Abstract:In this work, a highly sensitive label-free paper-based electrochemical immunosensor employing screen-printed working electrode (SPWE) for detection of carcinoembryonic antigen (CEA) was fabricated. In order to raise the detection sensitivity and immobilize anti-CEA, amino functional graphene (NH2-G)/thionine (Thi)/gold nanoparticles (AuNPs) nanocomposites were synthesized and coated on SPWE. The principle of the immunosensor determination was based on the fact that the decreased response currents of Thi were proportional to the concentrations of corresponding antigens due to the formation of antibody–antigen immunocomplex. Experimental results revealed that the immunoassay enabled the determination of standard CEA solutions with linear working ranges of 50pgmL−1 to 500ngmL−1, the limit of detections for CEA is 10pgmL−1 (S/N=3) and its corresponding correlation coefficients were 0.996. Furthermore, the proposed immunosensor could be used for the determination of clinical serum samples. A large number of clinical serum samples were detected and the relative errors between measured values and reference concentrations were calculated. Results showed that this novel paper-based electrochemical immunosensor could provide a new platform for low cost, sensitive, specific, and point-of-care diagnosis in cancer detection.

Abstract:With the advancement of nanotechnology, various types of nanomaterials have been integrated into electrochemical immunoelectrodes to enhance their performance. Among these, MXene stands out as a promising candidate due to its high electron transfer capacity and abundant surface chemical groups. However, the improvement in electrode performance is often hindered by the self-restacking and agglomeration of MXene. To address this issue, multi-walled carbon nanotubes (MWCNTs) were selected to form composites with MXene. Subsequently, a label-free immunosensor, BSA/Ab/AuNPs/MXene-MWCNTs-Nafion/ITO, was fabricated for specific detection of carcinoembryonic antigen (CEA), a widely used tumor marker. The results demonstrated that the incorporation of MWCNTs can effectively prevent the self-stacking of MXene. Moreover, the composites enhanced the loading of gold nanoparticles (AuNPs) to connect the antibodies, thereby improving electronic transmission signals and sensitivity. The sensor exhibited excellent analytical performance towards CEA with a wide linear range (0.050 to 200 ng mL-1) and a low limit of detection of 0.015 ng mL-1 (S/N = 3). The possibility of it being applied in clinical trials was verified by using ELISA and differential pulse voltammetry (DPV) assays to detect CEA in serum samples. The recoveries ranged from 95.34 to 102.09% with relative standard deviations (RSDs) below 5.00%. Furthermore, the sensor displayed satisfactory selectivity, repeatability, and stability. We hope the findings highlight promising prospects for advanced immunosensor development and alternative strategies in cancer diagnosis.

Abstract:The convenient construction of carbon-based electrochemical immunosensors with high performance is highly desirable for the efficient detection of tumor biomarkers. In this work, an electrochemical immunosensor was fabricated by integrating a biofunctionalized mesoporous silica nanochannel film with a carbon-based electrode, which can enable the sensitive determination of carcinoembryonic antigen (CEA) in serum. The commonly used carbonaceous electrode, glassy carbon electrode (GCE), was employed as the supporting electrode and was pre-treated through electrochemical polarization to achieve the stable binding of a vertically ordered mesoporous silica film with amino groups (NH2-VMSF) without the use of any adhesive layer. To fabricate the immunorecognition interface, antibodies were covalently immobilized after the amino groups on the outer surface of NH2-VMSF was derivatized to aldehyde groups. The presence of amino sites within the high-density nanochannels of NH2-VMSF can facilitate the migration of negatively charged redox probes (Fe(CN)63-/4-) to the supporting electrode through electrostatic adsorption, leading to the generation of electrochemical signals. In the presence of CEA, the formation of immunocomplexes on the recognitive interface can reduce the electrochemical signal of Fe(CN)63-/4- on the supporting electrode. Based on this principle, the sensitive electrochemical detection of CEA was achieved. CEA can be determined to range from 0.01 ng mL−1 to 100 ng mL−1 with a limit of detection of 6.3 pg mL−1. The fabricated immunosensor exhibited high selectivity, and the detection of CEA in fetal bovine serum was achieved.

