Abstract:We present a novel FMT endoscope by using a MEMS scanning mirror and an optical fiberscope. The diameter of this highly miniaturized FMT device is only 5 mm. To our knowledge, this is the smallest FMT device we found so far. Several phantom experiments based on indocyanine green (ICG) were conducted to demonstrate the imaging ability of this device. Two tumor-bearing mice were systematically injected with tumor-targeted NIR fluorescent probes (ATF-PEG-IO-830) and were then imaged to further demonstrate the ability of this FMT endoscope for imaging small animals.

Abstract:Existing super-resolution imaging technology relies on rigid and bulky systems, which limits its application in narrow space. Endoscopy based on multimode fibers (MMF) has emerged as a significant solution for in vivo imaging. There is a high requirement to observe samples inside the lumen of the body simultaneously break through the diffraction limit. We propose the frequency-shift mechanism for universal super-resolution imaging with light-field encoded modulation. In this paper, we demonstrate our method for a single MMF in vivo imaging at subcellular resolution through light-field encoded, which can also ensure stable imaging under various operating conditions.

Abstract:Point-of-care medical diagnosis demands immediate feedback on tissue pathology. Confocal endomicroscopy can provide real-time in vivo images with histology-like features. The working channel in medical endoscopes are becoming smaller in dimension. Microsystems methods can produce tiny mechanical scanners. We demonstrate a flexible fiber instrument for in vivo imaging as an endoscope accessory. The optical path is folded on-axis to reduce length while allowing the beam to expand and achieve a numerical aperture of 0.41. A high-speed parametric resonance mirror produces large deflection angles > 13°, and is mounted on a 2 mm diameter chip designed with clamp structures for reduced space. A compact lens assembly provides diffraction-limited lateral and axial resolution of 1.5 and <inline-formula xmlns:mml="http://www.w3.org/1998/Math/MathML" xmlns:xlink="http://www.w3.org/1999/xlink"> <tex-math notation="LaTeX">$12~\mu \text{m}$ </tex-math></inline-formula> , respectively. A working distance of <inline-formula xmlns:mml="http://www.w3.org/1998/Math/MathML" xmlns:xlink="http://www.w3.org/1999/xlink"> <tex-math notation="LaTeX">$50~\mu \text{m}$ </tex-math></inline-formula> and field-of-view of <inline-formula xmlns:mml="http://www.w3.org/1998/Math/MathML" xmlns:xlink="http://www.w3.org/1999/xlink"> <tex-math notation="LaTeX">$350\,\,\mu \text{m}\,\,\times 350\,\,\mu $ </tex-math></inline-formula> m are achieved. Miniature apparatus is fabricated to assemble and align the scanhead components. The optics and scanner are packaged in a distal tip with 2.4 mm diameter and 10 mm rigid length. These dimensions allow the endomicroscope to pass forward easily through the 2.8 mm diameter working channel in medical endoscopes commonly used in clinical practice. Fluorescence images are collected in vivo at 10 frames per second in the colon of genetically-engineered mice that spontaneously develop adenomas. A FITC-labeled peptide heterodimer is administered intravenously to provide specific contrast. Sub-cellular structures are visualized to distinguish pre-malignant from normal mucosa. These results demonstrate use of microsystems methods to produce an ultra-compact instrument with sufficiently small dimensions for broad use.

Abstract:A novel handheld probe based on a microelectromechanical systems (MEMS) scanning mirror for three-dimensional (3D) fluorescence molecular tomography (FMT) is described. The miniaturized probe consists of a MEMS mirror for delivering an excitation light beam to multiple preselected points at the tissue surface and an optical fiber array for collecting the fluorescent emission light from the tissue. Several phantom experiments based on indocyanine green, an FDA approved near-infrared (NIR) fluorescent dye, were conducted to assess the imaging ability of this device. Tumor-bearing mice with systematically injected tumor-targeted NIR fluorescent probes were scanned to further demonstrate the ability of this MEMS-based FMT for imaging small animals.

Abstract:A confocal laser scanning endomicroscopy is developed,which employs a telecentric endomicroscopic optical system to achieve long-distance relay transmission of images,telecentric f-theta optical scanning and endomicroscopic imaging functions.Two-dimensional confocal scanning is realized through two scanning mirrors,and low-noise control signals are generated by the embedded system.Miniaturized design is adopted in the confocal laser scanning endomicroscopy in order to easily use for portable applications.Firstly,the outer diameter of the optical system is 8 mm,and working length of which is 250.3 mm,so in-vivo endomicroscopic imaging can be realized through standard laparoscopic surgery port.Secondly,two small mirrors with aperture of 3mm are employed to achieve in vitro confocal scanning,with the maximum frequency of 100 Hz.Finally,laser system and fluorescence detection are connected with the distal end of the confocal laser scanning endomicroscopy through an optical fiber and a cable,and the size and the weight of the distal end are minimized by this method.The experimental results show that the system′s filed of view reaches φ600 μm,and its optical resolution is 2.2 μm.This system can be used under handheld operation or other ways to realize minimally invasive in-vivo fluorescence endoscopic surgery since its miniature size and low weight.

