Abstract:Various geometric shapes and structures self-assembled of amphiphilic lipids when present in an aqueous environment, as active delivery vehicles, are becoming one of focuses of drug delivery system. Lipid-based cubic liquid crystalline nanoparticles (or Cubosomes) consisting of "honeycombed (cavernous)" structure spontaneously formed when a certain concentration of amphiphilic lipids dispersed in aqueous solution has curved bicontinuous lipid bilayer in three dimensions, separating two congruent networks of water channels. Its unique structure consists of internal double water channels and large interfacial areas, which reveal great flexibility in encapsulation efficiency of various polarities and amount of drugs, and has variegated range of drugs encapsulated. As a drug delivery vehicle, high drug payloads, stabilization of peptides or proteins and simple preparation process are also its advantages. The ability of cubic phase to incorporate and control release of drugs of varying size and polar characteristics, and biodegradability of lipids make it an interesting drug delivery system for various routes of administration, including oral, topical (or mucosal) and intravenous administrations, with extensive application in a multitude of dosage forms. Furthermore, a number of different proteins in cubic phase appear to retain their native conformation and bioactivity, and are protected against chemical and physical inactivation. In this paper, investigations of lipid-based cubic liquid crystalline nanoparticles are reviewed and summarized, with a hope to provide a reference for its in-depth study. At the end, the authors made a development prospect of this novel excellent candidate for active ingredients delivery vehicle.
A novel cubic phase of medium chain lipid origin for the delivery of poorly water soluble drugs

Abstract:The existence of a novel cubic liquid crystalline phase is described within the pseudo-ternary system comprising lauric acid, monolaurin, and simulated endogenous intestinal fluid (SEIF). This phase behaviour has been characterized using cross-polarizing light microscopy (CPLM), and the structure of the cubic phase identified by small angle X-ray scattering (SAXS). The presence of the cubic phase was found to be temperature sensitive within the 20-37 degrees C range making it putative material for in situ gelation purposes. The cubic phase was shown to have a high capacity to solubilise a model poorly water-soluble drug, cinnarizine, and initial in vitro release data highlight the potential of this phase to provide sustained release. Absorption of cinnarizine from the cubic phase was studied in an unconscious rat model via duodenal administration and blood sampling via the carotid artery. The rate of absorption was significantly reduced when compared to a simple suspension formulation, a likely combination of retarded erosion of the cubic phase together with hindered drug release from the cubic matrix. The results of this study suggest that this cubic phase may potentially be of benefit in the delivery of poorly water-soluble compounds due to its high loading capacity and potential for sustained release. The ability to manipulate this system using temperature may warrant further interest in delivery applications via other routes of administration.

Abstract:Backgraound: Novel drug delivery systems have always been of immense interest for research. Majority of the drugs suffers from solubity related constraints. Lipid based drug delivery systems provide a promising approach to address this issue.Amogst various approaches, liquid crystalline systems offers distinct advantages over other available options. It maintains the phase stablility between solid and liquid phases. Objective: Lyotropic Liquid Crystals (LLCs) have highly ordered internal structure forming capabilities thereby assisting the controlled release of a therapeutic agent of varying sizes and polarities. Methods: The present article covers the structural features of dispersed mesophases (cubosomes and hexosomes), components used to prepare various mesophases, method of preparation, characterization techniques and potential applications. Results: LLCsoffers a distinct choice for delivering hydrophobic as well as hydrophilic drug. They are also suitable for the delivery of macromolecules such as peptide and protiens through various routes of administration.Lyotropic liquid crystals are extremely versatile drug delivery systems that can be used for delivering drugs across topical, oral, buccal, pulmonary and intravenous routes. Their use in the delivery of tough to deliver drugs, hormones, peptides, biomolecules and vaccines is particularly important. Conclusion: This review discussed the composition, methods of preparation and characterization of LLCs. The LLCs offer promsing delivery choice for the drugs with lower solubility and higher toxicity.

