Abstract:5-[2-(ethylthio)propyl]-1,3-cyclohexanedione, the title compound of this study, is an important intermediate for preparation of 3-hydroxy-cyclohex-2-enones herbicides, such as clethodim, sethoxydim, cloproxydim, etc.. The optimization of the synthesis route and the preparation method of the title compound were studied and the results were described concisely as follows. Firstly, 3-ethylthiobutanal was prepared with high yield by reacting crotonaldehyde with ethanethiol in triethylamine. Secondly, methyl acetoacetate was converted to 6-ethylthiohept-3-en-2-one via three steps, hydrolysis, condensation with 3-ethylthiobutanal and dehydration, then the 6-ethylthiohept-3-en-2-one was cyclocondensed with di-Me malonate by Michael addition and Clasisen condensation, and further via saponification and decarboxylation , the title compound was obtained. The products of each steps and the final title compound obtained were compared with each its own standerd samples and were confirmed by LC/MS.

Abstract:2 - O - Acetylation of 2 - (4-allyloxyphenyl) ethyl 4,6-O - benzylidene-�� - D -glucopyranoside (1) gave 2, which reacted with triacetylrhamnopyranosyl bromide to yield 3. Protective Osmanthuside B 6(5) was synthesized by reaction of p - acetyloxycinnamoyl chloride and 4 at low temperature, which was obtained by removing benzylidene of 3. Selective acetylation of 4 gave 6. Compound 6 reacted with p - acetyloxycinnamoyl chloride and 3, 4 - di - O - allylcaffeic acid respectively to afford two corresponding protective disaccharide phenylpropanoid glycosides,7 and 8.

Abstract:The (3S,5R) and (3R,5R) isomers of the title compound were synthesized from enantiopure epoxide chiral building blocks readily accessible from D-gluconolactone. The former (a syn-1,3-diol) gave H-1 and C-13 NMR spectroscopic data and optical rotation in good agreement with those reported for the natural product, whereas distinct differences were found with the data for the latter (an anti-1,3-diol). These results unambiguously showed that the absolute configuration of the natural product must be (3S,5R).

Abstract:Objective: To prepare a natural phenylprop anoid glycoside - Syringalide A, 2-(4-hydroxy- phenyl)ethyl-4-O-［( E )-caffeoyl］-β- D -gluco pyranoside. Methods: C hemical synthetic method was used and the allyloxycarbonyl (Aoc) and allyl group s were used for the protection of hydroxy groups in the sugar ring. The se lectio n of these two protective groups successfully avoided the migration of caffeoyl group occurring in the deblocking procedure under basic conditions. Result s: The total synthesis of natural product phenylpropanoid glycoside Syringalide A was realized for the first time. C onc lusion: The allyloxycarbonyl (Aoc) group may be one of the bes t alternative protective groups for the total synthesis of phenylpropanoid glyco sides.

Abstract:Phenylpropanoid natural products containing a cinnamaldehyde motif were easily synthesized from allylarenes mediated by 2,3-dichloro-5,6-dicyano-1,4-benzoquinone (DDQ) oxidation. Representative examples described herein are five types of 4-hydroxycinnamaldehyde derivatives from monolignols biosynthesis, Boropinal C, and 7-methoxywutaifuranal from plant extracts. Especially, simple synthesis of 7-methoxywutaifuranal was exploited through selective mono-oxidation and subsequent isomerization–ring-closing-metathesis strategy.

Abstract:Grayanoside A, 2-(4-hydroxyphenyl)ethyl 6-O-feruloyl-beta-D-glucopyranoside, was synthesized for the first time by using chloroacetyl groups for the protection of hydroxy functions. 2-(4-Allyloxyphenyl)ethyl 4-O-[(4-O-allyl)feruloyl]-2,3,6-tri-O-chloroacetyl-beta-D-glucopyranoside (12) was synthesized from 2-(4-allyloxyphenyl)ethyl 4,6-O-benzylidene-beta-D-glucopyranoside (8) in four steps with the goal of preparing syringalide B. Abstract:Methyl 3-deoxy-3-(diphenylphosphino)-4,6-O-benzylidene-α-D-altropyranoside (1) and methyl 2-deoxy-2-(diphenylphosphino)-4,6-O-benzylidene-α-D-altropyranoside (2) were prepared from methyl 2,3-anhydro-4,6-O-benzylidene-O-D-mannopyranoside and methyl 2,3-anhydro-4,6-O-benzyl-idene-α-D-allopyranoside,respectively,via regioselective and stcreospecific ring-opening reactions in high yields.Compounds 1 and 2 were oxidized to give the corresponding phosphine oxides (3 and 4).