Abstract:Micro-endoscopes are widely used for detecting and visualizing hard-to-reach areas of the human body and for in vivo observation of animals. A micro-endoscope that can realize 3D imaging at the camera framerate could benefit various clinical and biological applications. In this work, we report the development of a compact light-field micro-endoscope (LFME) that can obtain snapshot 3D fluorescence imaging, by jointly using a single-mode fiber bundle and a small-size light-field configuration. To demonstrate the real imaging performance of our method, we put a resolution chart in different z positions and capture the z-stack images successively for reconstruction, achieving 333-μm-diameter field of view, 24 μm optimal depth of field, and up to 3.91 μm spatial resolution near the focal plane. We also test our method on a human skin tissue section and HeLa cells. Our LFME prototype provides epi-fluorescence imaging ability with a relatively small (2-mm-diameter) imaging probe, making it suitable for in vivo detection of brain activity and gastrointestinal diseases of animals.
Fast volumetric fluorescence imaging with multimode fibers

Abstract:In this Letter, we propose a compact multimode fiber endoscope which employs wavefront shaping with a digital micromirror device (DMD). An automated single calibration step allows us to correct for optical misalignment, and the method achieves accurate focusing at various depths in the sample through rapid switching of holographic patterns by the DMD. The speed of calibration is one or two orders of magnitude faster than existing methods. The method, single calibration multimode fiber imaging (SCMFI), is compared with existing methods, and its performance is validated. We show a near diffraction limited focusing capability at imaging depths up to 110 µm with near constant lateral resolutions of 1.4 µm. Finally, we demonstrate the method for the imaging of small fluorescent beads embedded in a 3D matrix. The results indicate excellent power penetration and focusing performance. Combined with the high speed of SCMFI, this paves the way for volumetric tissue endoscopy at depth.

Abstract:Two-photon endomicroscopy is a promising technique with the ability to achieve in situ imaging and diagnosis at subcellular resolution. The large field-of-view capability is essential and useful to locate and image suspicious areas of biological tissue. In this work, we report objective-lens-free, lensed fiber-optic two-photon endomicroscopy for field-of-view enhancement. The field of view of this two-photon endomicroscopic probe is 750 μm with a resolution of 3.03 μm. This 1.6 g miniature probe has an integrated outer diameter of 5.8 mm and a rigid length of 33.5 mm. The imaging performance of the lensed-fiber-optic two-photon endomicroscopy was validated by examining an ex vivo mouse heart, kidney, brain, stomach wall tissues, and in vivo brain tissue.

Abstract:In this article authors present a miniaturized scanning optical probe for multimodal nonlinear endomicroscopic imaging. The miniaturized scanning optical probe includes a two-axis gimbal-less MEMS scanner for beam scanning and a group of miniature optics for laser collimation and optical signal collection. Both second harmonic generation and two-photon excited autofluorescence signals are collected. The imaging rate is tested as 30 Hz and FOV is 130 um by 130 um. Different from traditional “needle” shape optical imaging catheters, our design highlights its “mushroom” shape and has been demonstrated to work with a dual-instrument channel commercially available gastroscope as an apparatus.

Abstract:Imaging of the cells and microvasculature simultaneously is beneficial to the study of tumor angiogenesis and microenvironments. We designed and built a fiber-optic based photoacoustic microscopy (PAM) and confocal fluorescence microscopy (CFM) dual-modality imaging system. To explore the feasibility of this all-optical device for future endoscopic applications, a microelectromechanical systems (MEMS) scanner, a miniature objective lens, and a small size optical microring resonator as an acoustic detector were employed trying to meet the requirements of miniaturization. Both the lateral resolutions of PAM and CFM were quantified to be 8.8μm. Axial resolutions of PAM and CFM were experimentally measured to be 19μm and 53μm, respectively. The experiments on ex vivo animal bladder tissues demonstrate the good performance of this system in imaging not only microvasculature but also cellular structure, suggesting that this novel imaging technique holds potential for improved diagnosis and guided treatment of bladder cancer.
Towards a miniatured all-fiber photoacoustic endoscope (Conference Presentation)

Abstract:We present the all-fiber approach towards building a miniatured, optical-resolution photoacoustic endoscope. The catheter encapsulates two optical fibers for optical excitation and ultrasound detection, respectively. The ultrasound waves are detected with the laser-based fiber optic sensor, with a diameter of 125 μm, instead of a focused piezoelectric transducer. Photoacoustic endoscopic images from a rat rectum have been acquired in vivo with a range of 6.3 mm, a lateral resolution of 10 μm, and a 285° angular field-of-view. The catheter has a diameter of 2.3 mm and can be further reduced by replacing the bulk prism reflector.

