Abstract:The potato (Solanum tuberosum L.) is one of the most important food crops worldwide. Nevertheless, due to food safety concerns, substantial amounts of potatoes are not utilized, resulting in losses. Dormancy development is affected by potato variety, temperature, light, humidity, and so forth. Traditional and novel techniques to detect potato dormancy have been developed based on infrared spectroscopy, hyperspectral, and multispectral imaging systems in combination with simulation verification. This review describes the ability of novel approaches to address the uncertain hazardous issue and delay or inhibit potato dormancy incidence and development. Further, this review describes the role of carbohydrate metabolism and phytohormone regulation involved in potato dormancy, the mechanism of action to address dormancy, and challenges and recommendations moving forward. More work is needed to develop novel approaches toward addressing the problem of potato germination after harvest.
Genes Controlling Seed Dormancy and Pre-Harvest Sprouting in A Rice-Wheat-Barley Comparison

Abstract:Pre-harvest sprouting results in significant economic loss for the grain industry around the world. Lack of adequate seed dormancy is the major reason for pre-harvest sprouting in the field under wet weather conditions. Although this trait is governed by multiple genes it is also highly heritable. A major QTL controlling both pre-harvest sprouting and seed dormancy has been identified on the long arm of barley chromosome 5H, and it explains over 70% of the phenotypic variation. Comparative genomics approaches among barley, wheat and rice were used to identify candidate gene(s) controlling seed dormancy and hence one aspect of pre-harvest sprouting. The barley seed dormancy/pre-harvest sprouting QTL was located in a region that showed good synteny with the terminal end of the long arm of rice chromosome 3. The rice DNA sequences were annotated and a gene encoding GA20-oxidase was identified as a candidate gene controlling the seed dormancy/pre-harvest sprouting QTL on 5HL. This chromosomal region also shared synteny with the telomere region of wheat chromosome 4AL, but was located outside of the QTL reported for seed dormancy in wheat. The wheat chromosome 4AL QTL region for seed dormancy was syntenic to both rice chromosome 3 and 11. In both cases, corresponding QTLs for seed dormancy have been mapped in rice.

Abstract:Seed dormancy is an important trait for the analysis and manipulation of pre-harvest sprouting resistance in rice. This study presents data on dissection of the genetic basis of the seed dormancy character in the indica cultivar N22, which exhibits levels of dormancy highly in excess of traditional rice cultivars. Three simple sequence repeat (SSR)-based linkage maps derived from two BC1-type populations of Nanjing35 (a japonica non-dormant breeding line)/N22//Nanjing35 and USSR5 (non-dormant japonica cultivar)/N22//USSR5, and one F2 population of USSR5/N22, were constructed to detect genes controlling seed dormancy. Five quantitative trait loci (QTLs) for seed dormancy, i.e., qSdn-1, qSdnj-3, qSdn-5, qSdn-7 and qSdn-11, were identified on chromosomes 1, 3, 5, 7 and 11, respectively. The N22-derived alleles were observed to increase the seed dormancy at all the loci except qSdnj-3. Genes conferring seed dormancy in N22 showed incomplete dominance effect. Two QTLs, qSdn-5 and qSdn-11 were detected in the two BC1 populations, while qSdnj-3 and qSdn-7 were only identified in the one BC1 population (Nanjing35/N22//Nanjing35), whereas the QTL qSdn-1 was common in all the three populations. The dormancy trait was expressed whenever any of the dormancy genes presents and can be further improved by pyramiding these genes. Of all QTLs, qSdn-1 provided the most obvious target for marker-assisted selection (MAS) due to its stability for improving rice pre-harvest sprouting tolerance.

