Abstract:Analysts are always interested in finding new functional nanomaterials and devices with good properties for electrochemical sensor applications. In this paper, hyperbranched polyester nanoparticles with carboxylic acid functional groups (HBPE-CA NPs) were synthesized and combined with chitosan wrapped around Au nanoparticles (CS-Au NPs) to prepare a novel and sensitive electrochemical immunosensor by adsorption of carcinoembryonic antibody (anti-CEA) on the (HBPE-CA)/CS-Au NPs modified glass carbon electrode (GCE). Under the optimized conditions, the proposed immunosensor displayed a good amperometric response to carcinoembryonic antigen (CEA). Moreover, based on the antibiofouling properties, the immunosensor could be used for the direct detection of CEA in whole blood, and exhibited a wide detection range (1-10(7) fg mL(-1)), and a low detection limit of 0.251 fg mL(-1) (signal/noise = 3). Control experiments were also carried out by using ascorbic acid (AA), uric acid (UA), human immunoglobulin G (IgG), BSA and glucose in the absence of CEA. The good stability and repeatability of this immunosensor were also proven. Importantly, the results of the detection of clinical whole blood specimens with the proposed immunosensor showed good consistency with the data determined by enzyme-linked immunosorbent assay (ELISA) in serum samples. Furthermore, the developed immunosensor could provide a promising immunoassay strategy for clinical applications, since the values we measured in whole blood directly are likely closer to the real values.

Abstract:Sandwich-type electrochemical immunosensor was one of the main methods for detecting carcinoembryonic antigen (CEA). In this work, using Ce-MoF as the skeleton precursor, hyaluronic acid (HA) was coated on the surface of Ce-metal organic framework (Ce-MoF), which loaded with silver nanoparticles (Ag NPs) and horseradish peroxidase (HRP) to catalyze H2O2 and double amplified the current signal. Thus, a sensitive sandwich-type electrochemical immunosensor (Ce-MoF@ HA/Ag-HRP) was designed to detect carcinoembryonic antigen (CEA). The designed immunosensor used Au NPs to enhance the ability of attach more the first antibody (Ab1). This was due to Au NPs had good electrical conductivity and biocompatibility to accelerate electron transfer on the surface of the electrode. HA was riched in -COOH, -OH and had excellent biocompatibility, which can carry more Ag NPs to catalyze H2O2. Finally, the prepared sandwich-type electrochemical immunosensor had excellent biocompatibility and great catalytic performance. The immunosensor can be tested within 30 min and the logarithm of the current signal and CEA concentration showed a broad linear response range of 1 pg ml-1-80 ng ml-1, and the detection limit of CEA was 0.2 pg ml-1. More importantly, the proposed immunosensor had good reproducibility, selectivity, stability and without matrix effect. This confirmed that the proposed immunosensor had broad prospects in early clinical trials.

Abstract:Herein, a novel and sensitive sandwich-type electrochemical immunosensor was fabricated for quantitative detection of carcinoembryonic antigen (CEA). In order to construct the base of the immunosensor, the gold nanoparticles (Au NPs) were immobilized on the surface of bare glassy carbon electrode (GCE) through electrochemical reduction of HAuCl4 solution. The electrodeposited gold nanoparticles (D-Au NPs) not only effective improve immobilization of primary anti-CEA antibody (Ab(1)) but also accelerate the electron transfer on the electrode interface due to the high specific surface area, good biocompatibility and superior electrical conductivity. Moreover, the amino functionalized magnetic graphene loaded gold and silver core-shell nanoparticles to adsorb nickel ion (GS-Fe3O4/Au@Ag/Ni2+), which were used as a novel label to load the secondary anti-CEA antibody (Ab(2)). The resultant nanocomposites possess high specific surface area, excellent electrochemical property, good biocompatibility and superior auxiliary catalytic activity due to the synergetic effect. The signal amplification strategy, using the synergetic effect present in Au@Ag/Fe3O4-GS/Ni2+ to improve immobilization of Ab(2) and increase the reduction ability of the nanocomposites towards H2O2, improved the sensitivity of the immunosensor. Under the optimal conditions, a linear relationship between current signals and the concentrations of CEA was obtained in the range from 0.1 pg/mL to 100 ng/mL and the detection limit of CEA was 0.0697 pg/mL (signal-to-noise ratio of 3). Furthermore, the as-proposed immunosensor showed excellent performance in detection of human serum samples. The results suggest that the proposed immunosensor will be promising in the diagnostics application for accurately quantitative detection of CEA.

