Abstract:The Catellani reaction has received substantial attention because it enables rapid multiple derivatization on aromatics. While using alkyl electrophiles to achieveortho-alkylation was one of the earliest applications of the Catellani reaction,ipso-alkylation-terminated reactions with beta-H-containing reactants has not been realized to date. Herein, we report alkylation-terminated Catellani reaction using alkyl carbagermatranes (abbreviated as alkyl-Ge) as nucleophiles. The reactivity of alkyl-Geand alkyl-B(OH)(2)in this reaction is discussed. This approach enables efficient dialkylation with beta-H-containing reactants, which was previously inaccessible by Catellani reactions.

Abstract:Synthesis of seven-membered carbocycle-fused aromatics was realized by Catellani reaction using terminally brominated alkyl carbagermatranes through intermolecular cyclization manner. Various functional groups were well tolerated, and this transformation was also expanded to the synthesis of carbocycles of other size. The utility of this method was demonstrated by modification of natural product derivatives and synthesis of bioactive molecules.

Abstract:Pd-catalyzed cross-coupling reactions have achieved tremendous accomplishments in the past decades. However, C(sp3)-hybridized nucleophiles generally remain as challenging coupling partners due to their sluggish transmetalation compared to the C(sp2)-hybridized counterparts. While a single-electron-transfer-based strategy using C(sp3)-hybridized nucleophiles had made significant progress recently, fewer breakthroughs have been made concerning the traditional two-electron mechanism involving C(sp3)-hybridized nucleophiles. In this report, we present a series of unique alkyl carbagermatranes that were proven to be highly reactive in cross-coupling reactions with our newly developed electron-deficient phosphine ligands. Generally, secondary alkyl carbagermatranes show slightly lower, yet comparable activity to its Sn analogue. Meanwhile, primary alkyl carbagermatranes exhibit high activity, and they were also proved stable enough to be compatible with various reactions. Chiral secondary benzyl carbagermatrane gave the coupling product under base/additive-free conditions with its configuration fully inversed, suggesting that transmetalation was carried out in an "SE2(open) Inv" pathway, which is consistent with Hiyama's previous observation. Notably, the cross-coupling of primary alkyl carbagermatranes could be performed under base/additive-free conditions with excellent functional group tolerance and therefore may have potentially important applications such as stapled peptide synthesis.

Abstract:Addition of interelement compounds across alkynes is a straightforward strategy for the synthesis of densely functionalized alkenes, which are versatile units in numerous biologically active compounds. Of these processes the bis-silylation of alkynes is of particular interest, due to the synthetic use of the alkenyl silane reaction products. While cis -bis-silylation of alkynes is well known, trans -bis-silyation of alkynes is relatively underdeveloped. Here, we report the intermolecular trans -bis-silylation of terminal alkynes, using Pd-catalysis and disilane reagent 8-(2-substituted-1,1,2,2-tetramethyldisilanyl)quinoline (TMDQ), to selectively form trans -bis-silylated alkenes. The reaction process was found to be compatible with aryl and alkyl alkynes as well as those bearing electron-withdrawing groups and other alkenyl or alkynyl functionalities. The use of the reaction was displayed through late-stage functionalization of terminal alkynes bearing natural product or drug motifs and onward synthetic transformations of the trans -bis-silylated alkenes reaction products. Experimental and computational mechanistic studies reveal that the reaction probably proceeds through a combined cis -bis-silylation and Z / E isomerization process and that the use of TMDQ as the limiting reagent is key to the desired reactivity.

Abstract:Here, we reported a cobalt-hydride-catalyzed Markovnikov-type hydroallylation of terminal alkynes with allylic electrophile to access valuable and branched skipped dienes (1,4-dienes) with good regioselectivity. This operationally simple protocol exhibits excellent functional group tolerance and exceptional substrate scope. The reactions could be carried out in gram-scale with TON (turn over number) up to 1160, and the products could be easily derivatized. The preliminary mechanism of electrophilic allylation of α-selective cobalt alkenyl intermediate was proposed based on deuterium labeling experiment and kinetic studies.