Abstract:Seed dormancy in rice is an important trait related to the pre-harvest sprouting resistance. In order to understand the molecular mechanisms of seed dormancy, gene expression was investigated by transcriptome analysis using seeds of the strongly dormant cultivar N22 and its less dormant mutants Q4359 and Q4646 at 24 days after heading (DAH). Microarray data revealed more differentially expressed genes in Q4359 than in Q4646 compared to N22. Most genes differing between Q4646 and N22 also differed between Q4359 and N22. GO analysis of genes differentially expressed in both Q4359 and Q4646 revealed that some genes such as those for starch biosynthesis were repressed, whereas metabolic genes such as those for carbohydrate metabolism were enhanced in Q4359 and Q4646 seeds relative to N22. Expression of some genes involved in cell redox homeostasis and chromatin remodeling differed significantly only between Q4359 and N22. The results suggested a close correlation between cell redox homeostasis, chromatin remodeling and seed dormancy. In addition, some genes involved in ABA signaling were down-regulated, and several genes involved in GA biosynthesis and signaling were up-regulated. These observations suggest that reduced seed dormancy in Q4359 was regulated by ABA-GA antagonism. A few differentially expressed genes were located in the regions containing qSdn-1 and qSdn-5 suggesting that they could be candidate genes underlying seed dormancy. Our work provides useful leads to further determine the underling mechanisms of seed dormancy and for cloning seed dormancy genes from N22.
Current Insights into Weak Seed Dormancy and Pre-Harvest Sprouting in Crop Species

Abstract:During the domestication of crops, seed dormancy has been reduced or eliminated to encourage faster and more consistent germination. This alteration makes cultivated crops particularly vulnerable to pre-harvest sprouting, which occurs when mature crops are subjected to adverse environmental conditions, such as excessive rainfall or high humidity. Consequently, some seeds may bypass the normal dormancy period and begin to germinate while still attached to the mother plant before harvest. Grains affected by pre-harvest sprouting are characterized by increased levels of α-amylase activity, resulting in poor processing quality and immediate grain downgrading. In the agriculture industry, pre-harvest sprouting causes annual economic losses exceeding USD 1 billion worldwide. This premature germination is influenced by a complex interplay of genetic, biochemical, and molecular factors closely linked to environmental conditions like rainfall. However, the exact mechanism behind this process is still unclear. Unlike pre-harvest sprouting, vivipary refers to the germination process and the activation of α-amylase during the soft dough stage, when the grains are still immature. Mature seeds with reduced levels of ABA or impaired ABA signaling (weak dormancy) are more susceptible to pre-harvest sprouting. While high seed dormancy can enhance resistance to pre-harvest sprouting, it can lead to undesirable outcomes for most crops, such as non-uniform seedling establishment after sowing. Thus, resistance to pre-harvest sprouting is crucial to ensuring productivity and sustainability and is an agronomically important trait affecting yield and grain quality. On the other hand, seed color is linked to sprouting resistance; however, the genetic relationship between both characteristics remains unresolved. The identification of mitogen-activated protein kinase kinase-3 (MKK3) as the gene responsible for pre-harvest sprouting-1 (Phs-1) represents a significant advancement in our understanding of how sprouting in wheat is controlled at the molecular and genetic levels. In seed maturation, Viviparous-1 (Vp-1) plays a crucial role in managing pre-harvest sprouting by regulating seed maturation and inhibiting germination through the suppression of α-amylase and proteases. Vp-1 is a key player in ABA signaling and is essential for the activation of the seed maturation program. Mutants of Vp-1 exhibit an unpigmented aleurone cell layer and exhibit precocious germination due to decreased sensitivity to ABA. Recent research has also revealed that TaSRO-1 interacts with TaVp-1, contributing to the regulation of seed dormancy and resistance to pre-harvest sprouting in wheat. The goal of this review is to emphasize the latest research on pre-harvest sprouting in crops and to suggest possible directions for future studies.

Abstract:Pre-harvest sprouting (PHS), the germination of seeds on the plant prior to harvest, poses significant challenges to agriculture. It not only reduces seed and grain yield, but also impairs the commodity quality of the fruit, ultimately affecting the success of the subsequent crop cycle. A deeper understanding of PHS is essential for guiding future breeding strategies, mitigating its impact on seed production rates and the commercial quality of fruits. PHS is a complex phenomenon influenced by genetic, physiological, and environmental factors. Many of these factors exert their influence on PHS through the intricate regulation of plant hormones responsible for seed germination. While numerous genes related to PHS have been identified in food crops, the study of PHS in vegetable crops is still in its early stages. This review delves into the regulatory elements, functional genes, and recent research developments related to PHS in vegetable crops. Meanwhile, this paper presents a novel understanding of PHS, aiming to serve as a reference for the study of this trait in vegetable crops.

