Abstract:Objective: To prepare a natural phenylprop anoid glycoside - Syringalide A, 2-(4-hydroxy- phenyl)ethyl-4-O-［( E )-caffeoyl］-β- D -gluco pyranoside. Methods: C hemical synthetic method was used and the allyloxycarbonyl (Aoc) and allyl group s were used for the protection of hydroxy groups in the sugar ring. The se lectio n of these two protective groups successfully avoided the migration of caffeoyl group occurring in the deblocking procedure under basic conditions. Result s: The total synthesis of natural product phenylpropanoid glycoside Syringalide A was realized for the first time. C onc lusion: The allyloxycarbonyl (Aoc) group may be one of the bes t alternative protective groups for the total synthesis of phenylpropanoid glyco sides.

Abstract:Grayanoside A, 2-(4-hydroxyphenyl)ethyl 6-O-feruloyl-beta-D-glucopyranoside, was synthesized for the first time by using chloroacetyl groups for the protection of hydroxy functions. 2-(4-Allyloxyphenyl)ethyl 4-O-[(4-O-allyl)feruloyl]-2,3,6-tri-O-chloroacetyl-beta-D-glucopyranoside (12) was synthesized from 2-(4-allyloxyphenyl)ethyl 4,6-O-benzylidene-beta-D-glucopyranoside (8) in four steps with the goal of preparing syringalide B. It was found, however, that the feruloyl group migrated from the 4- to the 6-position of the glycopyranoside during the deprotection of 12. (C) 1997 Elsevier Science Ltd.

Abstract:A facile approach for the first total synthesis of naturally occurring geranylated flavanoids sepicanin A has been obtained with total yield 16% starting from 2,4,6-trihydroxyacetophenone after four steps.The key step was the protic acids(HCl or p-TsOH)-catalyzed benzopyrone formation in a protic polar solvent by deprotection and cyclization of chalcone in one step.All structures of new compounds were confirmed by IR,1H NMR and MS.

Abstract:Comprehensive Summary Syringin is found in the root of Acanthopanax senticosus (Rupr. Maxim.) Harms and belongs to the lignin chemical compound with many biological activities. In this study, we employed commercially available starting materials and accomplished the total synthesis of syringin in 5 steps with an overall yield of 58%. Palladium‐catalyzed C(O)–C bond activation and subsequent cross coupling reactions is the key to construct syringin and its natural analogues. This article is protected by copyright. Abstract:Leontopodioside A, isolated from the whole plants of Leontopodium leontopodioides, possesses significant alpha-glucosidase inhibitory activity. In this work, we studied the total synthesis of leontopodioside A by two strategies for the first time. The optimized strategy involved nine linear steps and has an overall yield of 16.1%. The key feature of the strategy is that glycosylation of chalcone acceptor first followed by the cyclization to construct the flavone scaffold, which has general applicability for the synthesis of flavonoid glycosides. Abstract:Eutigoside A, 2 - (4 - hydroxyphenyl) ethyl - 6 - O - coumaroyl - beta - D - glucopyranoside, was synthesized for the first time. 2 - (4 - Allyloxyphenyl) ethyl - 4 - O - (4 - O - acetyl) coumaroyl -2, 3, 6 - tri - O - acetyl - beta - D - glucopyranoside (10) was synthesized from 2 - O - (4 - allyloxyphenyl) ethyl-beta-D-glucopyranoside (3) in 5 steps with the aim of the synthesis of Osmanthuside A. It was found surprisingly that the coumaroyl group migrated from the 4-to 6-position of the glucopyranoside during the deacetylation of 10 with NH3/MeOH.

Abstract:Hybridaphniphylline B (1) is a Daphniphyllum alkaloid possessing 11 rings and 19 stereocenters. Here we report the first total synthesis of 1 featuring a late stage intermolecular Diels-Alder reaction of a fully elaborated cyclopentadiene and asperuloside tetraacetate. The diene was prepared on the basis of a scalable route to daphnilongeranin B (4). Claisen rearrangement of an ally! dienol ether was exploited as a key step; the subtle variation of the substrate and use of protic solvents suppressed the undesired Cope rearrangement. Daphniyunnine E (6) and dehydrodaphnilongeranin B (7), two congeners of 4, were also synthesized. The dienophile arose from (+)-genipin through glycosylation and lactonization. A one-pot protocol was developed for the diene formation and Diets-Alder reaction; one of the cycloadducts was converted into 1 through reductive desulfurization and global deacetylation.

