The clinical value of the FTA cartridge in cervical cancer screening

Abstract:Objective To investigate the clinical application value of the solid FTA cartridge (FTA card) as a sample cartier of storage and transshipment in detection and genotyping of HPV DNA in cervical cancer screening.Methods 102 cases of patients with colposcopy and cervical biopsy at cervical outpatient clinic were included.Cervical exfoliated cell specimens were collected by the gynecologist and each specimen was stored and transferred by solid FTA cards and cell preservation solution as a carrier.PCR-based MALDI-TOF method were used to test the two samples for high-risk HPV genotyping.FTA Cards and conventional liquid mediums were compared for the clinical value in cervical cancer screening.Results HPV infection rate was 69.6% in the specimens stored in liquid medium,66.7% in FTA cards group.There was no significant difference between two groups (x2=0.250,P>0.05),with a good consistency (Kappa =0.932,P<0.001) in detecting high-risk HPV.There was no significant difference in HPV genotyping between two groups.(x2=0.250,P>0.05) with a good consistency (kappa 0.930,P<0.001).Taken the colposcopy and cervical pathological diagnosis as the gold standard,comparing the screening efficiency of HPV in CIN 2 and CIN 3 cervical lesions by the two sample preservation methods,the area under the AUC curve of ≥ CIN2 lesions were 0.718 and 0.699 respectively (P <0.05).For ≥ CIN3 lesions were 0.695 and 0.678 respectively (P<0.05).Conclusions There were good consistency and highly similar diagnostic efficiency using FTA cards and liquid media as storage media for HPV genotyping.Due to a series of advantages,such as small specimen size,the convenience of transit,low risk of contamination and the simplified DNA extraction,the solid FTA cards could be regarded as a new HPV testing sample storage method for cervical cancer screening.
