Abstract:Molecular imprinting technique is a borderline subject which combines the advantages of the subjects of high polymer chemistry, analytical chemistry and science of material. Molecular imprinting technique is a novel technique based on mimicking specification of antibody-antigen. The study of fluorescent molecular imprinting polymer sensors is a significant direction in the development of current sensors with a bright prospect. Fluorescent molecular imprinting polymer sensors combine the recognition ability and predetermined selectivity of molecular imprinting polymers with the high sensitivity of fluorescence analysis. This review summarizes the progress of fluorescent molecular imprinting polymer sensors in recent years. The mechanism of preparation, methods for monitoring binding sites and applications on organic small molecules and ions of fluorescent molecular imprinting polymer sensors are described in detail. Additionally, the development trend of fluorescent molecular imprinting polymer sensors is also discussed.

Abstract:Fluorescence immunoassay technology is widely recognized as the most sensitive detection technology, now widely used in clinical testing. The research of quantitative analyzer for fluorescence immune chromatography strips is of great significance. In this thesis, a quantitative analyzer for fluorescence immune chromatography strips has realized, based on photoelectric detection principle and immunochromatographic. The system uses LPC1788 and MSP430 as processors. Using some algorithms such as low-pass filtering, least square method and area method to analyze the detected waveform and eliminate noise with high accuracy. In our experiment, the minimum detection concentration is at several mIU/ml levels, the relative standard deviation is less than 2% and it has good linear response characteristics in a concentration range from 0.5125 to 952mIU/ml while the correlation coefficient R2 is 0.981. The instrument has a good prospect.

Abstract:In vivo fluorescence molecular imaging plays a more and more important role in the observation of diseases, drug research and biology research because of its low cost, simplicity and no ionizing radiation to biological tissue. Herein, the most important parts of the optical fluorescence molecular imaging and their advances are summarized, including fluorescent molecular probes, imaging systems and reconstruction algorithms. The application and development trend of this technology are also introduced in this paper.

But there are many differences in labeling and immunoassay of nanoparticles from conventional fluorescent labels. This restrains their broad application in diagnosis. The labeling methods, immunological configurations and influence factors of fluorescent nanoparticles in recent years are reviewed.

Abstract:A time-resolved fluorescence immunoassay method based on the quantification of the new europium chelator, ((chlorosulfonyl)thenoyl)trifluoroacetone (CTTA), in the presence of excess EU(3+) in solution with a novel detection principle is described. CTTA-labeled solid-phase immunocomplexes obtained by conventional immunoassay procedures were transferred into solution with a dissociation solution, and then fluorescent complexes were formed. The dissociating solution was optimized and consisted of sodium dodecyl sulfate, Eu3+, and tri-n-octylphosphine oxide, which acted as dissociating reagent, fluorescent lanthanide ion, and enhancement reagent, respectively. The new dissociation solution was very simple and available, and it was applied to a competitive immunoassay of cortisol, which had a detection limit of 0.5 ng/mL of cortisol with a dynamic range up to 2500 ng/mL. The average recovery in urine was 105%.

Abstract:To apply the chemiluminescence immunoassay method in clinical applications, some key technologies were investigated based on deeply analyzing the chemiluminescence immunoassay process. Firstly, three key steps about the immunoassay were indicated: filling-up, washing and measuring. Accordingly, three key technologies were summarized: accurate filling-up of microcontent liquid, quick and effective separation of immune complex, and accurate measurement of weak optical signal. In the aspect of filling-up, based on the analysis and simulation of milliliter level liquid filling-up process, the principle for optimizing relative sampling parameter was obtained. In the aspect of washing, the principle for designing the magnetic field was then obtained based on the analysis of separating immune complex process. In the aspect of measuring, the principles for designing measuring room and processing optical signal were given. After investigating the above-mentioned key technologies, chemiluminescence immunoassay analyzer Autolumis 3000 has been developed and applied in hospital. The results show that the coefficient of variance (CV) is 5% and the maximum speed is 180 time/h. This analyzer can achieve similar performance comparing with the current high-end equipment in abroad, which validates our investigation of the key technologies.