Abstract:3,7-Dimethyl-7-hydroxy-2-octen-1,6-olide (1) and 3,7-dimethyl-2,6-octadien-1,6-olide (2), the natural bioactive compounds isolated from the fruit of Litsea cubeba and the liverwort Plagiochila rutilans, were totally synthesized using easily available cis-geraniol as raw material in short, convenient, and low-cost, five-step reactions including three steps of oxidation, cyclization, and dehydration, with an overall yield of 47.5% and 37.3%.

Abstract:AbstractThe first total synthesis of liangshanone, a hexacyclic ent‐kaurane diterpenoid alkaloid, has been completed. Its intricate cagelike framework was assembled through several key transformations, including an oxidative dearomatization/Diels–Alder (OD/DA) cycloaddition sequence, a tandem alkene cleavage/Mannich cyclization, a Robinson‐type annulation, and an intramolecular aldol reaction. Notably, an organocatalytic enantioselective α‐hydroxymethylation process allowed the preparation of an enantiomerically enriched tricyclic intermediate that should enable asymmetric access to the target natural product.

Abstract:(-)-Ceforalide B (1) and (-)-cephanolides B-D (2-4) are benzenoid cephanolide diterpenoids possessing the same pentacyclic skeleton, which contains three C13-C15 substituent patterns and different benzylic oxidation states. An olefination/67r-electrocyclization/oxidative aromatization cascade has been verified as divergent access to three C13-C15 patterns. The benzylic aerobic oxidations enabled by the Co(OAc)2middot4H2O/ bromide salt/O2/PPh3/N-hydroxyphthalimide system have been developed to deliver expected site-selectivity and different oxidation states. Through the divergent strategy, total synthesis of (-)-ceforalide B and (-)-cephanolides B-D is accomplished.

Abstract:Total synthesis of natural product (±)-bruguierol A was accomplished in 10-steps and with an overall 16.8% yield. The embedded unique 8-oxabicyclo[3.2.1]octane core skeleton in this natural product was constructed via a novel Sc(OTf)3-catalyzed intramolecular [3+2] cycloaddition of cyclopropane, which was developed recently in this laboratory. This general synthetic strategy can be potentially applied to the synthesis of a broad range of structurally related natural products.

Abstract:We report a step-economical, enantioselective total synthesis of (-)-robustanoid B and (-)-robustanoid A and four novel natural product-like compounds. Our strategy relied on our biosynthetic hypothesis and on a novel complexity generation methodology, namely, the one-pot hydroxylative double cyclization reaction. The latter consists of a modified 3,3-dimethyldioxirane-triggered epoxidation-epoxide-ring-opening cyclization reaction cascade and Trost's regioselectivity umpolung methodology ("anti-Michael addition").

Abstract:Three novel natural compounds, 3-farnesyl-2,4,6-trihydroxy-acetophenone (1), 4-farnesyloxy-2,6-dihydroxyacetophenone (2) and 6-farnesyl-3 ',4 ',5,7-tetrahydroxy-flavanone (3) isolated from the aerial part of Boronia ramosa in Australia, were firstly synthesized by starting from 2,4,6-trihydroxyacetophenone and farnesyl bromide with total yield 52.0%, 6.5% and 19.8%, respectively, The synthetic steps of compound 3 contained six reactions of C-farnesylation, hydroxy-protection, aldol-condesation, cyclization and group-deprotection all together.

Abstract:Starting from readily available vanillin and α-D-acetobromo glucose, two natural acylated phenolic glycosides vitexnegheteroin A and ovatoside D were synthesized for the first time in 4 steps with overall yields of 54% and 65%, respectively. The key steps involve the directly regioselective O-6 acylation of vanillin β-D-glucopyranoside with acyl chlorides, and simultaneous removal of the allyl protecting groups on the phenolic acid moiety and reduction of the aldehyde in the aglycon moiety by using Pd(PPh)3-NaBH4 system in one pot.