Abstract:A novel immunoassay was designed for ultrasensitive electrochemical detection of carcinoembryonic antigen (CEA) using the polyaniline-Au nanoparticles (PANi-Au NPs) as labels and a gap-based interdigitated electrode (IDEs) as sensor platform. In this strategy, the synthesized adamantine-modified primary antibodies (ADA-Ab(1)) were immobilized on the IDEs functionalized with carboxymethyl-beta-cyclodextrin (cm-beta-CD). After the immuno reaction between Ab(1) and CEA, the polyaniline-Au nanoparticles-labeled antibody (PANi-Au NPs-Ab(2)) as a signal tag was captured onto the immunosensor and connected the microgap of IDEs, which led to the resistance value decrease of IDEs greatly. In addition, the bottom of the immuno platform decorated with CD, Ab(1) and CEA was fully insulated and the electrochemical response only depended on the number of PANi-Au NPs-Ab(2) which was exactly proportional to CEA. The fabricated immunosensor detected CEA with a wide linear range from 0.1 ng/mL to 1000 ng/mL and low detection limit (0.007 ng/mL), and provides an enormous application potential for clinical analysis. Abstract:In this study, a sandwich-type electrochemical immunosensor was developed for detecting carcinoembryonic antigen (CEA) effectively. The proposed electrochemical immunosensor was based on gold nanocrystals (Au NDs) as the substrate material for capturing primary antibodies and using palladium nanoscale cubics (Pd NCs) loaded on amino-functionalized MoO 2 nanospheres (Pd NCs/NH 2 -MoO 2 NSs) as a secondary antibody label. Au NDs had dendritic structures that were more conducive to capturing Ab 1 and can accelerate electron transfer. MoO 2 had the good catalytic capacity for reduction of H 2 O 2 and favourable electrical conductivity. Similarly, Pd NCs had excellent catalytic performance for hydrogen peroxide reduction. Hence, the obtained Pd NCs/NH 2 -MoO 2 NSs were more effective than Pd NCs and NH 2 -MoO 2 NSs in catalytic reduction of hydrogen attribute to a synergistic effect, showing excellent catalysis. The proposed electrochemical immunosensor had a lower detection limit of 3.3 fg ml −1 jand a wider detection range from 10 fg ml −1 to 100 ng ml −1 (S/N = 3) for the detection of CEA under optimal experimental conditions. The proposed electrochemical immunosensor showed good sensitivity, stability, selectivity, reproducibility and its good detection performance of the immunosensor indicated that it had a broad application prospect in clinical detection.

Abstract:Using carcinoembryonic antigen (CEA) imprinted polymerized ionic liquid hydrogel as a recognition element and hollow gold nanoballs/MoSe2 nanosheets as a photoactive element, a photoelectrochemical (PEC) sensing platform was successfully fabricated. To accomplish the imprinted process under room temperature to maintain the biological activity and the configuration of CEA, and thus to improve the sensing performances, 3-{[{4-N,N-Bis[(carbamoyl)ethylmethacrylate]butyl}((carbamoyl)amino)ethyl methacrylate] -propyl}-1-ethenyl-1H-imidazol-3-ium bromide (BCCPEimBr) ionic liquid was synthesized. By using BCCPEimBr ionic liquid as the functional monomer, CEA as the template, a molecularly imprinted hydrogel film was prepared on a hollow gold nanoballs/MoSe2 nanosheets modified glassy carbon electrode surface. After removing the template, a CEA imprinted photoelectrochemical sensor was successfully fabricated. The imprinted PEC sensor shows good selectivity, sensitivity and stability towards CEA, produces a linear response in the concentration range from 0.05 to 5.0 ng mL(-1) and shows a detection limit of 11.2 pg mL(-1) (S/N = 3) under the optimized conditions. The imprinted PEC sensor was used to determine CEA in clinical human serum samples accurately. Abstract:In this study, an ultrasensitive and specific photoelectrochemical (PEC) immunosensor was designed for carcinoembryonic antigen (CEA) detection. Benefitting from the ascorbic acid (AA) used as an electron donor that led to a notable change in the photocurrent density, pillar[5]arene functionalized Au and Polyaniline-Bismuth oxybromide heterojunction (5/PANI-BiOBr) displayed superior PEC performance for CEA sensing. In detail, the localized surface plasmon resonance (LSPR) of the Au NPs and host-guest complexation between WP5 and AA increased the photocurrent signal, the photogenerated holes on BiOBr accelerated the oxidation of AA, and the fast electron transfer of the hollow PANI tubes benefited an increase in the photocurrent. The antibody effectively bound with CEA when bovine serum albumin blocked the residual sites on the /PANI-BiOBr electrode, generating a clear decrease in photocurrent density in AA solution. Under the optimum condition, the developed PEC immunosensor exhibited a sensitive response to CEA which was linear in the range of 0.01 ng·mL-1 to 50 ng·mL-1 with a detection limit of 3 pg·mL-1. The proposed PEC immunosensor displayed high specificity, good stability, and excellent reproducibility, paving a new way to construct sensitive and specific photoactive heterojunction materials for biomarker immunosensing.

