The Study of Stirring Influence Factors on Pteris Vittata L. Spore Solution for Sowing Operation Based on CFD

Abstract:In view of the current low efficiency and high labor intensity of Pteris Vittata L. artificial sowing, a kind of automatic sowing device was designed. In order to improve the sowing uniformity of the sowing device, the multiple reference frame (MRF) method and the Euler-Euler model were used to simulate the suspension characteristics of the spore particles in the stirred tank. Then, the effects of blade numbers, blades spacing and stirring speed on particle velocity distribution and particle mixing were obtained. The simulation results showed that with the increase of blade numbers, the fluid velocity increased, and the solid-liquid mixing efficiency improved. In the process of blades spacing increased from 50mm to110mm, the movement trend of fluid changed from the overall axial circulation flow to the layered circulation flow, which had negative effect on the overall mixing in the stirred tank. In addition, larger stirring speed could widen the movement range of the spore particles, which is helpful for particle mixing, but the power consumption increased significantly. This study could provide theoretical references for the design and application of micro seeds sowing device.

Abstract:Fern spore germination gives rise to the rhizoid and protonemal cell through asymmetric cell division, and then develops into a gametophyte. Spore germination is also a representative single-cell model for the investigation of nuclear polar movement, asymmetrical cell division, polarity establishment and rhizoid tip-growth. These processes are affected by various environmental factors, such as light, gravity, phytohormones, metal ions, and temperature. Here, we present a catalog of spore germination in response to different environmental factors. They are as follows: (1) Representative modes of light affecting spore germination from different fern species include red light-stimulated and far red light-inhibited spore germination, far red light-uninhibited spore germination, blue light-inhibited spore germination, and spore germination in the dark. The optimal light intensity and illumination time for spore germination are different among various fern species. Light response upon spore germination is initiated from the cell mitosis that regulated by phytochromes (PHYs) and cryptochromes (CRYs). AcPHY2, AcCRY3 and/or AcCRY4 are hypothesized to be involved in spore germination; (2) Gravity and calcium are crucial to early nuclear movement and polarity establishment of spores; (3) Gibberellin and antheridiogen can initiate and promote spore germination in many species, but abscisic acid, jasmonic acid, and ethylene pose only minor effects; (4) Spores can obtain the maximal germination rate in their favorable growth medium. Moreover, metal ions, pH, and spore density in the culture medium also affect spore germination; (5) Most fern spores germinate at 25°C, and an optimal CO2 concentration is necessary for spore germination of certain fern plants. These provide valuable information for understanding fern spore germination in response to environmental factors.
A Comparative Researches on Seed Germination of 15 Plant Species from Alpine Meadow

Abstract:Germination test of 15 alpine plant species showed that stratification, temperature fluctuation and light conditions could improve or decrease germination percentage of the most of alpine meadow plant species. In which, 13 plant species responded to stratification significantly, 11 species responded to light conditions, and 14 plant species responded to temperature fluctuations. Of all the 15 plant species, single factor or combinations of factors had significant effects on germination of 14plant species. Aster flaccidus is the only species that did not response to any factor or combination of factors. Herein seeds of alpine plant species could be categorized to different response types according to their germination performance under different conditions. Study is essential to understand the germination behavior of plant species under specific conditions for the restoration of degraded alpine meadow communities from seed regeneration.

The work done on the dormancy of algal spores and on the role of vegetative cells in tolerating environmental stress is also incorporated.

Abstract:This paper compared the methods of anther and microspore-isolated cultures in Chinese cabbage.The results showed that the difference of genotype between varieties was the key factor affecting production of the pollen embryo.The embryo inducement efficiency of microspore-isolated culture was 10 times greater than that of pollen culture.But the regeneration rate of pollen plants of microspore-isolated culture was much lower than that of pollen culture.Many key techniques of microspore-isolated culture should be improved in order to widely apply to breeding work.

