Abstract:Rap-D1,the isomer of sirolimus,was isolated from the culture broth of sirolimus producing strain Streptomyces hygroscopicus mutant FC904-25. The chemical structure was determined by spectral data such as UV,IR,LC-MS,NMR and X-ray single crystal diffraction. Rap-D1 was identical with 28-epirapamycin.

Abstract:Objective To isolate and identify a chemical compound,FIM-R24,extracted from the fermentation broth of rapamycin producing strain Streptomyces hygroscopicus FC904,which showed a RRT of 0.88 in the rapamycin HPLC analysis.Methods FIM-R24 was isolated by silica gel chromatography and preparative liquid chromatography,and its chemical structurewas determined by the spectral analysis including NMR,UV,IR,and HRMS.The anti-cancer activity of FIM-R24 was determined by the SRB method in vitro,and the antifungal activity was evaluated by the serial two-foldbroth dilution method.Result and Conclusion FIM-R24 was identical with rapamycin dialdehyde.By comparison,the anti-cancer activity of FIM-R24 was slightly weaker than that of rapamycin,and its anti-fungal activity was 8～15 times weaker than that of rapamycin.

Abstract:A reliable method to separate and identify glucosinolates in rapeseed has been reported. Roughed product would be got after extraction and ionic exchange, and then the product was purified with liquid chromatography. One of the best separated component would be confirmed as 4-pentenyl glucosinolate by means of mass spectrometry, nuclear magnetic resonance spectrometry, ultraviolet spectroscopy and infrared spectrometry. This research will be used as an effective method for getting special glucosinolate compounds.

Abstract:A complete and detailed characterization of Rapamycin ( 1 ) and Prolylrapamycin ( 2 ) has been conducted by homo- and hetero-nuclear NMR experiments in DMSO- d 6 along with HRMS and FT-IR spectra and DSCs analyses. The NMR experiments allowed the assignment of every single proton and carbon atom belonging to the two structures and the definitive confirm of the presence of a pyrrolidine ring in Prolylrapamycin ( 2 ) in place of the piperidine ring that characterizes the structure of Sirolimus.

Abstract:Rapamycin is a triene macrolide antibiotic produced by Streptomyces hygroscopicus. Besides its wide application as an effective immunosuppressive agent, other important bioactivities have made rapamycin a potential drug lead for novel pharmaceutical development. However, the low titer of rapamycin in the original producer strain limits further industrialization efforts and restricts its use for other applications. Predicated on knowledge of the metabolic pathways related to rapamycin biosynthesis in S. hygroscopicus, we have rationally designed approaches to generate a rapamycin high producer strain of S. hygroscopicus HD-04-S. These have included alleviation of glucose repression, improved tolerance towards lysine and shikimic acid, and auxotrophy of tryptophan and phenylalanine through the application of stepwise UV mutagenesis. The resultant strain produced rapamycin at 450 mg/L in the shake flask scale. These fermentations were further scaled up in 120 and 20,000 L fermentors, respectively, at the pilot plant. Selected fermentation factors including agitation speed, pH, and on-line supplementation were systematically evaluated. A fed-batch strategy was established to maximize rapamycin production. With these efforts, an optimized fermentation process in the larger scale fermentor was developed. The final titer of rapamycin was 812 mg/L in the 120 L fermentor and 783 mg/L in the 20,000 L fermentor. This work highlights a high rapamycin producing strain derived by mutagenesis and subsequent screening, fermentation optimization of which has now made it feasible to produce rapamycin on an industrial scale by fermentation. The strategies developed here should also be applicable to titer improvement of other important microbial natural products on an industrial scale. Biotechnol. Bioeng. 2010; 107: 506-515. Isolation and purification of novel antifungal antibiotic from streptomyces ahygroscopicus

Abstract:A technically feasible purification process for a novel pyrimidine nucleoside antifungal antibiotic produced by a Streptomyces strain (Streptomyces ahygroscopicus ZJUIB001) was conducted. The centrifugated culture broth was extracted with chloroform and then with petroleum ether. The water phase solution was adjusted to pH=2 and further isolated by cation-exchange chromatography, which was followed by macroporous resin adsorption. Dowex 50×16 was screened as the most suitable cation-exchange resin for removing the impurity from the crude extract under the optimized operating conditions as follows: sample loading flow rate of 2 bed volume (BV) per hour and eluting flow rate of 1 BV per hour using 0.6 mol/L NH3·H2O as the mobile phase. Amberlite XAD7HP was selected as the most suitable macroporous adsorption resin for the further purification of the antibiotic. The optimized desorbing condition of macroporous resin adsorption chromatography was multi-step gradient elution with ethanol solutions of 10% and 70% volume fraction, respectively. Then the purified antifungal compound was subjected to high-performance liquid chromatographu (HPLC) system with a YMC-C18 (5 μm, 4.6 mm×250 mm) column. The mobile phase was 0.01 mol/L ammonium acetate buffer (pH 4.3). The flow rate was 1 mL/min and the elution was detected at 220 nm. HPLC results show that the purified antibiotic compound has a purity of 93.0% with antifungal activity of 21 632 U/mg.

