Abstract:Microbial transformation was used to prepare novel cytotoxic bufadienolides. Twelve products (3–14) were obtained from bufalin (1) by the fungus Mucor spinosus. Their structures were elucidated by high-resolution mass spectroscopy (HR-MS) and extensive NMR techniques, including 1H NMR, 13C NMR, DEPT, 1H–1H correlation spectroscopy (COSY), two dimensional nuclear Overhauser effect correlation spectroscopy (NOESY), heteronuclear multiple quantum coherence (HMQC), and heteronuclear multiple bond coherence (HMBC). Compounds 3, 4, 9 and 11–14 are new mono- or dihydroxylated derivatives of bufalin with novel oxyfunctionalities at C-1β, C-7β, C-11β, C-12β and C-16α positions. The in vitro cytotoxic activities against human cancer cell lines of 3–14, together with 16 biotransformed products derived from cinobufagin (15–30) were determined by the MTT method, and their structure–activity relationships (SAR) were discussed.

Abstract:The biotransformations of cinobufagin (1), an animal-originated bufadienolide, by cell suspension cultures of Catharanthus roseus and Platycodon grandiflorum were investigated. Incubation for 6 days of 1 with C. roseus yielded four products, desacetylcinobufotalin (2), 3-epi-desacetylcinobufagin (3), 1β-hydroxyl desacetylcinobufagin (4) and 3-epi-desacetylcinobufotalin (5), among which 4 is a new compound. Time course investigation revealed that the biotransformation rates of two major products, 2 and 3, reached their highest levels of 44.7 and 16.5%, respectively, on the third day after substrate administration. Compounds 2–5 showed more potent cytotoxic activities against HL-60 cell lines than the parent compound 1. A plausible biotransformation pathway is proposed to account for the formation of the observed products. From the culture supernatant of P. grandiflorum, 2 was isolated in 37.8% yield after 8 days of incubation with 1. Cinobufotalin (6) and desacetylcinobufagin (7) were also obtained as minor products. The two plant suspension cultures exhibited similar transformation patterns on compound 1.

Abstract:The biotransformation of three bioactive bufadienolides, namely, bufotalin (1), telocinobufagin (2), and gamabufotalin (3) by cell suspension cultures of Saussurea involucrata yielded 11 products. Bufotalin yielded 3-epi-bufotalin (1a), 3-epi-desacetylbufotalin (1b), 3-epi-bufotalin 3-O-β-d-glucoside (1c), 1β-hydroxybufotalin (1d), and 5β-hydroxybufotalin (1e); telocinobufagin yielded 3-dehydroscillarenin (2a), 3-dehydrobufalin (2b), and 3-epi-telocinobufagin (2c); and gamabufotalin yielded 3-epi-gamabufotalin (3a), 3-dehydrogamabufotalin (3b), and 3-dehydro-Δ1-gamabufotalin (3c), respectively. Among these 11 products, 1a, 1b, 1c, 1d, 3a and 3c are previously unreported. The structures of these metabolites were elucidated based on NMR spectroscopic analyses and mass spectrometry. Most metabolites showed significant cytotoxic activities against human hepatoma (HepG2) and breast cancer (MCF-7) cell lines. In addition, the time course for the biotransformation of 3 was investigated.

Abstract:The microbial transformation of a cytotoxic bufadienolide, bufalin (1), was carried out using two strains of filamentous fungi. Cunninghamella blakesleana catalyzed the specific 12alpha-hydroxylation of bufalin and produced 12alpha-hydroxybufalin (2) and 7beta,12alpha-dihydroxybufalin (3) as the major metabolites, together with 7beta-hydroxybufalin (4) and 12beta-hydroxybufalin (5) in low yields. Two minor products were isolated from the culture broth of Mucor spinosus and were identified as 7beta,15alpha-dihydroxybufalin (6) and 5beta,7beta-dihydroxybufalin (7), respectively. Metabolites 2, 3, 6, and 7 are new compounds, and their structures were fully characterized by NMR and MS spectroscopy.