Abstract:Seed dormancy and germination play pivotal roles in the agronomic traits of plants, and the degree of dormancy intuitively affects the yield and quality of crops in agricultural production. Seed priming is a pre-sowing seed treatment that enhances and accelerates germination, leading to improved seedling establishment. Seed priming technologies, which are designed to partially activate germination, while preventing full seed germination, have exerted a profound impact on agricultural production. Conventional seed priming relies on external priming agents, which often yield unstable results. What works for one variety might not be effective for another. Therefore, it is necessary to explore the internal factors within the metabolic pathways that influence seed physiology and germination. This review unveils the underlying mechanisms of seed metabolism and germination, the factors affecting seed dormancy and germination, as well as the current seed priming technologies that can result in stable and better germination.

Abstract:Seeds are unique time capsules that can switch between 2 complex and highly interlinked stages: seed dormancy and germination. Dormancy contributes to the survival of plants because it allows to delay germination to optimal conditions. The switch between dormancy and germination occurs in response to developmental and environmental cues. In this review we provide a comprehensive overview of studies that have helped to unravel the molecular mechanisms underlying dormancy and germination over the last decades. Genetic and physiological studies provided a strong foundation for this field of research and revealed the critical role of the plant hormones abscisic acid and gibberellins in the regulation of dormancy and germination, and later natural variation studies together with quantitative genetics identified previously unknown genetic components that control these processes. Omics technologies like transcriptome, proteome, and translatomics analysis allowed us to mechanistically dissect these processes and identify new components in the regulation of seed dormancy and germination.

Abstract:The mung bean, or Vigna radiata (L.) Wilczek, is a significant grain legume with enormous global economic value. Kharif is the most significant of the three seasons and during this time pre-harvest sprouting (PHS) is a major issue that results in significant production losses. Due to PHS, yield losses in green gram have been estimated to be between 60 and 70 per cent. Chemical treatments that induce seed dormancy may prevent premature germination and enable a timely harvest. This study covers the effectiveness of inducing dormancy in mung bean plants with various chemicals at critical developmental stages before harvest. Treatment with various chemicals like maleic hydrazide, sodium azide and plant growth regulators (abscisic acid, ethylene, gibberellins) following chemical administration at pre-arranged intervals, physiological parameters such as seed moisture content, germination rates and antioxidant enzyme activities can be considered to prevent PHS. Mitigating pre-harvest sprouting losses and increasing mung bean output can be achieved by optimising dormancy induction by prudent chemical use. To increase mung bean output, PHS-tolerant cultivars must be developed due to the significant losses incurred by PHS. This review discusses various aspects, including how different traits affect pre-harvest sprouting.

Abstract:We provided a gene pool of moderate size for selecting or manipulating the candidate genes that favour the acquisition of seed dormancy, shedding light on the elevation of seed oil content in oilseed rape by blocking lipid degradation in developing seeds. In oilseed rape, the association between the germination potential of premature seeds and the final level of seed lipids, and the underlying mechanism, is elusive. Here, we investigated phenotypic differences in the germination percentage of premature seeds in a collection of oilseed rape cultivars. We compared the dynamic lipid accumulation between the deep-, moderate- and non-dormant genotypes and compared the transcriptomes of the seeds at 40 days after pollination between multiple pairs of deep- and non-dormant genotypes. We identified a wide range of differences in germination percentage of premature seeds and the association between the germination potential and the change of fatty acid content at late stage of seed maturation. The comparisons of transcriptomes between deep- and non-dormant seeds revealed the genetic basis for the dormant difference, e.g. the different expression levels of the genes involved in gibberellic and abscisic acid biosynthesis and/or signalling, fatty acid metabolic pathways, and the structure of seed cell wall. We provided a gene pool of moderate size for selecting or manipulating the candidate genes that favour the acquisition of seed dormancy, shedding light on the elevation of seed oil content in oilseed rape by blocking lipid degradation in developing seeds.

Abstract:Seed dormancy is an adaptive trait that does not allow the germination of an intact viable seed under favorable environmental conditions. Non-dormant seeds or seeds with low level of dormancy can germinate readily under optimal environmental conditions, and such a trait leads to preharvest sprouting, germination of seeds on the mother plant prior to harvest, which significantly reduces the yield and quality of cereal crops. High level of dormancy, on the other hand, may lead to non-uniform germination and seedling establishment. Therefore, intermediate dormancy is considered to be a desirable trait as it prevents the problems of sprouting and allows uniformity of postharvest germination of seeds. Induction, maintenance, and release of seed dormancy are complex physiological processes that are influenced by a wide range of endogenous and environmental factors. Plant hormones, mainly abscisic acid (ABA) and gibberellin (GA), are the major endogenous factors that act antagonistically in the control of seed dormancy and germination; ABA positively regulates the induction and maintenance of dormancy, while GA enhances germination. Significant progress has been made in recent years in the elucidation of molecular mechanisms regulating ABA/GA balance and thereby dormancy and germination in cereal seeds, and this review summarizes the current state of knowledge on the topic.

