Since a solid dispersion is basically a drug-polymer two-component system, the drug-polymer interaction is the determining factor in its design and performance. In this review, we summarize our current understanding of solid dispersions both in the solid state and in dissolution, emphasizing the fundamental aspects of this important technology.

Abstract:Background: In the development of drug delivery systems, drugs' solubility remains the most challenging constraint. Many newly synthesized chemical compounds are available, but they involve low solubility and poor permeability restrictions. Among various drug delivery systems, the utilization of solid dispersion technologies has become more focused due to their promising benefits. Objective: This technology has attracted extensive attention for dissolution rate improvement along with substantial bioavailability enhancement of poorly water-soluble drug candidates. Methods: Many approaches have been employed for preparing solid dispersions, such as the melting method, hot melt extrusion, solvent evaporation process, fusion and kneading method, spray drying technique, co-grinding and freeze drying, supercritical fluid technology, etc. Result: A wide variety of hydrophilic and hydrophobic materials are available as carriers, which are employed in the formulation of solid dispersions. Depending on the carrier characteristics, immediate- release solid dispersions and/or controlled-release solid dispersions can be formulated. Multiple hydrophilic materials have been explored for heightening dissolution features with enhanced bioavailability of poorly water-soluble drug molecules. The availability of commercially available products further validates the utility of solid dispersion technology in drug delivery systems. Conclusion: In the current manuscript, an attempt has been made to highlight the comprehensive development techniques, characterization techniques, recent solid dispersion technologies, clinical trial studies, and patented technology, along with studies heightening the dissolution behavior of numerous poorly aqueous soluble drugs. The major stability issues affecting the suitability of solid dispersions are also discussed.

Abstract:OBJECTIVE To summary of progress in the study on physical stability and anti-aging of soild dispersion.METHODS The relevant articles in recent years were referred and summarized.RESULTS Thermodynamic and kinetic factors for the physical stability of solid dispersion were reviewed.Suggestions about the formulation design,preparation method and storage conditions to improve the physical stability of solid dispersion were also introduced.CONCLUSION The intensive study of physical stability mechanism of soild dispersion will promote the development of this technology in pharmaceutical research.

Abstract:The concentrated ceramic suspension with good dispersity is the key to colloidal forming process.We have introduced the stabilization mechanism of polymer dispersants and discussed the investigation and application of the dispersants in single-phase and multiphase concentrated ceramic suspensions.Further more,the various factors on dispersing effect are also analyzed.Such factors include intrinsic characters of polymer dispersants,Physical-Chemical properties of ceramic dispersed phase and dispersion medium,as well as operation technology of colloidal forming process.Finally the research fronts of the polymer dispersant which can optimize properties of the concentrated ceramic suspension are pointed out.

Abstract:Hyperdispersant is a kind of super-efficient polymer dispersants. Due to the anchoring group and solvatable chain instead of hydrophilic groups and lipophilic groups of the traditional dispersant, it has more efficient dispersion and wide range of applications with uniqueness mechanism and diversity of species. In this paper, the hyperdispersant application in the solid-liquid suspension system was described in detail, and future research directions were predicted.

Abstract:Aim The objective of this study was to prepare and characterize quercetin-polyvinylpyrrolidone (Qurc-PVP)solid dispersion with the intention of improving its dissolution properties. Methods Qurc-PVP sclid dispersion was prepared by solvent method. The release rate of quercetin was determined from dissolution studies and the physicochemical properties of solid dispersion were investigated by differential scanning calorimetry (DSC), infrared spectroscopy (IR), powder X-ray diffraction (PXRD) and scanning electron microscopy (SEM). Results The results showed that the dissolution rate of quercetin was significantly improved by solid dispersion compared to that of the pure drug and physical mixture. Solubility studies revealed a markedly increase in the solubility of quercetin. The results of DSC and PXRD showed that the quercetin in solid dispersion was amorphous form. From SEM analysis, there was no quercetin crystal observed in the solid dispersions.Conclusion The solubility and dissolution of quercetin were improved by solid dispersion technique.

