Abstract:Macrocyclic polyamines constitute a significant class of macrocyclic compounds that play a pivotal role in the realm of supramolecular chemistry. They find extensive applications across diverse domains including industrial and agricultural production, clinical diagnostics, environmental protection and other multidisciplinary fields. Macrocyclic polyamines possess a distinctive cavity structure with varying sizes, depths, electron-richness degrees and flexibilities. This unique feature enables them to form specific supramolecular structures through complexation with diverse objects, thereby attracting considerable attention from chemists, biologists and materials scientists alike. However, there is currently a lack of comprehensive summaries on the synthesis methods for macrocyclic polyamines. In this review article, we provide an in-depth introduction to the synthesis of macrocyclic polyamines while analysing their respective advantages and disadvantages. Furthermore, we also present an overview of the recent 5-year advancements in using macrocyclic polyamines as non-viral gene vectors, fluorescent probes, diagnostic and therapeutic reagents as well as catalysts. Looking ahead to future research directions on the synthesis and application of macrocyclic polyamines across various fields will hopefully inspire new ideas for their synthesis and use.

Abstract:A series of novel carbohydrate-based iminophosphinite ligands have been synthesized for the first time and successfully applied in Pd-catalyzed asymmetric allylic alkylation. The substituent effects on the catalytic reaction were also investigated, and 92% ee was achieved when the p-nitro-substituted ligand 10d was used.

Abstract:An efficient synthesis of novel phenoxyimine ligands containing a difluorovinylbiphenyl group was based on the Suzuki coupling coupling of 4-bromo-2,6-difluoroaniline with 4-styrylboronic acid followed by a condensation reaction with various arylaldehydes. The structures were established by IR, 1H NMR, 13C NMR spectra and HRMS. The advantages of this synthetic route were its simple operation, mild reaction conditions and good yields.

Abstract:Poly(α-amino acid)s, containing amino acid repeat units, are remarkable biomaterials with similar structures to protein, which are mainly synthesized by solid phase peptide synthesis, ring opening polymerization of N-carboxyanhydrides, polymerization of activated urethane derivatives of α-amino acids and so on. α-Amino acid N-thiocarboxyanhydride (NTA) and N-substituted glycine N-thiocarboxyanhydride (NNTA) have recently emerged as a subject of scientific interest due to their easy synthesis and purification in open air, long shelf-life and well controlled polymerization to produce poly(α-amino acid)s. In this feature article, we review the history and the recent development on the design of NTA monomers, and summarize the recent advances in poly(α-amino acid)s synthesis through ring-opening polymerization of NTA and NNTA by using different initiators and reaction mediums. We also highlight the applications of poly(α-amino acid) materials resulting from NTA polymerization.

Abstract:A couple of enantiomers of novel chiral macrocyclic imines based on R/S-camphor were asymmetrically synthesized. Through the chiral macrocyclic ligands, enantiomers of nickel(II) coordination compounds were obtained. The chirality of the ligand was transferred to the nickel atom upon coordination, leading to predetermined configuration. The chiral ligands and corresponding nickel coordination compounds were characterized by UV, IR, CD, VCD spectra and single crystal X-ray crystallography. The structural and spectroscopic properties were further investigated by DFT calculations.

Abstract:A series of novel salts of heterocyclic polyamines with 5-sulfosalicylic acid (C4H7N4+)(C7H5O6S−)∙2(H2O) (1), (C4H6ClN4+)(C7H5O6S−)∙H2O (2), (C5H8N3+)(C7H5O6S−)∙H2O (3), (C5H7N6+)(C7H5O6S−)∙H2O (4), (C6H14N22+)(C7H4O6S2−)∙H2O (5), and (C14H19N2+)(C7H5O6S−) (6) have been successfully synthesized. Their crystal structures have been determined by single-crystal X-ray diffraction. Overall, compounds adopt a layered structure with aminium cations and 5-sulfosalicylic anions linked via water molecules. The solid-state architectures of these compounds are dominated by O(N,H)-H⋯O and N-H⋯N hydrogen bonds and stabilized by weak interconnects. C-Cl⋯π and S-O⋯π interactions, apart from π⋯π and C-H(O)⋯π, were reported. Diverse approaches were used to study the effect of substituents in the polyamines in solid-state arrangement. A Hirshfeld surface analysis, with associated 3D Hirshfeld surface maps and 2D fingerprint plots, molecular electrostatic potential, and energy frameworks were used to comprehensively investigate the nature and hierarchy of non-covalent interactions and inspect supramolecular differences. The contact enrichment ratio calculations provided deeper insight into the propensity of interconnects to influence crystal packing. The evaluation of the effects of H-bonding synthons resulting from different substituents in the polyamines on self-assemblies is also presented. In the context of crystal engineering, a specific intramolecular synthon via O-H⋯O observed in nearly all crystals can be employed in the pseudo-cyclic replacement strategy in the design of new molecules.