Abstract:This paper reviews our recent developments of ultrathin fiber-optic endomicroscopy technologies for transforming high-resolution noninvasive optical imaging techniques to in vivo and clinical applications such as early disease detection and guidance of interventions. Specifically we describe an all-fiber-optic scanning endomicroscopy technology, which miniaturizes a conventional bench-top scanning laser microscope down to a flexible fiber-optic probe of a small footprint (i.e. similar to 2-2.5 mm in diameter), capable of performing two-photon fluorescence and second harmonic generation microscopy in real time. This technology aims to enable real-time visualization of histology in situ without the need for tissue removal. We will also present a balloon OCT endoscopy technology which permits high-resolution 3D imaging of the entire esophagus for detection of neoplasia, guidance of biopsy and assessment of therapeutic outcome. In addition we will discuss the development of functional polymeric fluorescent nanocapsules, which use only FAD approved materials and potentially enable fast track clinical translation of optical molecular imaging and targeted therapy.
A Miniature Endoscopic Optical Coherence Tomography Probe Based on C-Lens

Abstract:We present a novel miniature endoscopic optical coherence tomography (OCT) probe based on an electrothermal microelectromechanical (MEMS) mirror and a C-lens. The MEMS mirror has a relatively large mirror plate of 0.5 mm × 0.5 mm on a small chip size of 1.5 mm × 1.3 mm, leading to the outer diameter of the endoscopic probe down to only 2.5 mm so that the probe is capable of being inserted through the biopsy channel of a conventional endoscope. A long focal length of 12 mm is achieved by properly designing the C-lens. Compared to commonly used GRIN lenses, C-lenses have advantages of much smaller sensitivities of the working distance and spot size to machining variations. The C-lens based probe can scan a large field of view (FOV) of 6 mm × 6 mm at only 5-V drive voltage. A swept source endoscopic OCT system is constructed using this miniature probe. Experiments show that the corresponding lateral and axial resolutions are about 50 and 14 μm, respectively. Various samples including human fingers, onions, and cancerous colon tissue are imaged and the result suggests this large-working-distance, large-FOV endoscopic probe is promising for applications in large tubular organs and large-area lesions.
Confocal Fluorescence Microendoscopy Using A Digital Micro-Mirror Device

Abstract:A design of confocal fluorescence microendoscopy utilizing a digital micro-mirror device (DMD) is described. Laser beams of the microendoscope are coupled into the body through a telescopic optics system, rather than through fibers or fiber bundles which are widely used in existing microendoscopes. Each micro-mirror of the DMD is used as a confocal pinhole. The DMD not only couples the laser beams into the body by a random time-varying speckle pattern and performs the scanning mechanism of the body tissue with different positions, but also couples the fluorescent signal emitted from the markers out to the CCD camera. Because of the CCD's integration feature and DMD's rapid parallel scanning feature, a complete predetermined depth tomography image accumulated by different scanning patterns of DMD can be acquired through only one CCD exposure procedure. The objective lens to realize high resolution and high sensitivity fluorescence imaging is the other function of the telescopic optics, with a numerical aperture of 0.35. The resolution of confocal microendoscope is superior to 228 lp/mm determined by 1951USAF resolution test target. Images of a tendon specimen are also shown to demonstrate practical application of the design. The confocal microendoscope using a DMD permits the acquisition of high-resolution real-time confocal images of epithelial tissue in vivo organ and realizes the aim of non-invasive diagnosis and treatment.

