Experimental Study of the Protective Effect of Creatine Phosphate in Immature Myocardium

Abstract:Using isolated perfused immature rabbit hearts, we studied the protective effect of St. Thomas'Hospital cardioplegic solution No. 2 on the immature myocardial cells with creatine phosphate (CP). Methods Twenty immature rabbits were randomized either into the control group, which received St. Thomas'Hospital cardioplegic solution No. 2, or into the CP- treated group, receiving the same cardioplegic solution with CP(10mmol/L). Degree of the left ventricular function, myocardial ATP content and water content, coronary artery flow(CF), myocardial enzyme contents (CK, CK - MB, LDH) and SOD, MDA content in coronary venous blood,and myocardial ultrastructural changes were assessed. Results Concerning to recovery of left ventricular function, 30 minutes after reperfusion, there was no significant difference between two groups. Sixty minutes after reperfusion, the left ventricular systolic pressure (LVSP) and left ventricular developed pressure (LVDeP) and rate of left ventricular pressure rise during systole ( + dp/dt) were much more improved in the CP - treated group than in the control group. The myocardial ATP content was much more improved in the CP- treated group than in the control group. The myocardial water content was significantly reduced compared with the control group. The CF, SOD and MDA content were much more improved in CP - treated group than in the control group. The leakage of CK, CK - MB, LDH in the CP- treated group was significantly reduced compared with the control group. There was less damage of myocardial ultrastructure in the CP - treated group than in thecontrol group. Conclusion CP significantly enhances the protective effect of St. Thomas' Hospital car-dioplegic solution No. 2 on the immature myocardium.
