Abstract:ABSTRACTBiotransformation of natural products has great potential for producing new drugs and could provide in vitro models of mammalian metabolism. Microbial transformation of the cytotoxic steroid cinobufagin was investigated. Cinobufagin could be specifically hydroxylated at the 12β-position by the fungusAlternaria alternata. Six products from a scaled-up fermentation were obtained by silica gel column chromatography and reversed-phase liquid chromatography and were identified as 12β-hydroxyl cinobufagin, 12β-hydroxyl desacetylcinobufagin, 3-oxo-12β-hydroxyl cinobufagin, 3-oxo-12β-hydroxyl desacetylcinobufagin, 12-oxo-cinobufagin, and 3-oxo-12α-hydroxyl cinobufagin. The last five products are new compounds. 12β-Hydroxylation of cinobufagin byA. alternatais a fast catalytic reaction and was complete within 8 h of growth with the substrate. This reaction was followed by dehydrogenation of the 3-hydroxyl group and then deacetylation at C-16. Hydroxylation at C-12β also was the first step in the metabolism of cinobufagin by a variety of fungal strains. In vitro cytotoxicity assays suggest that 12β-hydroxyl cinobufagin and 3-oxo-12α-hydroxyl cinobufagin exhibit somewhat decreased but still significant cytotoxic activities. The 12β-hydroxylated bufadienolides produced by microbial transformation are difficult to obtain by chemical synthesis.

Abstract:Hydroxylation is an important route to synthesize more hydrophilic compounds of pharmaceutical significance. Microbial hydroxylation offers advantages over chemical means for its high specificity. In this study, a fungal strain Alternaria alternata AS 3.4578 was found to be able to catalyze the specific 12β-hydroxylation of a variety of cytotoxic bufadienolides. Cinobufagin and resibufogenin could be completely metabolized by A. alternata to generate their 12β-hydroxylated products in high yields (>90%) within 8h of incubation. A. alternata could also convert 3-epi-desacetylcinobufagin into 3-epi-12β-hydroxyl desacetylcinobufagin as the major product (70% yield). C-3 dehydrogenated products were detected in these reactions in fair yields, while their accumulation was relatively slow. The 12β-hydroxylation of bufadienolides could be significantly inhibited by the substitution of 1β-, 5-, or 16α-hydroxyl groups, and the 14β,15β-epoxy ring appeared to be a necessary structural requirement for the specificity. For the biotransformation of bufalin, a 14β-OH bufadienolide, this reaction was not specific, and accompanied by 7β-hydroxylation as a parallel and competing metabolic route. The biotransformation products were identified by comparison with authentic samples or tentatively characterized by high-performance liquid chromatography-diode array detection-atmospheric pressure chemical ionization-mass spectrometry analyses.
Concise Synthesis of Bufadienolide Cinobufagin via Late-Stage Sin-glet Oxygen Oxidation/Rearrangement Approach

Abstract:This manuscript describes a concise synthesis of cinobufagin, a natural steroid of bufadienolide family, from readily available dehydroepiandrosterone (DHEA), as well as its a5-epimer derived from 3-epi-andosterone. This synthesis features expe-dient installation of the b17-pyrone moiety with the b14,b15-epoxide and the b16-acetoxy group using a photochemical re-gioselective singlet oxygen [4+2] cycloaddition followed by CoTPP-promoted in situ endoperoxide rearrangement to provide a b,b-bis-epoxide in 64% yield with a 1.6:1 d.r. This b,b-bis-epoxide intermediate was subsequently subjected to a regioselective scandium(III) trifluoromethanesulfonate catalyzed House-Meinwald rearrangement to establish the b17-configuration. The synthesis of cinobufagin is achieved in 12 steps (LLS) and 7.4% overall yield, and we demonstrate that it could be used as a platform for the subsequent medicinal chemistry exploration of cinobufagin analogs such as cinobufagin a5-epimer.

Abstract:The biotransformation of cinobufagin by cell suspension cultures of Catharanthus roseus was investigated. After 6 days incubation, four new glucosylated derivatives, desacetylcinobufagin 16-O-β-d-glucoside, 3-epi-desacetylcinobufagin 16-O-β-d-glucoside, 3-oxo-desacetylcinobufagin 16-O-β-d-glucoside, and cinobufagin 3-O-β-d-glucoside, respectively, were isolated and identified on the basis of their physical and chemical data. This is the first report of bufadienolides with a glucosyl substituent at the C-16 position.