Abstract:The biotransformation of arenobufagin (1) and cinobufotalin (2), isolated from the natural medicine Chan Su, by Alternaria alternata AS 3.4578 was carried out. Incubation of 1 and 2 afforded six metabolites: 3-oxo-arenobufagin (1a), psi-bufarenogin (1b), 3-oxo-psi-bufarenogin (1c), 3-oxo-Delta(4)-derivative of cinobufotalin (2a), 3-oxo-cinobufotalin (2b) and 12 beta-hydroxycinobufotalin (2c). Among them, metabolites 1a, 1c and 2c are new compounds and their structures were characterized on the basis of their spectroscopic data (NMR, MS and IR). Compounds 1, 2, 1b, 2a and 2b were evaluated for their cytotoxicity against HepG2 and MCF-7 human cancer cells, and all of them showed significant inhibitory activities.

Abstract:Resibufogenin is a cytotoxic steroid isolated from the Chinese drug ChanSu. The biotransformation of resibufogenin by Mucor polymorphosporus afforded 22 products, and 15 of them were new. The transformation reactions involved hydroxylations at C-1β, C-5, C-7α, C-7β, C-12α, C-12β and C-16α, as well as epimerization or dehydrogenation of 3-OH. Hydroxylations at C-12α, C-12β and C-16α were the major reactions, each giving products in >5% yields, whereas the other products were obtained in fairly low yields. Some of the products showed decreased but still potent cytotoxicities. This investigation provided a useful approach to prepare new bufadienolides and most of them were difficult to obtain by chemical means.

Abstract:Four biotransformed products of resibufogenin were obtained after 4 days of incubation with cell suspension cultures of Platycodon grandiflorum. On the basis of chemical and spectral data, their structures were elucidated as 3-epi-resibufogenin. marinobufagin, 1beta-hydroxy-resibufogenin, and 3-epi-marinobufagin respectively, among which the latter two compounds are new. These compounds were found to exhibit potent cylotoxic activities against Bel-7402. BGC-823. HeLa, and HL-60 human cancer cell lines.

Abstract:Resibufogenin, cinobufagin, and bufalin are cytotoxic steroids isolated from the Chinese drug Chan'su. Biotransformation of these three bufadienolides by Nocardia sp. NRRL 5646 was investigated. Notably, resibufogenin was converted to 3-acetyl 15 beta-hydroxyl bufotalin, via an unprecedented 14 beta,15 beta-epoxy ring cleavage and a regio-selective acetoxylation. This product showed significantly increased cytotoxic activity. The regio-selective acetylation of the 3-OH was also involved in the other reactions. The structures of metabolites were established by ESI-LC/MS and 2D NMR techniques. The in vitro cytotoxic activities against human cancer cell lines of the substrates and the transformed products were determined by the MTT method and their structure-activity relationship (SAR) was discussed. Abstract:Twenty strains of filamentous fungi (from nine genera) and four species of bacteria were screened for their capabilities to transform bufadienolides. The highly stereo- and regiospecific isomerization and dehydrogenation of three major bufadienolides (resibufogenin, cinobufagin and bufalin) at 3-OH by Fusarium solani AS 3.1829 were reported. Three major transformed products were obtained and their structures were identified as 3-epi-resibufogenin (1a), 3-epi-bufalin (2a), 3-keto-cinobufagin (3a), respectively, on the basis of their spectral data. The reaction kinetics and the effects of pH, substrate concentration and incubation phase on the biotransformation were also investigated.