Abstract:The convenient construction of carbon-based electrochemical immunosensors with high performance is highly desirable for the efficient detection of tumor biomarkers. In this work, an electrochemical immunosensor was fabricated by integrating a biofunctionalized mesoporous silica nanochannel film with a carbon-based electrode, which can enable the sensitive determination of carcinoembryonic antigen (CEA) in serum. The commonly used carbonaceous electrode, glassy carbon electrode (GCE), was employed as the supporting electrode and was pre-treated through electrochemical polarization to achieve the stable binding of a vertically ordered mesoporous silica film with amino groups (NH2-VMSF) without the use of any adhesive layer. To fabricate the immunorecognition interface, antibodies were covalently immobilized after the amino groups on the outer surface of NH2-VMSF was derivatized to aldehyde groups. The presence of amino sites within the high-density nanochannels of NH2-VMSF can facilitate the migration of negatively charged redox probes (Fe(CN)63-/4-) to the supporting electrode through electrostatic adsorption, leading to the generation of electrochemical signals. In the presence of CEA, the formation of immunocomplexes on the recognitive interface can reduce the electrochemical signal of Fe(CN)63-/4- on the supporting electrode. Based on this principle, the sensitive electrochemical detection of CEA was achieved. CEA can be determined to range from 0.01 ng mL−1 to 100 ng mL−1 with a limit of detection of 6.3 pg mL−1. The fabricated immunosensor exhibited high selectivity, and the detection of CEA in fetal bovine serum was achieved.

Abstract:In this work, a highly sensitive label-free paper-based electrochemical immunosensor employing screen-printed working electrode (SPWE) for detection of carcinoembryonic antigen (CEA) was fabricated. In order to raise the detection sensitivity and immobilize anti-CEA, amino functional graphene (NH2-G)/thionine (Thi)/gold nanoparticles (AuNPs) nanocomposites were synthesized and coated on SPWE. The principle of the immunosensor determination was based on the fact that the decreased response currents of Thi were proportional to the concentrations of corresponding antigens due to the formation of antibody–antigen immunocomplex. Experimental results revealed that the immunoassay enabled the determination of standard CEA solutions with linear working ranges of 50pgmL−1 to 500ngmL−1, the limit of detections for CEA is 10pgmL−1 (S/N=3) and its corresponding correlation coefficients were 0.996. Furthermore, the proposed immunosensor could be used for the determination of clinical serum samples. A large number of clinical serum samples were detected and the relative errors between measured values and reference concentrations were calculated. Results showed that this novel paper-based electrochemical immunosensor could provide a new platform for low cost, sensitive, specific, and point-of-care diagnosis in cancer detection.
Label-free Photoelectrochemical Immunosensing Platform for Detection of Carcinoembryonic Antigen Through Photoactive Conducting Poly(5-Formylindole) Nanocomposite.

