Abstract:Rapamycin is a triene macrolide antibiotic produced by Streptomyces hygroscopicus. Besides its wide application as an effective immunosuppressive agent, other important bioactivities have made rapamycin a potential drug lead for novel pharmaceutical development. However, the low titer of rapamycin in the original producer strain limits further industrialization efforts and restricts its use for other applications. Predicated on knowledge of the metabolic pathways related to rapamycin biosynthesis in S. hygroscopicus, we have rationally designed approaches to generate a rapamycin high producer strain of S. hygroscopicus HD-04-S. These have included alleviation of glucose repression, improved tolerance towards lysine and shikimic acid, and auxotrophy of tryptophan and phenylalanine through the application of stepwise UV mutagenesis. The resultant strain produced rapamycin at 450 mg/L in the shake flask scale. These fermentations were further scaled up in 120 and 20,000 L fermentors, respectively, at the pilot plant. Selected fermentation factors including agitation speed, pH, and on-line supplementation were systematically evaluated. A fed-batch strategy was established to maximize rapamycin production. With these efforts, an optimized fermentation process in the larger scale fermentor was developed. The final titer of rapamycin was 812 mg/L in the 120 L fermentor and 783 mg/L in the 20,000 L fermentor. This work highlights a high rapamycin producing strain derived by mutagenesis and subsequent screening, fermentation optimization of which has now made it feasible to produce rapamycin on an industrial scale by fermentation. The strategies developed here should also be applicable to titer improvement of other important microbial natural products on an industrial scale. Biotechnol. Bioeng. 2010; 107: 506-515. Abstract:Sirolimus, a powerful immunosuppressant, is produced by Streptomyces hygroscopicus FC904. In an attempt to improve its production, mutation and screening were carried out. Spore suspensions and the forming buds were treated with UV, nitrosoguanidine (NTG) and ethylmethane sulfonate (EMS), some of survivors were selected and detected their resistance to sirolimus. A mutant FC904-33 with higher productivity of sirolimus was obtained and the features of the mutant such as fewer by-products and the susceptibilities to antibiotics as well as its cultural characteristics were presented.

Abstract:A Streptomyces strain which could produce a novel kind of antifungal-antibiotics was screened out.The fermentation conditions were optimized: initial pH 6.5,inoculum percentage 10%,at 30℃,fermentation time (72 h) using conical beaker.And with the same K_(L)a,the reversed jet loop reactor(JLR) was more fit for antifungal-antibiotics fermentation than continuous stirred tank reactor(CSTR): the potency of fermentation liquor and the morphology of mycelium with JLR was much higher than those with CSTR.
Microbial Conversion of Sirolimus by Actinoplanes Sp. ATCC 12065

Abstract:A powerful immunosuppressant sirolimus,a mTOR inhibitor,was transformed into four fractions(fraction1,2,3 and 4) by Actinoplanes sp.ATCC 12065.The results of HPLC showed the retention time of the four fractions were 25.41min,34.13min,48.46min and 52.24min,respectively.All four fractions possessed a UV spectra typical of sirolimus.The ESI mass spectra of fractions 1 and 2 were shown M.W.916,which increased in 2 mass units in comparison with sirolimus,however,fractions 3 and 4 were shown M.W.900,which reduced 14 mass units.In this paper,the primary results of microbial conversion of sirolimus by Actinoplanes sp.ATCC 12065 had been discussed.
Fermentation process and kinetics of novel antifungal antibiotic production

Abstract:A strain Bacillus amyloliquefaciens 091,which shows a strong inhibitory effect against Candida albicans ATCC 64548,was screened from soil samples of East Sea,Zhejiang Province,China.The strain can produce a kind of novel macrolide antibiotic with relative molecular mass 410.In order to increase antibiotic potency,single-factor methodology was applied in the experiment with 250 mL shake flask to optimize the culture conditions.The results are as follows: temperature 28 ℃,initial pH=6.5,rotation speed 200 r/min,inoculation 8%(volume fraction),medium volume 40 mL,and fermentation time 36 h.Under the aforementioned culture conditions,the potency activity of antifungal metabolites is up to 7 026.79 U/mL,which is increased by 2.49 folds compared with that under original culture conditions.According to Logistic,Luedeking-Piret and Luedeking-Piret-like equations,the kinetic models describing cell growth,antifungal metabolites production and substrate consumption were constructed,and the model parameters were fitted by MATLAB software based on the experimental data in 5 L fermentor.The calculation results are in a good agreement with the experimental data and this model can provide the theoretical basis for the antibiotic production in pilot scale.
Optimization of fermentation conditions for production of a novel antifungal metabolite by response surface methodology

