionalized thiaphenalene derivatives are of high synthetic value, yet their preparation remains underexplored. Herein, we report an efficient approach for the synthesis of 1-thiaphenalene derivatives through radical cyclization of 1-naphthalenesulfonyl chlorides with alkynes. A variety of 1-thiaphenalene derivatives are readily furnished that are otherwise difficult to prepare by present methods. The protocol features mild photocatalytic conditions, broad functional group compatibility, and high product diversity.

Abstract:The incorporation of the (trifluoromethyl)thio group into the isocoumarin scaffold through a Lewis-acid-mediated electrophilic cyclization reaction of 2-(2-alkynyl)benzoates with trifluoromethanesulfanylamide is reported. The transformation proceeds well in the presence of BiCl3 and BF3 center dot Et2O under mild conditions to give 4-[(trifluoromethyl)thio]-1H-isochromen-1-ones regioselectively and in good yields.

Abstract:1,4-Bis(tert-butylthio)-3,6-diphenyl- and 3,6-di(2-thienyl)thieno[3,4-c]thiophenes (2a,b)were synthesized from 2-(tert-butylthio)-3-phenyl- and 3-(2-thienyl)cyclopropenethiones (1a,b) and triphenylphosphine in dry benzene at 50 degrees C, although similar treatment of 2,4,6-triisopropylphenyl, N,N-diethylamino, pyrrolidino, and diphenyl-substituted cyclopropenethiones (1c-h) did not result in the production of the corresponding thieno[3,4-c]thiophene derivatives. The possible reaction pathway for the formation of 2a,b is described. The protonation of 2a with trifluoroacetic acid (TFA) gave 4-(tert-butylthio)-3,6-diphenylthieno[3,4-c]thiophene-1(3H)-thione (13a), the treatment of which with sodium hydride and then isopropyl iodide led to 4-(tert-butylthio)-3,6-diphenyl-1-(isopropylthio)thieno[3,4-c]thiophene (16) by the regeneration of the thieno[3,4-c]thiophene ring system, this making possible the synthesis of other alkylthio-substituted thieno[3,4-c]thiophene derivatives. The reactions of 2a,b with N-phenylmaleimide (NPM) gave predominantly the endo-cycloadducts (17a,b) at the 1- and 3-positions, and that of 2a with dimethyl acetylenedicarboxylate (DMAD) led to the benzo[c]thiophene derivative (19) by desulfurization.

Abstract:A new strategy for the preparation of 3-halobenzo[b]thiophenes via a photo-driven halocyclization/demethylation of 2-alkynylthioanisoles with simple alkyl halides was developed. The reaction can proceed smoothly at room temperature under visible-light irradiation without any external photocatalyst, and the protocol has a range of advantages, including simplicity and mildness of the reaction conditions, good functional-group tolerance, and excellent yields of the products.

Abstract:An efficient one-pot method is proposed for the synthesis of 2-aroylbenzo[ b ]thiophen-3-ols from 2-mercaptobenzoic acid and various substituted aryl bromomethyl ketones in the presence of triethylamine. The reaction is likely to proceed through S N 2-type nucleophilic attack of the sulfhydryl group in thiosalicylic acid on bromomethyl ketone in the presence of a base to afford sulfanylbenzoic acid, which undergoes an intramolecular cyclization in situ to furnish 2-aroylbenzo[ b ]thiophen-3-ol in high yield. To investigate the utility of the synthesized benzothiophene scaffold, an alkyne moiety was introduced at the 3-hydroxy position and subsequently subjected to a click reaction to form novel benzothiophene-triazole hybrids in good yields. A simple and straightforward approach to synthesizing 2-aroylbenzo[ b ]thiophen-3-ols can open new avenues for discovering novel biological and pharmaceutical compounds.