Abstract:Pollen germination is a prerequisite for double fertilization of flowering plants. A comprehensive understanding of the structural and molecular basis of pollen germination holds great potential for crop yield improvement. The pollen aperture serves as the foundation for most plant pollen germination and pollen aperture formation involves the establishment of cellular polarity, the formation of distinct membrane domains, and the precise deposition of extracellular substances. Successful pollen germination requires precise material exchange and signal transduction between the pollen grain and the stigma. Recent cytological and mutant analysis of pollen germination process in Arabidopsis and rice has expanded our understanding of this biological process. However, the overall changes in germination site structure and energy-related metabolites during pollen germination remain to be further explored. This review summarizes and compares the recent advances in the processes of pollen aperture formation, pollen adhesion, hydration, and germination between eudicot Arabidopsis and monocot rice, and provides insights into the structural basis and molecular mechanisms underlying pollen germination process.

Abstract:Germination is a key event in plant life cycle and imbibition temperature is an important factor for the major reorganization pro- cesses in the germinating seeds. Sub-physiological temperature at this stage affects virtually all aspects of cellular function including protein folding, kinetic parameters of membrane fluidity, protein assembly, and general metabolic processes. In this article we will review the re- search which has been carried out to decipher the basic mechanisms of low temperature (LT) stress with special emphasis on germination. With the better understanding of LT tolerance in some plant species we will also discuss how these attributes can be transferred in the im- portant food crops to attain better germination stamina at sub-physiological temperatures. At germination stage, the cellular machinery in the embryo is not at its proper place and is going through reorganisation and any environmental severity has devastating effects on the fragile plant. But LT resistant species have evolved the ability to acclimatise, with the remodelling of cell and tissue structures and the reprogram- ming of metabolism and gene expression. Metabolic networks are redirected towards the synthesis of cryoprotectant molecules, which in association with other proteins bring about physical and biochemical restructuring of cell membranes through changes in the lipid composi- tion and induction of other non-enzymatic proteins that alter the freezing point of water. Genetic engineering of crops for enhanced seed germination performance will certainly help to achieve optimum agricultural yields..
Seed Germination Ecology of Semiparasitic Weed Pedicularis kansuensis in Alpine Grasslands

Abstract:The semiparasitic weed Pedicularis kansuensis Maxim. has rapidly spread in the alpine grasslands of northern China over the past twenty years and has caused serious ecological problems. In order to effectively halt the spread of this weed, a thorough understanding of the dormancy type and the seed-germination ecology of P. kansuensis is required. We have conducted a series of experiments to investigate the effects of plant growth regulators (gibberellin (GA3) and strigolactone synthesis (GR24)), as well as different abiotic (temperature, light, cold stratification, and drought) and biotic (aqueous extracts of three native dominant plants) factors on the seed-germination characteristics of P. kansuensis. The seed-germination percentages ranged from 2% to 62% at all of the temperatures that were examined, with the highest occurring at 25/10 °C. The light conditions did not significantly affect the germination percentage. The seed germination was greatly improved after two to eight weeks of cold stratification. The seed germination decreased dramatically with an increasing polyethylene glycol (PEG-6000) concentration, from 55% to 0%, under 10% and 20% PEG-6000. The seed germination was improved at a proper concentration of GA3, GR24, and the aqueous extracts of Festuca ovina L., Stipa purpurea L., and Leymus secalinus (Georgi) Tzvel. Furthermore, in the pot experiment, the seedling emergence of P. kansuensis was also improved by the cultivation of these three dominant grasses. These findings indicate that the dormancy type of P. kansuensis seeds is non-deep physiological dormancy, and such findings will help in paving the way for the creation of effective weed management strategies, based on a thorough knowledge of germination ecology.

Abstract:Seed dormancy and germination play pivotal roles in the agronomic traits of plants, and the degree of dormancy intuitively affects the yield and quality of crops in agricultural production. Seed priming is a pre-sowing seed treatment that enhances and accelerates germination, leading to improved seedling establishment. Seed priming technologies, which are designed to partially activate germination, while preventing full seed germination, have exerted a profound impact on agricultural production. Conventional seed priming relies on external priming agents, which often yield unstable results. What works for one variety might not be effective for another. Therefore, it is necessary to explore the internal factors within the metabolic pathways that influence seed physiology and germination. This review unveils the underlying mechanisms of seed metabolism and germination, the factors affecting seed dormancy and germination, as well as the current seed priming technologies that can result in stable and better germination.