Abstract:A strain ZS09-33, antagonising human pathogenic fungi-Candida albicans, was screened from soil samples of East Sea, Zhejiang Province, China. The strain was identified as Bacillus amyloliquefaciens based on sequencing 16S rRNA gene and blasting in Genbank. Then a two-level Plackett-Burman design was established to determine which factors in the fermentation medium influence the potency activity of metabolites. The concentrations of soybean bran, starch soluble and (NH4)2SO4 were optimized using the response surface methodology (RSM). The optimum composition of medium was as follows (per liter): soybean flour 2.72, soluble starch 26.67, (NH4)2SO4 3.95, K2HPO4 1.8, NaCl 2, MgSO4 0.5, CaCO3 0.72, CuSO4 0.02, FeSO4 0.02, MnSO4 0.02, initial pH 6.5. Under the RSM-optimized conditions, the potency activity of culture broth was 16 806.48 U/mL, which accorded well with the model-predicted value and resulted in a 2.39-fold increase in antifungal activity compared to the original culture conditions obtained by single-factor methodology. After analyzed by ESI/MS, 1H-NMR and 13C-NMR, the substance was determined as a macrolides antibiotic with relative molecular weight 402.

Abstract:Bacterium strain 287 isolated from the soil samples in Fuzhou, China was identified as Bacillus megaterium based on the taxonomic properties and 16s rRNA gene sequence. Bacillus megaterium 287 biotransformed sirolimus to three compounds: 287-P1, 287-P2 and 287-P3. Among them, 287-P1 was identified as 29,42-bis-O-demethylrapamycin by physico-chemical properties and spectral analyses.

Abstract:AIM: To investigate the possible relationship between the yield of sirolimus and the FKBP25 in streptomyces hygroscopicus. METHODS: The mycelial proteins and PPIase activities were compared between the three higher sirolimus producer mutants and wild strain. RESULTS: The quantity of FKBP25 in wild strain was higher than that in all three mutant stains. CONCLUSION: The initial results showed that there may be a relevance between the yield of sirolimus and the quantity of FKBP25 in S hygroscopicus.

Abstract:Objective To optimize the fermentation process of Sanglifehrin A(SFA) and increase yield.Methods Estimated optimum values for the production of SFA were as follows:the age of inoculm 24 h;inoculation amount 6%(v/v);temperature 30 ℃,initial pH 6.5,shaking speed 300 rpm,liquid volume in flask 25 mL/250 mL,fermentation period 108 h.Results Under the above conditions,the yield of SFA(6.12 mg·L-1) was 3 times higher than that with the original fermentation conditions.Conclusion The method was simple and reliable.It can increase yield for the large-scale industrial production of Sanglifehrin A.

Abstract:Rapamycin is a clinically important macrocyclic polyketide with immunosuppressive activity produced by Streptomyces hygroscopicus. To rationally guide the improvement of rapamycin production, comparative metabolic profiling analysis was performed in this work to investigate the intracellular metabolic changes in S. hygroscopicus U1-6E7 fermentation in medium M1 and derived medium M2. A correlation between the metabolic profiles and rapamycin accumulation was revealed by partial least-squares to latent structures analysis, and 16 key metabolites that most contributed to the metabolism differences and rapamycin production were identified. Most of these metabolites were involved in tricarboxylic acid cycle, fatty acids, and shikimic acid and amino acids metabolism. Based on the analysis of key metabolites changes in the above pathways, corresponding exogenous addition strategies were proposed as follows: 1.0 g/L methyl oleate was added at 0 h; 1.0 g/L lysine was added at 12 h; 0.5 g/L shikimic acid was added at 24 h; 0.5 g/L sodium succinate, 0.1 g/L phenylalanine, 0.1 g/L tryptophan, and 0.1 g/L tyrosine were added at 36 h, successively, and a redesigned fermentation medium (M3) was obtained finally on the basis of M2. The production of rapamycin in M3 was increased by 56.6 % compared with it in M2, reaching 307 mg/L at the end of fermentation (120 h). These results demonstrated that metabolic profiling analysis was a successful method applied in the rational guidance of the production improvement of rapamycin, as well as other industrially or clinically important compounds.

Abstract:Strains producing β-glucosidase were isolated by solid medium containing esculin and ferric.Then six strains having high β-glucosidase activity were selected with p-nitropheny l-β-D-glucopyranoside as substrate.Strain having high ginsenoside bioconversion was finally chosen with Rbl as substrate and converted products were determined by TLC and HPLC.β-glucosidase activity of this strain was 0.803 U/mL and ginsenoside Rd conversation rate was 97.234 2％.The selected strain was identified as Saccharomycopsisfibuligera based on the morphology and nucleotide sequence analysis of the 18S rDNA.Fermentation medium was optimized to contain lactose as carbon source and corn flour as nitrogen source,and Ca2+ as metal ion.Finally inducer was optimized to promote the ginsenoside bioconversion.Results indicated that inducer had little impact on the bioconversion but rutin,a kind of inducer,could promote the bioconversion of Rd to CK.The selected strain with high ginsenoside bioconversion had good potential application.

