Abstract:We report a 38-year-old man who presented with bilateral conjunctival congestion, hoarseness, and progressively growing pruritic, infiltrated skin lesions that had first begun over the face and neck, and later spread to the trunk and the limbs in 4 months. The clinical appearance of the lesions mimics granulomatous rosacea, acne vulgaris, or pityrosporum folliculitis. Histopathologic examination of the lesions from the face and chest both revealed dense dermal nodular lymphohistiocytic infiltrates which were positive for CD68 and S-100, but negative for CD1a. A systemic work-up for him detected no lymphadenopathy or other systemic involvement. A diagnosis of extranodal Rosai-Dorfman disease was made, and the patient received systemic glucocorticoids, with considerable improvement after 4 months of therapy.

Abstract:A case of cutaneous Rosai-Dorfman disease (CRDD) is reported. The 47-year old female patient presented with painless, brown-red mass on the face and neck for 3 years, without lymphadenopathy and other systemic abnormalities. Microscopically, many histiocytes, lymphocytes and plasma cells were noted intradermally and subcutaneously. The major feature was the phagocytosis of intact lymphocytes and plasma cells by unique histiocytes. Immunohistochemical staining showed that these histiocytes were positive for S-100 and CD68. Polymerase chain reaction (PCR) for mycobacteria and cultures for both mycobacteria and fungi were all negative. CRDD is a rare, benign proliferating disease of histiocytes. The understanding of the clinical manifestations, histopathological features and immunohistological findings of this disease will be helpful for accurate diagnosis.

Abstract:Three cases of Rosai-Dorfman Disease (RDD) are reported. Patient 1 was a 47-year-old woman, presenting with painless, brownish plaque on the face and neck. No lymphadenopathy and systemic involvement were found. Patient 2 was a 70-year-old-man, presenting with fresh-colored nodules on the chest without systemic involvement. Patient 3 was a 47-year -old-man, started with a red nodule on his left cheek. The lesions scattered, overthe whole body rapidly,and complicated with lymphadenopathy of mediastinum, uveitis and hydropericardium. The skin histopathology of these 3 cases showed typical changes of RDD. Patient 1 and patient 2 were diagnosed as cutaneous Rosai-Dorfman disease (CRDD). Patient 3 was diagnosed as systemic RDD, and the lesions responded well to chemotherapy. However, he died suddenly shortly after chemotherapy.

Abstract:Rosai-Dorfman disease is a rare, idiopathic, benign, and self-limiting histiocytic proliferative disorder. A 26-year-old man presented with a single massive cutaneous nodule (reaching 30 cm in diameter) on the left shoulder and back for 15 months. The routine hematological and biochemical tests were normal. Magnetic resonance scanning showed the lesion involved the skin, subcutaneous tissue, and subjacent muscle group, accompanied by obvious lymph node enlargement in the left part of the neck, supraclavicular fossa, and axillary fossa. The histopathology of the left cervical lymph node revealed diffuse effacement of the normal nodal architecture, with patchy chronic inflammatory cell infiltrates comprising lymphocytes and sheets of histiocytes. Some histiocytes contained lymphocytes within their pale cytoplasm. Many multinucleated giant cells were found; however, caseating granulomas were not seen. The skin and muscle biopsy specimen obtained from the back revealed infiltrating lymphocytes and histiocytes diffusely distributed in the dermis, subcutaneous tissue, and crevices of the muscle fibers. The phenomenon of emperipolesis and the presence of multinucleated giant cells were also seen. Immunohistochemical staining revealed that the histiocytes were positive for S-100 protein and CD68 but negative for CD1a. Immunophenotyping of the infiltrating lymphocytes indicated positive reactions to CD3, CD45RO, CD5, CD7, CD4, CD8 (partly), CD79a, CD20 (partly), and Ki-67 (<1%). The final diagnosis was Rosai-Dorfman disease. Owing to the extensive and deep involvement of the subcutaneous tissue and muscles, the patient did not undergo surgery to excise the massive skin nodule. The lesion showed no obvious change at the 12-month follow-up.
Cutaneous rosai-dorfman disease: a clinical and histopathologic study of 25 cases in China.