Abstract:Blue phase liquid crystals (BPLCs) self-assembling into soft cubic crystalline structures possess significant applications in the field of dynamic photoelectron spectroscopy. However, three scientific challenges limit their broader application, namely, the difficulty to form a free-standing structure due to fluidity, dependence on nanoparticles or polymer matrix to widen the BP temperature range, and high working voltages to reorientate the LCMs. Herein, we proposed a new concept of free-standing BPLC microcapsules (BPLCMs) prepared by microfluidic emulsification and interfacial polymerization. The temperature range of BPLCs could be widened by surface anchoring and physical confinement. The BPLCMs built a color library in the visible region in the thermal response process. Most importantly, the stable BPLCMs exhibited a reversible surface-switch mode driven by an electric field lower than 0.55 V mu m(-1), which was the lowest record for BPLCs. The BPLCMs demonstrate intriguing prospects in the fields of smart display and reversible sensors.

Abstract:This paper reviews liposomes with crystalline phase and polymersomes exhibiting crystalline and thermotropic liquid crystalline phases in the membrane. Intriguing morphologies of vesicles are described, including spherical, ellipsoidal and faceted vesicles, produced by a large variety of amphiphilic molecules and polymers with nematic phase, smectic phase or crystalline phase. It is highlighted how the phase transitions and the phase grain boundaries could be used ingeniously to destabilize the vesicular structure and to achieve cargo-release under the action of external stimulation. These liposomes and polymersomes are responsive to physical stimuli, such as temperature variation, shear stress, light illumination, and magnetic and electric fields. These stimuli-responsive properties make them promising candidates as new smart drug delivery systems.

Abstract:<p class="a-plus-plus">Liquid crystalline phases (LCPs) are generated upon lipolysis of ingested lipids in the gastrointestinal tract. The breaking off and subsequent evolution of LCPs produce more advanced vesicular and micellar structures which facilitate oral absorption of lipids, as well as co-loaded drug entities. Owing to sustained or controlled drug release, bioadhesiveness, and capability of loading drugs of different properties, LCPs are promising vehicles to implement for enhancement of oral bioavailability. This review aims to provide an overview on the classification, preparation and characterization, <em class="a-plus-plus">in vivo</em> generation and transformation, absorption mechanisms, and encouraging applications of LCPs in enhancement of oral bioavailability. In addition, we comment on the merits of LCPs as oral drug delivery carriers, as well as solutions to industrialization utilizing liquid crystalline precursor and preconcentrate formulations.</p>

Abstract:Due to their distinctive structural features, lyotropic nonlamellar liquid crystalline nanoparticles (LCNPs), such as cubosomes and hexosomes, are considered effective drug delivery systems. Cubosomes have a lipid bilayer that makes a membrane lattice with two water channels that are intertwined. Hexosomes are inverse hexagonal phases made of an infinite number of hexagonal lattices that are tightly connected with water channels. These nanostructures are often stabilized by surfactants. The structure's membrane has a much larger surface area than that of other lipid nanoparticles, which makes it possible to load therapeutic molecules. In addition, the composition of mesophases can be modified by pore diameters, thus influencing drug release. Much research has been conducted in recent years to improve their preparation and characterization, as well as to control drug release and improve the efficacy of loaded bioactive chemicals. This article reviews current advances in LCNP technology that permit their application, as well as design ideas for revolutionary biomedical applications. Furthermore, we have provided a summary of the application of LCNPs based on the administration routes, including the pharmacokinetic modulation property.

Abstract:Cubosomes are lipid vesicles that are comparable to vesicular systems like liposomes. Cubosomes are created with certain amphiphilic lipids in the presence of a suitable stabiliser. Since its discovery and designation, self-assembled cubosomes as active drug delivery vehicles have drawn much attention and interest. Oral, ocular, transdermal, and chemotherapeutic are just a few of the drug delivery methods in which they are used. Cubosomes show tremendous potential in drug nanoformulations for cancer therapeutics because of their prospective advantages, which include high drug dispersal due to the structure of the cubic, large surface area, a relatively simple manufacturing process, biodegradability, ability to encapsulate hydrophobic, hydrophilic, and amphiphilic compounds, targeted and controlled release of bioactive agents, and biodegradability of lipids. The most typical technique of preparation is the simple emulsification of a monoglyceride with a polymer, followed by sonication and homogenisation. Top-down and bottom-up are two different sorts of preparation techniques. This review will critically analyse the composition, preparation techniques, drug encapsulation approaches, drug loading, release mechanism and applications relevant to cubosomes. Furthermore, the challenges faced in optimising various parameters to enhance the loading capacities and future potentialities are also addressed.