Combining Metabolomics and Network Analysis to Improve Tacrolimus Production in Streptomyces Tsukubaensis Using Different Exogenous Feedings

Abstract:Tacrolimus is widely used as an immunosuppressant in the treatment of various autoimmune diseases. However, the low fermentation yield of tacrolimus has thus far restricted its industrial applications. To solve this problem, the time-series response mechanisms of the intracellular metabolism that were highly correlated with tacrolimus biosynthesis were investigated using different exogenous feeding strategies in S. tsukubaensis . The metabolomic datasets, which contained 93 metabolites, were subjected to weighted correlation network analysis (WGCNA), and eight distinct metabolic modules and seven hub metabolites were identified to be specifically associated with tacrolimus biosynthesis. The analysis of metabolites within each metabolic module suggested that the pentose phosphate pathway (PPP), shikimate and aspartate pathway might be the main limiting factors in the rapid synthesis phase of tacrolimus accumulation. Subsequently, all possible key-limiting steps in the above metabolic pathways were further screened using a genome-scale metabolic network model (GSMM) of S. tsukubaensis . Based on the prediction results, two newly identified targets ( aroC and dapA ) were overexpressed experimentally, and both of the engineered strains showed higher tacrolimus production. Moreover, the best strain, HT- aroC/dapA , that was engineered to simultaneously enhanced chorismate and lysine biosynthesis was able to produce 128.19 mg/L tacrolimus, 1.64-fold higher than control (78.26 mg/L). These findings represent a valuable addition to our understanding of tacrolimus accumulation in S. tsukubaensis , and pave the way to further production improvements.

Abstract:Objective To improve geldanamycin production by studying fermentation process of geldanamycin-producing strain Streptomyces hygroscopicus SIPI.A.2039.Method Fermentation process was optimized by a geldanamycin-producing strain Streptomyces hygroscopicus SIPI.A.2039 reserved in our laboratory in aspects of shaker's rotate speed,flask capacity,carbon source,and 50L-fermentor's parameters.Results The results showed that the productivity of geldanamycin could reach 2500μg/mL at the fermentation period of 144 hours using optimized fermentation process in flask while the control was only 230μg/mL.Further studies were performed in 50L-fermentor and the results showed that under optimized dissolve oxygen supply and feed batch process,the maximum productivity could reach 3700μg/mL and the time cause could be shortened by 48 hours in comparison of condition in the flask.Conclusion This result is twice higher than that reported in the literatures or patents at home and abroad up to now and the process is easy to scale up further.
Sirolimus formulation with improved pharmacokinetic properties produced by a continuous flow method

Abstract:The oral bioavailability of Sirolimus is limited by poor dissolution of the compound in the gastrointestinal tract resulting in a low bioavailability and large inter-individual differences in blood levels. Several different formulation approaches were applied to overcome these disadvantageous pharmacokinetic properties including the marketed oral solution and a tablet form containing wet milled nanocrystals. These approaches deliver improved pharmacokinetics, yet, they share the characteristics of complex production method and composition. We have developed a nanostructured Sirolimus formulation prepared by the controlled continuous flow precipitation of the compound from its solution in the presence of stabilizers. We have shown that contrary to the batch production the process could be easily intensified and scaled up; apparently the uniformity of the precipitation is heavily dependent on the production parameters, most likely the mixing of the solvent and antisolvent. We compared the physicochemical and pharmacokinetic properties of the nanostructured formula with the marketed nanoformula. We found that our method produces particles in the size range of less than 100nm. The solid form redispersed instantaneously in water and in biorelevant media. Both the solid form and the redispersed colloid solution showed excellent stability even in accelerated test conditions. The oral administration of the nanostructured formula resulted in faster absorption, higher exposure and higher trough concentrations when compared to the marked form. These advantageous properties could allow the development of solid oral Sirolimus formulae with lower strength and gel based topical delivery systems.
Separation and identification of Rap-D1,the isomer of sirolimus

Abstract:Rap-D1,the isomer of sirolimus,was isolated from the culture broth of sirolimus producing strain Streptomyces hygroscopicus mutant FC904-25. The chemical structure was determined by spectral data such as UV,IR,LC-MS,NMR and X-ray single crystal diffraction. Rap-D1 was identical with 28-epirapamycin.

