Abstract:This study focuses on the aggregation behavior of the biodegradable amphiphilic block copolymers based on methoxy poly(ethylene glycol) (mPEG) as a hydrophilic block and either crystalline poly(caprolactone-b-l-lactide) (P(CL-LLA)) or amorphous poly(caprolactone-b-D,L-lactide) (P(CL-DLLA)) as a hydrophobic block. These block copolymers have a strong tendency to form micelles in aqueous medium, with very low critical micelle concentrations (CMCs). The CMC of P(CL-LLA)-b-mPEG is higher than that of P(CL-DLLA)-b-mPEG when the mPEG block has the same molecular weight. Furthermore, the partition equilibrium coefficient (K(v)) of pyrene in the micellar solution of P(CL-LLA)-b-mPEG copolymer was lower than that of P(CL-DLLA)-b-mPEG copolymer when the mPEG block was the same length. These differences were believed to be related to the physical state of the core-forming blocks, i.e., the crystalline P(CL-LLA) block and the amorphous P(CL-DLLA) block. The TEM images showed that micelles formed by P(CL-LLA)-b-mPEG assembled in a cylindrical morphology, whereas those formed by P(CL-DLLA)-b-mPEG took a classical spherical shape. In addition, with differential scanning calorimetry (DSC) and wide-angle X-ray diffraction (WAXD) analyses, it is believed that the crystallization tendency of the core-forming blocks is the main factor governing the morphology of micelles in water. A possible mechanism for the cylindrical assembly morphology was discussed.

Abstract:In this paper, the main factors which may influence the micelle morphology and behavior of amphiphilic block copolymers in selective solvents are reviewed. These factors mainly include the solution temperature, the type of selective solvents, the lengths of blocks, the crystalline block, the hydrogen-bond interaction between the blocks and the solvent as well as the concentration of the block copolymers. The experimental methods for controlling the micelle morphology of amphiphilic block copolymers in selective solvents are then summarized. The recent advances in the micellar behaviors of a special type of amphiphilic block copolymers in selective solvents, which are capable to response to the external stimuli such as the environmental temperature and pH value, are also presented. Finally, the existing problems and future possible development directions of this research field are discussed.

Abstract:The synthesis of a new kind of amphiphilic poly(vinyl-co-ester)s via simple one-pot radical ring-opening polymerization (RROP) is reported. A series of biodegradable, biocompatible, and cross-linkable copolymers P(MDO-co-PEGMA-co-CMA) with different compositions were synthesized by copolymerization of 2-methylene-1,3-dioxepane (MDO), poly(ethylene glycol) methyl ether methacrylate (PEGMA) and 7-(2-methacryloyloxyethoxy)-4-methylcoumarin methacrylate (CMA). All the copolymers were well characterized by H-1 NMR, C-13 NMR, and gel permeation chromatography (GPC). The hydrolytic degradation and enzymatic degradation of the copolymers were investigated. The amphiphilic P(MDO-co-PEGMA-co-CMA) can self-assemble into micelles in aqueous solution. Polymeric micelles with different sizes were obtained through tuning the compositions of the copolymers, i.e., the hydrophilic/hydrophobic ratio. The size and morphology of the micelles were measured by dynamic light scattering (DLS) and transmission electron microscopy (TEM). Because of the coumarin side groups in the micellar core, the micelles can be cross-linked upon exposure to long-wavelength UV light (lambda = 365 nm). These micelles also showed excellent biocompatibility by the in vitro cytotoxicity experiments. Furthermore, the biodegradable micelles were applied for the delivery of an anticancer drug doxorubicin (DOX). The cell viability of DOX-loading micelles compared to free DOX was discussed. The effects of cross-linking and the copolymer composition on the drug release behavior were well studied. The results indicate that this new kind of amphiphilic poly(vinyl-co-ester) s could serve as promising nanocarriers for the delivery of anticancer drugs.

