Abstract:Background: In the development of drug delivery systems, drugs' solubility remains the most challenging constraint. Many newly synthesized chemical compounds are available, but they involve low solubility and poor permeability restrictions. Among various drug delivery systems, the utilization of solid dispersion technologies has become more focused due to their promising benefits. Objective: This technology has attracted extensive attention for dissolution rate improvement along with substantial bioavailability enhancement of poorly water-soluble drug candidates. Methods: Many approaches have been employed for preparing solid dispersions, such as the melting method, hot melt extrusion, solvent evaporation process, fusion and kneading method, spray drying technique, co-grinding and freeze drying, supercritical fluid technology, etc. Result: A wide variety of hydrophilic and hydrophobic materials are available as carriers, which are employed in the formulation of solid dispersions. Depending on the carrier characteristics, immediate- release solid dispersions and/or controlled-release solid dispersions can be formulated. Multiple hydrophilic materials have been explored for heightening dissolution features with enhanced bioavailability of poorly water-soluble drug molecules. The availability of commercially available products further validates the utility of solid dispersion technology in drug delivery systems. Conclusion: In the current manuscript, an attempt has been made to highlight the comprehensive development techniques, characterization techniques, recent solid dispersion technologies, clinical trial studies, and patented technology, along with studies heightening the dissolution behavior of numerous poorly aqueous soluble drugs. The major stability issues affecting the suitability of solid dispersions are also discussed.

Abstract:The new methods of physicochemical characterization, release model and release mechanism, stability study of solid dispersion were reviewed. The application of three new analytical methods in physicochemical characterization, including Raman spectroscopy,FTIR spectroscopic imaging and Hyper differential scanning calorimetry, was introduced. Drug release mathematical model, carrier-controlled drug dissolution and drug-controlled dissolution, and Lacoulonche thermodynamic model of solid dispersion were discussed. The facts that they influence the stability of solid dispersion were also summarized.

Abstract:Studies on polysaccharide microgels being used as the carrier for sustained and controlled drug delivery systems has become an important research field in polymeric science.In this paper,three kinds of polysaccharide microgels and their related application are reviewed.The physical method,chemical precipitation,reversed phase microemulsion,microemulsion-solvent volatilization and total aqueous phase method are used to synthesize polysaccharide microgels,including starch microgels,cellulose microgels and dextran microgels.The method of total aqueous phase without toxic solvents is the most promising way.The microgels show many advantages in drug sustained and controlled release,decreasing toxic,improving bioavailability in vivo,etc.Moreover,they are successively used to load and control the release of the medicine such as Insulin,Brufen,and so on.

Abstract:Introduction: Solid dispersion has been considered to be one of the most promising methods for improving the solubility and bioavailability of insoluble drugs. However, the physical stability of solid dispersions (SDs), including its aging and recrystallization, or phase separation, has always been one of the most challenging problems in the process of formulation development and storage. Areas covered: The high energy state of SDs is one of the primary reasons for the poor physical stability. The factors affecting the physical stability of SDs have been described from the perspective of thermodynamics and kinetics, and the corresponding theoretical model is put forward. We briefly summarize several commonly used techniques to characterize the thermodynamic and kinetic properties of SDs. Specific measures to improve the physical stability of SDs have been proposed from the perspective of prescription screening, process parameters, and storage conditions. Expert opinion: The separation of the drug from the polymer, the formation, and migration of drug crystals will cause the SDs to shift toward the direction of energy reduction, which is the intrinsic cause of instability. Furthermore, computational simulation can be used for efficient and rapid screening suitable for the excipients to improve the physical stability of SDs.

Abstract:<p>Amorphous solid dispersions (ASDs) are popular for enhancing the solubility and bioavailability of poorly water-soluble drugs. Various approaches have been employed to produce ASDs and novel techniques are emerging. This review provides an updated overview of manufacturing techniques for preparing ASDs. As physical stability is a critical quality attribute for ASD, the impact of formulation, equipment, and process variables, together with the downstream processing on physical stability of ASDs have been discussed. Selection strategies are proposed to identify suitable manufacturing methods, which may aid in the development of ASDs with satisfactory physical stability.</p>

