Effects of calcium/calmodulin dependent protein kinase Ⅱ δ on proliferation and invasion of prostate cancer cells via Wnt/β-catenin signaling

Abstract:Objective To study the effects of calcium/calmodulin dependent protein kinase Ⅱ δ (CaMK Ⅱ δ) on proliferation and invasion of human prostate cancer PC-3 cells.Methods Human CaMK Ⅱ δ siRNA was transfected into PC-3 cells.CaMK Ⅱ δ mRNA and protein expression levels were detected by reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blotting respectively.Cell proliferation affected by CaMK Ⅱ δ was determined by methyl thiazol tetrazolium (MTT) assay,and cell cycle was analyzed by flow cytometry.The migration and invasion abilities of PC-3 cells were measured by wound healing assay and Transwell chamber assays,respectively.The protein expression levels of Nuclear β-catenin,c-myc,Cyclin D1,survivin,lymphoid enhancing factor 1 (Lef1) and axis inhibition protein 2 (Axin2) were detected by Western blotting.Results CaMK Ⅱ δ was stably transfected into PC-3 cells.As compared with untreated cells,the protein and mRNA expression levels of CaMK Ⅱ δ in PC-3 cells transfected with CaMK Ⅱ δ siRNA were decreased by 78.5% (P < 0.01) and 80% respectively (P < 0.01).MTT assay revealed that the proliferation of PC-3 cells was significantly inhibited after transfection with CaM Ⅱδ siRNA.The knock-down of CaMK Ⅱ δ gene expression significantly decreased abilities of PC-3 cell migration [(0.37 ±0.03) mm vs.(0.29±0.02) mm and (0.28±0.03) mm,P< 0.01)] and invasion [(35.00±3.35) vs.(75.25 ±6.35) and (67.23 ±4.32) cells,P<0.01].CaMK Ⅱ δ siRNA could down-regulate nuclear β-catenin,c-myc,Cyclin D1,survivin and Lef1 protein levels,but significantly up-regulate Axin2 expression levels.Conclusion CaMK Ⅱ δ gene silencing could significantly inhibit proliferation,migration and invision of human prostate cancer PC-3 cells in vitro,which may be associated with inhibition of Wnt/β-catenin signaling pathway.