Abstract:Detecting diseases at the molecular level aids in early diagnosis and treatment. However, traditional immunological detection techniques, such as enzyme-linked immunosorbent assay (ELISA) and chemiluminescence, have detection sensitivities between 10 −16 and 10 −12 mol/L, which are inadequate for early diagnosis. Single-molecule immunoassays can reach detection sensitivities of 10 −18 mol/L and can detect biomarkers that are difficult to measure using conventional detection techniques. It can confine molecules to be detected in a small spatial area and provide absolute counting of the detected signal, offering the advantage of high efficiency and accuracy. Herein, we demonstrate the principles and equipment of two single-molecule immunoassay techniques and discuss their applications. It is shown that the detection sensitivity can be improved by 2–3 orders of magnitude compared to common chemiluminescence or ELISA assays. The microarray-based single-molecule immunoassay technique can test 66 samples in 1 h, which is more efficient than conventional immunological detection techniques. In contrast, microdroplet-based single-molecule immunoassay techniques can generate 107 droplets in 10 min, which is more than 100 times faster than a single droplet generator. By comparing the two single-molecule immunoassay techniques, we highlight our personal perspectives on the current limitations of point-of-care applications and future development trends.

Abstract:A novel immunoaffinity column used as an immunosensor for flow-injection chemiluminescent (CL) immunoassay was prepared by immobilizing antibody on carboxylic resin beads. The immunosensor could fast recognize and trap the immunocomplex of horseradish peroxidase (HRP)-labeled antibody and antigen, which was firstly formed with a micro-bubble accelerated pre-incubation process, to produce a sandwich immunocomplex. The HRP introduced in the immunoaffinity column could catalyze the CL reaction to produce enzyme-enhanced emission. With α-fetoprotein (AFP) as a mode, a flow-injection CL immunoassay was proposed. The whole assay for one sample, including the pre-incubation and the regeneration of immunoaffinity column, could be performed within 16min. The linear range was 1.0–80ng/ml with a correlation coefficient of 0.998 and a detection limit of 0.1ng/ml at a signal/noise ratio of 3. The intra- and inter-assay coefficients of variation at 20ng/ml AFP were 1.2% and 8.5%, respectively. The storage stability of the immunoaffinity column and the accuracy for sample detection were acceptable. This flexible, sensitive, low-cost, and rapid method is valuable for clinical immunoassay.

Abstract:A new sensor system based on the total internal reflection fluorescence and the immunoassay was studied. It may be applied for monitoring the organic pollutants in water environment, for example 2, 4-dichlorphenoxy-acetic;Acid and microcystin etc. Cyanine 5.5 (Cy 5.5) dye and He-Ne laser were used in this system. Integrated with flow-injection system, the sensor can detect the immuno-reaction on the transducer rapidly and conveniently. We mainly discussed the detection principle, system structure, modification of the transducer rapidly and conveniently. We mainly discussed the de I transducer, immobilization of ligand and testing of the system. The result of the experiment shows that the system is very stable. Detection limit to Cy 5.5 dye is I nmol/L. The modified transducer have little non-specific adsorption to protein, and response to a series of concentration of antibody fit the logistic formula. The same transducer was used for more than 20 test cycles without much loss of performance.

Abstract:Time-resolved fluorescent nanospheres (TRFNs) are polymer nanospheres with special functional groups on the surface loaded with rare earth complexes which emit fluorescence when irradiated by ultraviolet light. TRFNs offer spherical morphology, homogeneity, good dispersity, large specific surface area, easy chemical modification, high loading capacity and good biocompatibility. The rare earth complexes possess unique properties that render them highly sensitive and capable of effectively excluding unspecific fluorescence. Hence, they constitute an excellent labeling material for the time-resolved fluorescence immunochromatographic assay (TRFIA). The developed method has a linear response to ferritin from 6 to 1000 ng/mL with a detection limit of 3.5 ng/mL, good reproducibility, and good accuracy compared with commercial chemiluminescence measurements of serum. These results suggest that the reported TRFN has utility for time-resolved fluorescence immunochromatographic assays.