Abstract:The post-harvest phase of potato tuber dormancy and sprouting are essential in determining the economic value. The intricate transition from dormancy to active growth is influenced by multiple factors, including environmental factors, carbohydrate metabolism, and hormonal regulation. Well-established environmental factors such as temperature, humidity, and light play pivotal roles in these processes. However, recent research has expanded our understanding to encompass other novel influences such as magnetic fields, cold plasma treatment, and UV-C irradiation. Hormones like abscisic acid (ABA), gibberellic acid (GA), cytokinins (CK), auxin, and ethylene (ETH) act as crucial messengers, while brassinosteroids (BRs) have emerged as key modulators of potato tuber sprouting. In addition, jasmonates (JAs), strigolactones (SLs), and salicylic acid (SA) also regulate potato dormancy and sprouting. This review article delves into the intricate study of potato dormancy and sprouting, emphasizing the impact of environmental conditions, carbohydrate metabolism, and hormonal regulation. It explores how various environmental factors affect dormancy and sprouting processes. Additionally, it highlights the role of carbohydrates in potato tuber sprouting and the intricate hormonal interplay, particularly the role of BRs. This review underscores the complexity of these interactions and their importance in optimizing potato dormancy and sprouting for agricultural practices.

Abstract:The ability of a seed to germinate and establish a plant at the right time of year is of vital importance from an ecological and economical point of view. Due to the fragility of these early growth stages, their swiftness and robustness will impact later developmental stages and crop yield. These traits are modulated by a continuous interaction between the genetic makeup of the plant and the environment from seed production to germination stages. In this review, we have summarized the established knowledge on the control of seed germination from a molecular and a genetic perspective. This serves as a “backbone” to integrate the latest developments in the field. These include the link of germination to events occurring in the mother plant influenced by the environment, the impact of changes in the chromatin landscape, the discovery of new players and new insights related to well-known master regulators. Finally, results from recent studies on hormone transport, signaling, and biophysical and mechanical tissue properties are underscoring the relevance of tissue-specific regulation and the interplay of signals in this crucial developmental process.

Abstract:Germination inhibitors, which inhibit the germination of seeds, spores and other plant reproductive material, are abundant in the plant kingdom and include phenols, cyanides, alkaloids, essential oils, amino acids, etc. These inhibitors can be classified as germination destructors and germination retarders depending on whether they harm the morphology, structure and physiology of the seed. Germination retarders are closely related to seed dormancy, and exogenous retarders can be used to extend the “dormancy” period of non-dormant seeds or perishable seeds by applying the proper dosage. They have significant potential applications as preservatives for seed preservation following harvest or for the storage of long-term germplasm resources. Germination destructors, as a type of relatively high-efficiency, low-specificity “toxic chemicals”, are of significant benefit in the application of effective and environmentally benign herbicides. At present, the main problems related to the research methods of germination inhibitors include difficulty in determining the specific endogenous substances and the minimum inhibitory concentration to induce dormancy, as well as whether the application of exogenous inhibitors will cause physiological damage to seeds. In the future, we should strengthen the tracking of germination inhibitors, explore the mechanisms of action of specific substances and deeper molecular mechanisms and finally explore new developments and new applications of different inhibitors.

Abstract:In recent years, with the accelerated pace of life and the high incidence of chronic diseases, the demand for functional food has been increasingly urgent. Under such circumstances, germinated edible foods have attracted much attention because of their unique physiological effects. This review mainly summarizes recent studies about the physiological and biochemical changes during edible seeds germination. Further, the different germination treatments and the enrichment of γ-amino butyric acid are discussed. Finally, the applications of germinated seeds in the food sector are mentioned. Germination of edible seeds is a complex process of material transformation, which not only improves the nutritional value of raw seeds, but also accumulates a wide variety of bioactive compounds with health benefits. As a potential source of numerous functional ingredients, germinated edible foods have attracted widespread attention for its physiological benefits. Therefore, germinated grain and legume seeds are expected to be functional dietary supplements with a variety of health benefits and medicinal values. Appropriate germination treatments can enrich bioactive components, which is conducive to the efficient production of bioactive functional food. Research in this area is expected to serve the purpose of preventing and alleviating chronic diseases.

