Abstract:The occurrence development and progression of infectious diseases involves the two aspects: the immune response of both the pathogens and the hosts. At present, pathogen-based testing is the gold standard and the most commonly used method for the diagnosis of infectious diseases. The purpose of the immune system is to identify and eliminate invading pathogens. Testing based on host reactions has become an effective auxiliary means adjunct of traditional pathogen-based tests, with the potential to improve the accuracy and efficiency of diagnosis. The combined application of host-based tests and pathogen-based tests is a new field worth exploring.

Abstract:The adaptive immune system uses several strategies to generate a repertoire of T- and B-cell antigen receptors (TCR/BCR) with sufficient diversity to recognize the wide array of potential pathogens. The Immunoglobulin and TCR repertoires are a mirror of the human immune system, which reflects processes caused by infections, cancer, autoimmunity and aging. The rapid advances in sequencing technologies have provided improvements in read length, throughput and cost. Nowadays, high-throughput sequencing (HTS) of lymphoid receptor genes is an emerging technology that could comprehensively assess the diversity of the immune system, and has been applied to vaccine development and efficacy assessment, biomarker discovery, minimal residual disease detection, autoimmune diseases, transplant rejection and tolerance. As biotechnology progresses, immune repertoire sequencing will applied more and more. The following review details some of the technologies and recent achievements in this field.

Abstract:Although the incidence and mortality of lung cancer have decreased significantly in the past decade, it is still one of the leading causes of death, which greatly impairs people's life and health. Proteomics is an emerging technology that involves the application of techniques for identifying and quantifying the overall proteins in cells, tissues and organisms, and can be combined with genomics, transcriptomics to form a multi-omics research model. By comparing the content of proteins between normal and tumor tissues, proteomics can be applied to different clinical aspects like diagnosis, treatment, and prognosis, especially the exploration of disease biomarkers and therapeutic targets. The applications of proteomics have promoted the research on lung cancer. To figure out potential applications of proteomics associated with lung cancer, we summarized the role of proteomics in studies about tumorigenesis, diagnosis, prognosis, treatment and resistance of lung cancer in this review, which will provide guidance for more rational application of proteomics and potential therapeutic strategies of lung cancer.

Abstract:Proteomics is a subject that aims to study the expression, translation and location of proteins, the relationship between proteins and proteins as well. Two-dimensional gel electrophoresis and mass spectrometry are traditional methods, the former serves to separate proteins while the later to identify the different proteins. Some new techniques such as differential in-gel electrophoresis and Isotope coded affinity tag are more and more widely used these days. Plasma, pancreatic tissue and juice were taken from normal and sick individuals who were suffered from acute pancreatitis, chronic pancreatitis or pancreatic cancer. Comparing the different proteins between normal and sick individuals could be a useful way for early diagnosis and effective treatment for pancreatic disease.

Abstract:Introduction: Genetics combined with proteomics allows for a better understanding of parasite-host interactions and host immune responses. Immunomics elucidates that antigens are targets of induced or naturally acquired immunity (NAI), a promising solution to the challenge of eradicating human infections. High-throughput protein microarrays enhance rapid antigen discovery for the development of serodiagnostic tests/vaccines.Areas covered: This review systematically analyzes the emergence of protein microarrays as a powerful technology for parasite antigen discovery and subsequently summarizes some of the attributes and disadvantages of these approaches. Major insights on novel/validated serological biomarkers or vaccine candidates against malaria and Neglected Tropical Diseases (NTDs) are highlighted. We conclude with a brief description of the processes involved in immunomic protein microarrays.Expert commentary: Interesting discoveries have been made using protein microarrays. However, there is a need to evaluate targets that elicit strong immunogenicity and correlates of protective efficacy to aid prioritization and guide further clinical development. The goal of parasitic disease elimination will be best achieved through an integrated strategy that will incorporate and implement the different control components.

Abstract:Proteins are the key players in many cellular processes. Their composition, trafficking, and interactions underlie the dynamic processes of life. Furthermore, diseases are frequently accompanied by malfunction of proteins at multiple levels. Understanding how biological processes are regulated at the protein level is critically important to understanding the molecular basis for diseases and often shed light on disease prevention, diagnosis, and treatment. With rapid advances in mass spectrometry (MS) instruments and experimental methodologies, MS-based proteomics has become a reliable and essential tool for elucidating biological processes at the protein level. Over the past decade, we have witnessed great expansion of knowledge of human diseases with the application of MS-based proteomic technologies, which has led to many exciting discoveries. Herein we review the recent progress in MS-based proteomics in biomedical research, including that in establishing disease-related proteomes and interactomes. We also discuss how this progress will benefit biomedical research and clinical diagnosis and treatment of disease.