Abstract:Cutaneous Rosai-Dorfman disease (CRDD) is a rare proliferative disorder of histiocytes with unknown etiology, broadly different from systemic Rosai-Dorfman disease. We present the largest series of CRDD, describing the clinical manifestation, histopathology, immunohistochemistry, and follow-up course of 25 cases in China. Clinically, 39 skin lesions in 25 patients were divided into 3 main types: papulonodular type (79.5%), indurated plaque type (12.8%), and tumor type (7.7%). Extremities were the most frequently involved, followed by trunk and face. None of the patients was found to have visceral organ involvement or lymphadenopathy. Microscopically, CRDD was characterized by scattering, clusters or sheets of large polygonal histiocytes intermingled with a florid, mixed inflammatory infiltrate. The most important feature was emperipolesis, which can be highlighted by S-100 protein stain. Patch and bandlike infiltrate of numerous mature plasma cells around glands and vessels was a constant finding in all lesions. Neutrophils existed in all cases to a variable degree with 2 cases forming microabscess. Four cases were remarkable for fibrosis, and xanthomatous change was observed in 2 cases. Coexistence of localized Langerhans cell histiocytosis and CRDD was interestingly found in case 7, which was evidenced by CD1a stain. Clinical follow-up in 22 patients, ranging from 2 to 55 months, indicated that surgical excision was the exclusive effective treatment for CRDD. Partial or complete spontaneous remission was achieved in 7 patients within 6 to 55 months. Owing to its favorable outcome, CRDD should be differentiated from a variety of benign and malignant lesions. Recognition of its wide clinical spectrum and histologic features combined with S-100 protein stain can help to establish the correct diagnosis.

Abstract:Recently, we described a case of generalized pure cutaneous Rosai-Dorfman disease in a 43-year-old Asian man in JAMA. The lesions distributed on nearly all of the skin of the whole body, except for mucous sites. Molecular, immunophenotypic, and sequencing analyses seem to define it as a histiocytic-mesenchymal transition and intermediate proliferative histiocytosis not associated with mtDNA large deletion and pathogenic mutation, as well as the SLC29A3 gene mutation.

Abstract:Objective: To improve the diagnosis of cutaneous Rosai-Dorfman disease (CRDD) by investigating the clinical, pathological and immunophenotypic features of this disease. Methods: The data of 14 cases of CRDD were collected. The clinical category was classified according the features of clinical manifestations of these patients. The histopathologic and immunchistochemical studies were performed in all cases. Results: The skin lesions of 14 patients were divided into 3 main types: papulonodular type (64.7%), infiltrating plaque type (5.9%) and tumor type (29.4%). No other systemic disease such as lymphadenopathy was found. Mixed inflammatory cell infiltration in the dermis and subcutis, mainly lymphocytes, plasma cells, and a variable number of S-100 positive histiocytes, was shown by histopathology and immunohistochemistry. The presence of lymphocytes engulfed within the histiocytic cytoplasm, namely emperipolesis, was seen. Few histiocytes between vascular spaces were found in one case. Lymph follicle, fibrosis, panniculitis-like change and vasculitis-like change were seen in a few cases. Conclusions: The classification of the skin lesions into three types, combined with histopathologic and immunophenotypic features, are helpful for the diagnosis of CRDD, especially for the differentiation of it from various other cutaneous neoplastic and non-neoplastic skin diseases.

Abstract:Rosai-Dorfman disease (RDD) is rare and characterized by histiocytic proliferation and massive cervical lymphadenopathy. About 40% of patients have extra-nodal involvement. Ophthalmic involvement is seen in 10% of cases. A case of orbital Rosai Dorfman disease in a 58 years old woman is presented here, who was misdiagnosed as orbital inflammatory disease initially. The patient did not respond to a course of oral prednisolone. Then complete surgical excision of the mass was performed and the histopathological examination was consistent with a diagnosis of RDD.

Abstract:Rosai-Dorfman disease (RDD) is a rare disorder characterized by excessive growth of histiocytes. We present a case of a 14-year-old female with cutaneous RDD who had a subcutaneous lump on her left arm for three years. The lump became tender and progressively larger over the past year. She had no systemic symptoms, and her physical examination revealed a mobile, tender lump. Laboratory tests were normal. Surgical excision of the lump was performed, and histopathological examination confirmed RDD with the presence of epithelioid histiocytes with eosinophilic and clear cytoplasm, along with emperipolesis and positive staining for CD68, CD163, S100, and OCT2. The patient was referred for follow-up and required no further treatment. RDD can present with subcutaneous masses without systemic symptoms, and it is important to consider RDD in the differential diagnosis of such cases. Surgical excision is the main treatment, and long-term monitoring is necessary due to the potential for disease recurrence. Awareness of cutaneous RDD presentations is crucial for accurate diagnosis and management.