Abstract:Transition metal-catalyzed hydrostannylation of alkynes is an atom-economic way towards valuable α-vinylstannanes. However, directing-group-free examples for the construction of α-adducts are rare and mostly undergo the M-Sn species insertion process. Here, we report an efficient cobalt-catalyzed highly α-selective hydrostannylation of terminal alkynes via M-H species, using readily available Bu3SnH and easily accessible as well as bench-stable ligand. The robust catalytic system, bearing good functional group tolerance and operational simplicity, provides the α-vinylstannanes in moderate to excellent yields with ⩾97:3 rr averagely. The efficiency of the reaction is investigated with turnover number (TON) up to 10,000 and turnover frequency (TOF) up to 144,000 h−1, respectively. The synthetic utility of the α-vinylstannanes is proved by further derivatizations towards the 1,1-disubstituted alkenes via Kosugi-Migita-Stille cross-coupling, conjugated alkene, α-vinyliodine, α-vinylazide, and allyl alcohol. Several control experiments, deuterium-labeling experiment and density functional theory (DFT) calculations are conducted to reveal the preliminary mechanism.

Abstract:A highly regioselective Cu(I)-catalyzed anti-carbometallation of secondary terminal propargylic alcohols with 1 degrees alkyl or aryl Grignard reagents affording 2-substituted allylic alcohols was developed. By using this method, optically active allylic alcohols can be prepared from the optically active propargylic alcohols without obvious loss of the enantiopurity. The cyclic organometallic intermediate formed may undergo an iodination or a Pd(0)-catalyzed coupling reaction to afford stereo-defined allylic alcohols.

Abstract:We reported a Pd/NBE cooperative catalyzed ortho C−H methylation and trideuteromethylation of arylthianthrenium salts, enabling the efficient synthesis of a wide variety of (trideutero)methylated arenes in moderate to good yields. The method demonstrates excel-lent tolerance towards functional groups, scalability, and potential extension to the late-stage functionalization of biorelevant mole-cules. In combined with C−H thianthrenation of arenes, this approach provides an effective method for the site-selective C−H (trideutero)methylation of arenes. Additionally, this reaction represents the first example of a Catellani reaction involving aryl sul-fonium salts.

Abstract:In the past few decades, palladium-catalyzed cross-coupling reactions have taken root in the construction of a complex synthetic community. The development of organometallics has been an important objective in this field. Our group has focused on exploiting new germanium-based reagents and the corresponding catalytic processes. In the past three years, we have established new methods for the synthesis of structure-modified (hetero)aryl germatranes and alkyl carbagermatranes. Particularly for alkyl carbagermatranes, the stability to be compatible with various derivatization reactions and the high activity for transmetallation (e.g. base/additive-free for primary alkyl carbagermatranes) distinguish them from many reported nucleophiles. In this article, we would introduce (1) the development process of organogermanium reagents in palladium-catalyzed cross-couplings; (2) the history of germatrane-type systems and the breakthrough we have made in the field; (3) the outlook for (carba)germatranes and alkyl-GeMe3.

Abstract:The N-heterocyclic carbene (NHC)-catalyzed enantioselective Petasis-like alkenylation of o-hydroxycinnamaldehydes or hydroxyl-tethered alpha,beta-unsaturated aldehydes with styryl, dienyl, or trienyl boronic acids is disclosed. This method involves the addition of pi-system-containing boronic acids to NHC-bounded alpha,beta-unsaturated acyl azoliums and allows access to divergent assembly of beta-alkenyl substituted dihydrocoumarin and gamma- and delta-lactones. DFT calculations suggest that an unprecedented zwitterionic intermediate and 1,4- or 1,5- migration of alkenyl groups play a crucial role in the reaction. More in-depth studies of orbital and noncovalent interaction analysis provide more detailed explanations for pathways and stereoselectivity control.