Abstract:An ultra-sensitive electrochemical immunosensor for detecting carcinoembryonic antigen was prepared. Graphene oxide-gold was used as the substrate modifier of the sandwich immunosensor. The high conductivity of graphene oxide effectively enhanced the electron transfer rate. The large specific surface area of graphene and the biocompatibility of gold nanoparticles increased the number of primary antibodies. The bovine serum albumin coated silver – platinum composite was used as a secondary antibody tag to prepare signal probes. The excellent biocompatibility of bovine serum albumin increased the number of secondary antibody connections. Silver nanoparticles increased the speed of electron transfer, and platinum further enhanced electrochemical sensing capabilities by catalyzing hydrogen peroxide. The immunosensor showed good performance for detecting carcinoembryonic antigen. Under the optimal conditions, the dynamic response range for carcinoembryonic antigen was from 0.005 ng·mL−1 to 100 ng·mL−1 with a detection limit of 0.76 pg·mL−1. The immunosensor also shown high selectivity, suitable stability, and good reproducibility, which provides new capabilities for clinical carcinoembryonic antigen detection.

Abstract:A simple and controllable one-step electrodeposition method for the preparation of a chitosan–carbon nanotubes–gold nanoparticles (CS–CNTs–GNPs) nanocomposite film was used to fabricate an immunosensor for detection of carcinoembryonic antigen (CEA). The porous three-dimensional CS–CNTs–GNPs nanocomposite film, which offered a large specific surface area for immobilization of antibodies, exhibited improved conductivity, high stability and good biocompatibility. The morphology of the formed nanocomposite film was investigated by scanning electron microscopy (SEM), and the electrochemical behaviors of the immunosensor were characterized by electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). Under the optimal conditions, the proposed immunosensor could detect CEA in two linear ranges from 0.1 to 2.0ngmL−1 and from 2.0 to 200.0ngmL−1, with a detection limit of 0.04ngmL−1. The immunosensor based on CS–CNTs–GNPs nanocomposite film as the antibody immobilization matrix could exhibit good sensitivity, stability, and reproducibility for the determination of CEA.

Abstract:The excellent direct electron transfer (DET) of enzyme labeled to antibody immobilized in designer organically modified silicate (ormosil) sol–gel was achieved at an electrode, which was used to construct a novel reagentless immunosensor for antigen determination. The synthesized ormosil architecture provided a hydrophilic interface for retaining the activity of immobilized enzyme labeled immunocomponent. The proposed immunosensor for carcinoembryonic antigen (CEA) prepared by immobilizing horseradish peroxidase-labeled CEA antibody (HRP-anti-CEA) in the architecture showed a surface-controlled electrode process attributed to the DET between electrode and HRP with a rate constant of 5.94 ± 0.40 s−1. The formation of immunocomplex upon incubation in CEA or sample solution led to block of DET and linearly decrease in voltammetric response over CEA concentration ranging from 0.5 to 3.0 and 3.0 to 120 ng ml−1. The limit of detection for CEA was 0.4 ng ml−1. The immunosensor showed good accuracy and acceptable storage stability, precision and reproducibility. The proposed method was simple, low-cost and potentially attractive for clinical immunoassays.

Abstract:A new signal amplification strategy employing nanocomposites composed of gold nanoparticles (GNPs) with DNA and methylene blue (MB) as labels has been developed to construct a novel immunosensor for the highly sensitive bioanalysis of carcinoembryonic antigen (CEA). To fabricate the label, DNA was first linked to GNPs through the formation of an Au–S bond. Next, the resulting bioconjugates were sequentially bound with MB via the guanine (G) bases in DNA specifically and then with secondary CEA antibody (Ab2) via the crosslinking of glutaraldehyde (GA). The receptor–analyte complex is formed by the biorecognition of the as-obtained MB labeled Ab2 with the primary CEA antibody (Ab1) immobilized on the surface of a modified GNP/Chitosan (Chits) composite electrode. After the receptor–analyte complex has formed, square wave voltammetry (SWV) was employed to oxidize the labeled MB in order to detect the CEA. This immunosensor shows a linear response from 0.10 to 2.0pgmL−1 CEA with an improved detection limit of 0.05pgmL−1 as compared to the conventional immunoassay. In addition, this new protocol shows acceptable stability, reproducibility and good recovery (97.5–110.0%) for CEA in human serum with great potential for clinical applications.