Abstract:Phenylethanoid glycosides are a well-studied class of bioactive compounds found throughout the plant kingdom. In contrast, research on the synthesis and pharmacological activity of phenacyl glycosides, a specific subgroup of phenylethanoid glycosides with a ketone functionality at the alpha position of the phenol ring, has been limited. In this study, we report the synthesis, cytotoxic, antiviral, and anti-inflammatory evaluation of a series of 18 4'-hydroxyphenacyl glycosides. These compounds consist of six different sugar residues (β-d-glucose, β-d-galactose, α-l-arabinose, β-d-xylose, α-l-rhamnose, and β-d-glucuronic acid) and display three distinct methoxylation patterns at the phenacyl ring, similar to the substitution motifs of anthocyanins. We obtained the target phenacyl glycosides in high yield and stereoselectivity through the coupling of benzoyl-protected trichloroacetimidate glycosyl donors and corresponding acetophenones. Our work represents the first total synthesis of the natural products 4'-hydroxyphenacyl-β-d-glucopyranoside (1) and 4'-hydroxy-3'-methoxyphenacyl-β-d-glucopyranoside (2). None of the phenacyl glycosides showed cytotoxicity against the tested cell lines. Notably, several of the synthesized compounds exhibited antiviral activity, with natural product 2 being the most active against herpes simplex virus type 1, while phenacyl arabinoside 9 and natural product 2 were the most active against human coronavirus OC43. Natural product 2 significantly inhibited the production of interleukin-6 in lipopolysaccharide-stimulated microglia cells. Overall, our findings highlight the importance of the sugar residue and phenacyl ring substitution pattern in modulating the antiviral activity of phenacyl glycosides. Natural product 2 and phenacyl arabinoside 9 emerge as promising leads for the development of antiviral agents.

Abstract:Allyl 2-O-benzoyl-4,6-O-benzylidene-alpha-D-glucopyranoside (3), obtained by selective benzoylation of allyl 4,6-O-benzylidene-alpha-D-glucopyranoside (2) with benzoyl chloride-imidazole in anhydrous chloroform, reacted with 2, 3, 4-tri-O-benzoyl-alpha-L-rhamnopyranosyl bromide to give a disaccharide derivative (4), and an important intermediate (5) was obtained by cleavage of its acetal. Treatment of 5 with a series of glycopyranosyl bromides, protected by acetyl or benzoyl groups in benzene-nitromethane in the presence of Hg(CN)(2) as a catalyst afforded four trisaccharides (6-9). The disaccharide (5) and trisaccharides (6-9) constitute the sugar cores of phenylpropanoid glycosides. A new glycosyl anomeric leaving group, trichloroacetoxy, was employed to prepare the disaccharide (4) and trisaccharides residue (8) efficiently and with high stereoselectivity.

Abstract:Concise total syntheses of the natural phytoalexins 2-hydroxy-8-(4-hydroxyphenyl)phenalen-1-one (1), 2-hydroxy-8-(3,4-dihydroxyphenyl)phenalen-1-one (2), and hydroxyanigorufone (4), together with regioisomer 3 are accomplished in 11 or 12 steps. The synthetic strategy features a Friedel Crafts acylation to construct the 1H-phenalen-1-one tricyclic core followed by a Suzuki cross-coupling to obtain the target compounds.

Abstract:A new phenolic glycoside (1), named methyl 2-phenylpropanoate-2-O-β-D-apiofuranosyl-(1→6)-O-β-D–glucopyranoside, was isolated from the barks of Cinnamomum cassia, along with three known phenolic glycosides and four known lignan glycosides. The structure of 1 was elucidated by extensive interpretation of spectroscopic data and chemical method. Selected compounds were evaluated for their immunosuppressive activities against murine lymphocytes. Compounds 1, 2, 6 and 8 exhibited differential inhibition against ConA-induced T cells proliferation.