Abstract:In higher plants, seed is a propagule which ensures dissemination and survival of species. Developmental phases of a seed comprise embryogenesis, maturation and germination paving a way to its final fate i.e. seedling establishment. The final stage of seed maturation is marked by dehydration, acquisition of dessication tolerance and induction of dormancy. A precise Abscisic acid (ABA) to Gibberellins (GA) ratio, accumulation of miRNA 156, low level of reactive oxygen species (ROS) and enzyme inactivity govern seed dormancy. This also prevent pre harvest sprouting of the seeds. Overtime, stored seed mRNAs and proteins are degraded through oxidation of specific nucleotides in response to ROS accumulation. This degradation alleviates seed dormancy and transforms a dormant seed into a germinating seed. At this stage, ABA catabolism and degradation accompanied by GA synthesis contribute to low ABA to GA ratio. GA as well as ROS acts downstream, to mobilize reserve food materials, rupture testa, enhance imbibition and protrude radicle. All these events mark seed germination. Further, seedling is established under the governance of auxin and light. ABA and GA are master regulators while auxin, cytokinins, ethylene, jasmonic acid, brassinosteroids act through interdependent pathways to tightly regulate seed dormancy, germination and seedling establishment. In this review, the role of phytohormones and ROS in accordance with environmental factors in governing seed dormancy, promoting seed germination and thus, establishing a seedling is discussed in detail.

Abstract:Abstract With the growth of the global population and the increasing frequency of natural disasters, crop yields must be steadily increased to enhance human adaptability to risks. Pre-harvest sprouting (PHS), a term mainly used to describe the phenomenon in which grains germinate on the mother plant directly before harvest, is a serious global problem for agricultural production. After domestication, the dormancy level of cultivated crops was generally lower than that of their wild ancestors. Although the shortened dormancy period likely improved the industrial performance of cereals such as wheat, barley, rice, and maize, the excessive germination rate has caused frequent PHS in areas with higher rainfall, resulting in great economic losses. Here, we systematically review the causes of PHS and its consequences, the major indicators and methods for PHS assessment, and emphasize the biological significance of PHS in crop production. Wheat quantitative trait loci functioning in the control of PHS are also comprehensively summarized in a meta-analysis. Finally, we use Arabidopsis as a model plant to develop more complete PHS regulatory networks for wheat. The integration of this information is conducive to the development of custom-made cultivated lines suitable for different demands and regions, and is of great significance for improving crop yields and economic benefits.

Abstract:Freshly matured seeds exhibit primary dormancy, which prevents germination until environmental conditions are favorable. The establishment of dormancy occurs during seed development and involves both genetic and environmental factors that impact on the ratio of two antagonistic phytohormones: abscisic acid (ABA), which promotes dormancy, and gibberellic acid, which promotes germination. Although our understanding of dormancy breakage in mature seeds is well advanced, relatively little is known about the mechanisms involved in establishing dormancy during seed maturation. We previously showed that the SPATULA (SPT) transcription factor plays a key role in regulating seed germination. Here we investigate its role during seed development and find that, surprisingly, it has opposite roles in setting dormancy in Landsberg erecta and Columbia Arabidopsis ecotypes. We also find that SPT regulates expression of five transcription factor encoding genes: ABA-INSENSITIVE4 ( ABI4 ) and ABI5 , which mediate ABA signaling; REPRESSOR-OF-GA ( RGA ) and RGA-LIKE3 involved in gibberellic acid signaling; and MOTHER-OF-FT-AND-TFL1 ( MFT ) that we show here promotes Arabidopsis seed dormancy. Although ABI4 , RGA , and MFT are repressed by SPT, ABI5 and RGL3 are induced. Furthermore, we show that RGA , MFT , and ABI5 are direct targets of SPT in vivo. We present a model in which SPT drives two antagonistic “dormancy-repressing” and “dormancy-promoting” routes that operate simultaneously in freshly matured seeds. Each of these routes has different impacts and this in turn explains the opposite effect of SPT on seed dormancy of the two ecotypes analyzed here.