Abstract:Rosai–Dorfman disease (RDD) is a rare subtype of non–Langerhans cell histiocytosis. It is a benign disease with variable presentations, with breast involvement being an uncommon manifestation. In this paper, we present a case of breast RDD in a 59‐year‐old Chinese woman who presented with a painless, enlarging mass in her right breast, accompanied by skin changes and no palpable axillary lymph node. A mammogram and breast ultrasonography revealed a progressively enlarging lesion with a suspicious axillary lymph node, leading to an upgrade from Breast Imaging Reporting and Data System (BIRADS) score of 3 to 4A on interval imaging. Core biopsy of the breast mass and cytology of the right axillary lymph node confirmed extranodal RDD. After a 5‐year period of observation, excision was performed due to persistent symptoms. Local recurrence of RDD, with skin discolouration and thickening, was confirmed by skin biopsy 1 year and 9 months after the operation. To the best of our knowledge, this is the first reported case of breast RDD in Hong Kong.

Abstract:Rosai-Dorfman disease (RDD) is a rare non-Langerhans cell histiocytic proliferation that occurs in nodal and extranodal sites. Rare examples of the disease involving the digestive system have been described. To characterize the digestive tract manifestations of this disease, 12 specimens from 11 patients with extranodal RDD affecting the digestive organs were analyzed. Hematoxylin and eosin sections and available immunohistochemical stains were reviewed, and the clinical information was obtained from patients’ electronic or submitted records. Eight patients were female and 3 male (median age, 65 y; range, 17 to 76 y). Abdominal pain was the most frequent symptom. Six patients had an associated immunologic or malignant disease. Nine lesions arose in the gastrointestinal tract (1 involving the appendix, 2 right colon, 6 left colon), 2 in the pancreas, and 1 in the liver. Two patients had the coexistent nodal disease, and 1 had bone and soft-tissue involvement. The lesions were generally composed of polygonal to spindle-shaped histiocytes with eosinophilic to clear cytoplasm admixed with lymphoplasmacytic cells. The inflammatory cells formed lymphoid aggregates in 7 cases and included focally scattered or small collections of neutrophils in 6 cases. Fibrosis was variable, and 4 cases had a storiform pattern. Vasculopathy in the form of a thickened capillary wall, medium-sized arterial wall infiltration by lesional and inflammatory cells and phlebitis was seen in 10, 5, and 2 cases, respectively. All cases were reactive for S100-protein. Of the 5 patients with follow-up, 1 developed immunoglobulin A nephropathy and died of renal failure.

Intracranial Rosai-Dorfman disease mimicking melanoma: a case report and review of the literature

Abstract:Intracranial Rosai-Dorfman disease is quite rare. Here, we report a 67-year-old man with a 90 day history of pain and numbness in his right limbs. The patient was suspected of suffering from melanoma. Then he received craniotomy and was finally diagnosed with intracranial Rosai-Dorfman disease. MEDLINE was used to search the related literature; and the diagnosis, mechanism, treatment and prognosis of this rare tumor are discussed.

Abstract:Background:Rosai–Dorfman disease was commonly characterized as massive, painless, bilateral, symmetric cervical lymphadenopathy, with fever, leukocytosis, and elevated sedimentation rate. However, soft tissue Rosai–Dorfman disease (STRDD) is a rare benign tumor. Methods:We hereby present 1 case of a 17-month-old girl, an isolated subcutaneous mass was detected on her right forearm, and no signs of pain, swelling, or erythema were observed at the site. Results:The patient underwent an excisional biopsy for the mass. Immunohistochemistry results showed that it was positive for S-100 protein and CD68, whereas negative for CD1a, which supported the diagnosis of STRDD. Conclusions: The patient showed no evidence of recurrence or metastasis 2 years after the surgery.Some multifocal masses were reported to be much more prone to recurrence. Further follow-up of STRDD is necessary.