Abstract:A powerful immunosuppressant sirolimus,a mTOR inhibitor,was transformed into four fractions(fraction1,2,3 and 4) by Actinoplanes sp.ATCC 12065.The results of HPLC showed the retention time of the four fractions were 25.41min,34.13min,48.46min and 52.24min,respectively.All four fractions possessed a UV spectra typical of sirolimus.The ESI mass spectra of fractions 1 and 2 were shown M.W.916,which increased in 2 mass units in comparison with sirolimus,however,fractions 3 and 4 were shown M.W.900,which reduced 14 mass units.In this paper,the primary results of microbial conversion of sirolimus by Actinoplanes sp.ATCC 12065 had been discussed.

Abstract:Objective:To develop a rapidaly method for the identification of rapamycin and its analoges.Methods:Electrospray ionization mass spectrometric method in negative mode was applied.Analysis was performed at the flow rate of dry gas 5.98 L·min-1 with the temperature at 300 ℃ and the pressure of nebulizing gas 137.93 kPa.The capillary voltage was set at 4 kV.Results:The fragmentation patterns and structural assignments of these substances were discussed.Under the experimental conditions,the main fragmentation pathways started by primary splitting between C27-C28.Some specific fragment ions were appeared in the mass spectra.Conclusion:The method with simplicity and specificity and sensitivity has been established for the rapidly identification of rapamycin and its analoges.

Abstract:Sirolimus, a powerful immunosuppressant, is produced by Streptomyces hygroscopicus FC904. In an attempt to improve its production, mutation and screening were carried out. Spore suspensions and the forming buds were treated with UV, nitrosoguanidine (NTG) and ethylmethane sulfonate (EMS), some of survivors were selected and detected their resistance to sirolimus. A mutant FC904-33 with higher productivity of sirolimus was obtained and the features of the mutant such as fewer by-products and the susceptibilities to antibiotics as well as its cultural characteristics were presented.

Abstract:Bacterium strain 287 isolated from the soil samples in Fuzhou, China was identified as Bacillus megaterium based on the taxonomic properties and 16s rRNA gene sequence. Bacillus megaterium 287 biotransformed sirolimus to three compounds: 287-P1, 287-P2 and 287-P3. Among them, 287-P1 was identified as 29,42-bis-O-demethylrapamycin by physico-chemical properties and spectral analyses.

Abstract:In the course of searching for new rapamycin derivatives by microbial conversion, a microorganism, which was able to biotransform rapamycin into four products, was obtained and classified as Micromonospora chersina FIM03-712. One conversion product from the converted broth of the strain was isolated and purified. By means of physico-chemical properties and spectral analyses, it was identified with 14-deoxorapamycin.

Abstract:The present study was designed to identify the difference between two rapamycin biosynthetic gene clusters from Streptomyces hygroscopicus ATCC29253 and Actinoplanes sp. N902-109 by comparing the sequence and organization of the gene clusters. The biosynthetic gene cluster for rapamycin in Streptomyces hygroscopicus ATCC29253 was reported in 1995. The second rapamycin producer, Actinoplanes sp. N902-109, which was isolated in 1995, could produce more rapamycin than Streptomyces hygroscopicus ATCC29253. The genomic map of Actinoplanes sp. N902-109 has been elucidated in our laboratory. Two gene clusters were compared using the online software anti-SMASH, Glimmer 3.02 and Subsystem Technology (RAST). Comparative analysis revealed that the organization of the multifunctional polyketide synthases (PKS) genes: RapA, RapB, RapC, and NRPS-like RapP were identical in the two clusters. The genes responsible for precursor synthesis and macrolactone modification flanked the PKS core region in N902-109, while the homologs of those genes located downstream of the PKS core region in ATCC29253. Besides, no homolog of the gene encoding a putative type II thioesterase that may serve as a PKS "editing" enzyme accounted for over-production of rapamycin in N902-109, was found in ATCC29253. Furthermore, no homologs of genes rapQ (encoding a methyltransferase) and rapG in N902-109 were found in ATCC29253, however, an extra rapM gene encoding methyltransferase was discovered in ATCC29253. Two rapamycin biosynthetic gene clusters displayed overall high homology as well as some differences in gene organization and functions.

