Abstract:Objective To examine the effect of capsaicin on glucose metabolism in type 1 diabetes rats and its potential mechanism. Methods (1) Normal rats were divided into three treatment groups with different dosages of capsaicin, and one blank control group (n=8). In the treatment groups, rats were treated by 3, 6, 9 mg/(kg·bw) of capsaicin respectively for 28 days. Then, postprandial blood glucose, intestinal amylase and glycosidase enzymes were observed; (2) The diabetic rats model was induced by streptozotocin, and the rats were divided into three treatment groups, blank control group and model group (n=8). In the treatment groups, rats were fed capsaicin (3, 6, 9 mg/kg·bw) for 28 days. Subsequently, the levels of postprandial blood sugar, glucose tolerance, glycosylated serum protein, hepatic glycogen, serum insulin and the activity of digestive enzymes were examined. Results Medium and high dosages of capsaicin significantly inhibited the activity of amylase and glucosaccharase enzymes in rat intestine, and significantly reduced the postprandial blood sugar levels in normal rats. The medium dosage of capsaicin had the greater effects than the high dosage of capsaicin. Moreover, the high dosage of capsaicin significantly reduced the level of serum glycated albumin. Conclusion The decrease of blood glucose by capsaicin may be achieved by two ways:(1) Inhibiting the enzyme activity and intestinal absorption of carbohydrate; (2) Increasing insulin secretion and hepatic glycogen synthesis.