Effect of felodipine on renal haemodynamics and tubular sodium handling after single-dose cyclosporin infusion in renal transplant recipients treated with azathioprine and prednisolone.

Abstract:A total of 25 renal transplant recipients, treated solely with prednisolone and azathioprine, were investigated in a randomized, double-blind, placebo-controlled, cross-over study. The effect of a single oral dose of felodipine 5 mg or placebo on: glomerular filtration rate (GFR); renal plasma flow (RPF); renal vascular resistance (RVR); renal tubular sodium and water handling, measured by the lithium clearance technique; plasma levels of angiotensin II (AngII), aldosterone (Aldo), atrial natriuretic factor (ANF) and arginine vasopressin (AVP); blood pressure (BP), and heart rate (HR) was studied before, during, and after an intravenous infusion of cyclosporin (CyA). Three consecutive clearance periods were performed, each lasting 1 h. During the second period, CyA (0.75 mg kg-1 body weight) was infused. Before infusion of CyA, felodipine caused a significant rise (6.7%) in RPF and lowered RVR, but did not change GFR significantly. The rise in RPF was abolished by infusion of CyA. After infusion, both GFR (7.8%) and RPF (9.4%) were significantly higher and RVR lower after felodipine than after placebo. Proximal tubular output and total sodium excretion were higher on the felodipine day before and after, but not during CyA infusion. In all three periods felodipine reduced both systolic and diastolic BP. In conclusion, a single dose of felodipine increases RPF and decreases blood pressure in renal transplant recipients not treated with CyA. Although some of these changes are abolished by an acute intravenous infusion of CyA, the effects of felodipine are present again also during the 1st hour after the infusion and thereby indicate at least in part some renal protective effect of felodipine. It is suggested that a higher dose of felodipine might also have been preventive against CyA renal side-effects during the acute infusion.

Abstract:Lyotropic liquid crystals (LLCs) have garnered attention as a promising nanocarrier delivery system for enhancing skin permeation owing to their unique structural properties, such as improved drug loading and controlled drug release and versatility. LLCs are greatly explored for topical drug delivery owing to their strong bio-adhesive nature and structural similarity to the biological membranes when applied topically, which improves skin retention and permeation of the drugs. This review explores the functionality of structural design, especially the optimization of micellar systems, hexagonal and lamellar phases, and bicontinuous cubic and sponge phases, to achieve efficient skin permeation of therapeutic loaded LLCs. Furthermore, customization of surfactant to achieve skin compatibility and permeation is explored along with incorporating molecular modifications, functionalization, and the influence of surfactant hydrophilic–lipophilic balance. Additionally, the review illuminates the impact of LLCs curvature and the engineering of ordered and disordered phases for optimal drug release. Emerging advanced techniques such as surface modification with various ligands, stimuli-responsive LLCs for on-demand drug release, and combination therapies with multifunctional LLCs for synergistic effects that can promote targeted and site-specific drug delivery have also been discussed. The detailed findings have been discussed in this review with appropriate case studies. Challenges and future perspectives in designing and formulating LLCs for skin permeation are addressed to ignite future research. The knowledge and advancements presented in this review pave the way for developing next-generation LLCs-based systems, enabling enhanced permeation through the skin and opening new possibilities in the treatment of various dermatological conditions.

Abstract:The success of many drugs in ophthalmic treatments is hindered by their physico-chemical properties and the limited precorneal retention time. Here, lyotropic liquid crystals are proposed as a new ophthalmic drug delivery system. Acyclovir was chosen as model drug for its solubility and its controlled release from cubic phase was achieved. We demonstrated the effortless application of lamellar phase on corneal surface and its ability to convert itself in cubic phase in situ. While the complex viscosity of lamellar phase was affected by temperature (5.1 ± 1.4 kPa·s at 25 °C and 0.12 ± 0.001 Pa·s at 35 °C, respectively), the cubic phase shown no changes in viscosity values and shear thinning behaviour at both temperatures and even in presence of the drug The degradation kinetic of drug-loaded cubic phase was slightly slower than the empty formulation, recording 27.92 ± 1.43% and 33.30 ± 3.11% of weight loss after 8 h. Ex vivo studies conducted on porcine eyeballs and isolated cornea confirmed the instantaneous transition to cubic phase, its ability to resist to gravity force, and forced dripping of simulated tear fluid. Histopathological investigation showed how treated cornea did not report changes in epithelial and stroma structures. In summary, lyotropic liquid crystals could represent an advantageous ophthalmic drug delivery system.