Abstract:Microbial transformation was used to prepare novel cytotoxic bufadienolides. Twelve products (3–14) were obtained from bufalin (1) by the fungus Mucor spinosus. Their structures were elucidated by high-resolution mass spectroscopy (HR-MS) and extensive NMR techniques, including 1H NMR, 13C NMR, DEPT, 1H–1H correlation spectroscopy (COSY), two dimensional nuclear Overhauser effect correlation spectroscopy (NOESY), heteronuclear multiple quantum coherence (HMQC), and heteronuclear multiple bond coherence (HMBC). Compounds 3, 4, 9 and 11–14 are new mono- or dihydroxylated derivatives of bufalin with novel oxyfunctionalities at C-1β, C-7β, C-11β, C-12β and C-16α positions. The in vitro cytotoxic activities against human cancer cell lines of 3–14, together with 16 biotransformed products derived from cinobufagin (15–30) were determined by the MTT method, and their structure–activity relationships (SAR) were discussed.

Abstract:Twenty strains of filamentous fungi (from nine genera) and four species of bacteria were screened for their capabilities to transform bufadienolides. The highly stereo- and regiospecific isomerization and dehydrogenation of three major bufadienolides (resibufogenin, cinobufagin and bufalin) at 3-OH by Fusarium solani AS 3.1829 were reported. Three major transformed products were obtained and their structures were identified as 3-epi-resibufogenin (1a), 3-epi-bufalin (2a), 3-keto-cinobufagin (3a), respectively, on the basis of their spectral data. The reaction kinetics and the effects of pH, substrate concentration and incubation phase on the biotransformation were also investigated.

Abstract:The biotransformation of arenobufagin (1) and cinobufotalin (2), isolated from the natural medicine Chan Su, by Alternaria alternata AS 3.4578 was carried out. Incubation of 1 and 2 afforded six metabolites: 3-oxo-arenobufagin (1a), psi-bufarenogin (1b), 3-oxo-psi-bufarenogin (1c), 3-oxo-Delta(4)-derivative of cinobufotalin (2a), 3-oxo-cinobufotalin (2b) and 12 beta-hydroxycinobufotalin (2c). Among them, metabolites 1a, 1c and 2c are new compounds and their structures were characterized on the basis of their spectroscopic data (NMR, MS and IR). Compounds 1, 2, 1b, 2a and 2b were evaluated for their cytotoxicity against HepG2 and MCF-7 human cancer cells, and all of them showed significant inhibitory activities.

Abstract:Microbial transformation of a cytotoxic bufadienolide, cinobufagin (1), was performed by Syncephalastrum racemosum. The six metabolites obtained were identified as 7 beta-hydroxycinobufagin (2), 12 beta-hydroxycinobufagin (3), cinobufotalin (4), 5,12 beta-dihydroxycinobufagin (5), 4 beta, 11 alpha-dihydroxycinobufagin (6), and 4 beta, 12 alpha-dihydroxycinobufagin (7), respectively, on the basis of spectroscopic studies. Metabolites 2 and 5-7 were characterized as new compounds, and 2-7 proved to be cytotoxic against Bel-7402 human hepatoma cells.

Abstract:The biotransformations of cinobufagin (1), an animal-originated bufadienolide, by cell suspension cultures of Catharanthus roseus and Platycodon grandiflorum were investigated. Incubation for 6 days of 1 with C. roseus yielded four products, desacetylcinobufotalin (2), 3-epi-desacetylcinobufagin (3), 1β-hydroxyl desacetylcinobufagin (4) and 3-epi-desacetylcinobufotalin (5), among which 4 is a new compound. Time course investigation revealed that the biotransformation rates of two major products, 2 and 3, reached their highest levels of 44.7 and 16.5%, respectively, on the third day after substrate administration. Compounds 2–5 showed more potent cytotoxic activities against HL-60 cell lines than the parent compound 1. A plausible biotransformation pathway is proposed to account for the formation of the observed products. From the culture supernatant of P. grandiflorum, 2 was isolated in 37.8% yield after 8 days of incubation with 1. Cinobufotalin (6) and desacetylcinobufagin (7) were also obtained as minor products. The two plant suspension cultures exhibited similar transformation patterns on compound 1.

Cunninghamella blakesleana catalyzed the specific 12alpha-hydroxylation of bufalin and produced 12alpha-hydroxybufalin (2) and 7beta,12alpha-dihydroxybufalin (3) as the major metabolites, together with 7beta-hydroxybufalin (4) and 12beta-hydroxybufalin (5) in low yields. Two minor products were isolated from the culture broth of Mucor spinosus and were identified as 7beta,15alpha-dihydroxybufalin (6) and 5beta,7beta-dihydroxybufalin (7), respectively. Metabolites 2, 3, 6, and 7 are new compounds, and their structures were fully characterized by NMR and MS spectroscopy.

