The Effect of Hypothermia on Splenic Lymphocyte Proliferation and Activity of Natural Killer Cell Following Traumatic Brain Injury in Rat

Abstract:Objective To investigate the effect of hypothermia on the function of splenic immune competent cells following traumatic brain injury(TBI) in rat. Methods After TBI,male SD rats were divided randomly into moderate hpothmic(27℃) group,mild hypothmic(33℃) group and normothermic(37℃) group. Sham operative northermic group served as coutrol.Using 3H-TdR uptade and LDH release assay, dynamic changes on splenic lymphocyte proliferation and activity of nature killer cell (NK cell) were determined in rat following TBI. Results Compared with sham control group, proliferation of T splenic lymphocytes [(45.6±5.6)×10 3/min] and activity of NK cell [(17.19±3.83)%] were decreased in normothermic traumatic group (37℃), proliferation of T, B splenic lymphocytes [( 44.0 ±4.5)×10 3/min, (25.4±3.6)×10 3/min] and activity of NK cell [(13.21±4.62)%] decreased furtherly in moderate hypothermic group (27℃), proliferation of B splenic lymphocytes [(25.0±4.9)×10 3/min]decreased only in mild hypothermic traumatic groups(33℃). Conclusions Moderate hypothermia (27℃) inhibits the function of T,B lymphocyte and activity of NK and mild hypothermia (33℃) inhibits the function of B lymphocyte in rat following TBI.