Abstract:Amphiphilic block copolymers can form polymeric micelles through self-assembly in aqueous solution,in which the hydrophobic blocks constitute the core of micelle which allows to encapsulate hydrophobic drugs,while the hy-drophilic blocks constitute the shell of micelle which benefits the stability of micelle and long circulation in the blood-stream.Polymeric micelles have been widely studied as drug carriers,and some of them have entered the stages of clinical trials and application.We summarize the characteristics of polymeric micelles,and introduce the composition of polymeric micelles,the preparation methods of drug-loading micelles,and examples of polymeric micelles as drug delivery systems. Finally,we prospect the research direction of polymeric micelle drug-loading system in the future.

Abstract:Polymeric micelles self-assemblied by amphiphilic block copolymers in aqueous media are thought to be promising drug carriers with nanometer size.Polymeric micelles possess several advantages as compared with other drug carriers,such as high loading capacity,small particle size,long-circulation time,targeting ability and so on.Recently,the research for polymeric micelles has been focused on the combination of the above properties to construct multifunctional polymeric micelles.This review highlights the recent progress in multifunctional polymeric micelles,including the classification and their applications.

Abstract:Two model drugs, indomethacin and vitamin E (alpha-tocopherol), which have different physical stae and similar hydrophobicity, were incorporated respectively into polymerc micelles in aqueous media by dialysis method. The micelles were formed from two types of biodegradable amphiphilic block copolymers based on methoxy poly(ethylene glycol) (mPEG) as a hydrophilic block and either crystalline poly(epsilon-caprolactone-b-L-lactide) (P (CL-b-LLA)) or amorphous poly(epsilon-caprolactone-b-D, L-lactide) (P (CL-b-DLLA)) as a hydrophobic block. The morphology and drug loading property of the polymeric micelles were characterized by transmission electron microscopy (TEM), laser light scattering (LLS), ultraviolet (UV) spectroscopy measureemnts. It was found that incorporating drugs into the micelles resulted in significant changes of the micellar morphologies. The different behaviors of the morphological change depended mainly on the crystalline property of the core-forming blocks. The morphology of micelles formed with P(CL-b-LLA)-b-mPEG changed from short rod-like cylinders to wormike ones; whereas those formed with P(CL-b-DLLA)-b-mPEG changed from spheres to cylinders. In addition, the transformation of the micellar morphologies upon the drug entrapment was influenced by the hydrophobicity of the drugs more significantly than by the physical state of the drugs, while the drug loading amount was affected by the comatibility between carriers and drugs. Much more liquid-like drugs were entrapped in the amphiphilic copolymers with non-crystalline core.

Abstract:Recently the crystallization-driven self-assembly of block copolymers into semicrystalline micelles in the selective solvent has been paid increasing attention due to the good controllability. In this paper we first review some factors controlling the morphology and size of the semicrystalline micelles of block copolymers, including solvent quality, structure of block copolymers, and crystallization temperature. The living growth of semicrystalline micelles with time and formation of "block co-micelles" using micelles as building blocks are also introduced. Finally, currently existing problems and outlook in this field are discussed.

Abstract:Multimolecular assemblies of block copolymers have received considerable attention from both fundamental and applied standpoints. These assemblies are formed in selective solvents based on the different solubilities of constitutent segments; for example, hydrophilic and hydrophobic segments in an aqueous medium. Although the assemblies may assume various morphologies, including spherical, rod and lamellar structures, spherical micelles have been most extensively investigated. Polymeric micelles have a fairly narrow size distribution and are characterized by their unique core–corona structure. Their physicochemical properties have been extensively studied and higher-order controlled association has also been attained. This review highlights the recent progress in physicochemical characterization of the polymeric micelles and their applications as functional materials. Particular focus is placed on their applications in drug delivery systems, capillary electrophoresis and surface modification. Further, novel polymeric micelle formation through electrostatic interaction in aqueous medium is reviewed from the standpoint of their utility as bioreactors, diagnostic tools and non-viral gene vectors.