Abstract:A wide-field endoscope that is sensitive to fluorescence can be used as an adjunct to conventional white light endoscopy by detecting multiple molecular targets concurrently. We aim to demonstrate a flexible fiber-coupled accessory that can pass forward through the instrument channel of standard medical endoscopes for clinical use to collect fluorescence images. A miniature scan mirror with reflector dimensions of 1.30 × 0.45 mm 2 was designed, fabricated, and placed distal to collimated excitation beams at λ ex = 488, 660, and 785 nm. The mirror was driven at resonance for wide angular deflections in the X and Y-axes. A large image field-of-view (FOV) was generated in real time. The optomechanical components were packaged in a rigid distal tip with dimensions of 2.6 mm diameter and 12 mm length. The scan mirror was driven at 27.6 and 9.04 kHz in the fast (X) and slow (Y) axes, respectively, using a square wave with 50% duty cycle at 60 V pp to collect fluorescence images at 10 frames per sec. Maximum total divergence angles of ± 27.4° and ± 22.8° were generated to achieve a FOV of 10.4 and 8.4 mm, respectively, at a working distance of 10 mm. Multiplexed fluorescence images were collected in vivo from the rectum of live mice using 3 fluorescently-labeled peptides that bind to unique cell surface targets. The fluorescence images collected were separated into 3 channels. Target-to-background ratios of 2.6, 3.1, and 3.9 were measured. This instrument demonstrates potential for broad clinical use to detect heterogeneous diseases in hollow organs.

Abstract:Since its inception, endoscopy has aimed to establish an immediate diagnosis that is virtually consistent with a histologic diagnosis. In the past decade, confocal laser scanning microscopy has been brought into endoscopy, thus enabling in vivo microscopic tissue visualization with a magnification and resolution comparable to that obtained with the ex vivo microscopy of histological specimens. The major challenge in the development of instrumentation lies in the miniaturization of a fiber-optic probe for microscopic imaging with micron-scale resolution. Here, we present the design and construction of a confocal endoscope based on a fiber bundle with 1.4-μm lateral resolution and 8-frames per second (fps) imaging speed. The fiber-optic probe has a diameter of 2.6 mm that is compatible with the biopsy channel of a conventional endoscope. The prototype of a confocal endoscope has been used to observe epithelial cells of the gastrointestinal tracts of mice and will be further demonstrated in clinical trials. In addition, the confocal endoscope can be used for translational studies of epithelial function in order to monitor how molecules work and how cells interact in their natural environment.

Abstract:In this paper, we present a miniaturized endoscopic probe, consisted of MEMS micromirror, silicon optical bench (SiOB), grade index (GRIN) lens, single mode optical fiber (SMF) and transparent housing, for optical coherence tomography (OCT) bioimaging. Due to the use of the MEMS micromirror, the endoscopic OCT system is highly suitable for non-invasive imaging diagnosis of a wide variety of inner organs. The probe engineering and proof of concept were demonstrated by obtaining the two-dimensional OCT images with a cover slide and an onion used as standard samples and the axial resolution was around 10µm.

Abstract:Over the last decade, optical fiber-based forms of microscopy and endoscopy have extended the realm of applicability for many imaging modalities. Optical fiber-based imaging modalities permit the use of remote illumination sources and enable flexible forms supporting the creation of portable and hand-held imaging instrumentations to interrogate within hollow tissue cavities. A common challenge in the development of such devices is the design and integration of miniaturized optical and mechanical components. Until recently, microelectromechanical systems (MEMS) sensors and actuators have been playing a key role in shaping the miniaturization of these components. This is due to the precision mechanics of MEMS, microfabrication techniques, and optical functionality enabling a wide variety of movable and tunable mirrors, lenses, filters, and other optical structures. Many promising results from MEMS based optical fiber endoscopy have demonstrated great potentials for clinical translation. In this article, reviews of MEMS sensors and actuators for various fiber-optical endoscopy such as fluorescence, optical coherence tomography, confocal, photo-acoustic, and two-photon imaging modalities will be discussed. This advanced MEMS based optical fiber endoscopy can provide cellular and molecular features with deep tissue penetration enabling guided resections and early cancer assessment to better treatment outcomes.

Abstract:In this paper, we report a miniaturized laser scanning microscope. Unlike conventional microscopes, the optical fiber used here for the excitation laser and signal’s transmission largely improves our microscope’s mobility, and thus eliminates the space constraints of the sample stage. This miniaturized microscope can be motorized along XYZ axis and rotated in theta direction. Based on this modified microscope, we take images of test targets and a micrometer, and compare the resolution and signal to background ratio between its confocal and non-descanned detection modules. We also present the imaging results of Hela cells on a tilted sample stage to simulate the imaging of large and irregular samples. Therefore, this miniaturized microscope has the potential to be utilized for in vivo imaging of large animals.

Abstract:We demonstrate a miniature fiber-optic two two-photon endomicroscopy with microsphere-spliced double-cladding antiresonant fiber for resolution enhancement. An easy-to-operate process for fixing microsphere permanently in an antiresonant fiber core, by arc discharge, is proposed. The imaging ability is verified using ex-vivo mouse kidney, heart, stomach, tail tendon, and in-vivo brain neural imaging.
