Abstract:A convenient and efficient synthesis of 2-substituted benzo[b]thiophenes is described. This one-step reaction of alkylthiobenzaldehyde and activated electrophiles proceeded smoothly in toluene at 120 degrees C with magnesium oxide as a catalyst. The target 2-substituted benzo[b]thiophenes are obtained in moderate yields and the side reaction is inhibited via azeotropic distillation. According to the experiments, a plausible mechanism is presented.

Abstract:An improved procedure for the synthesis of 3,4-ethylenedioxythiophene is reported starting from ethyl chloroacetate. Reaction with sodium sulfide gave diethyl thiodiglycolate which was then reacted with diethyl oxalate, and then 1,2-dibromoethane to give 2,5-dicarbethoxy-3,4-ethylenedioxythiophene. Hydrolysis, and decarboxylation gave 3,4-ethylenedioxythiophene in 16% overall yield. The structures of the key intermediates in synthetic routes were confirmed, and every step was optimised, to give a procedure suitable for large-scale industrial production.

Abstract:A one-pot two-step reaction of 4-aminocoumarin with arylglyoxal monohydrates and p-toluenesulfonates has been developed, which offers 3-alkoxy-substituted chromeno[4,3-b]pyrrol-4(1H)-ones in moderate to good yields. At first, arylglyoxal monohydrates and 4-aminocoumarin were converted into 3-hydroxychromeno[4,3-b]pyrrol-4(1H)-one intermediates in AcOH under reflux. Then the title compounds in an in situ reaction of the intermediates with p-toluenesulfonates catalyzed by DBU in refluxing toluene were obtained.

Abstract:An efficient synthetic approach to variously substituted 2,3-dihydrothiophenes has been developed. The reaction proceeded by copper-catalyzed tandem S-alkylation and S-alkenylation of sodium sulfide with 1,4-diiodobut-1-enes to afford the corresponding 2,3-dihydrothiophene derivatives in high yields. The 2,3-dihydrothiophenes could bear alkyl, aryl as well as heteroaryl substituents. The resulted 2,3-dihydrothiophenes could be further transferred into the corresponding thiophenes by oxidation.

Abstract:A convenient methodology for the synthesis of mono- and di-halogenated benzo[b]thiophenes is described herein, which utilizes copper(II) sulfate pentahydrate and various sodium halides in the presence of substituted 2-alkynylthioanisoles. The proposed method is facile, uses ethanol as a green solvent, and results in uniquely substituted benzo[b]thiophene structures with isolated yields up to 96%. The most useful component of this methodology is the selective introduction of bromine atoms at every available position (2-7) around the benzo[b]thiophene ring, while keeping position 3 occupied by a specific halogen atom such as Cl, Br or I. Aromatic halogens are useful reactive handles; therefore, the selective introduction of halogens at specific positions would be valuable in the targeted synthesis of bioactive molecules and complex organic materials via metal-catalyzed cross coupling reactions. This work is a novel approach towards the synthesis of dihalo substituted benzo[b]thiophene core structures, which provides a superior alternative to the current methods discussed herein.

Abstract:A new strategy for the preparation of 3-halobenzo[b]thiophenes via a photo-driven halocyclization/demethylation of 2-alkynylthioanisoles with simple alkyl halides was developed. The reaction can proceed smoothly at room temperature under visible-light irradiation without any external photocatalyst, and the protocol has a range of advantages, including simplicity and mildness of the reaction conditions, good functional-group tolerance, and excellent yields of the products.

Abstract:1,4-Bis(tert-butylthio)-3,6-diphenyl- and 3,6-di(2-thienyl)thieno[3,4-c]thiophenes (2a,b)were synthesized from 2-(tert-butylthio)-3-phenyl- and 3-(2-thienyl)cyclopropenethiones (1a,b) and triphenylphosphine in dry benzene at 50 degrees C, although similar treatment of 2,4,6-triisopropylphenyl, N,N-diethylamino, pyrrolidino, and diphenyl-substituted cyclopropenethiones (1c-h) did not result in the production of the corresponding thieno[3,4-c]thiophene derivatives. The possible reaction pathway for the formation of 2a,b is described. The protonation of 2a with trifluoroacetic acid (TFA) gave 4-(tert-butylthio)-3,6-diphenylthieno[3,4-c]thiophene-1(3H)-thione (13a), the treatment of which with sodium hydride and then isopropyl iodide led to 4-(tert-butylthio)-3,6-diphenyl-1-(isopropylthio)thieno[3,4-c]thiophene (16) by the regeneration of the thieno[3,4-c]thiophene ring system, this making possible the synthesis of other alkylthio-substituted thieno[3,4-c]thiophene derivatives. The reactions of 2a,b with N-phenylmaleimide (NPM) gave predominantly the endo-cycloadducts (17a,b) at the 1- and 3-positions, and that of 2a with dimethyl acetylenedicarboxylate (DMAD) led to the benzo[c]thiophene derivative (19) by desulfurization.