Abstract:In this paper, gold nanoparticle-thionine-reduced graphene oxide (GNP-THi-GR) nanocomposites were prepared to design a label-free immunosensor for the sensitive detection of carcinoembryonic antigen (CEA). The nanocomposites with good biocompatibility, excellent redox electrochemical activity and large surface area were coated onto the glassy carbon electrode (GCE) surface and then CEA antibody (anti-CEA) was immobilized on the electrode to construct the immunosensor. The morphologies and electrochemistry of the formed nanocomposites were investigated by using scanning electron microscopy (SEM), ultraviolet-visible (UV-vis) spectrometry, electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). CV and differential pulse voltammetry (DPV) studies demonstrated that the formation of antibody-antigen complexes decreased the peak current of THi in the GNP-THi-GR nanocomposites. The decreased currents were proportional to the CEA concentration in the range of 10-500 pg/mL with a detection limit of 4 pg/mL. The proposed method was simple, fast and inexpensive for the determination of CEA at very low levels.

Abstract:We present a novel label-free electrochemical immunosensor for detecting carcinoembryonic antigen (CEA) based on three-dimensional (3D) gold nanoparticles/prussian blue-poly(3,4-ethylenedioxythiophene) (AuNPs/PB-PEDOT) nanocomposite, which was firstly synthesized by a simple redox reaction between the PB precursors and EDOT in an aqueous solution, followed by the electrochemical reduction of HAuCl4. The 3D nanocomposite not only possessed large surface area and favorable microenvironment, but also exhibited remarkable conductivity, stability and excellent biocompatibility. In addition, PB showed excellent redox properties. Then AuNPs/PB-PEDOT was used as both electron mediators and 3D matrices in the fabrication of immunosensor. FT-IR spectra were employed to confirm the formation of PB, PEDOT and PB-PEDOT. Significantly, the AuNPs/PB-PEDOT exhibited a 3D and hierarchically porous nanostructure, while the PB-PEDOT showed core-shell structure. The AuNPs/PB-PEDOT modified immunosensor showed good linearity with the concentration of CEA ranging from 0.05 to 40ngmL−1, and the detection limit was 0.01ngmL−1. Moreover, the prepared electrode displayed good selectivity, high stability and good repeatability, and showed great potential for application in real sample analysis.

Abstract:This work designed an ultrasensitive non-enzymatic immunosensor for microcystin-LR (MC-LR) detection using carbon nanofibers (CNFs) as immobilization matrix and gold nanoparticles (AuNPs) as an electrochemical label. The carboxylic group functionalized CNFs could well disperse in water for at least three months, and CNFs modified electrode showed high conductivity and enhanced electron transfer. The immunosensor was constructed by immobilizing MC-LR antigen on CNFs modified electrode. With a competitive immunoassay format, the coated MC-LR antigen competed for MC-LR antibody with added target MC-LR to form antibody-antigen immunocomplex. Subsequently, the immunocomplex reacted with AuNPs labeled secondary antibodies, and the bound AuNPs were finally electro-oxidized in 0.1M HCl to produce AuCl4− for differential pulse voltammetric detection of MC-LR. Under optimal conditions, the immunosensor exhibited a wide linear response to MC-LR ranging from 0.0025 to 5μgL−1 with a detection limit of 1.68ngL−1 at a signal to noise of 3. This method showed good accuracy, acceptable precision and reproducibility. The assay results of MC-LR in polluted water were in a good agreement with the reference values. The proposed ultrasensitive immunoassay showed the promising application in environmental monitoring and public health.

Abstract:Nanomaterial-based signal-amplification strategies hold a great promise in realizing sensitive biological detection. A simple label-free electrochemical immunosensor for sensitive detection of carcinoembryonic antigen (CEA) was developed by immobilizing anti-CEA antibodies onto the Au-F127 strawberry-like nanospheres modified glassy carbon electrode (Au-F127/GCE). The Au-F127 strawberry-like nanospheres offered a large surface and multifunctional substrate for the effective immobilization of anti-CEA and the existence of Au could accelerate electron transfer and make the electrochemical signal amplified. The Au-F127 nanocomposites and anti-CEA were characterized by transmission electron microscopy (TEM), polycrystalline electron diffraction ring pattern, ultra-violet visible (UV-vis) spectra and attenuated total reflection-Fourier transform infrared (ATR-FTIR) spectra. Electrochemical impedance spectroscopy (EIS) and differential pulse voltammetry (DPV) were employed to verify the stepwise assembly of the immunosensor and evaluated the analytical performance of the fabricated immunosensor, respectively. The immunosensor showed a wide liner response range between 0.01 and 80 ng mL(-1) with a low detection limit of 0.24 pg mL(-1) at a signal-to-noise (S/N) ratio of 3. Additionally, the proposed method was successfully applied to determine CEA in human serum samples with satisfactory results.

