An experimental study on concanavalin A-induced acute hepatic injury in mice

Abstract:Objective The aim of the study was to investigate the effect of different doses of concanavalin A (ConA) on hepatic injuries in mice.Methods 60 mice were randomly divided into 6 groups,which included a control group and five experimental groups (3,6,9,12,15 mg/kg of BW).The control group was tail vein-injected with saline,while the experimental group was injected with 3-15 mg/kg BW of ConA.After 8 hours of fasting,blood was taken from eyes of the mice and hepatic tissues were removed for examination of all biochemical indicators and histopathological examinations.Results With the increase in ConA dosage,splenohepatomegalia of the experimental mice became more severe,and hepatosplenic indicators of the mice became significantly different from those of the control mice (P<0.01).Plasma AST and ALT showed significant increase at higher ConA dosages (P<0.01),and the increase in ConA strongly impacted the level of reduced glutathione (GSH) (P<0.01) but not superoxide dismutase (SOD).In liver homogenates,glucogen and GSH-PX levels and SOD activity exhibited different degrees of reduction,which did not show to correlate with the dosage of ConA.Liver lesions were not significant under the above-mentioned dosage range.Conclusion High dosages of ConA can cause significant changes in biochemical indicators in plasma and liver of mice,but has little effect on lesions of the liver.
