Efficacy of venlafaxine in treatment of selective serotonin reuptake inhibitors non-responders with major depressive disorder

Abstract:Objective The purpose of this study was to assess the efficacy of venlafaxine in outpatients who did not respond to selective serotonin reuptake inhibitors (SSRI) treatment.Methods~Thirty-four depressive outpatients who had inadequate or unsustained response to at least 8-week treatment of SSRI were switched to venlafaxine treatment for 6 weeks. And another 34 outpatients were firstly given venlafaxine treatment for their depression as controls. The efficacy was measured with the Hamilton Depression Scale (HAMD), while the side effects were recorded.Results The HAMD scores of both groups decreased significantly from the first weekend to the sixth weekend (P0.05 or P0.01). After 6 weeks of treatment, 68% (23/34) of SSRI-resistant patients and 77% (26/34) in control group were responded (HAMD,17 criteria) (P0.05). The side effects as dry mouth, nausea and vomit were more common in SSRI-resistant patients than in the controls (P0.05).Conclusion Venlafaxine may be effective in SSRI-resistant depressive patients.