Abstract:Poly(5-formylindole)/electrochemically reduced graphene oxide (P5FIn/erGO) nanocomposite is firstly used to construct a label-free photoelectrochemical (PEC) immunosensor to detect carcinoembryonic antigen (CEA). As photoactive material and electroactive mediator, the prepared P5FIn/erGO nanocomposite exhibits high photocurrent intensity under visible-light irradiation due to the synergistic effect of P5FIn and erGO. The anti-CEA is connected to the P5FIn/erGO modified electrode surface, and gold nanoparticles (AuNP) is used as cross-linking in the process. The linear decrease of photocurrent is caused by the specific recognition of anti-CEA and CEA. This PEC immunosensor shows a wide linear response to CEA ranging from 0.0005 to 50 ng ml(-1) with a low detection limit of 0.14 pg mL(-1). The proposed immunosensor has good stability, reproducibility and high specificity. The satisfied results are also obtained when this immunosensor is used to detect CEA in actual human serum samples analysis, thus opening up a new promising PEC analysis platform based on conducting polymers.

Abstract:Fabrication of functional electrochemical biosensor is a hot topic; however, precise and sensitive cancer detection in early clinical diagnosis is still a great challenge. Continuous efforts have been devoted to explore functional materials for this issue. In this work, we developed a dual binding sites and dual signal-amplifying electrochemical aptasensor of self-polymerized dopamine-decorated Au and coordinated with Fe-MOF (Au@PDA@Fe-MOF) for the detection of carcinoembryonic antigen (CEA). Remarkably, Au@PDA@Fe-MOF features high sensitivity, multiple active sites, good biocompatibility, and excellent selectivity, which is attributed to abundant -COOH in porous Fe-MOF and unsaturated Fe3+ sites on the surface of Fe-MOF as the active binding sites grafting more NH2-functionalized CEA-specific aptamer and redox PDA and Fe-MOF accelerating the movement of electrons for dual signal amplifying. Meanwhile, the electrochemical aptasensor shows favorable repeatability with 1.82% relative standard deviation (RSD) under five independent aptasensors and strong stability with only 3.3% degradation after 12 days of storage. In addition, the aptasensor has wide CEA detection range from 1 fg mL-1 to 1 μg mL-1 with a low detection limit of 0.33 fg mL-1 (S/N = 3). Furthermore, the aptasensor is feasible for accurate and quantitative detection of CEA in serum samples with RSD below 2.32%. The satisfying results demonstrate promising applications of the CEA aptasensor in practical sample analysis and lay an important foundation for other biomarker detection in early clinical diagnosis.

Abstract:In this study, due to the oxidative and strong adhesion characteristics of mussel-inspired polydopamine, the aimed antibody-anti-CEA could be directly attached to the polydopamine-coated glassy carbon electrode surface. The immobilization method and sandwich immunoreactions strategy were applied to form a facile and effective electrochemical immunosensor. Anti-CEA was verified by attenuated total reflection Fourier-transform infrared spectroscopy (ATR-FTIR), X-ray photoelectron spectroscopy (XPS), and atomic force microscopy (AFM). Under optimum conditions, a linear response range of CEA from 0.01 to 80 ng/mL with a low detection limit of 1.58 pg/mL was obtained by the differential pulse voltammetry determination. Assay results of clinical serum samples using the standard ELISA and reference methods gave an excellent correlation and dependence. Moreover, the preparation of immunosensor with low cost, convenient operation, and all steps processing at room temperature which possesses great potentials for practical applications.

Abstract:Carcinoembryonic antigen (CEA) is a well-known cancer biomarker for the detection of several malignancies. The development of ultrasensitive CEA diagnostic tools is crucial for early detection and progression observation of tumors. Herein, a dual signal amplified sandwich-type electrochemical immunoassay was developed based on dual-labeled mesoporous silica nanospheres as a signal amplifier, combined with NiO@Au decorated graphene as a conductive layer for ultrasensitive and rapid determination of CEA. The dual-labeled mesoporous silica (DLMS) nanosphere, which was synthesized by entrapping Au nanorod (Au NR) and horseradish peroxidase (HRP) in the channels of amine-functionalized SBA-15 followed by subordinate antibody (Ab2) conjugation which was denoted as Au NR@SBA-15/Ab2-HRP. The dual signal amplification from Au NR@SBA-15 and HRP enhanced the sensitivity of the proposed immunoassay. Consequently, the developed DLMS based immunosensor displayed ultra-low limits of detection of 5.25 fg/mL and a wide range of linearity (0.1-5 pg/mL), which was extended for CEA determination in real-time samples with improved recoveries of >98%. Therefore, this dual amplification prototype would cater to the clinical requirements for the ultrasensitive detection of CEA biomarkers.