Abstract:To review the research of block copolymer micelles as delivery system for poorly soluble(antineoplastic) carrier over the last 10 years,including the composition,preparation methods and factors which influence the loading efficiency,physicochemical properties,and targeting characteristics of block copolymer micelles.Block copolymer micelles are self-assembled structures formed from amphiphilic block copolymers by dispersing in aqueous media.The hydrophilic blocks of the copolymer form the outer shell of the micelle,while the hydrophobic blocks form the inner core,and the proper coreshell micellar architecture was constituted.Block copolymer micelles have a whole set of unique properties,such as small sizes,narrow particle size distribution,high drug loading capacities,and available disposition characteristics in the body.Block copolymer micelles have been found as promising drug carriers due to making poorly soluble antineoplastic lysis,toxicities and side effects decrease,bioavailability increase,and targeting the drugs to specific sites in a passive way or attaching ligands in an active way,which can be specifically(recognized) by receptors onto the surface of copolymers.

The synthesis and properties of more complicated block and micellar architectures such as triblock- and graft copolymers, Janus micelles, and core-shell cylinder brushes are reviewed as well. Investigations reveal details of the interactions of polyelectrolyte layers and electro-steric stabilization forces.

Abstract:Amphiphilic copolymers that can assemble into core-shell micelles in aqueous media have potential application as drug carriers for controlled-release. In this article an amphiphilic triblock copolymer poly(ethylene oxide)-block-poly(lactic acid)-block-poly(2-hydroxyethyl methacrylate) (PEG-b-PLA-b-PHEMA) was synthesized by ring-opening reaction and atom transfer radical polymerization. The polymers were characterized by H-1 NMR, FT-IR and gel permeation chromatography. In aqueous solution, PEG-b-PLA-b-PHEMA can self-assemble into micelles. Characterized by DLS and TEM, micelles are found to be spherical with uniform size. Large numbers of hydroxyl groups on the PHEMA chain offer reaction sites for subsequent functional modification, which make it a promising platforms for drug delivery systems.

Abstract:A linear amphiphilic multiblock copolymer (PNIPAm-PtBA-PNIPAm)m was successfully synthesized by a two-step reversible addition–fragmentation transfer (RAFT) polymerization in the presence of a cyclic trithiocarbonate as RAFT agent. The micelle behavior of (PNIPAm-PtBA-PNIPAm)m multiblock copolymer in aqueous solution was then investigated by means of normal TEM, cryo-TEM, static and dynamic light scattering. The morphology, size, and size distribution of (PNIPAm-PtBA-PNIPAm)m micelles were found to be dependent on the initial concentration of multiblock copolymer in THF. Spherical micelles, associated aggregates of spherical micelles, cage-like micelles, layered structures, and vesicular micelles were experimentally observed, which were in good agreement with the prediction of theory and simulations on linear amphiphilic multiblock copolymer in selective solvent. The (PNIPAm-PtBA-PNIPAm)m micelles also exhibit thermo-sensitive behavior in aqueous solution because of the PNIPAm blocks.

Abstract:The complex microstructures assembled by amphiphilic diblock copolymer in dilute solution have been investigated by the application of real-space self-consistent field theory in two-dimensional space. Different micelles, such as spherelike micelles, rodlike micelles, and vesicles are produced. The simulations show that vesicles are metastable in dilute solution within a certain range of some parameters. The mechanism for the formation of micromorphologies, especial vesicles, is discussed.

Abstract:This outlook paper focuses on micelles formed by introducing more than one kind of polymer blocks with different properties into the same micelle.It can be a plausible candidate that is capable of tuning the resulting properties and meeting various requirements for specific applications.Many parameters may anticipate to affect the formation of stable mixed micelles.Several typical forming types and structures are discussed.There are micelles with different internal structure such as micelles with a heterogeneous core and a homogeneous corona or micelles with a homogeneous core and a mixed corona or core-shell-corona micelles.They possess characteristic stability,drug delivery and stimuli-responsibility.Finally,potential applications of these objects are discussed.Composite structures are expected to realize synergistic drug loading,channel release and drug controlled release.It will play a more important role in the field of drug controlled release.

