Influence of Cold Stress Response on Cell-mediated Immunity in Rats

Abstract:Objective This study was to investigate the influence of cold stress on Toll-like receptor 4 (TLR4) expression of immunocompetent cells in the spleen and regulatory T cells (Treg) in the peripheral blood in rats.Methods 108 male SD rats were divided into three ambient temperature groups,namely,a 20℃ group,a 4℃ group and a-12℃ group,respectively.In each group there were three subgroups of non-stimulation,lipopolysaccharide (LPS) stimulation and concanavalin A (Con-A) stimulation.In each subgroup there were four observation time points of 1h,12h,48h and 168h.The TLR4,CD4+CD25+ and CD4+CD25+Foxp3+expressions were determined by flow cytometry.Results The TLR4 expression rate of immunocompetent cells in the spleen was low in the 4℃ and-12℃ groups compared to the 20℃ group (P<0.05).The lower the temperature was,the lower TLR4 expression rate,and the TLR4 expression rate failed to increase in SD rats even if they were stimulated by LPS or Con-A.The CD4+CD25+ and CD4+CD25+Foxp3+ Treg in the peripheral blood in rats were low in the-12℃ group compared to the 20℃ group (P<0.05).Even when the rats were stimulated by the intraperitoneal injection of LPS or Con-A,their expression was still low in the 4℃ and-12℃groups at 168 h compared to the 20℃ group (P<0.05).Conclusion Findings in this study show that the innate and adaptive immunity functions in rats are damaged after a dramatic drop in ambient temperature,and the degree of the damage is positively related to the drop in ambient temperature.
