Changes in the apoptosis pathway of hippoca mpal neurons during the process of ketamine exerting antide-pressant effect

Abstract:[Abstract ] Objective Ketamine, as a general anesthetic, produces a rapid and robust antidepressant effect , but its underlying mechanisms are not fully elucidated .The aim of this study was to observe the changes in the apoptosis pathway of hippocampal neurons during the process of ketamine exerting antidepressant effect . Methods Forty-eight healthy male adult SD rats were equally randomized into three groups:control, chronic unpredicted stress (CUS), and ketamine treatment.CUS was used to establish a rat depression mod-el.The animals in the control and CUS groups were intraperitoneally injected with 1 mL saline and those in the treatment group with ket-amine at 10 mg/kg, respectively.At 1 h after injection, 8 rats were taken from each group for observation of their behavioral changes in the forced swimming test and sucrose preference test .Western blot was used to determine the levels of Bcl-2, Bax, and caspase-3 in the hippocampus of the other 8 animals in each group . Results The forced swimming time was remarkably longer in the CUS group ([165.33 ±4.59]s) than in the treatment ([115.83 ±9.17]s) and control group ([110.50 ±12.28]s) (both P<0.01), but with no statistically significant differences between the latter two (P>0.05).The sucrose consumption was markedly lower in the CUS group <br> ([58 ±5]%) than in the treatment ([80 ±6]%) ( P<0.01) and control group ([86 ±10]%) (P <0.05), but with no statistically significant differences between the latter two (P >0.05).The CUS group, in comparison with the treatment and control groups , showed obviously decreased expression of Bcl-2 ([73 ±3]%vs [157 ±21]%and [100 ±4]%, P<0.01 and P<0.05) and increased expressions of Bax ([175 ±4]%vs [114 ±6]%and [100 ±5]%, P<0.01 and <br> P<0.05) and Caspase-3 ([165 ±6]%vs [78 ±13]%and [100 ±1]%, P<0.01 and P<0.05). Conclusion Ketamine exerts its antidepressant effect probably by inhibiting the apoptosis pathway of hippocampal neurons in rats .
