Genomic analysis of apolipoprotein E gene in patients with lipoprotein glomerulopathy

Abstract:Objective:Lipoprotein glomerulopathy (LPG) is characterized by lipoprotein thrombi in the glomerular capillary lumen and hyperlipoproteinemia similar to the familial type Ⅲ hyperlipidaemia (hyperlipoproteinemia). Association of apoE gene mutation and LPG was implicated for the elevated apoE plasma level and the presence of apoE in the lipoprotein thrombi in immunofluorescence studies. Results of the studies in this field varied and no consensus concluded. In this study, we analyzed the full sequences of apoE in LPG patients and controls, and discussed the role of apoE gene mutation in the pathogenesis of LPG. Methodology:Five normal controls and 17 LPG patients were involved in this study. Full sequences of apoE were analyzed by gene sequencing after PCR. The design of PCR primers was based on the genomic sequence of the apoE published in GenBank. Direct comparison were made between the genomic sequences of apoE in the subjects of this study and those published in GenBank. Results:Compared the sequences of apoE in the 5 controls and the 17 patients with the those published in GenBank (M10065), we found 10 differences between. But we found no mutant forms of apoE in any of the 17 LPG patients as compared to the normal controls, and to those reported mutations such as apoE Sendai. Conclusion:In this study, we found no mutation form of apoE in this group of LPG patients, and we could not find any relationship between apoE mutations and the pathogenesis of LPG. Ethnic variations might be present in the gene loci for apoE.