A New Method of Total Urinary Testosterone Determination with a Highly Sensitive Time-resolved Fluorescence Immunoassay

Abstract:Objective To develop a method of highly sensitive and accurate time-resolved fluorescence immunoassay(TrFIA) for total urinary testosterone determination in our laboratory.Methods High affinity anti-testosterone antibody and Eu3+ labeled monkey anti-sheep IgG as tracer were used to develop the TrFIA method.The assay was evaluated for specificity,sensitivity,recovery rate,precision and dilution linearity on 260 urine specimens and the results compared with those from conventional RIA.Results A satisfactory standard curve for testosterone TrFIA was developed with good sensitivity(5.1pmol/L).Specificity,recovery rate,precision and dilution linearity studies were determined and all found to be satisfactory.Male urinary total testosterone excretion ranged from 40 to 233.82 nmol/L,which was about 4 times of the value for women(9.44~67.1 nmol/L).The validity of urinary testosterone assay was confirmed by the good correlation between the results obtained by TrFIA(X) and conventional RIA(Y)(Y=0.075+0.971X,r=0.992).Conclusion A rapid,reliable,easy to automate,highly sensitive and specific TrFIA type assay for the measurement of total testosterone in urine specimens was developed originally in our laboratory.The advantage of this novel assay is that it does not require an initial extraction step nor involve time consuming procedures such as chromatography.

Abstract:Traditionally,polymerase is assayed by denaturing gel electrophoresis and autoradiography,which are complex and discontinuous. Based on the fact that the dye SYBR Green I can intercalate double-stranded DNA to emit fluorescence,we report a new fluorescence-based DNA polymerase assay that can translate the polymerization process into a fluorescent signal in real-time. Comparatively,the method is simple,convenient,less time consuming and inexpensive without the need of gel electrophoresis,radial labeling,fluorescent labeling and polymerase chain reaction (PCR). Based on this assay,the influence of anti-tumour drugs on the activity of DNA polymerase is investigated. This method will provide a potential in diagnosis and therapy of disease and selecting drugs.

Abstract:Abstract Immunoassay is a powerful technique that uses highly specific antigen‐antibody interactions to detect biochemical targets such as proteins and toxins. As a diagnostic tool, immunoassay is employed in the screening, diagnosis, and prognosis of diseases, which are crucial for the grasp and control of patient conditions in clinical practice. With the rapid development of nanotechnology, immunoassays based on nanoprobes have attracted more and more attention due to the advantages of high sensitivity, specificity, stability, and versatility. These nanoprobes are nanoscale particles that can act as signal carriers or targeting agents for immunoassays. In this paper, we review the recent advances in various types of nanoprobes for immunoassays, such as colloidal gold, quantum dots, magnetic nanoparticles, nanozymes, aggregation‐induced emission, and up‐conversion nanoparticles. The effect of the nanoprobe construction and synthesis methods on their detection performance deserves to be studied in depth. We also compare their detection ranges and limits in different immunoassay methods, such as lateral flow immunoassays, fluorescent immunoassays, and surface‐enhanced Raman scattering immunoassays. Moreover, we discuss the benefits and challenges of nanoprobes in immunoassays and provide insights into their future development. This study aims to offer a comprehensive and critical perspective on the role of nanoprobes in the field of immunoassays.

Abstract:Time-resolved fluorescence has been developed for immunoassay to obtain higher sensitivity than usual immunoassays. In this paper, a simple, sensitive and specific method was developed for immunoassay of serum carcinoembryonic antigen (CEA) by combining time-resolved fluoroimmunoassay (TRFIA) and flow injection analysis. Based on a sandwich immunoassay format, a monoclonal antibody immobilized immunoaffinity column inserted in a flow system was used for immunoreactions. The cleaved solution was collected after the reaction between the immunocomplex in the immunoaffinity column and the enhancement solution that was used to cleave the Eu-labels from the immunocomplex, and then detected by time-resolved fluorescence. Serum CEA could be detected in the linear range from 2.5 to 100 ng/ml with a correlation coefficient of 0.997 and the detection limit of 1.0 ng/ml. Twenty human serum samples detected by this method were in good agreement with the results obtained by the electrochemiluminescence immunoassay. This method could be further developed for fast practical clinical detection of serum CEA levels.
Novel Fluorescent Europium Chelate-Doped Silica Nanoparticles: Preparation, Characterization and Time-Resolved Fluorometric Application