Abstract:Seed germination is a critical process in the life cycle of higher plants. During germination, the imbibed mature seed is highly sensitive to different environmental factors.However, knowledge about the molecular and physiological mechanisms underlying the environmental effects on germination has been lacking. Recent proteomic work has provided invaluable insight into the molecular processes in germinating seeds of Arabidopsis, rice (Oryza sativa), soybean (Glycine max), barley (Hordeum vulgare), maize (Zeamays), tea (Camellia sinensis), European beech (Fagus sylvatica), and Norway maple (Acer platanoides) under different treatments including metal ions (e.g. copper and cadmium), drought, low temperature, hormones, and chemicals (gibberellic acid, abscisic acid, salicylic acid, and α-amanitin), as well as Fusarium graminearum infection. A total of 561 environmental factor-responsive proteins have been identified with various expression patterns in germinating seeds. The data highlight diverse regulatory and metabolic mechanisms upon seed germination, including induction of environmental factor-responsive signaling pathways, seed storage reserve mobilization and utilization, enhancement of DNA repair and modification, regulation of gene expression and protein synthesis, modulation of cell structure, and cell defense. In this review, we summarize the interesting findings and discuss the relevance and significance for our understanding of environmental regulation of seed germination.

Study on the Seed Germination Experiment of Pediculariss spicata Pall.

Abstract:[Objective] The aim was to research the method of improving the seed germination percentage of Pedicularis spicata Pall.[Method] With the P.spicata seeds as the tested materials,they were taken with the H2SO4(dense) liquor treatment,the water soaking treatment and stratification treatment for the study on the experiment of seed germination percentage of P.spicata.[Result] With the increase of H2SO4(dense) concn,the germination percentage of P.spicata seeds was decreased gradually.The germination percentage of P.spicata seeds was highest with the water soaking treatment under the normal temperature(20 ℃).The seed germination percentage had not large difference under 10-50 ℃,but that was decreased greater under 50-60 ℃ when the seeds were treated with the water soaking treatment.The germination percentage was highest under the normal temperature with the leach treatment for 24 h,reaching 73%.When the seed soaking exceed 60 h,the germination percentage was decreased obviously.The effect of cold stratification treatment for 60 d was remarkable and the germination percentage could reach 85%.[Conclusion] The treatment effect of H2SO4(dense) liquor was not obvious and the operation had some difficulty;the effect of water soaking treatment was greater;the stratification treatment for 60-90 d could get the germination percentage of 85%-86%,which was a accelerating germination method of safe,effective,low cost and easy operation.

Abstract:We investigated the factors influencing teliospore germination of Ustilago esculenta,such as surface characters,temperature,pH value and stimulants.The results showed that hydrophobic surfaces promoted the germination of teliospore,and the germination rates were higher on the hydrophobic polyethylene Petri dishes and surfaces of stem epidermis of Zizania latifolia.The highest germination rate occurred at 27 to 30??,whereas it was inhibited by higher or lower temperature.The acidic condition promoted the germination of teliospore,with the optimum pH value of 3 to 4,and sucrose with the concentration of 1% was best fit for the germination of teliospore,but other carbon sources,such as mannitol,glucose,trehalose,D-fructose,inhibited teliospore germination.

Abstract:Spores are highly resistant dormant cells, adapted for survival and dispersal, that can withstand unfavourable environmental conditions for extended periods of time and later reactivate. Understanding the germination process of microbial spores is important in numerous areas including agriculture, food safety and health, and other sectors of biotechnology. Microfluidics combined with high-resolution microscopy allows to study spore germination at the single-cell level, revealing behaviours that would be hidden in standard population-level studies. Here, we present a microfluidic platform for germination studies where spores are confined to monolayers inside microchambers, allowing the testing of four growth conditions in parallel. This platform can be used with multiple species, including non-model organisms, and is compatible with existing image analysis software. In this study, we focused on three soil dwellers, two prokaryotes and one fungus, and revealed new insights into their germination. We studied endospores of the model bacterium Bacillus subtilis and demonstrated a correlation between spore density and germination in rich media. We then investigated the germination of the obligate-oxalotrophic environmental bacterium Ammoniphilus oxalaticus in a concentration gradient of potassium oxalate, showing that lower concentrations result in more spores germinating compared to higher concentrations. We also used this microfluidic platform to study the soil beneficial filamentous fungus Trichoderma rossicum, showing for the first time that the size of the spores and hyphae increase in response to increased nutrient availability, while germination times remain the same. Our platform allows to better understand microbial behaviour at the single-cell level, under a variety of controlled conditions.