Abstract:This study aimed to expand extant Alzheimer's disease (AD) research on cluster size and switching strategies in semantic verbal fluency (SVF). First, it addressed a significant shortcoming in research, that is, the use of a single semantic category (i.e., Animal). Second, it examined subjects of lower education levels, as research to date has primarily focussed on tertiary education. Subjects were 26 mild to moderate AD patients and 26 healthy elderly, 65 years old and over, with less than 12 years of schooling. The study examined the cluster size and switching variables, together with the number of subcategories, in two semantic categories (i.e., Animal and Supermarket). Furthermore, it investigated the error patterns (i.e., repetitions and categorical errors) across three semantic categories (i.e., Animal, Supermarket and Furniture). The findings provide preliminary support for category-specific effects on qualitative aspects of SVF highlighting the need to incorporate multiple semantic categories to research and clinical practice. This paper drew on preliminary analyses undertaken during the first author's PhD on ‘Language use in dementia and normal ageing', and the results were reported as an appendix body of the thesis. The PhD was funded by the Australian Postgraduate Award. Abstract:In this study, a sandwich-type electrochemical immunosensor was developed for detecting carcinoembryonic antigen (CEA) effectively. The proposed electrochemical immunosensor was based on gold nanocrystals (Au NDs) as the substrate material for capturing primary antibodies and using palladium nanoscale cubics (Pd NCs) loaded on amino-functionalized MoO 2 nanospheres (Pd NCs/NH 2 -MoO 2 NSs) as a secondary antibody label. Au NDs had dendritic structures that were more conducive to capturing Ab 1 and can accelerate electron transfer. MoO 2 had the good catalytic capacity for reduction of H 2 O 2 and favourable electrical conductivity. Similarly, Pd NCs had excellent catalytic performance for hydrogen peroxide reduction. Hence, the obtained Pd NCs/NH 2 -MoO 2 NSs were more effective than Pd NCs and NH 2 -MoO 2 NSs in catalytic reduction of hydrogen attribute to a synergistic effect, showing excellent catalysis. The proposed electrochemical immunosensor had a lower detection limit of 3.3 fg ml −1 jand a wider detection range from 10 fg ml −1 to 100 ng ml −1 (S/N = 3) for the detection of CEA under optimal experimental conditions. The proposed electrochemical immunosensor showed good sensitivity, stability, selectivity, reproducibility and its good detection performance of the immunosensor indicated that it had a broad application prospect in clinical detection.

Abstract:Sandwich-type electrochemical immunosensor was one of the main methods for detecting carcinoembryonic antigen (CEA). In this work, using Ce-MoF as the skeleton precursor, hyaluronic acid (HA) was coated on the surface of Ce-metal organic framework (Ce-MoF), which loaded with silver nanoparticles (Ag NPs) and horseradish peroxidase (HRP) to catalyze H2O2 and double amplified the current signal. Thus, a sensitive sandwich-type electrochemical immunosensor (Ce-MoF@ HA/Ag-HRP) was designed to detect carcinoembryonic antigen (CEA). The designed immunosensor used Au NPs to enhance the ability of attach more the first antibody (Ab1). This was due to Au NPs had good electrical conductivity and biocompatibility to accelerate electron transfer on the surface of the electrode. HA was riched in -COOH, -OH and had excellent biocompatibility, which can carry more Ag NPs to catalyze H2O2. Finally, the prepared sandwich-type electrochemical immunosensor had excellent biocompatibility and great catalytic performance. The immunosensor can be tested within 30 min and the logarithm of the current signal and CEA concentration showed a broad linear response range of 1 pg ml-1-80 ng ml-1, and the detection limit of CEA was 0.2 pg ml-1. More importantly, the proposed immunosensor had good reproducibility, selectivity, stability and without matrix effect. This confirmed that the proposed immunosensor had broad prospects in early clinical trials.