Abstract:Background: Solid dispersions are an effective formulation technique to improve the solubility, dissolution rate, and bioavailability of water-insoluble drugs for oral delivery. In the last 15 years, increased attention was focused on this technology. There were 23 marketed drugs prepared by solid dispersion techniques. Objective: This study aimed to report the big picture of solid dispersion research from 1980 to 2015. Method: Scientific knowledge mapping tools were used for the qualitative and the quantitative analysis of patents and literature from the time and space dimensions. Results: Western Europe and North America were the major research areas in this field with frequent international cooperation. Moreover, there was a close collaboration between universities and industries, while research collaboration in Asia mainly existed between universities. The model drugs, main excipients, preparation technologies, characterization approaches and the mechanism involved in the formulation of solid dispersions were analyzed via the keyword burst and co-citation cluster techniques. Integrated experimental, theoretical and computational tools were useful techniques for in silico formulation design of the solid dispersions. Conclusions: Our research provided the qualitative and the quantitative analysis of patents and literature of solid dispersions in the last three decades.
Amorphous Solid Dispersions: Implication of Method of Preparation and Physicochemical Properties of API and Excipients

Abstract:The present study investigated the effect of different polymers and manufacturing methods (hot melt extrusion, HME, and spray drying, SD) on the solid state, stability and pharmaceutical performance of amorphous solid dispersions. In the present manuscript, a combination of different binary amorphous solid dispersions containing 20% and 30% of drug loadings were prepared using SD and HME. The developed solid-state properties of the dispersions were evaluated using small- and wide-angle X-ray scattering (WAXS) and modulated differential scanning calorimetry (mDSC). The molecular interaction between the active pharmaceutical ingredients (APIs) and polymers were investigated via infrared (IR) and Raman spectroscopy. The in vitro release profile of the solid dispersions was also evaluated to compare the rate and extend of drug dissolution as a function of method of preparation. Thereafter, the effect of accelerated stability conditions on the physicochemical properties of the solid dispersions were also evaluated. The results demonstrated higher stability of Soluplus® (SOL) polymer-based solid dispersions as compared to hydroxypropyl methylcellulose (HPMC)-based solid dispersions. Moreover, the stability of the solid dispersions was found to be higher in the case of API having high glass transition temperature (Tg) and demonstrated higher interaction with the polymeric groups. Interestingly, the stability of the melt-extruded dispersions was found to be slightly higher as compared to the SD formulations. However, the down-processing of melt-extruded strands plays critical role in inducing the API crystal nuclei formation. In summary, the findings strongly indicate that the particulate properties significantly influence the performance of the product.

Abstract:Although there was a great interest in solid dispersion systems during the past four decades to increase dissolution rate and bioavailability of poorly water-soluble drugs, their commercial use has been very limited, primarily because of manufacturing difficulties and stability problems. Solid dispersions of drugs were generally produced by melt or solvent evaporation methods. The materials, which were usually semisolid and waxy in nature, were hardened by cooling to very low temperatures. They were then pulverized, sieved, mixed with relatively large amounts of excipients, and encapsulated into hard gelatin capsules or compressed into tablets. These operations were difficult to scale up for the manufacture of dosage forms. The situation has, however, been changing in recent years because of the availability of surface-active and self-emulsifying carriers and the development of technologies to encapsulate solid dispersions directly into hard gelatin capsules as melts. Solid plugs are formed inside the capsules when the melts are cooled to room temperature. Because of surface activity of carriers used, complete dissolution of drug from such solid dispersions can be obtained without the need for pulverization, sieving, mixing with excipients, etc. Equipment is available for large-scale manufacturing of such capsules. Some practical limitations of dosage form development might be the inadequate solubility of drugs in carriers and the instability of drugs and carriers at elevated temperatures necessary to manufacture capsules.