Abstract:Homotemsirolimuses A, B, and C (2a, 2b, 2c), were found to be minor components of a temsirolimus preparation made from rapamycin. These three temsirolimus analogues are derived from the Corresponding rapamycin analogues, homorapamycins A, B, and C. (1a, 1b, 1c) produced by the strain Streptomyces hygroscopicus. The structures of homotemsirolimuses A, B, and C were determined by spectroscopic methods. These compounds were tested for mTOR kinanse inhibition and in two proliferation assays, using LNCap prostate and MDA468 breast cancer cells; The results suggested that the mTOR inhibition and antiproliferation potencies for 2a,, 2b, and 2c are comparable to those of rapamycin (1) and temsirolimus (2).

Abstract:Quantification of the new immunosuppressant sirolimus (syn. rapamycin; Rapamune) in whole blood by chromatography is essential for its clinical use since no immunoassay is available although monitoring is mandatory. Here we report on a rapid and convenient liquid chromatography (LC-tandem mass spectrometry method and describe our practical experience with its routine use. Whole blood samples were hemolyzed and deproteinized using an equal volume (150 microl) of a mixture of methanol/zinc sulfate solution containing the internal standard desmethoxy-rapamycin. After centrifugation, the clear supernatants were submitted to an on-line solid-phase extraction procedure using the polymeric Waters Oasis HLB material, with elution of the extracts onto the analytical column in the back-flush mode by column switching. For analytical chromatography a RP-C18 column was used with 90/10 methanol/2 mM ammonium acetate as the mobile phase. A 1:10 split was used for the transfer to the mass spectrometer, a Micromass Quattro LC-tandem mass spectrometry system equipped with a Z-spray source and used in the positive electrospray ionization mode. The following transitions were recorded: sirolimus, 931>864 m/z, and desmethoxy-rapamycin (I.S.), 901>834 m/z. The analytical running time was 5 min, including on-line extraction. The method has a linear calibration curve (r>0.99; range 1.6-50 microg/l) and is rugged and precise with monthly CVs <7% at a sirolimus concentration of 13.1 microg/l in routine use; the instrumentation proved to be reliable and required minimal maintenance. Abstract:Rapamycin is a macrolide antibiotic with antifungal, immunosuppressive, antitumor, and lifespan-extension activities etc. In order to increase its structural diversity, microbial transformation of rapamycin was conducted. More than 3 converted compounds were shown in the microbial conversion culture of rapamycin by Bacillus subtilis CGMCC7764. By extensive NMR and MS spectroscopic analyses, the major compound was identified as 43-O-(beta-D-glucoside)-rapamycin. This compound has an inhibitory effect on tumour growth, and its activity is similar to rapamycin.

Abstract:The natural product rapamycin, produced during fermentation by Streptomyces hygroscopicus, is known for its potent antifungal, immunosuppressive, and anticancer activities. During rapamycin biosynthesis, the amino acid l-pipecolate is incorporated into the rapamycin molecule. We investigated the use of precursor-directed biosynthesis to create new rapamycin analogs by substitution of unusual l-pipecolate analogs in place of the normal amino acid. Our results suggest that the l-pipecolate analog (+/-)-nipecotic acid inhibits the biosynthesis of l-pipecolate, thereby limiting the availability of this molecule for rapamycin biosynthesis. We used (+/-)-nipecotic acid in our precursor-directed biosynthesis studies to reduce l-pipecolate availability and thereby enhance the incorporation of other pipecolate analogs into the rapamycin molecule. We describe here the use of this method for production of two new sulfur-containing rapamycin analogs, 20-thiarapamycin and 15-deoxo-19-sulfoxylrapamycin, and report measurement of their binding to FKBP12.

Abstract:The title compound, C51H78N4O12, is a derivative of rapamycin, a triene macrolide antibiotic molecule isolated from Streptomyces hygroscopicus. The macrocyclic ring structure has 15 chiral centres, with one of the substituent hydroxy groups giving an intramolecular hydrogen bond to a ketone O-atom acceptor. The molecules also form intermolecular hydroxy-ketone O-H center dot center dot center dot O hydrogen-bonding associations, giving one-dimensional chains extending along (010).
