Effects of Long-Term High-Fat Diet on Skeletal Muscle Global Gene Expression in C57BL/6 Mice

Abstract:Objective To explore the molecule mechanisms in the development of obesity,insulin resistance (IR),type 2 diabetes (T2DM) which are induced by long-term high-fat diet.Gene microarray was performed to analyze the changes of global gene expression in the skeletal muscle of long-term high-fat diet fed C57BL/6 mice.Methods 20 of 4-week old,male C57BL/6 mice were randomly divided into normal diet group and high-fat diet group,fed by normal food and high-fat diet respectively.After 16 weeks,body weight of each group were measured.The serum glucose,fasting serum insulin (FINs),serum high density lipopro-tein (HDL),triglyceride (TG),total cholesterol (TC) and free fatty acid (FFAs) levels were measured.Four mice randomly selected from each group were put to death and quadriceps femoris were removed for RNA extraction.The RNA samples were then analyzed by gene microarray.Results Compared with normal diet group,body weight,the levels of FINs,TC,TG and FFAs were significantly increased in high-fat diet group,while the level of HDL had no significant difference.There were 590 differentially expressed genes in high-fat diet group compared with normal diet group,including 321 up-regulated and 269 down-regulated genes.Conclusion Long-term high-fat diet can induce significant changes in global gene expression in the skeletal muscle of C57BL/6 mice.
