Global Gene Expression Responses to 16-Week High Fat Diet in Skeletal Muscle of C57bl/6 Mice

Abstract:The expression of multiple genes can be measured simultaneously by gene microarray. Compared with traditional biological technology, gene array represents a simple, fast and high flux. It has been well recognized that human dietary habits have changed in the past decades and long-term high-fat diet can cause energy imbalance between intake and consumption, which could cause the abnormal signal pathways and promote the development of metabolic disorders such as obesity, insulin resistance, and type 2 diabetes. The high-fat diet is increasingly recognized as an important cause for metabolism syndrome. PURPOSE: To study the molecule mechanisms in the development of insulin resistance induced by long-term high-fat diet, gene microarray was performed to analyze the changes of global gene expression in the skeletal muscle of C57BL/6 mice. The differentially expressed genes were analyzed to provide the experimental evidences and potential targets for the cure of metabolic disorders. METHODS: 20 of 4-week old, C57BL/6 male mice were randomly divided into two groups, normal diet group (C) and high fat diet group (H), and then the mice were feed by normal chow or high fat diet for 16 weeks respectively. Six mice were randomly selected from each group and skeletal muscle was removed for RNA extraction. The RNA samples were then analyzed by gene microarray and data were evaluated by statistics software SPSS. Significance Analysis of Microarrays Software provided by Capital-Bio Corporation was used to dissect the differentially expressed genes between two groups. RESULTS: There are 590 differentially expressed genes in group H comparing with group C after 16 -week high-fat diet, including 321 up-regulated and 269 down-regulated genes. The 590 differentially expressed genes were further analyzed by gene ontology (GO) and the results indicated that these genes play 727 different molecular functions, participate 412 biological processes and 229 cellular component. The differentially expressed genes between the C and H groups were further tested by Disease Categorization and the result showed 10 genes intimately correlated with T2DM, including IDE,CASQ1,APOE,APOC3,PDK4,KLF13,AKT1,UCP2,GSTT1 and ADM. Furthermore, APOE,APOC3 and UCP2 were also responsible for the cause of other diseases including hypertension, atherosclerosis, obesity and liver disease. CONCLUSION: Long-term high-fat diet can produce significant changes in global gene expression in the skeletal muscle of C57BL/6 mice.