Abstract:Carcinoembryonic Antigen (CEA), an acidic glycoprotein with human embryonic antigen properties, is found on the surface of cancer cells that have differentiated from endodermal cells. This paper presents a label-free electrochemical immunoassay for the dual amplification detection of CEA using gold nanoparticles loaded with polypyrrole polydopamine (Au/PPy-PDA) and polymerized polycaprolactone (Ng-PCL) prepared by ring-opening polymerization (ROP). First, the composite Au/PPy-PDA was adhered to the electrode surface. Then, gold nanoparticles form a Au-S bond with the sulfhydryl group in Apt1 to secure it on the electrode surface. Subsequently, the non-specific binding sites on the electrodes surface are closed by bovine serum albumin (BSA). Next, CEA is dropped onto the electrode surface, which is immobilized by antigen-antibody specific recognition, and the carboxyl-functionalized Apt2 forms a "sandwich structure" of antibody-antigen-antibody by specific recognition. Polymeric Ng-PCL is adhered to the electrode surface, leading to an increase in the electrochemical impedance signal, resulting in a complete chain of signal analysis. Finally, the response signal is detected by electrochemical impedance spectroscopy (EIS). Under optimal experimental conditions, the method has the advantages of high sensitivity and wide linear range (1 pg mL-1∼100 ng mL-1), and the lower limit of detection (LOD) is 0.234 pg mL-1. And it has the same high sensitivity, selectivity and interference resistance for the real samples detection. Thus, it provides a new way of thinking about biomedical and clinical diagnosis.

Abstract:Biosensors based on organic field-effect transistors(OFETs) are one of the most promising electronic devices for emerging bioanalytical applications. The selection of organic semiconductors(OSCs) is essential to improve the sensitivity and reliability of this kind of biosensors. Given the good field effect performance and tunable structures of D-A type conjugated polymers, here, we design two D-A type copolymers[P(BDT- co -DPP2T-ThC2) and P(BDT- co -DPP2T-Th)], which are applied as the OSC layers. With carcinoembryonic antigen antibody(anti-CEA) adsorbed onto the OSC layers as the recognition sites, OFETs based biosensors for CEA detection are developed. The experimental findings support that the attachment of ester side groups onto the polymer backbone[as for P(BDT- co -DPP2T-ThC2)] is favorable for improved solubility and filming properties of the polymer. The introduction of ester side groups affects molecular stacking and enhances intermolecular forces. The resultant devices show high charge mobility and antibody adsorption ability, both of which are critical for sensitive and facile detection of CEA biomarkers. The reliable determination of CEA down to the picomolar level is determined. It is expected that this kind of biosensors fabricated by D-A type conducting polymers will open new avenues toward the early diagnosis, real-time monitoring and treatment of future cancer diseases.

Abstract:An electrochemical immunosensor for quantitation of carcinoembryonic antigen (CEA) based on Au-Ag/rGO@PDA nanocomposites for dual signal amplification strategy was designed. Polydopamine-coated Ag/graphene hybrids were synthesized by one-step reduction and functionalization, and the dopamine was introduced to replace some reducing agents that are harmful to organisms. Silver nanoparticles modified on the electrodes produced sensitive electrochemical redox peaks. When different amounts of protein (antigen, antibody, bovine serum protein, etc.) were adsorbed on the surface of nano-silver, the surface of nano-silver was covered. The electrochemical redox peak of silver particles decreased due to the change of the effective area of bare silver particles which could react with electrodes. In addition, CEA-antibody (anti-CEA) was immobilized into a biocompatible nanocomposite of Au nanoparticles as a conductive matrix, and CEA-antibody (anti-CEA) was combined with Ag/rGO@PDA to form a dual signal amplifying element. Under optimal condition, the linear relationship was obtained from 0.001 ng mL−1 to 80 ng mL−1 between current signal and CEA concentration with a detection limit of 0.286 pg mL−1 (signal-to-noise ratio is 3). The label-free electrochemical immunosensor has been successfully applied to human serum detection due to good selectivity, stability and reproducibility.