Abstract:We developed a method for synthesizing aryl alkyl thioether compounds via a three-component reaction involving aryldiazonium salts, 1,4-diazabicyclo[2.2.2]octane bis(sulfur dioxide), and alkyl bromides. Abstract:A facile and efficient approach for the synthesis of the CF3-containing dioxodibenzothiazepines has been developed via copper-catalyzed trifluoromethylation/cyclization of alkynes utilizing a radical relay strategy. Abstract:A Lewis acid-catalyzed tandem nucleophilic addition/C-S cleavage/intramolecular annulation of ketene 1,3-dithietanes with aldehydes and photo-induced cycloaddition of ketene 1,3-dithietanes with alkynes were developed in this work. The readily available ketene 1,3-dithietanes were demonstrated as a promising sulfur-containing synthon for the facile construction of the thiophene ring. Moreover, these processes feature simple operation, good functional tolerance and mild reaction conditions. Also, the possible mechanisms are proposed in this study.

Abstract:A highly efficient and simple route for the synthesis of 2-alknyl benzo[ d ]thiazoles has been developed by palladium and copper catalyzed the cross-coupling reaction of 2-thiobenzo[ d ]thiazoles with (hetero)aromatic and aliphatic alkynylaluminum reagents. Various 2-alkynyl benzo[ d ]thiazole derivatives can be obtained in 19–97 % isolated yields using 1 mol% PdCl 2 (dppf)/5 mol% xanthpos as the catalyst and CuI (3 equiv.) as desulfurization agent. The cross-coupling reaction can be carried out smoothly with electron-donating or -withdrawing groups on the aromatic ring of the organo aluminum reagents or the 2-thiobenzo[ d ]thiazoles. Furthermore, the broad substrate scope and the maintenance of high efficiency on a gram scale make this protocol a potentially practical method to synthesize 2-alkynylbenzo[ d ]thiazole derivatives. Abstract:An AcOH-mediated three-component reaction of 2-aminothiophenols, alpha,beta-unsaturated aldehydes, and thiophenols to prepare the high value-added 2-thioalkylbenzothiazoles was developed. The reaction was conducted under metal-free conditions, where oxygen served as the terminal oxidant. In addition, this strategy is tolerant of kinds of functional groups, providing efficient access to a variety of functionalized benzothiazole derivatives in moderate to good yields. Abstract:Reaction between 3,3′-dibromothiophene and sodium methylate/methanol in the presence of KI/CuO afforded 3-bromo-4-methoxythiophene as a major product. It was transformed into 4,4′-dimethoxy-3,3′-bithiophene by using Pd(OAc) 2 as a catalyst and agarose as a ligand. The conditions of the reaction were mild with an acceptable yield of 20 %. The product was characterized by MS, 1 H NMR, and its spectra of UV–Vis and cyclic voltammetry were given. Graphical Abstract 4,4′-dimethoxy-3,3′-bithiophene was synthesized from 3-bromo-4-methyloxy thiophene in the homogeneous system formed by water and agarose.

Abstract:Abstract 3H-Benzo[1,2]-dithiole-3-thiones were prepared from potassium sulfide and 2-halobenzaldehydes in moderate-to-good yields, and a plausible mechanism for this catalyst-free intramolecular heteroannulation reaction has been proposed. The Knoevenagel condensation reactions of 3H-benzo[1,2]dithiole-3-thiones with active methylene compounds such as ethyl 2-cyanoacetate and diethyl malonate, and the three-component one-pot reactions of potassium sulfide, 2-halobenzaldehydes, and ethyl 2-cyanoacetate, affording the corresponding products, have been developed.