Abstract:<p>Since the foundation of macromolecular science by Staudinger, advances in polymer chemistry have unveiled enormous avenues for controlling chemical structures at the molecular scale. As a typical self-assembling polymer covalently linked by chemically distinct polymer blocks, amphiphilic block copolymers (BCPs) spontaneously form a variety of nanostructures with diverse functional applications, especially in the fields of nanomedicine and biomaterials. Self-assembling processes could occur at single chain level (i.e., intrachain folding) or involve multiple chains with similar and dissimilar chemical structures (i.e., interchain association). This review article discusses recent trends concerning solution self-assembly of BCPs, including the formation of inverted mesophases, self-folding into single-chain nanoparticles (SCNPs), compartmentalized nanoparticles with sophisticated functions, and nanostructures of amphiphilic copolymers with precise chain lengths and predetermined sequences, by highlighting selected examples.</p>

Abstract:Self-assembled micro-paricles have been proven to be efficient microcarriers for delivering various therapeutic agents. Polycarbonates have demonstrated excellent biodegradability and biocompatibility as material for polymeric microcarriers. A diblock polycarbonate with tunable block length and active sites is synthesized via one-pot sequential ring-opening polymerization (ROP). The polymer acts as a material platform for versatile modifications to prepare a series of amphiphilic diblock polycarbonates which can self-assemble into microparticles. The effect of different hydrophilic pendent groups controlling the morphology and surface charge of the assemblies is systematically investigated. The non-equilibrium spherical particles with varying sizes are formed in one day, and the spheres can further assemble into multi-compartments vesicles (MCVs). Cationic, anionic or neutral carriers can be prepared depending on the type of pendant groups attached. The versatile amphiphilic copolymer assemblies have good potential in biomedical applications.

Abstract:At the end of the last century, Professor Jiang Ming et al. succeeded in fabricating regular nanostructures through localizing interactions in the systems of polymer complexes. Through continuous expansion and deepening of this principle and method, a new strategy of macromolecular self-assembly was established. This review summarizes recent developments and applications of non-covalently connected micelles (NCCMs), including: Introducing of new driving forces such as supramolecular interactions to drive the formation of NCCMs, providing the resultant NCCMs with new features and functions; Applying the principle to the self-assembly of biological macromolecules, thereby developing an entirely green route to self-assembly of macromolecules; Also, through localizing covalent crosslinking reaction of one block of a diblock copolymer, we realized micellization of the block copolymer in its common solvent induced by the crosslinking reaction, resulting in highly efficient fabrication of core-stabilized micelles as well as a new pathway for controlling the kinetics of polymer micellization, the structures and the properties of the polymeric micelles.

Abstract:The self-assembly of polymeric micelles and colloidal dispersions into intricate structures represents a transformative approach in contrast to traditional fabrication methods for creating new functional materials. Despite remarkable advancements in the field, there exists substantial potential for further progress. In the past two decades, scientists have innovated and applied sophisticated methods for synthesizing nanocrystals of diverse geometries and polymeric materials with varying molecular characteristics. These synthetic pathways play a pivotal role in the facile fabrication of a multitude of nanostructures, which also greatly facilitates our understanding of the governing principles of meso and nano object self-assembly. This review focuses on the complex packing structures of block copolymer micelles and colloidal particles, with a particular emphasis on the Frank-Kasper phase. A comprehensive exploration of the recent theoretical and experimental advances in this interdisciplinary field is presented. By drawing parallels between the self-organized formation of both hard particles and soft micelles, this review aims to provide a fundamental understanding of the underlying physics and chemistry that contribute to the emergence of complex packing behavior. This knowledge is instrumental in the design and engineering of structures tailored to specific applications, fostering further innovation in the realm of nanomaterials.