Abstract:A palladium(II)-catalyzed 1,2-dicarbofunctionalization reaction of unactivated alkenes has been developed, wherein a cleavable bidentate directing group is used to control the regioselectivity and stabilize the putative alkylpalladium(II) intermediate. Under the optimized reaction conditions, a broad range of nucleophiles and electrophiles were found to participate in this transformation, providing moderate to high yields. 3-Butenoic acid derivatives containing internal alkenes and α-substituents were reactive substrates, offering a powerful platform for preparing β,γ-substituted carbonyl compounds with multiple stereocenters.

Abstract:Cycloaddition and annulation strategies are among the most powerful methods for creating molecular complexity in organic molecules. In this manuscript, we report a highly site-selective palladium-catalyzed decarbonylative [4+2] cyclization of carboxylic acids with terminal alkynes by a sequential C–C/C–H bond activation. Most notably, this method represents the first use of carboxylic acids as the ubiquitous and underdeveloped synthons for intramolecular cycloadditions by decarbonylative C–C bond cleavage. The method provides a solution to the long-standing challenge of the regioselective synthesis of substituted naphthalenes by cycloaddition. Mechanistic studies show that this reaction occurs through a sequential process involving the formation of key palladacycle by a sequential C–C/C–H bond activation and highly regioselective alkyne insertion enabled by cluster catalysis. Wide substrate scope for both carboxylic acids and terminal alkynes is demonstrated with high functional group tolerance. Moreover, this reaction is scalable and applicable to the synthesis of functionalized molecules featuring bioactive fragments. This reaction advances the toolbox of redox-neutral carboxylic acid interconversion to cycloaddition processes. We anticipate that this approach will find broad application in organic synthesis, drug discovery and functionalized material research fields.

Abstract:Four novel N-heterocyclic carbene (NHC) silver complexes, , have been synthesized and characterized. The single X-ray crystal diffraction data indicate a dinuclear solid-state structure for and and a mononuclear structure for and . These complexes have been successfully used as efficient catalysts for the C-H activating carboxylation of terminal alkynes with CO2. A wide range of substrates with various functional groups afforded the corresponding aryl or alkyl substituted propiolic acids in good yields under mild conditions. Moreover, the role of bases and the reaction mechanism is thoroughly discussed.

Abstract:Controlling the regioselectivity for the alkoxycarbonylation of unsymmetric internal alkynes is challenging. Herein, a palladium-catalyzed ligand-controlled regiodivergent alkoxycarbonylation of internal trifluoromethylthiolated alkynes was achieved. A series of α- or β-SCF3 acrylates from the same trifluoromethylthiolated alkyne were obtained with moderate to high yield and regioselectivity.

Abstract:One of the central goals of synthetic chemistry is to develop efficient methods for constructing heterocyclic architectures, given their broad distribution in a wide range of natural compounds, pharmaceuticals, agrochemicals, and materials. As a result, methods that allow for the modular and diverse synthesis of heterocyclic compounds via one single approach are in high demand. Here, we report a novel strategy for the preparation of diverse heterocycles via a Pd-catalysed migratory 1,1-cycloannulation reaction (MCAR) of alkenes. Starting from readily available alkenyl amines and alkenyl alcohols, this approach allows the formation of a wide range of heterocycles, including five to seven-membered azaheterocycles and oxaheterocycles with high efficiency and good functional group tolerance. The key to the realisation of this reaction is the use of 4-iodophenol or 2-iodophenol derivatives, where the phenolic hydroxyl group plays a critical role in controlling the direction of migration and the ring-size of the heterocycles through the formation of a quinone methide intermediate. The utility of this strategy in synthetic chemistry and medicinal chemistry were preliminarily demonstrated by the late-stage introduction of heterocycle scaffolds into complex drug molecules, and the efficient preparation of serval bioactive compounds.

Abstract:Rh(acac)(CO)(2) catalyzes addition of (N,N-diisopropylcarbamoyl)trimethylstannane to terminal alkynes to give (Z)-beta-stannyl-alpha,beta-unsaturated amides with terminal attachment of the amide group, while the use of Ni(COd)(2) results in the reversal of the regioselectivity.