Abstract:Bufadienolide is a kind of natural steroids with significant anti-cancer activities. Hitherto, the number of structural modification on the bufadienolides has reached more than 100 . The purifica-tion of sulfated,a trace cytotoxic component bufadienolides from toad skin is a tough work due to its low content and unstable structures. However,there is no report on synthesis of bufadienolides sul-fonation though they have promising cytotoxic activities. In this paper,the arenobufagin was firstly taken as an example,to develop an effective method for the synthesis of arenobufagin-3-O-sulfonate. In order to enrich the arenobufagin-3-O-sulfonate quickly and mildly,a reversed phase/hydrophilic solid-phase extraction method based on our homemade hydrophilic Click TE -Cys was developed. The structures of arenobufagin-3-O-sulfonate and arenobufagin were both identified by MS,1 H NMR and 13 C NMR spectra. In addition,the total bufogenins were sulfonated by this method and the sul-fated bufogenins were also successfully enriched via the reversed phase/hydrophilic solid-phase extrac-tion approach. Finally,a LC-MS system was employed to test the result,and about 20 sulfated bu-fogenins were primarily identified according to their molecular weight. Based on this, a systemic method was first developed for the synthesis and enrichment of the sulfated total bufogenins and a large number of potential anticancer new compounds were obtained,which provided lots of agents for fur-ther screening to achieve high effective and low toxic compounds. The efficient reversed phase/hydro-philic solid-phase extraction method for the enrichment of sulfated bufogenins could be used to enrich sulfated bufogenins in toad skin extract.

Abstract:Twelve compounds were isolated from the venom of Bufo bufo gargarizans. On the basis of their physical and chemical properties and spectral data, their structures were identified as resibufagenin (1), bufotalin (2), desacetylcinobufagin (3), 19-oxodesacetylcinobufotalin (4), cinobufotalin (5), 1beta-hydroxylbufalin (6), 12alpha-hydroxybufalin (7), bufotalinin (8), Hellebrigenin (9), telocinobufagin (10), hellebrigenol (11) and cinobufagin-3-hemisuberate methyl ester (12), respectively. Compounds 7 and 12 are new natural products.

Abstract:To study the chemical composition of the skin of Bufo bufo gargarizans, many kinds of chromatography methods were used in the isolation procedures, while the structures of isolated compounds were determined on the basis of their NMR and MS spectral analysis. As a result, two new compounds were isolated from its ethanolic extract and characterized as cinobufotalin 3-nonanedioylarginine ester (8) and bufotalin 3-pimeloylarginine ester (14). Furthermore, 13 known compounds were obtained. Isolated bufadienolides showed significant inhibition effect against SMMC-7721 cell lines in vitro.

Abstract:Bufo gargarizans Cantor (B. gargarizans) i s the most widely distributed and abundant species of toad in China. Bufadienolides and indole alkaloids have cardiotonic and anti-tumor activities and are important pharmacological components of B. bufo gargarizans . In this experiment, a novel compound ( 1 ) and two known compounds ( 2 and 3 ) were isolated and identified from the dry skin of B. bufo gargarizans , both of which are bufadienolides. Various column chromatography methods were used to separate and purify the extract from the dried skin of B. bufo gargarizans . Accurate molecular weights were measured by HR-ESI-MS, and the chemical structure of the compounds was determined by NMR spectrometers. The structures were named as (2 β ,5 β ,16 α )-2,5,16-trihydroxide bufa-14,20,22 dienolide ( 1 ), gamabufotalin ( 2 ) and desacetylbufotalin ( 3 ). In vitro cytotoxic activity assay indicated that compound 1 showed a moderate cytotoxicity against A549 cells with IC 50 value of 12.65 μM.