Abstract:Solid dispersions have attracted considerable interest as an efficient means of improving the dissolution rate and hence bioavailability of a range of hydrotrophic drugs. Up to 40% of new chemical entities discovered by the pharmaceutical industry today are poorly soluble or lipophilic compounds. Solid dispersions of poorly water-soluble drugs with water-soluble carriers reduce the incidence of these problems and enhanced dissolution. Solid dispersion is one of the most promising approaches for solubility enhancement. The term solid dispersion refers to a group of solid products consisting of at least two different components, generally a hydrophilic matrix and a hydrophobic drug. The matrix can be either crystalline or amorphous. As per biopharmaceutical classification system class II drugs are with low solubility and high permeability and are the promising candidates for improvement of bioavailability by solid dispersion. Some of the practical aspects to be considered for the preparation of solid dispersions, such as selection of carrier, molecular arrangement of drugs in solid dispersions are discussed in this article. This article reviews the various preparation techniques for solid dispersion, characterization and compiles some of the recent technology transfers. Availability of a wide variety of polymers that are themselves poorly soluble or which swell under aqueous conditions suggests that solid dispersions have tremendous potential in the area of controlled release dosage forms. Abstract:Amorphous solid dispersion (ASD) is one of the most effective approaches for delivering poorly soluble drugs. In ASDs, polymeric materials serve as the carriers in which the drugs are dispersed at the molecular level. To prepare the solid dispersions, there are many polymers with various physicochemical and thermochemical characteristics available for use in ASD formulations. Polymer selection is of great importance because it influences the stability, solubility and dissolution rates, manufacturing process, and bioavailability of the ASD. This review article provides a comprehensive overview of ASDs from the perspectives of physicochemical characteristics of polymers, formulation designs and preparation methods. Furthermore, considerations of safety and regulatory requirements along with the studies recommended for characterizing and evaluating polymeric carriers are briefly discussed. Abstract:Drug-polymer solid dispersion has been demonstrated as a feasible approach to formulate poorly water-soluble drugs in the amorphous form, for the enhancement of dissolution rate and bioperformance. The solubility (for crystalline drug) and miscibility (for amorphous drug) in the polymer are directly related to the stabilization of amorphous drug against crystallization. Therefore, it is important for pharmaceutical scientists to rationally assess solubility and miscibility in order to select the optimal formulation (e.g., polymer type, drug loading, etc.) and recommend storage conditions, with respect to maximizing the physical stability. This commentary attempts to discuss the concepts and implications of the drug-polymer solubility and miscibility on the stabilization of solid dispersions, review recent literatures, and propose some practical strategies for the evaluation and development of such systems utilizing a working diagram.

Abstract:Solid dosage forms are better than liquid dosage forms in many ways, such as improved physical and chemical stability, ease of storage and transportation, improved handling properties, and patient compliance. Therefore, it is required to transform dosage forms of liquid origins into solid dosage forms. The functional approaches are to absorb the liquids by solid excipients or through drying. The conventional drying technologies for this purpose include drying by heating, vacuum-, freeze-and spray-drying, etc. Among these drying technologies, fluidbed drying emerges as a new technology that possesses unique advantages. Fluid-bed drying or coating is highly efficient in solvent removal, can be performed at relatively low temperatures, and is a one-step process to manufacture formulations in pellet forms. In this article, the status of the art of manufacturing solid dosage forms from bulk liquids by fluid-bed drying technology was reviewed emphasizing on its application in solid dispersion, inclusion complexes, self-microemulsifying systems, and various nanoscale drug delivery systems.
Applications of supercritical fluid transport technology in preparation of controlled-release drug delivery systems

Abstract:Among the research of new pharmaceutic dosage forms, controlled-release drug delivery system is a very important issue nowdays. In this field, processes using supercritical fluid technology are mostly "clean" process leading to "clean" products. Meanwhile, inorganic porous materials are emerging as a new category of host/guest systems due to some interesting features such as their biological stability and their drug-releasing properties. This review summarizes the applications of supercritical fluid transport technology in preparation of controlled-release drug delivery system in recent years and pays more attention on the method using supercritical fluid as the solvent and inorganic mesoporous materials as the support to prepare this controlled-release drug delivery system. The technical principle, development of technological process and the main influence factors are discussed here besides the drug release experiments and the comparison with the traditional methods. It shows clearly the advantages and disadvantages of various processes, and sums up the superiority of the supercritical transport technology in preparing controlled-release drug delivery system. Although this technique has lots of advantages, as for the papers delivered at present, the research on supercritical fluid transport technology is just at its initial stage of development because there are so many factors influencing the experimental results and these factors are sometimes link-coupled. It is still challenging to make the preparation controllable. It indicates that the diffusion and penetration of the supercritical carbon dioxide drug solution in porous materials, the surface chemical and physical adsorption mechanism should be focused on as well as the controlled drug release mechanism, thermodynamic model and process dynamic.