Abstract:Herein, a method was developed for the sensitive monitoring of carcinoembryonic antigen (CEA) by gold nanoparticles dotted prussian blue@polyaniline core-shell nanocubes (Au NPs/PB@PANI). First, a facile low-temperature method was used to prepare the uniform PB@PANI core-shell nanocubes with the assistance of PVP, where PB acted as the electron transfer mediator to provide electrochemical signals, and the PANI with excellent conductivity and desirable chemical stability not only played the role of a protective layer to prevent etching of PB in basic media but also effectively improved electron transfer. Importantly, to further enhance the electrical conductivity and biocompatibility of PB@PANI and to further enhance the electrochemical signal and capture a large amount of Ab2, Au NPs were doped on the surface of PB@PANI to form Au NPs/PB@PANI nanocomposites. Subsequently, benefiting from the advantages of core-shell structure nanoprobes and gold-platinum bimetallic nanoflower (AuPt NF), a sandwich-type electrochemical immunosensor for CEA detection was constructed, which provided a wide linear detection range from 1.0 pg·mL-1 to 100.0 ng·mL-1 and a low detection limit of 0.35 pg·mL-1 via DPV (at 3σ). Moreover, it displayed a satisfactory result when the core-shell structure nanoprobe-based immunosensor was applied to determine CEA in real human serum samples.

Abstract:AbstractA novel carbon encapsulated Fe3O4 nanoparticles embedded in two-dimensional (2D) porous graphitic carbon nanocomposites (Fe3O4@C@PGC nanocomposites) were synthesized by situ synthesis strategy, which provided a sensor platform owing to a large aspect ratio and porous structure. Polydopamine (PDA) were modified on the surface of Fe3O4@C@PGC nanocomposites through self-polymerization of dopamine, acting as both the reductant and template for one-step synthesis of gold nanoparticles. The prepared Au/PDA/Fe3O4@C@PGC nanocomposites show ferromagnetic features, extremely excellent electron transfer, large specific surface area and excellent dispersing property. These are conducive to the electrochemical signal output and the immobilization of antibody. In this work, a highly label-free sensitive magnetic immunosensor was developed based on Au/PDA/Fe3O4@C@PGC nanocomposites for the detection of carcino-embryonic antigen (CEA). The magnetic glassy carbon electrode was used to fix the Au/PDA/Fe3O4@C@PGC nanocomposites with the help of magnetic force. Under the optimal conditions, the immunosensor exhibited a wide linear range (0.001 ng/mL–20.0 ng/mL), a low detection limit (0.33 pg/mL), good reproducibility, selectivity and acceptable stability. The proposed sensing strategy may provide a potential application in the detection of other cancer biomarkers.

Abstract:Highly sensitive and selective method of determination for carcinoembryonic antigen (CEA) is of important significance for the effective early diagnosis and treatment of cancer. In this work, we report an ultrasensitive sandwich photoelectrochemical (PEC) immunosensor for CEA detection. To be precise, cobalt-borate nanosheet arrays (Co-Bi NSs) with the larger specific surface area are prepared by electrodeposition and used as the basal material to capture the primary antibody (Ab1) for the first time. And the other photoactive material Mn2O3@Au nanocube is exploited for labeling the secondary antibody (Ab2). The coulomb force of Mn2O3@Au nanocube and Co-Bi NSs photoelectrodes as the driving force effectively improves the photoelectric response. Under the light irradiation, charge separation occurs at the same time between AuNPs and Mn2O3, and the energy levels of Mn2O3@Au and Co-Bi NSs are effectively matched, achieving the high PEC performance of the biosensor. The synthesized nanomaterials are physically characterized by a series of characterization techniques. Besides, ascorbic acid (AA) is employed as an excellent electron donor, which can inhibit the recombination process of photogenerated electrons and holes, then obtaining enhanced and stable photocurrent signals. Under the optimal experimental conditions, the immunosensor shows a lower detection limit of 5.0 pg mL−1 (S/N = 3), a wider linear range of 0.1 ng mL−1 ∼ 1000 ng mL−1, as well as excellent specificity, stability and reproducibility. Additionally, as-constructed PEC immunosensor can be applied to detect CEA in human serum, this proposed strategy may afford a promising approach for the biomarker sensing and clinical applications.