Abstract:Nanogold functionalized mesoporous carbon foam (Au/MCF) coupling with a signal amplification by C–Au synergistic silver enhancement was designed for sensitive electrochemical immunosensing of biomarker. The Au/MCF was prepared by in situ growth of nanogold on carboxylated MCF and used as a tracing tag to label signal antibody via the inherent interaction between protein and nanogold. The immunosensor was prepared by covalently immobilizing capture antibody on an electrochemically reduced graphene oxide/chitosan film modified glassy carbon electrode. Through a sandwich-type immunoreaction, Au/MCF tags were captured on the immunoconjugates to induce a silver deposition process. The electrochemical stripping signal of the deposited silver was used to monitor the immunoreaction. The Au/MCF-mediated silver enhancement along with the graphene-promoted electron transfer led to high detection sensitivity of carcinoembryonic antigen. Under optimal conditions, the proposed immunoassay method showed wide linear range from 0.05pgmL−1 to 1ngmL−1 and a detection limit down to 0.024pgmL−1. The newly designed amplification strategy holds great potential for ultrasensitive electrochemical biosensing of other analytes.

Abstract:Sixty-two patients with ninety injuries to the radial or the ulnar artery, or to both of these arteries, had arterial repairs using microvascular techniques in three medical centers. Twenty-six of the twenty-eight patients who had an ischemic hand secondary to injuries to both arteries and a successful operative repair had at least on patent vessel at follow-up. Of the other two patients, one had an amputation through the forearm for ischemia of the hand and the other had a viable hand supplied by collateral vessels, although both the radial and the ulnar arteries were thrombosed. The over-all success rate for all repairs was 54 per cent. After 47 per cent of the one-artery and 57 per cent of the two-artery lacerations, the vessels were patent at follow-up. Repairs of acute sharp transections of the radial artery within thirty-six hours of injury were most successful. Of the factors other than operative technique that strongly influence vessel patency after repair, back pressure in the distal arterial stump and the extent of the ischemia of the hand relative to its normal blood supply (a function of the completeness of the palmar arch) appear to be the most important.

Abstract:A new amperometric immunosensor for the determination of carcinoembryonic antigen (CEA) was constructed. First, the uniform nanomultilayer film was fabricated via layer-by-layer (LBL) assembly of positively charged carbon nanotubes wrapped by poly(diallyldimethylammonium chloride) and negatively charged poly(sodium-p-styrene-sulfonate), which could provide a high accessible surface area and a biocompatible microenvironment. Subsequently, gold nanoclusters were electrodeposited on the electrode to immobilize anti-CEA. The fabricated process and electrochemical behaviors of the immunosensor were characterized by cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS), and scanning electron microscopy (SEM). Under optimal conditions, the proposed immunosensor could detect CEA in two linear ranges from 0.1 to 2.0ngmL−1 and from 2.0 to 160.0ngmL−1, with a detection limit of 0.06ngmL−1.
Development of a high-performance label-free electrochemical immunosensor for early cancer diagnosis using anti-CEA/Ag-MOF/GO/GCE nanocomposite

Abstract:In order to detect carcinoembryonic antigen (CEA) as a tumor marker in lung cancer for early cancer diagnosis, this study aimed to develop a label-free electrochemical immunosensor based on the immobilization of an Anti-CEA antibody on a metal-organic framework (MOF)-graphene oxide nanocomposite modified glassy carbon electrode (Anti-CEA/Ag-MOF/GO/GCE). Ag-MOF/GO nanocomposite was prepared on the GCE surface using the ultrasonic irradiation method, and Anti-CEA antibody was subsequently immobilized on the surface. Analysis of the crystal structure and morphology of the modified electrode using FE-SEM and XRD revealed that the correct combination of GO nanosheets and Ag-MOF nanoparticles produced a high surface area to trap the antibodies. Electrochemical tests utilizing the CV and DPV methods revealed that the immunosensor's sensitivity, stability, and selectivity were improved by Anti-CEA/Ag-MOF/GO/GCE. Results showed that, with a detection limit of 0.005 ng/mL, the change in the reduction peak current was inversely correlated with the logarithm concentration of CEA in the range of 10-3 to 5000 ng/mL. The suggested CEA immunosensor's applicability in a human serum sample was investigated, and findings of analytical studies via standard addition technique for both ELISA and DPV assays revealed that significant agreement existed between the outcomes of the two assays. Additionally, the recoveries ranged from 99.00% to 99.25%, and all relative standard deviations (RSDs) for the sample detections were below 5.01%, indicating satisfactory accuracy in results measured with the proposed CEA immunosensor, indicating that the prepared CEA immunosensor in this study can be used in clinical applications and human fluids.