Abstract:FK506, a widely used immunosuppressant, is produced by industrial fermentation processes using various Streptomyces species. However, the low titer becomes a bottleneck for its application and industrialization. It urgently required a full understanding of the biological mechanisms for FK506 overproduction. Towards this end, comparative metabolomics approach was employed to analyze metabolite concentrations difference of Streptomyces tsukubaensis cultivated in two media with low and high productivities. Initially, 98 intracellular metabolites were identified and 13 metabolites involved in five pathways were determined to be directly correlated with FK506 biosynthesis. Then in-depth analysis elucidated how those key factors exerted influence on FK506 biosynthesis. Many previously unreported metabolites were shown to play an important role in FK506 biosynthesis and provided potential regulation points for external manipulation. Based on such key information, rationally designed feeding strategy was carried out. Results showed that the FK506 yield increased from 251 to 405mg/L, whereas, by-products FK520 and 37,38-dihydro-FK506 decreased by 31% and 39%, respectively, compared with the values of control. To our knowledge, it is the first study to apply the comparative metabolomics method to identify key metabolites to promote the FK506 production. Abstract:FK506 (tacrolimus) is a 23-membered macrolide antibiotic with immunosuppressive activity, which is now widely used clinically in bone marrow, liver, and kidney transplantation. However, large-scale industrial use of FK506 is limited due to its low yield. In this study, we employed a series of strategies to enhance FK506 production. First, a strong promoter gapdhp was identified in S. tsukubaensis L19 through the XylE reporter system. Then two cluster-situated regulators (CSRs), FkbN and Tcs7, were co-overexpressed under the control of gapdhp to generate strain L24, which showed significant improvement in FK506 titer from 171 mg/L to 434 mg/L. Supplementing the growth medium with alanine and isoleucine further improved production to 493 mg/L and 542 mg/L, respectively. Next, the significant components of the medium were determined by Plackett-Burman (PB) design, with optimum values of yeast extract, cottonseed meal and KH2PO4 found to be 15.9 g/L, 17.2 g/L, and 5.0 g/L, respectively. Using Box-Behnken (BB) design based on response surface methodology (RSM), a model was developed to predict the maximum titer with 747 mg/L. With the optimized medium, scale-up production of FK506 by strain L24 in a 5 L fermenter reached a maximum titer of 756 mg/L after 192 h fermentation.
[Rational Design of a 500 M3 Fermenter for Erythromycin Production by Saccharopolyspora Erythraea].

Abstract:Erythromycin is a macrolide antibiotic produced by Saccharopolyspora erythraea. Its yield is greatly affected by the fermentation conditions and the bioreactor configurations. In this study, a novel scale-up method for erythromycin fermentation was developed based on computational fluid dynamics (CFD) and time constant analysis. Firstly, the dissolved oxygen (DO) was determined as a key parameter according to the physiological properties of S. erythraea cultivated in a 50 L bioreactor. It was found that the time constant of oxygen supply (tmt) in a 500 m3 bioreactor should be less than 6.25 s in order to satisfy the organism's oxygen uptake rate (OUR). Subsequently, a 500 m3 bioreactor was designed using the time constant method combined with empirical correlations. The impeller combination with one BDT8 impeller at bottom and two MSX4 impellers at upper part was determined, and then validated by numerical simulation. The results indicated that the tmt of the bioreactor (< 6.25 s) and the fluid properties, including gas hold-up, shear stress and fluid vector, met the requirements of erythromycin fermentation. Finally, the industrial production of erythromycin in the 500 m3 showed the design method was applicable in large scale fermentation.

Abstract:The cultivation system requires that the approach providing biomass for all types of metabolic analysis is of excellent quality and reliability. This study was conducted to enhance the efficiency and yield of antifungal substance (AFS) production in Streptomyces yanglinensis 3–10 by optimizing operation conditions of aeration, agitation, carbon source, and incubation time in a fermenter. Dissolved oxygen (DO) and pH were found to play significant roles in AFS production. The optimum pH for the production of AFS in S. yanglinensis 3–10 was found to be 6.5. As the AFS synthesis is generally thought to be an aerobic process, DO plays a significant role. The synthesis of bioactive compounds can vary depending on how DO affects growth rate. This study validates that the high growth rate and antifungal activity required a minimum DO concentration of approximately 20% saturation. The DO supply in a fermenter can be raised once agitation and aeration have been adjusted. Consequently, DO can stimulate the development of bacteria and enzyme production. A large shearing effect could result from the extreme agitation, harming the cell and deactivating its products. The highest inhibition zone diameter (IZD) was obtained with 3% starch, making starch a more efficient carbon source than glucose. Temperature is another important factor affecting AFS production. The needed fermentation time would increase and AFS production would be reduced by the too-low operating temperature. Furthermore, large-scale fermenters are challenging to manage at temperatures that are far below from room temperature. According to this research, 28°C is the ideal temperature for the fermentation of S. yanglinensis 3–10.
