Influence of Synthetic Osteogenic Growth Peptide on Hematopoiesis in Irradiated Mice

Abstract:Purpose: To evaluate the effect of synthetic osteogenic growth peptide (sOGP) on hematopoiesis in irradiated mice. Methods: sOGP was administered to mice sc or im on days 0-8 after 4.0 or 7.5 Gy 137Cs γ-irradiation at the dose range of 0. 039-640 nmol/(kg · d). Peripheral blood cell counts and bone marrow nucleated cell counts were measured by usual method. The bone marrow was assayed for clonogenic activity. The differential cell counts and histological features of bone marrow were evalua-ted. Results: sOGP exhibited a dose-dependent effect on accelerating the recovery of numbers of white blood cell (WBC) and bone marrow nucleated cell(BMNC). The number of colony forming unit-granulocyte(CFU-G) in sOGP-treated mice increased twice in compared with granulocyte colony stimulating factor(G-CSF)-treated mice. The injuries of bone marrow hematopoietic tissue were markedly alleviated and the proliferation of bone marrow granulocyte was stimulated. The number of endogenous CFU-S and spleen coefficient were significantly increased in comparison to irradiated mice. Conclusions: sOGP could effectively promote the recovery of hematopoietic funtions and hematopoiesis of spleen in irradiated mice. Its mechanism is possibly due to affect the early stage of hematopoiesis and/or improve its microenvironment.
