Abstract:Objective:To investigate the effects of repeated neonatal administration of dizociplinemaleate (MK-801),the glutamate N-methyl-D-aspartate (NMDA) receptor antagonist,on recognition memory and expression of NMDA receptor subunits including GluN1,GluN2A and GluN2B at different developmental stages in female rats. Methods:Neonatal female Sprague-Dawley (SD) rats were randomly divided into model group and control group.Rats were administrated subcutaneously with MK-801 (0.25 mg·kg-1,bid) or normal saline from postnatal day (PND) 5 to PND14.Object recognition test was performed on adolescence and adulthood in rats; The expression levels of GluN1,GluN2A,and GluN2B on neonatal period, adolescence and adulthood in the medial prefrontal cortex and hippocampus were measured. Results:Neonatal exposure to MK-801 elicited recognition memory deficitat both adolescence and adulthood (P < 0.05,P < 0.01,respectively),especially in adult female rats.At three different development stages, the expression of GluN1 in both brain regions were significantly higher than that of control group (P < 0.05).GluN2A protein levels in the hippocampus were higher in model group on PND15 and PND42 only, while GluN2B protein expression in both brain areas were significantly upregulated on PND42 and PND70.Conclusion:Early NMDA receptor inhibition produces long-term deleterious effects on cognitive function in rats, which may related to the change expression of NMDA receptor subunits in relevant brain regions.