Abstract:Resibufogenin is a cytotoxic steroid isolated from the Chinese drug ChanSu. The biotransformation of resibufogenin by Mucor polymorphosporus afforded 22 products, and 15 of them were new. The transformation reactions involved hydroxylations at C-1β, C-5, C-7α, C-7β, C-12α, C-12β and C-16α, as well as epimerization or dehydrogenation of 3-OH. Hydroxylations at C-12α, C-12β and C-16α were the major reactions, each giving products in >5% yields, whereas the other products were obtained in fairly low yields. Some of the products showed decreased but still potent cytotoxicities. This investigation provided a useful approach to prepare new bufadienolides and most of them were difficult to obtain by chemical means.

Abstract:Resibufogenin, cinobufagin, and bufalin are cytotoxic steroids isolated from the Chinese drug Chan'su. Biotransformation of these three bufadienolides by Nocardia sp. NRRL 5646 was investigated. Notably, resibufogenin was converted to 3-acetyl 15 beta-hydroxyl bufotalin, via an unprecedented 14 beta,15 beta-epoxy ring cleavage and a regio-selective acetoxylation. This product showed significantly increased cytotoxic activity. The regio-selective acetylation of the 3-OH was also involved in the other reactions. The structures of metabolites were established by ESI-LC/MS and 2D NMR techniques. The in vitro cytotoxic activities against human cancer cell lines of the substrates and the transformed products were determined by the MTT method and their structure-activity relationship (SAR) was discussed. Abstract:From the fruits of Celastrus orbiculatus Thunb. a new beta-dihydroagarofuran sesquiterpene ester named 1beta,2beta,13-triacetoxy-9alpha-cinnamoyloxy-beta-dihydroagarofuran (1) has been isolated along with two known compounds, 6alpha-acetoxy-1beta, 9beta-dibenzoyloxy-8beta-hydroxy-beta-dihydroagarofuran (2), and 1beta, 6alpha-diacetoxy-9beta-benzoyloxy-8beta-hydroxy-beta-dihydroagarofuran ( 3). Their structures were elucidated on the basis of spectroscopic data.

Abstract:1. Cinobufagin (1) is a major bufadienolide in ChanSu (a traditional Chinese medicine) with a wide range of pharmacological activities. In this paper, the in vivo metabolites of 1 in rats were studied. 2. Nine metabolites were isolated from the bile of rats, and their structures were identified as: desacetylcinobufagin (2), 3-ketodesacetylcinobufagin (3), 3-epi-desacetylcinobufagin (4); beta-hydroxy-3-epi-desacetylcinobufagin (5), 1 alpha-hydroxy-3-epi-desacetylcinobufagin (6), 12 beta-hydroxy-3-epi-desacetylcinobufagin (7), 1 beta-hydroxy-3-epi-desacetylcinobufagin (8), 1 alpha,5 alpha-dihydroxy-3-epidesacetylcinobufagin (9), and 2 alpha,5 beta-dihydroxy-3-epi-desacetylcinobufagin (10), respectively, on the basis of widely spectroscopic studies including two-dimensional-nuclear magnetic resonance (NMR). Among them, metabolites 6-10 are new compounds. 3. The results show that hydroxylation is the main reaction involved in metabolism of 1, and the preferred hydroxylation sites were C-1 and C-5.

Abstract:Mucor spinosus has been employed for the biotransformation of cinobufagin ( 1 ) to afford three metabolites. On the basis of their physico-chemical data, the structures of the transformation products have been characterized as 1beta-hydroxy-cinobufagin ( 2 ), 12beta-hydroxy-cinobufagin ( 3 ) and 1beta,12beta-dihydroxy-cinobufagin ( 4 ), of which metabolites 2 and 4 are new compounds. In vitro cytotoxic activities of the biotransformation products and the substrate-cinobufagin have been assayed against four tumor cell lines of Bel 7420, BGC 823, HeLa and HL 60; they all showed cytotoxic activities.

Abstract:The biotransformation of three bioactive bufadienolides, namely, bufotalin (1), telocinobufagin (2), and gamabufotalin (3) by cell suspension cultures of Saussurea involucrata yielded 11 products. Bufotalin yielded 3-epi-bufotalin (1a), 3-epi-desacetylbufotalin (1b), 3-epi-bufotalin 3-O-β-d-glucoside (1c), 1β-hydroxybufotalin (1d), and 5β-hydroxybufotalin (1e); telocinobufagin yielded 3-dehydroscillarenin (2a), 3-dehydrobufalin (2b), and 3-epi-telocinobufagin (2c); and gamabufotalin yielded 3-epi-gamabufotalin (3a), 3-dehydrogamabufotalin (3b), and 3-dehydro-Δ1-gamabufotalin (3c), respectively. Among these 11 products, 1a, 1b, 1c, 1d, 3a and 3c are previously unreported. The structures of these metabolites were elucidated based on NMR spectroscopic analyses and mass spectrometry. Most metabolites showed significant cytotoxic activities against human hepatoma (HepG2) and breast cancer (MCF-7) cell lines. In addition, the time course for the biotransformation of 3 was investigated.
