Mobilizing Properties of Synthetic Osteogenic Growth Peptide on Hematopoiesis in Experimental Aplastic Anemia Mice

Abstract:Purpose To investigate the promotive effects of synthetic osteogenic growth peptide on hematopoiesis in experimental aplastic anemia (EAA) mice. Methods EAA mice model was induced by sublethal irradiation of 5 Gy to BALB/C mice as receptor, then 1×105 per mice of mixed active lymphocyte from thymus and celiac lymph node of DBA/2 mice was injected through caudal vein within 4 hours after irradiation . From the next day on,doses of sOGP [0.05,0.5,5.0 nmol per mice] was administrated intraperitoneally for twelve consercutive days. Hematopoietic parameters including bone marrow smear,sternum slice and colony forming unit-granulocyte-macrophage(CFU-GM), were evaluated on the fourteenth day. Results Body weight,spleen weight,peripheral blood counting and bone marrow nuclear cells (BMNCs) had not been altered by sOGP treatment.However,the number of CFU-GM were significantly different in different treatment groups, effect of sOGP demonstrated an obviously positive linear relationship with its treatment dosage. Bone marrow smear and sternum slice showed that bone marrow proliferation recovered gradually as the dosage of sOGP increased, in the group of high dosage sOGP treatment(5.0 nmol/mice·day-1),both bone marrow smear and sternum slice were getting close to normal.Conclusions sOGP could effectively enhance the restoration of hematopoiesis on experimental aplastic anemia mice.