Abstract:Germination inhibitors, which inhibit the germination of seeds, spores and other plant reproductive material, are abundant in the plant kingdom and include phenols, cyanides, alkaloids, essential oils, amino acids, etc. These inhibitors can be classified as germination destructors and germination retarders depending on whether they harm the morphology, structure and physiology of the seed. Germination retarders are closely related to seed dormancy, and exogenous retarders can be used to extend the “dormancy” period of non-dormant seeds or perishable seeds by applying the proper dosage. They have significant potential applications as preservatives for seed preservation following harvest or for the storage of long-term germplasm resources. Germination destructors, as a type of relatively high-efficiency, low-specificity “toxic chemicals”, are of significant benefit in the application of effective and environmentally benign herbicides. At present, the main problems related to the research methods of germination inhibitors include difficulty in determining the specific endogenous substances and the minimum inhibitory concentration to induce dormancy, as well as whether the application of exogenous inhibitors will cause physiological damage to seeds. In the future, we should strengthen the tracking of germination inhibitors, explore the mechanisms of action of specific substances and deeper molecular mechanisms and finally explore new developments and new applications of different inhibitors.

The importance of mistakes: Variation in spores reveals trade-offs for range expansion in the xeric-adapted Australasian species (Pteridaceae)

Abstract:Biological trade-offs present a central issue for evolutionary biology: it has been a fundamental understanding within the field that limits exist on the phenotypic traits a species is able to exhibit in part due to trade-offs. Reproduction-with its myriad forms-has been studied extensively in the context of these dynamics. And while considerable literature has explored trade-offs between seed size and number and their associated environmental conditions, none has looked at spore size trade-offs in ferns. We can hypothesise potential trade-offs in spore size: smaller spores should be able to disperse farther, but may not have sufficient provisions to survive in environments that require them to remain at the gametophyte stage for longer periods if their germination cues are mismatched. Reproductive mode (sexual vs. asexual) and ploidy may also be playing a role. In order to study trade-offs related to spore size, I focus on the Australasian fern species Cheilanthes distans (Pteridaceae), which is most often found in xeric environments, growing in crevices or on top of rocks which are haphazardly scattered across their range. Apomictic diplospores in this species are formed through first division restitution, a meiotic pathway particularly prone to mistakes in chromosome pairing and cell division (as compared to premeiotic endomitosis). Rather than being problematic, these mistakes ultimately lead to considerable additional variation in spore size, spore products (through a range of aneuploid spores), and spore ploidy. In this study, I explore trade-offs between spore size, dispersal, and germination, taking into account effects from reproductive mode and ploidy. I carried out an extensive survey of C. distans specimens to establish the prevalence of sexual vs. apomictic (asexual) specimens, and to describe in greater depth the variation in ploidy across the species. I also collected data on spore size and sporogenesis forms. With these data I then asked: is spore size correlated with range area or with germination? And does spore form correlate with either spore size or germination? Ultimately, I find that variations in sporogenesis may be leading to large variation in spore sizes-especially since spores traditionally considered abortive are in fact viable-and that this variation may provide abundant fodder for evolution to act through trade-offs between dispersal into large ranges and germination leading to establishment. Especially in light of the fact that many spores that were historically considered abortive are fully viable and likely shaping evolution in important ways, it is worth remarking on what these results illustrate more broadly: the way in which we have constructed 'disability' ultimately affects how we perceive so-called 'genetic errors'-both in humans and in other species-and thus limits what we allow ourselves to imagine 'disabled' beings are capable of.