Abstract:Introduction Amorphous solid dispersion (ASD) technique has recently been used as an effective formulation strategy to significantly improve the bioavailability of insoluble drugs. The main industrialized preparation methods for ASDs are mainly hot melt extrusion and spray drying techniques; however, they face the limitations of being unsuitable for heat-sensitive materials and organic reagent residues, respectively, and therefore novel preparation processes and technology coupling for developing ASDs have received increasing attention.

Abstract:The new methods of physicochemical characterization, release model and release mechanism, stability study of solid dispersion were reviewed. The application of three new analytical methods in physicochemical characterization, including Raman spectroscopy,FTIR spectroscopic imaging and Hyper differential scanning calorimetry, was introduced. Drug release mathematical model, carrier-controlled drug dissolution and drug-controlled dissolution, and Lacoulonche thermodynamic model of solid dispersion were discussed. The facts that they influence the stability of solid dispersion were also summarized.

Abstract:Background: Solid dispersions are an effective formulation technique to improve the solubility, dissolution rate, and bioavailability of water-insoluble drugs for oral delivery. In the last 15 years, increased attention was focused on this technology. There were 23 marketed drugs prepared by solid dispersion techniques. Objective: This study aimed to report the big picture of solid dispersion research from 1980 to 2015. Method: Scientific knowledge mapping tools were used for the qualitative and the quantitative analysis of patents and literature from the time and space dimensions. Results: Western Europe and North America were the major research areas in this field with frequent international cooperation. Moreover, there was a close collaboration between universities and industries, while research collaboration in Asia mainly existed between universities. The model drugs, main excipients, preparation technologies, characterization approaches and the mechanism involved in the formulation of solid dispersions were analyzed via the keyword burst and co-citation cluster techniques. Integrated experimental, theoretical and computational tools were useful techniques for in silico formulation design of the solid dispersions. Conclusions: Our research provided the qualitative and the quantitative analysis of patents and literature of solid dispersions in the last three decades.
Facile preparation of solid dispersions by dissolving drugs in <i>N</i>-vinyl-2-pyrrolidone and photopolymerization

Abstract:Drug solid dispersions improve the dissolution of drugs in aqueous media for enhancement of oral bioavailability. The current preparation methods of drug solid dispersions mainly involve the evaporation of solvents or the melting of drugs and matrix. Here, we create a new and simple method for the preparation of drug solid dispersions by dissolving drugs in N-vinyl-2-pyrrolidone (NVP) and then NVP photopolymerization. A variety of drugs were explored to find whether they were suitable for this method and only some of them were soluble in NVP and formed transparent and hard solid dispersions, including fluconazole, ketoconazole, bifonazole, miconazole nitrate, sulfamethoxazole, aspirin, ibuprofen and artesunate. The formation of photocuring solid dispersions was highly related to the free radical scavenging function of drugs. Those drugs with strong free radical scavenging capability, including curcumin, resveratrol, quercetin, genistein, puerarin, nicergoline, olanzapine, indomethacin, did not form solid dispersions. They scavenged 2,2-diphenyl-1-picrylhydrazyl free radicals, which was demonstrated by ultraviolet spectrometry and electron spin resonance. The scavenging of free radicals stopped the chain polymerization of NVP. The Fourier transform infrared spectra, X-ray diffraction and differential scanning calorimetry of ibuprofen solid dispersions and artesunate solid dispersions showed the molecularly miscible state of the drugs and the hydrogen bonding between the drugs and polyvinyl pyrrolidone. The NVP-based solid dispersions of the two drugs had faster and more complete dissolution than their traditional solid dispersions. The NVP photopolymerization-based solid dispersion method provides a new choice for the production of solid dispersions in the research and industrial fields.

Abstract:There is a review on theory and practice of synthesis of super-high solid content aqueous polyurethane dispersions.From the mathematical model of close packing of sphere particles,the maximal volume fraction of dispersed phase was calculated.From the dispersion particle's structure,which contains large amount of swelling water,the difference between volume fraction and solid content was analyzed.The necessities to use emulsifiers and self-emulsifiers and high internal phase ratio dispersion process to prepare super-high solid content aqueous polyurethane dispersion were discussed.The development in higher solid content aqueous polyurethane dispersion was introduced.