Abstract:Bufadienolides are a class of steroids with a distinctive α-pyrone ring at C17, mostly produced by toads and consisting of over 100 orthologues. They exhibit potent cardiotonic and antitumor activities and are active ingredients of the traditional Chinese medicine Chansu and Cinobufacini. Direct extraction from toads is costly, and chemical synthesis is difficult, limiting the accessibility of active bufadienolides with diverse modifications and trace content. In this work, based on the transcriptome and genome analyses, using a yeast-based screening platform, we obtained eight cytochrome P450 (CYP) enzymes from toads, which catalyze the hydroxylation of bufalin and resibufogenin at different sites. Moreover, a reported fungal CYP enzyme Sth10 was found functioning in the modification of bufalin and resibufogenin at multiple sites. A total of 15 bufadienolides were produced and structurally identified, of which six were first discovered. All of the compounds were effective in inhibiting the proliferation of tumor cells, especially 19-hydroxy-bufalin (2) and 1β-hydroxy-bufalin (3), which were generated from bufalin hydroxylation catalyzed by CYP46A35. The catalytic efficiency of CYP46A35 was improved about six times and its substrate diversity was expanded to progesterone and testosterone, the common precursors for steroid drugs, achieving their efficient and site-specific hydroxylation. These findings elucidate the key modification process in the synthesis of bufadienolides by toads and provide an effective way for the synthesis of unavailable bufadienolides with site-specific modification and active potentials.
Structurally diverse bufadienolides from the skins of Bufo bufo gargarizans and their cytotoxicity

Abstract:Natural products, with their extensive chemical diversity, distinctive biological activities, and vast reservoirs, provide a robust foundation for advancing cancer therapeutics. A comprehensive phytochemical investigation of the skins from Bufo bufo gargarizans afforded two new bufadienolide derivatives identified as bufalactamides A and B ( 1–2 ), along with six known compounds: argentinogenin (3) , desacetylcinobufagin ( 4 ), desacetylcinobufaginol ( 5 ), cinobufaginol ( 6 ), bufalin ( 7 ) and gamabufalin ( 8 ). The structural elucidation of these compounds was meticulously carried out by analyses of spectroscopic data (1D and 2D NMR, HR-ESIMS), and comparison with the literature data. Plausible biosynthetic pathways for the new compounds were also discussed. Moreover, the cytotoxicity of the compounds was investigated using various cancer cell lines, including lung cancer (A549), colon cancer (HCT-116), liver cancer (SK-Hep-1), and ovarian cancer (SKOV3). Our research findings indicated that compounds 3 , and 6 – 8 exhibit potent cytotoxic activity (IC 50 < 2.5 μM). In contrast, compounds 4 and 5 display moderate cytotoxic activity (IC 50 < 50 μM) while compounds 1 and 2 show no cytotoxic activity (IC 50 > 100 μM). From this data, we conducted a comprehensive analysis of the structure-activity relationships among these compounds.

Abstract:Bioguided fractionation of the ethanol extracts obtained from Platycodon grandiflorum roots led to isolation of two new triterpenoid saponins, characterized as 3-O-beta-D-glucopyranosyl-2beta,12alpha,16alpha,23,24-pentahydroxyoleanane-28(13)-lactone (1) and 3-O-beta-D-glucopyranosyl-(1-->3)-beta-D-glucopyranosyl-2beta,12alpha,16alpha,23alpha-tetrahydroxyoleanane-28(13)-lactone (2) by 1D- and 2D-NMR and MS techniques, as well as chemical means. Both compounds showed cytotoxic activity against human ECA-109 cells.

Abstract:Bufadienolides, naturally occurring steroids primarily found in toads, have garnered attention for their pharmacological properties and ecological significance. In this study, we isolated and identified 21 bufadienolides from the gallbladders of Bufo gargarizans , comprising four new compounds and 17 known ones. Notably, the predominance of 15 bufadienolides with a 3α-OH configuration in toad bile differs significantly from the 3β-OH bufadienolides found in venom secreted by toad glands. Moreover, our investigation into the biotransformation of 3β-OH and 3α-OH bufadienolides in the liver and kidney tissues of toads revealed an irreversible conversion from 3β-OH to 3α-OH bufadienolides, suggesting a crucial role in toad self-detoxification. These findings provide valuable insights into the structural diversity of bufadienolides and advance our understanding of their medical and ecological significance. Graphical