Abstract:Objective: The aim of this case report is to investigate an uncommon presentation of Rosai–Dorfman–Destombes (RDD) disease, and discuss possible differential diagnoses and treatment options for this pathology. RDD is a rare disorder of histiocytes that typically presents in patients as painless cervical lymphadenopathy. However, this case involves a patient with the central nervous system (CNS) type of RDD who later developed cutaneous lesions. Method: Several differential diagnoses were examined, including hidradenitis suppurativa, pilonidal cyst and pressure ulcers. It is important to be able to exclude these diagnoses based on the presentation, patient demographic and wound location. Results: Biopsies verified the presence of RDD in the patient's suprasellar hypothalamic mass and skin lesions, confirming the patient had both CNS-RDD and cutaneous-RDD in the absence of lymphadenopathy. Conclusion: Recognising the unique manifestations of rare diseases such as RDD prevents delay of proper intervention and treatment.

Abstract:Rosai-Dorfman disease (RDD) is a rare non-Langerhans cell histiocytosis. The etiology and pathogenesis of RDD have not been fully explained, and its occurrence is even rarer in the maxillofacial region. This paper reported a patient who was first misdiagnosed as facial inflammation caused by oral disease and finally diagnosed as RDD. Clinical features, diagnosis, treatment, and prognosis were discussed based on the literature. This work provides reference for future diagnosis and differentiation of RDD in cheek.

Abstract:Abstract Background Sinus histiocytosis with massive lymphadenopathy, also known as Rosai-Dorfman disease, is a rare, self-limiting disease that predominantly affects children and young adults. Moreover, the disease is characterized by painless bilateral cervical lymphadenopathy in 95% of the patients. However, few reports are available on the Rosai-Dorfman disease of the thymus. Case presentation We report a rare case of thymic Rosai-Dorfman disease detected using computed tomography. During a medical examination, a 50-year-old man underwent a chest computed tomography scan, which revealed an anterior mediastinal single mass with fat in the thymus. A thymectomy was performed to completely remove the tumor using a thoracoscopic technique due to a clinical suspicion of thymoma. Furthermore, Rosai-Dorfman disease was confirmed using histological and immunohistochemical analyses. Conclusions To the best of our knowledge, this is the sixth case of thymus-affecting solitary Rosai-Dorfman disease with histological and immunohistochemical evidence. Fat in the thymus, as was present in this case, has never been described in Rosai-Dorfman disease previously. Our results highlight the challenge of diagnosing this uncommon tumor before surgery, and more cases need to be reported to help with the preoperative diagnosis of such a rare tumor.

Extranodal Rosai-Dorfman disease: a clinicopathological analysis of three cases

Abstract:Objective To study the clinicopathological features,diagnosis and differential diagnosis of extranodal RosaiDorfman disease(RDD).Methods Clinical feature,sections of hematoxylin-eosin and immunohistochemieal staining were analyzed in 3 cases of extranodal RDD.Results The patients were 1 males and 2 females,aged from 40 to 72 years(mean 57 years).The lesions were located in the right nasal cavity subcutaneous soft tissue of left thigh and endometrium.Histologically,the lesions were characterized by variable numbers of large histiocytes with emperipolesis,accompanied by infiltration of lymphocytes and plasma cells.The histiocytes were positive for S-100 protein and CD163.Conclusion Extranodal RDD is a very rare histiocytic proliferative disorder which has distinct morphologic feature.Systemic involvement is rare and its prognosis good,but patients have some of recurrence rate.It should be differentiated from other diseases such as Langerhans cell histiocytosis.
Successful treatment of large cutaneous facial Rosai-Dorfman disease using combination of subtotal resection and ALA-PDT: A case report

Abstract:<p>Rosai-Dorfman disease(RDD), also called sinus histiocytosis with massive lymphadenopathy, is a rare, benign and self-limited histiocytosis. We present a case of cutaneous Rosai-Dorfman disease (CRDD) on her right cheek that responded well to combination of subtotal resection and ALA-PDT. So we believe that combination of subtotal resection and ALA-PDT treatment for large cutaneous facial Rosai-Dorfman disease patients is more effective, highly satisfying, and quick and thus is worth promoting.</p>