Abstract:To develop novel biologically active organic compounds possessing a sugar moiety, a series of 2-phenylsulfonylhydrazono-3-(2′,3′,4′,6′-tetra-O-acetyl-β-d-glucopyranosyl)thiazolidine-4-one were synthesized via reaction of the thiosemicarbazide with ethyl bromoacetate. Their chemical structures were characterized by 1H and 13C NMR spectroscopy, elemental analysis and MS. The bioassay results indicated that some of these compound exhibit moderate fungicidal and herbicidal activities. Furthermore, the effect of various solvents at reflux temperature on the reactions of ethyl bromoacetate with the related thiosemicarbazides was investigated.

Abstract:A benzyl-protected analog of the phenylpropanoid glycoside arenarioside, (4-benzyloxyphenyl)ethyl α-l-rhamnopyranosyl-(1→3)-4-O-[(E)-3,4-di-O-benzyl-caffeoyl]-[β-d-xylopyranosyl-(1→6)]-β-d-glucopyranoside (22), was synthesized through two different routes from d-glucose. This is the first approach on the synthesis of a trisaccharide phenylpropanoid glycoside, although the benzyl-protecting group in the backbone of the arenarioside analog could not be removed by conventional debenzylation procedures.

Abstract:2-Ac-Osmanthuside B6 [2-(4-hydroxyphenyl)ethyl 2-O-acetyl-3-O-(alpha-L-rhamnopyranosyl)-6-O-(4-hydroxycinnamoyl)-beta-D-glucopyranoside 8], a derivative of Osmanthuside B6, was synthesized in 7 steps with the goal of preparing Osmanthuside B6. Abstract:A variety of functionalized 2‐pyrrolidinones bearing N‐trifluoromethylphenyl moieties were prepared in moderate to good yields with high diastereoselectivity through copper(II)‐catalyst and base co‐catalyzed formal [4+1] cycloaddition of cinnamaldehyde nitrones with 1‐ethynylnaphthalen‐2‐ols in the presence of m‐CPBA as the oxidant. The triple bond of 1‐ethynylnaphthalen‐2‐ol served as one‐carbon synthon for the pyrrolidinone ring formation. Mechanistic studies revealed that m‐CPBA played an important role to interrupt the intramolecular O‐cyclization to afford the 2‐pyrrolidinones. Moreover, the reaction could be easily performed at gram scales and the obtained products were selectively converted into diverse pyrroline scaffolds.

Abstract:From natural tartaric acid, (R)-2-benzyloxy-3-(2-tetrahydropyranyloxy) propanol 3 was designed and synthesized, and (R)-2-benzyloxy-3-(4-methoxybenzyloxy) propanol 7 was prepared in a new method. They can be used as chiral synthons of lysophosphatidic acid and other compounds with asymmetric propanetriol backbone.

Abstract:In this study, ten phenylpropionyl phenylethylamines, including five previously undescribed ones ( 1a / b , 2a / b , and 3 ), five known analogues ( 4 – 8 ), and two established phenylpropanoids precursors ( 9 , 10 ) were isolated from the aerial parts of Chloranthus henryi Hemsl. Their structures, including absolute configurations, were determined by high-resolution mass spectrometry, enantio-separation, electronic circular dichroism calculation, and single crystal diffraction. Compounds 1a and 1b were the first examples of natural hetero- [2 + 2] cycloaddition products between phenylpropionyl phenylethylamine and phenylpropene. The plausible hetero- [2 + 2] biosynthesis pathway was confirmed by a photocatalytic biomimetic synthesis in eight steps, which also led to the production of three other potential natural homo -[2 + 2] adducts ( 1′a / b , 2′ , and 3′ ). Bioactivity screening indicated that these adducts bear medium inhibitory activity on nitric oxide generation, with IC 50 values of 6 – 35 μ M in RAW 264.7 macrophages.

Abstract:4-Chlorobenzoyl hydrazine(2) reaction with arylaldehydes produced corresponding hydrazones(3a～3h).Then 3a～3h respectively cyclodehydrated with propanoic anhydride to afford 3-N-propanoyl-2-aryl-5-(2-chlorophenyl)-1,3,4-oxadiazolines(4a～4h).The structures of 4a～4h were confirmed by elementary analyses,IR,~1 H NMR and MS spectroscopy.