Abstract:The discovery of cubosomes is a classic story and deals with food science, differential geometry, biological membranes and digestive processes. Cubosomes are highly balanced and nanoparticles in design formed primarily from the lipid cubic state and protected by a polymer-based outer circle. Hydrating a surfactant that designates cubic phase and then disperses a solid state into smaller particles, typically forms cubosomes. They behave concretely like rheology with unique properties of practical interest. Cubosome formation can be optimized to engineer pore size or consist of bioactive lipids, polymers can be used for targeting to the outer circle and they are highly secure under physiological conditions. This type of network structure gives them greater drug trapping potential. Related to liposomes, the structure adds a significantly more enhanced membrane surface area to trap membrane proteins and small drug entities. Cubosome may increase the solubility of poorly soluble drugs. Due to modern advances, nanoparticles patterns include drug delivery, membrane bioreactors, artificial cells and biosensors and can be engineered both in vitro. This review, focused on modern advances in cubosome technology, not only facilitates their work but also contributes to standard procedures for the rational design of innovative systems for biomedical applications. Due to the nature of cubosome dispersion being bio-adhesive and biocompatible, as well as having different properties, cubosomes are functional systems, administered in a variety of ways, such as orally and parenterally. Cubosome structure is investigated through electron microscopy, light scattering, X-rays, and NMR, although some researchers are studying the potential of cubosomes as a delivery system.

Abstract:Liquid crystal is a state of matter being intermediate between solid and liquid. Liquid crystal materials exhibit both orientational order and fluidity. Whilst liquid crystals have long been highly recognized in the display industry, in recent decades, liquid crystals provide new opportunities into the cross‐field of material science and biomedicine due to its biocompatibility, multifunctionality, and responsiveness. In this review, we summarize the latest achievements of liquid crystal materials applied in biomedical fields. We start by introducing the basic concepts of liquid crystals, and then shift to the components of liquid crystals as well as functional materials derived therefrom. After that, we discuss the ongoing and foreseeable applications of liquid crystal materials in the biomedical field with emphasis put on several cutting‐edge aspects, including drug delivery, bioimaging, tissue engineering, implantable devices, biosensing, and wearable devices. We hope this review will stimulate ingenious ideas for the future generation of liquid crystal‐based drug development, artificial implants, disease diagnosis, health status monitoring, and beyond. Abstract:The development of a smart microencapsulation system programmed to actively respond to environmental stimuli is gaining increasing importance compared to the traditional forms of drug administration. In this study, we introduce two new concepts of the drug delivery system, based on the light and pH-sensitive liquid crystal (LC) microspheres. The proposed system represents an emulsion formed by the immiscibility between the LC micro-spheres and a water environment. Abstract:New amphiphilic liquid crystal (LC) polycarbonate block copolymers containing side-chain cholesteryl units were synthesized. Their structure, thermal stability, and LC phase behavior were characterized with Fourier transform infrared (FT-IR) spectrum, ¹H NMR, gel permeation chromatographic (GPC), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC), polarizing optical microscope (POM), and XRD methods. The results demonstrated that the LC copolymers showed a double molecular arrangement of a smectic A phase at room temperature. With the elevating of LC unit content in such LC copolymers, the corresponding properties including decomposition temperature (Td), glass temperature (Tg), and isotropic temperature (Ti) increased. The LC copolymers showed pH-responsive self-assembly behavior under the weakly acidic condition, and with more side-chain LC units, the self-assembly process was faster, and the formed particle size was smaller. It indicated that the self-assembly driving force was derived from the orientational ability of LC. The particle size and morphologies of self-assembled microspheres loaded with doxorubicin (DOX), together with drug release tracking, were evaluated by dynamic light scattering (DLS), SEM, and UV-vis spectroscopy. The results showed that DOX could be quickly released in a weakly acidic environment due to the pH response of the self-assembled microspheres. This would offer a new strategy for drug delivery in clinic applications.