Abstract:Novel fluorescent europium(III) chelate-doped silica nanoparticles were prepared and characterized as a new type of fluorescence probe for quantitative bioassay. The preparation was carried out in a water-in-oil (w/o) microemulsion consisting of a strongly fluorescent Eu3+ chelate, 4,4′-bis(1″,1″,1″,2″,2″,3″,3″-heptafluoro-4″,6″-hexanedion-6″-yl)-o-terphenyl–Eu3+ (BHHT–Eu3+), surfactant (Triton X-100), co-surfactant (n-hexanol, n-heptanol or n-octanol), aqueous phase (H2O or D2O) and oil phase (cyclohexane) by controlling the hydrolysis of tetraethylorthosilicate (TEOS). The effects of different co-surfactants and aqueous phases on the size and fluorescence lifetime of the nanoparticles were investigated. The results reveal that the size of the nanoparticles is decreased with a change of co-surfactants from n-hexanol to n-octanol, and the fluorescence lifetime of the nanoparticles is increased with a change of aqueous phase from H2O to D2O. A new method was established for the surface modification and bioconjugation of the nanoparticles. Nanoparticle-labeled streptavidin (SA) was used for the time-resolved fluoroimmunoassay of human hepatitis B surface antigen (HBsAg). The result shows that the new fluorescent europium(III) chelate-doped silica nanoparticles are suitable to be used as a fluorescence probe for highly sensitive bioassays.

Abstract:In this work, we propose fluorescence and scattering light cross-correlation spectroscopy (FSCCS) based on laser confocal configuration using silver nanoparticle (SNPs) and Alexa Fluor 488 (Alexa) as probe pairs. FSCCS is a single molecule (particle) method, and its principle is similar to that of fluorescence cross-correlation spectroscopy (FCCS). We established the setup of FSCCS using single wavelength laser and developed an immunoassay model of FSCCS. The reliability and adaptability of FSCCS method were evaluated by homogeneous sandwich immunoassay mode. In the study, liver cancer biomarker alpha-fetoprotein (AFP) was used as an assay model, two different antibodies were labeled with SNPs and fluorophore Alexa Fluor 488, respectively. In the optimal conditions, the linear range of AFP covers 5 pM to 580 pM and the detection limit is 3.1 pM. This method was successfully applied for direct determination of AFP levels in human serum samples, and the obtained results were in good agreement with data obtained via ELISAs. The advantage of this method lies in its simplicity, attractive SNPs probes, high sensitivity and selectivity and high efficiency. We believe that FSCCS method exhibits promising potential applications in homogeneous bioassays and study on the molecular interaction and nanoparticle-molecule interaction.

Abstract:Determination of low-abundance proteins in body liquid is the significant strategy for the early diagnosis of diseases and the discovery of signal paths in pathobiology. Here, the limit of detection of 113 aM for model human IgA is successfully obtained by the fluoroimmunoassay developed on an optical soda lime glass microsphere in micrometers that is illumed by the microsphere amplified fluorescence (MAF), which can be identified to the most sensitive immunoassay so far. In this case, the fluorescence signal of fluoroimmunoassay can be physically amplified by MAF while the large reacting area will be supplied by the glass microsphere to complete the fluoroimmunoassay. Therefore, the assay time of the fluoroimmunoassay can be shortened to 12 min via the optical glass microsphere-enabled ultrasensitive fluoroimmunoassay at the same time. The developed fluoroimmunoassay indicates a possibility to perform the ultrasensitive immunoassay on the optical glass microsphere in 10 min and more, which may greatly facilitate the development of the ultrasensitive immunoassay for the urgent measurements and promote the development of portable fluoroimmunoassay.