Abstract:A new family of positively charged and water soluble amino acid-based poly(ester amide)s (PEAs) consisting of nontoxic L-arginine, diols, and aliphatic dicarboxylic acids building blocks was synthesized and characterized. The L-arginine based PEAs (Arg-PEAs) were prepared by a solution polycondensation of two monomers: tetra-p-toluenesulfonic acids salts or hydrochloride acid salts of bis-(L-arginine) alpha, omega-alkylene diesters (monomer II), and di-p-nitrophenyl esters of saturated or unsaturated dicarboxylic acids (monomer I). Optimal reaction conditions were studied as functions of type of solvents and acid acceptors, concentrations of reactants. The molecular weights (Mn and Mw) of Arg-PEAs measured by GPC ranged from 20,000 to 60,000 g mol-1 with a rather narrow molecular weight distribution below 1.5. The chemical structures were confirmed by IR and NMR spectra. Arg-PEAs obtained were all amorphous materials with Tg from 33 to 125 degrees C, depending on the number and the type (saturated vs. unsaturated) of methylene groups in diols or diacids, and the type of counter-ions attached to the guanidine group of the Arg-based PEAs. The Arg-PEAs had a high solubility in all polar solvents, including water. Preliminary studies of cell morphology and DNA capture capability of Arg-PEAs indicated that this new family of cationic PEAs was nontoxic and more biocompatible than a commercial transfection agent (Superfect (R)), and can successfully capture plasma DNA. The strong positive charge of Arg-PEAs as well as their good water solubility could provide unique characteristics for potential gene transfection or other charge preferred biomedical applications. Abstract:In recent years, modulating amphiphilic assembly by macrocyclic receptors has been emerging as a fantastic topic. As we know, amphiphiles possess ability of self-assembly in aqueous solution due to their unique structures, resulting in important biological functions and various applications in our daily life and industry. Among methods to modulate their assembly behavior, using macrocyclic compounds as amphiphile adaptors owns advantages of convenience and efficiency. Preorganized structures and cavities of macrocyclic compounds enable them the ability of efficiently interacting with amphiphiles in different manners. These additional interactions alter amphiphilic assembly. In this review, we would like to provide a systemic summary of why and how macrocyclic compounds could behave as amphiphile adaptors. For this objective, we introduce related works from the viewpoint of intermolecular interaction. In general, addition of interactions which decrease electrostatic reputation of amphiphiles could promote amphiphilic assembly, and vice versa. Addition of interactions which prevent hydrophobic interaction could inhibit amphiphilc assembly. On the other hand, complexation of macromolecular compounds and amphiphiles could cause steric hindrance, which is unfavorable for assembly. We also take several classical amphiphiles as examples to clarify how different macrocyclic compounds adapt amphiphile assembly.

Abstract:Using highly dilute method and 5-fold excess of diamine a new acyclic diamine containing pyridine moiety, N,N′-bis(2-aminoethyl)-2,6-pyridinedicarbamide (1a) was synthesized. Six novel linear polydentate Schiff base ligands containing pyridine and amide moieties, N,N′-bis[β-(R-substitutedbenzalimino)ethyl]-2,6-pyridinedicar-bamide (2a-2e), where R=H, o-OH, p-OH, m-NO2, p-N(CH3)2, and N,N′-bis(γ-salicylideneaminopropyl)-2,6- pyridinedicarbamide (2f) have been synthesized and characterized by elemental analysis, IR, UV-Vis, MS and 1H NMR methods. The single-crystal analysis of 2e showed that it belongs to triclinic system with P-1 space group and the unit cell parameters: a=11.010(2) nm, b=13.865(3) nm, c=9.6537(19) nm, α=102.77(2) °, β=92.07(3)) °, γ=87.98(3)) °, V=1435.7(5) nm3, Z=2, Dc=1.230 mg·cm-3, Mr=531.66. In addition, microcalorimetric test of the inhibitory capacities of these compounds for E. coli has shown that they have antibacterial activity to different extent and that of salicylidene Schiff base is the best. The structure-activity relationship has been primarily discussed.

Abstract:Polyamines in naturally occurring compounds are biologically active molecules that do not display a great diversity but are very abundant in almost all organisms. Their functions comprise antibiotics, immunosuppressants, toxins, and many others. In most biologically active compounds the polyamine chain is coupled to other functionalities making the polyamine an excellent template for solid phase chemistry and combinatorial approaches. This review comprises common techniques for the synthesis of polyamines and polyamine derivatives on solid support. It includes the description of basic synthesis requirements such as of suitable resins, new linkers, reaction conditions for generation of biologically active polyamines and polyamines in natural products as well as the description of protection groups. Moreover, it will focus on the modular coupling of the amine building blocks using different synthesis strategies. Thus, the advantages of performing reactions on polymer-supported substrates in terms of avoiding side reactions and facilitating product/metal separation are highlighted.

Abstract:A one-pot synthetic protocol to access a diverse library of diamide–diester macrocycles from the same starting materials is reported. The molecular symmetry can be readily tuned based on the reaction sequence, while the core structure can be varied using amino acids and aromatic building blocks. The first class of macrocycles with C 2 axes in their molecular plane were obtained in 24 h with 40–70% yield, while another class with C 2 axes perpendicular to the plane were synthesized in 18 h in 10–30% yield. The phenyl- and serine-derived macrocycles of the first class could bind acetate, chloride, and phosphate ions. These macrocycles can be functionalized with hydrophobic groups and potentially be used as ion transporters. Abstract:A novel triphenylamine derivative of 4,4'-bis-[2-[4-[N, N-bis-(4-methoxyphenyl-amino)] phenyl-1-yl]-vinyl-1-yl]-1, 1'-biphenyl (DMPAVBI) was synthesized. The chemical structure was confirmed by IR, H-1 NMR spectroscopy and elemental analysis. Its properties were studied by UV-Vis spectroscopy, photoluminescence spectroscopy and cyclic voltammetry methods.

Abstract:To study the properties of hydrolysates of cyclic adenosine diphosphate ribose (cADPR), a series of novel acyclic analogues of cADPR with an ether chain as the northern moiety and 8-substituted adenine or hypoxanthine as the base moiety were synthesized via an N(1) substitution construction, followed by bisphosphorylation, phosphoramidition or pyrophosphorylation. These compounds also provide various precursors for synthesizing cADPR analogues.
